In order to evaluate the genetic diversity and discrimination among five Korean native chicken lines, a total of 86 individuals were genotyped using 150 microsatellite (MS) markers, and 15 highly polymorphic MS markers were selected. Based on the highest value of the number of alleles, the expected heterozygosity (He) and polymorphic information content (PIC) for the selected markers ranged from 6 to 12, 0.466 to 0.852, 0.709 to 0.882 and 0.648 to 0.865, respectively. Using these markers, the calculated genetic distance (Fst), the heterozygote deficit among chicken lines (Fit) and the heterozygote deficit within chicken line (Fis) values ranged from 0.0309 to 0.2473, 0.0013 to 0.4513 and -0.1002 to 0.271, respectively. The expected probability of identity values in random individuals (PI), random half-sib ($PI_{half-sibs}$) and random sibs ($PI_{sibs}$) were estimated at $7.98{\times}10^{-29}$, $2.88{\times}10^{-20}$ and $1.25{\times}10^{-08}$, respectively, indicating that these markers can be used for traceability systems in Korean native chickens. The unrooted phylogenetic neighbor-joining (NJ) tree was constructed using 15 MS markers that clearly differentiated among the five native chicken lines. Also, the structure was estimated by the individual clustering with the K value of 5. The selected 15 MS markers were found to be useful for the conservation, breeding plan, and traceability system in Korean native chickens.
Field experiments were conducted to evaluate the morphological characteristics and growth rates of 101 medium-leaf type zoysiagrasses (Zoysia spp.) collected at the major sod production area (Jang Seong Gun) in South Korea. Collected lines with distinctive morphology and visual growth rate were planted in plastic pots and measured morphological characteristics under the plastic house conditions. Variation of leaf width, plant height, leaf angle, length of leaf sheath, trichome, stolon length, and color were measured. Six lines were selected by evaluating growth rates from one hundred one collected lines. Eight standard cultivars and three other superior lines previously collected were compared to 7 selected lines form Jang seong area by checking growth rates and morphological characteristics. Average leaf blade width was 3.4 mm, leaf angle was 45.8 degree, plant height was 21.6 cm, height of lowest leaf was 5.0 cm, and length of leaf blade was 14.1 cm. Ground cover rates of selected lines 'CY6097' and 'CY6069' were 70% and 68.3%, respectively. These are believed to be faster than 60% ground cover rate of zoysiagrass 'Anyang', and also, twice as faster than the 31.7% ground cover rate of Z. matrella. Selected line 'CY6069' showed fast growth rate with shorter internode length (5.1 cm) compared to zoysiagrass 'Anyang'. Based on the results of this study, we could select useful fast growing zoysiagrass breeding lines from the major sod production area (Jang Seong Gun) in Korea.
In order to develop a rapid and simple method for testing the purity of radish hybrid seeds using a procedure based on the PCR(Polymerase chain reaction), eighty random primers were screened with the genomic DNA extracted from five day old seedlings of inbred parent lines and their F1 hybrids. Two primers, HRM-02 (5'-GAGACCAGAC-3') and HRM-19(5'-TGAGGCGTGT-3'), generate reproducible unique PCR patterns which can identify each parent lines as well as their hybrids. In actual test of randomly selected hybrid seeds using the two marker primers, the purity tested by one primer was exactly same as that of other primer. It suggests that one marker primer selected in this experiment is enough for the purity test of radish hybrid seeds. We demonstrates the use of RAPD(randomly amplified polymorphic DNAs) markers to identify each of inbred parent lines and hybrids by rapid and simple method.
Ginseng root explants and calli induced from selected cell lines were cultured on modified Murashige and Skoog's media supplemented with different concentrations of organic or inorganic compounds and plant growth requlators to clarify the effects of chemical composition and plant growth regulators in the medium on the growth of ginseng calli and the production of ginseng saponin. For optimum growth of calli, the concentrations of 2, 4-D and sucrose were the range of 1 to 3 mg/${\ell}$l and 1 to $3\%,$ respectively. And it was clarified that sucrose, nitrogen, phosphate, calcium, magmesian plant growth regulators and their concentrations influcenced the relative biosynthesis of saponin in tissue cultures of Panax ginseng. The patterns of ginsenosides, pharmacologically useful component, were different among the cell lines and contents of ginsenosides were much higher in selected cell lines than in original cell line.

Tiller angle, defined as the angle between the main stem and its side tillers, is one of the main target traits selected inbreeding to achieve the ideal plant type and increase rice yield. Therefore, the discovery and identification of tiller angle-related genes can provide architecture and yield. In the present work, using QTL analysis hence a total of 8 quantitative trait loci (QTLs) were detected based on the phenotype data of tiller angle and tiller crown width in two years. Among them, four QTLs (qTA9, qCW9, qTA9-1, qCW9-1) were overlapped at marker interval RM6235-RM24288 on chromosome 9 with a large effect value regarded as stable major QTL. Twenty tiller angle-related genes were selected from the target region and the relative gene expression levels were checked in five compact type lines, five spreading type lines, and their parental lines. Finally, OsSA URq9 which belongs auxin-responsive SMALL AUXIN UP RNA (SAUR) protein family was selected as a target gene. Overall, this work will help broaden our understanding of the genetic control of tiller angle and tiller crown width, and this study provides both a good theoretical basis and a new genetic resource for the breeding of ideal-type rice.
Nitrate reductase deficient (NR) mutant lines were selected indirectly by their resistance to 100mM chlorate in cell cultures of P. parviflora. A total of 585 chlorate resistant lines were confirmed by a second passage on a high concentration of chlorate. Frequency of spontaneous mutation was $9.7{\times}10^{-7}$ in 3 month old suspension-cultured cells, and in non-selective media containing amino acids as sole nitrogen source. The frequency of mutation could be increased up to 11-fold by culture for 12 months. Out of 40 randomly selected calli, 22 were fully deficient in NR. The rest of the clones contained a decreased level of NR activity. Further characterization was carried out in 13 mutant lines which were fully deficient in NR and in 5 mutant lines containing residual (0-7.0%) NR activity, as compared to wild-type cells cultured on the same medium. The $NR^-$ mutants were tentatively classified as defective in the NR apoenzyme (nia-type; 11 mutant lines including the 5 with residual NR activity) or in the molybdenum cofactor (cnx-type; 7 mutant lines) by the XDH activity. The cnx-type could be further classified into two groups. In one group (5 mutant lines) of these, the NR activity could be partially restored by nonphysiologically high (1.0mM) molybdate in the culture medium. Both types of $NR^-$ mutants were unable to grow on minimal medium containing nitrate as sole nitrogen source, but grew well on amino acids. They also proved to be extremely sensitive to the standard medium ($MSP_1$) containing nitrate and ammonium. Shoot regeneration was obtained only in the $NR^-$ mutants, which contained residual NR activity, but they so far have failed to grow into plants.

Recently, there are growing concerns about power frequency electric and magnetic fields coming out from the high voltage transmission lines, because of the wide spread perception of their probable harmful effect on human body. In connection with this trend, this paper describes the electric and magnetic fields measurement result around 154 kV and 345 kV transmission lines, a comparison of measured EMF to calculated one and the correlations between transmission lines currents and measured magnetic fields. Daily maximum and minimum magnetic field quantities under the selected 154 kV and 345 kV transmission lines had been measured for 1 year of 1997 and the average value of magnetic field exposure under the lines were calculated and presented based on the measured data.
The combination of radiation technique with an in vitro culture system was initiated to develop salt tolerant rice. We established an in vitro culture system to select tolerant lines against salt stress. NaCl tolerant cell lines were selected from the callus irradiated with gamma ray on N$_{6}$ medium with 1.5% NaCl and 2 mg/L 2,4-D. Regenerants (M$_1$) were obtained from the tolerant callus which was cultured for 30 days auxin-free medium. The M$_2$seeds were harvested from M$_1$plants on an individual plant basis. Thirty seedlings from each 450 M$_2$lines were transplanted in a field and total 5,000 M$_3$lines were harvested with an average 90 percent of fertile grain. M$_3$lines were utilized for selection of salt tolerance. Salinity-tolerant lines (225) were selected among 5,000 M$_3$lines. Of the 225 lines tested, the morphological traits of two lines (120-10 and -11) were far superior to control (Donagjinbyeo) in agromomic traits such as plant height, root length and no. of roots. Control and tolerant lines were analyzed by RAPD markers. Three polymorphic bands were presented in only tolerant lines, demonstrating a genetic difference between control and the tolerant lines. Such tolerant lines could be used as genetic resources to improve salt tolerance.e.

Because of computational burden and complex topology of substation, a transmission overload relief using circuit-breaker switching was very complex and difficult. However, a on-line algorithm for reducing the overloads in transmission lines has made progress due to the advance of IT technology. This paper describes the methodology for alleviating the overloads in transmission lines by circuit-breaker switching. First, the severe contingency lists and substations were selected from the results of contingency analysis. Then the switch combinations are determined using circuit-breakers of the selected substation. The topology changes are limited to equipment outage, bus split, island split, bus merge and island merge. Finally, the fast screening and full analysis methods are used to analyze the overload in transmission lines. To verify the performance of the proposed methodology, we performed a comprehensive test for both test system and large-scale power systems. The results of these tests showed that the proposed methodology can accurately alleviate the overloads in transmission lines from online data and can be applied to on-line applications.
To select new germplasm for salinity tolerance from new plant type (NPT) breeding lines, the sixty F$_4$ lines selected from the crosses between Korean cultivars and IRRI's NPT lines were evaluated for salinity tolerance at the seedling stage with salinized culture solution (EC=12 dS/m) in the controlled conditions. Two NTP lines derived from a cross between 'Ilmibyeo' and 'IR66152-AC5-1', 'HR15258-7-1' and 'HR15258-27-1', were found to have good tolerance. The salinity tolerance of the lines was compared to their parents and the sensitive ('IR29') and tolerant ('Pokkali') checks in three salinity levels, no salinity (control) and an EC of 12 and 16 dS/m. Visual salinity score, shoot Na+ and Na-K ratio in two NPT lines was significantly low compared with the parents and IR29. Indicating that salinity tolerance of the lines might be derived from a transgressive segregation. The relative water content of the lines was higher than Pokkali, and the dry weight of shoot and root was proportionally decreased to salinity score and salinizing concentration. The visual salinity scores were significantly correlated with shoot Na concentration, Na-K ratio, relative water content, and reduction of dry weight (P<0.01). Their tolerance was attributed to root and shoot characteristics that led to high shoot water content, thus diluting the toxic effect of salts.
