Morphological and Genetic Characterization of Caffeine-Rich and -Poor Tea Tree (Camellia sinensis L.) Lines

  • In this study, 160 tea tree (Camellia sinensis L.) lines were classified by caffeine content using colorimetric methods. Among them, caffeine-rich lines (HR-78, HR-137, HR-82 and HR-123) and poor lines (HP-85, HP-88, HP-19, and HP-131) were selected. To know the difference in morphological and genetic characters between caffeine-rich and poor lines, we used leaf/shoot growth and RAPD methods. Cluster pattern of morphological characters (leaf width, leaf length, leaf area and shoot length) showed that shoot length was longer in caffein-rich lines than in -poor lines. In genetic analysis, amplified DNA bands having various sizes were detected in RAPD analysis where 30 random primers were used. However, the discriminated primer set that distinguish caffein-rich tree line from -poor lines was not found. These results can be used as the basic data to determine the morphological and genetic differences among caffein-rich and -poor lines.

Culture Conditions for Mycelial Growth and Anti-Cancer Properties of Termitomyces

  • Termitomyces sp. that grow in symbiosis with fungus-farming Termites have medicinal properties. However, they are rare in nature, and their artificial culture is challenging. The expression of AXL receptor tyrosine kinase and immune checkpoint molecules favor the growth of cancer cells. The study evaluated the optimal conditions for the artificial culture of Termitomyces and their inhibitory activity on AXL and immune checkpoint molecules in lung adenocarcinoma and melanoma cell lines. The culture of 45 strains of Termitomyces was compared. Five strains with marked growth rates were selected. Four of the selected strains form a single cluster by sequence analysis. The mycelium of 4 selected strains produces more fungal mass in potato dextrose broth than in a mixed media. The bark was the most appropriate solid substrate for Termitomyces mycelia culture. The mycelium of all five selected strains showed a higher growth rate under normal CO2 conditions. The culture broth, methanol, and ethyl acetate of one selected strain (T-120) inhibited the mRNA relative expression of AXL receptor tyrosine kinase and immune checkpoint molecules in cancer cell lines. Overall, these results suggest the potential usefulness of Termitomyces extracts as a coadjuvant therapy in malignant diseases.

Selection of Pure Lines with Various Growth and Flowering Characteristics of Spreading Petunia, Petunia × hybrida (생육 및 개화 특성이 다양한 덩굴 페튜니아 순계 선발)

  • To obtain pure lines for breeding $F_1$ varieties of spreading surfinia and wave petunia, Petunia ${\times}$ hybrida, 20 lines of surfinia petunia and 28 lines of wave petunia that were considered uniform in growth and flowering characteristics were selected by self-pollination of the fifth($S_5$) or the seventh generation($S_7$). The 20 selected lines of surfinia petunia had the branch number ranged from 6.0 to 11.0 cm, and the internode length ranged from 2.0 to 4.2 cm. Among them, ten lines, including '$Pe99-017^7$' were above 60 cm of plant width, above 300 leaves in a plant. Fourteen lines including '$Pe99-017^7$' were more than 150 in the number of flower. In the petal color, thirteen lines, including '$Pe99-017^7$', were red-purple; three, including '$Pe99-007^7$', were purple; '$Pe04-086^7$' and '$Pe04-159^7$' were violet; and line '$Pe072-1^7$' was white. Eight lines including '$Pe02-205-2^5$' ranged from 4.0 to 5.0 cm of flowers diameter, and seven lines including '$Pe04-086^5$' ranged from 3.0 to 4.0 cm of leaf length, which is relatively low. Germination rate of the lines was more than 50%. In the wave petunia, the branch number of the 28 selected lines ranged from 8.0 to 13.0 cm, and the internode length ranged from 1.0 to 3.2 cm, which is relatively higher than surfinia petunias. Among them, ten lines, including '$Pe99-020^7$' were above 60 cm of plant width, above 200 leaves in a plant. Twelve lines including '$Pe04-034-2^5$' were more than 150 in the number of flower. In the petal color, eighteen lines, including '$Pe99-020^7$', were red-purple; three, including '$Pe04-113-4^5$', were red; three, including '$Pe04-263^5$', were white; '$Pe04-201^5$' and '$Pe04-263^5$' were violet blue; and line '$Pe04-072-5^5$' was purple. Nine lines including '$Pe04-201^5$' ranged from 4.0 to 5.0 cm of flowers diameter, and eleven lines including '$Pe04-263^5$' ranged from 3.0 to 4.0 cm of leaf length, which is relatively low. All the lines with various growth and flowering characteristics would be very promising to use as breeding materials for $F_1$ hybrids of spreading petunia, Petunia ${\times}$ hybrida.

Study on the morphological Characteristics of Local Cultivars in Paeonia spp. (지방 수집 작약의 유망계통에 대한 형태학적 특성에 관한 연구)

  • The major morphological characteristics of 97 lines selected from 1,510 local collection lines of Paeonia spp. were studied and classified into some groups according to their Q correlation. The Coefficient of Variation of 8 characteristics in 97 lines varied greatly with the range from 21.3% to 127.1%. The 22 lines with useful morphological characteristics selected from 97 lines had 2n=10 chromosomes, in which there were no difference among the number of chromosomes of 22 lines. The 97 lines were classified into 14 groups based upon the results of Q correlation analysis obtained from data with 16 quantitative factors; Group III and group VI were divided into two sub-groups. Group I was large flowered lines with a single flower, Group IIIa, doubled flowered lines, Group VII, late maturing lines and Group IX, early maturing lines flowering in 17th May. Group VIII were dwarf type lines which has the shortest stem length and leaf length among 14 groups, which are suitable for potted plants. Group XIII had a lot of stems per plant and a large type leaf with leaf length and width. A needle-type leaf was found in Group X, whereas a wide and oval-type leaf in group XII.

Identification of Correlative Transmission Lines for Stability Prediction

  • Power system stability is correlated with system structure, disturbances and operating conditions, and power flows on transmission lines are closely related with those conditions. This paper proposes a methodology to identify correlative power flows for power system transient and small-signal stability prediction. In transient stability sense, the Critical Clearing Time is used to select some dominant contingencies, and Transient Stability Prediction index is proposed for the quantitative comparison. For small-signal stability discusses a methodology to identify crucial transmission lines for stability prediction by introducing a sensitivity factor based on eigenvalue sensitivity technique. On-line monitoring of the selected lines enables to predict system stability in real-time. Also, a procedure to make a priority list of monitored transmission lines is proposed. The procedure is applied to a test system, and it shows capabilities of the proposed method.

  • In order to investigate the radiation characteristics of unmixed line noise from overhead high voltage AC transmission lines, a gap noise generator was manufactured. Disk size which decides capacitance between the noise generator and earth was selected through preliminary indoor experiments and analysis by charge simulation method. The capacitance is one of principal parameters related to inject a proper noise current into lines. The field experiments were performed with the noise generator installed in Kochang 765 kV full scale test line. As the results, the useful data which can be used to analysis the radiation characteristics of noise from transmission lines was obtained. Those are lateral profiles the directivity of antenna toward lines, frequency spectra and so on.

CALCULATION OF TELLURIC ABSORPTION SPECTRA (지구 대기 흡수선 스펙트럼 계산)

  • In ground-based astronomical spectroscopic observations, there are many telluric absorption lines that are laid on the spectra of celestial objects. To study the physical properties of the celestial objects with these contaminated spectra, the telluric lines should be removed. A conventional method for removing the telluric lines is using the standard stellar spectrum as telluric line. In this paper, we introduce a technique to calculate synthetic telluric spectra and use them to remove telluric lines from a spectrum of a celestial object. We used Line-by-Line Radiative Transfer Model (LBLRTM) for calculating a synthetic spectrum and selected Michelson Interferometer for Passive Atmospheric Sounding (MIPAS) model as atmospheric model. We apply our method to some spectra obtained by Bohyunsan Observatory Echelle Spectrograph (BOES) to show that the telluric lines are well removed from the observed spectra by our model within an accuracy of 2% which is close to the 1-sigma rms of the original spectra.

Induction and Selection of Citrus Mutant by Gamma-Irradiation (감마선조사를 통한 돌연변이 궁천조생 감귤 가지 유도 및 선발)

  • We have subjected to gamma-irradiation to citrus buds and then grafted onto mature citrus tree. Mutant citrus branch lines have been induced. As a result of first selection, we found the several mutant lines showing interesting phenotypes such as higher sugar content. We have selected several branches showing good qualities such as higher sweetness and/or lower acidity. Some branch lines showed over $13^{\circ}Brix$ sugar content and below 0.9% acidity. Other mutant branch lines showed the changes of shape, size, peel thickness, and fiber contents or distribution of fruits. The results suggest that gamma-irradiation is an effective tool for induction of citrus mutant lines.
