Identification of the Oligotrophic Bacteria Strain 7F Biocontrolling Phytophthora Blight Disease of Red-pepper (고추 역병 방제를 위한 저영양 길항세균 7F 균주의 동정)

  • A total of 10,753 oligotrophic bacteria were isolated from the cultivated soils of red-pepper infected by Phytophthora blight disease in various regions of Korea (Chungju, Anmyon, Taean, Andong, Eumsung and Goesan). Seven bacteria isolates among these collected resources were selected by the first screening of in vitro antagonistic assay against major several plant pathogenic fungi including Phytophthora capsici. Finally, strain 7F was selected by pot assay for a possible biological control agent against Phytophthora blight disease of pepper seedling in the greenhouse. Strain 7F was identified as Bacillus subtilis on the basis of its 16S rDNA sequence analysis and as standardized biochemical characteristics assay kits such as API20 NE. In the experiment of P. capsici zoospore infected red-pepper on the pot test, infection rate of red-pepper with nonetreatment to Phytophthora blight disease was 87%, while the rate was only 6% in the pot treated with strain 7F. This result indicated that the Bacillus subtilis strain 7F will be useful as a potential biocontrol agent for Phytophthora blight disease of red-pepper.

Symptom and Resistance of Cultivated and Wild Capsicum Accessions to Tomato Spotted Wilt Virus (토마토반점위조바이러스에 대한 재배 및 야생형 고추 수집종의 병징과 저항성 조사)

  • One hundred Capsicum accessions were screened for symptomatic response and resistance to Tomato spotted wilt virus-pb1 (TSWV-pb1). Symptom and its severity rating were checked by visual observation at 9, 12, 14, and 45 days after inoculation, respectively. Enzyme-linked immune-sorbent assay was performed all tested individuals on non-inoculated upper leaves after the third rating to indentify viral infection. Leaf curling was predominant in almost susceptible individuals of each accession. Stem necrosis was most frequent in wild species while yellowing in commercial hybrids and Korean land race cultivars. Ring spot, a typical symptom of TSWV, was rarely detected in some of a few accessions. Different levels of resistance to TSWV-pb1 were observed among the tested accessions. High level of resistance was detected in 4 commercial cultivars of Kpc-35, -36, -57, and -62, and 8 wild species of PBI-11, C00105, PBC076, PBC280, PBC426, PBC495, PBC537, and PI201238 through seedling test by mechanical inoculation.

Effects of Paper Mill Sludge-Fertilizer Application on the Growth Performances of Tree Seedlings (제지(製紙)슬러지 가공비료(加工肥料)의 시용(施用)이 수묘(樹苗)의 생육(生育)에 미치는 영향(影響))

  • Effects of paper mill sludge-fertilizers were investigated on the growth performances of tree seedlings under field conditions. Two types of sludge-fertilizers applied were an organic sludge compost and a processed sludge-pellet fertilizer strengthened with several components of organic and inorganic nutrients. Three species of tree seedlings studied were ibota privet(Ligustrum obtusifolium) and yellow poplar(Liriodendron tulipifera) of 1-0 year seedlings and also eastern white pine(Pinus strobus) of 2-2 year seedlings. Nitrogen and phosphorus contents of plot soil treated with sludge compost+sludge-pellet fertilizer+sludge-pellet fertilizer plot were increased to 8 times and 2.6 times as those of control plot, respectively. The growth performances of root collar diameter and seedling height were repeatedly measured up to five times through the growing season and the results were analyzed statistically by analysis of variance of randomized block design and Duncan's multiple range test. The growth performances of dry weight were measured after last fifth measurements.

Growth of Plug Seedlings of ‘Nokkwrang’ Pepper in Mixture of Used Rockwool and Woodchip Particles (폐암면과 목재 입자의 혼합비율에 따른 고추 플러그 묘의 생육)

  • Rockwool slabs used for hydroponic cultures are expected to have potential to be reused after breakdown as a growing medium component for hydroponic culture of vegetables and cut flowers, pot plants, or plug plants. This study was conducted to test the feasibility of using particles of used rockwool slabs (PURS) mixed with woodchips for growing plug seedlings of ‘Nokkufang’ pepper. In the first experiment growth of pepper seedlings was tested in media of steam sterilized PURS mixed with chestnut woodchips (CW), which was weatherized for six months and screened through 2.8 mm or 5.6 mm sieves, at 100:0, 75:25, 50:50, 28:75 or 0:100 (%, v/v). In the second experiment growth of pepper seedlings was tested in 20 different media containing steam sterilized PURS mixed with 4 mm chestnut woodchips (CW), 4 mm pinc woodchips (PW), coir, peatmoss, or perlite. In the first experiment, there were no significant growth differences as affectcd by particle size of CW, while the overall best growth was achieved il1 the control and l00% PURS. In the second experiment, seedling growth was enhanced as compared to the control in 100% PURS or PURS mixed with peatmoss, coir, or perlite. In contrast, growth was suppressed as the proportion of peatmoss or coir mixed with PURS decreased and the proportion of CW increased. Growth was better in the PURS + peatmoss than in the 100% PURS or PURS + coir, and when woodchips were mixed with coir than peatmoss.

  • The 167 plus tree clones of Pinus koraiensis in clone bank planted in 1983 were investigated for time of flowering, rate of flowering, and number of flowering from 1986 to 1989. The results were as follows : 1) There were clones in the minority which do not cross in natural pollination between earlest flowering female clones and latest shedding male clones. 2) The rate of male flowering appeared less than rate of female flowering and received influence of genetic more than rate of female flowering. 3) The numbers of flowering in female and male flower were affected by small number of clones. 4) The flowering number for female strobili was not related to the flowering number of male flower. The 42 clones among 167 clone had not male flower. 5) The flowering number of 167clones were classified three groups in female flower and four groups in male flower by L.S.D 5% test. 6) The clones with abundant female and male flower were selected based on the component analysis. 7) As Based on flowering rate and number of female, the juvenile phase of plus tree clones appeared to be until four years after grafting ; the transition period appeared to be from five to seven years after grafting ; the adult phase appeared to begin from eight years after grafting, 8) The grafted trees of Pinus koraiensis appeared early flowerring about six years compared with seedling trees.

Studies on the Seed Transmission of Colletotrichum spp. in Red Pepper (Capsicum annuum) (고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究))

  • Colletotrichum acutatum, C. coccodes, C. dematium and C. gloeosporioides were detected in seed samples of red pepper (Capsicum annuum). C. dematium and C. gloeosporioides were the predominant species, maximum seed infection of the species in some samples were 84% and 28%, respectively. C. acutatum and C. coccodes were recorded only in low percentages of 1-2. The blotter method proved more suitable for detecting Colletotricum spp. than the deep freezing blotter or agar plate methods. Plating of seed components showed that C. dematium and C. gloeosporioides were recovered more frequently from seed coat, and decreasing amounts of infection were observed in the endosperm and cotyledon. Seed-borne C. dematium and C. gloeosporioides caused seed rot, damping-off, seedling blight and brown discoloration of cotyledon and hypocotyl when infected seeds were sown in agar of test tube or in soil. Inoculation experiments showed that C. acutatum was pathogenic to red fruit of red pepper and C. coccodes was highly pathogenic to red fruit and weakly pathogenic to leaf of the plant. C. dematium was highly pathogenic to leaf and green fruit and C. gloeosporioides was pathogenic to not only leaf but also green and red fruits. Host range of the four seed-borne species of Colletotrichum was also investigated.

  • The study has been carried to confirm the pathotypes of Kresek inducing isolates of X. oryzae, and its reaction to the test plants when they were inoculated by the use of various methods. Induction of Kresek symptom by 23 isolates which randomly samplified from 123 Kresek areas through the country also were examined. The specificity of Pathotypes I, II, V in causing Kresek and the potenciality of infected rice straw is a first inoculum for Kresek epidemics in nature were tested. Fifteen out of 23 isolates from kresek area belonged to pathotype group I, while the rest of 8 isolates 5 howed group II reaction, and the most of isolates were originated from infected rice plant of Milyang #23. All of five pathotype groups were abled to produce 'Kresek' symptom although they showed some differences in each group of pathotype. The varieties Milyang #21, 22 and 23 were attacked by all isolates of group I, II and V while the Gogyoku group varieties including Yushin produced kresek symptom only by pathotype group II. Infected and dried straw maintained the pathogens during the winter period, and these straw acted as a first inoculum of Kresek disease when the straw were cutted and used as a manure just before transplanting of rice seedling.

The Influence of Temperature and Water Stress on the Varietal Reactions of Rice to the Inoculum of the Blast Infected Leaves (BIL) (도열병 이병엽접종원에 대한 수도 품종의 반응에 미치는 온도 및 Water Stress의 영향)

  • Using the milled, blast infected leaves (BIL) as an inoculum source on the screening for the resistance to blast of rice plant was a simple and useful technique. The temperature with high (25^\circ C\sim 35^\circ C) and low (15^\circ C\sim 28^\circ C) and the water stressed or not, was conditioned of to the inoculation with the BIL to the test varieties in seedling stage. In low temperature, most of the varieties were more infected with blast, however the Indica-Japonica hybrids were more infected in high temperature conditions. The water stressed was more infected with blast than the not stressed. The interaction of variety with water stress was not so much as that of variety with temperature. Resistant reaction to blast (BIL) was not affected by the temperature and water stress, but the moderately resistant or susceptible one was much affected by them. Inoculum of BIL was virulent to the newly bred Indica-Japonica hybrid cultivars such as Tongil, Nopung, etc, but not virulent to the Japonica cultivars such as Nongbaek, Jinheung, etc. The discrete, mixed or variable lesions were observed mainly in the moderately resistant or susceptible cultivars such as Kanto 51, Yashiromochi, Ishikari-shiroke, etc.

Development of Non-Destructive Sorting Technique for Viability of Watermelon Seed by Using Hyperspectral Image Processing (초분광 영상기술을 이용한 수박종자 발아여부 비파괴 선별기술 개발)

  • Seed viability is one of the most important parameters that is directly related with seed germination performance and seedling emergence. In this study, a hyperspectral imaging (HSI) system having a range of 1000-2500 nm was used to classify viable watermelon seeds from nonviable seeds. In order to obtain nonviable watermelon seeds, a total of 96 seeds were artificially aged by immersing the seeds in hot water ($25^{\circ}C$) for 15 days. Further, hyperspectral images for 192 seeds (96 normal and 96 aged) were acquired using the developed HSI system. A germination test was performed for all the 192 seeds in order to confirm their viability. Spectral data from the hyperspectral images of the seeds were extracted by selecting pixels from the region of interest. Each seed spectrum was averaged and preprocessed to develop a classification model of partial least square discriminant analysis (PLS-DA). The developed PLS-DA model showed a classification accuracy of 94.7% for the calibration set, and 84.2% for the validation set. The results demonstrate that the proposed technique can classify viable and nonviable watermelon seeds with a reasonable accuracy, and can be further converted into an online sorting system for rapid and nondestructive classification of watermelon seeds with regard to viability.

Scab Resistance of Some Pear Genetic Resources and Inter Specific Hybrid Seedlings in Field Condition (포장조건에서 주요 배 유전자원과 종간교배 집단의 검은별무늬병 저항성)

  • This study was accomplished to select breeding materials among the pear germplasm 6 species 43 accessions and check out the expression of scab incidence of inter specific hybrids of 5 crosses 609 seedlings for scab resistance breeding. Fruitlets scab incidence of P. pyrifolia, P. bretschneideri and P. ussuriensis were 59.5, 73.4 and 63.0% respectively, and also fruitlets scab incidence of interspecific hybrid cultivars between P. pyrifolia and P. bretschneideri were 77.7%. However, fruitlets scab incidence of P. communis and interspecific hybrid accessions between P. pyrifolia and P. communis were significantly less than 15%. As a result of the Duncan test, scab resistance of pear accessions were classified into two groups: P. communis and their hybrid were resistance and P. bretschneideri, P. pyrifolia, P. ussuriensis, and among their hybrid were susceptible. Also, it showed the same trend in the seedlings. Scab incidence of 'Seolwon' ${\times}$ 'Chuwhangbae' and 'Supergold' ${\times}$ 'Mantienhong' in P. pyrifolia seedlings showed more than 80%, while that of interspecific hybrid between P. pyrifolia and P. communis showed about 2%. It showed scab resistance of P. communis transferred to hybrid seedlings and expressed to be dominant in their seedlings. Namely, P. communis was good breeding materials for scab resistance.
