Fusarium spp. Isolated from Seed, Root and Cultivated Soil of Phaseouls vidissimus and Their Pathogenicity (녹두종자, 뿌리와 녹두 재배토양에서 분리된 Fusarium spp.와 병원성)

  • Fusarium oxysporum, F. moniliforme, F. solani, F. equiseti, F. semitectum, and F. sporotrichioides were detected from seeds, roots and cultivated soil of Phaseolous vidissimus collected from Kyung-gi Provincial Rural Development Administration. The rate of seedling desease incidence was $60\%$ by testing of seed germination using a large petri-dish. According to the blotter method, F. moniliforme showed $7\%$ infection at seed-coat and $2\%$ at cotyledon and embryo. Their pathogenicities of F. moniliforme, F. semitectum, F. equiseti, and F. sporotrichioides isolated from seeds were recognized on seedlings by water-agar test tube methods. F. oxysporum and F. solani isolated from infected-roots had their pathogenicity by water-agar test tube method but were weakly pathogenic by soil treatment method. Their pathogenicities of F. oxysporum. F. solani and F. uiseti isolated from cultivated-soil were recognized by water-agar test tube method. These F. oxysporum and equiseti isolates had their pathogenicities but F. solani was weakly pathogenic by soil treatment method.

Rural Survey on Agricultural Mechanization Project - Rice Transplantation Operation - (농업기계화(農業機械化)에 관(關)한 연구(硏究) - 수도이앙작업(水稻移秧作業)의 기계화(機械化)를 중심(中心)으로 -)

  • Mechanization of rice transplanting operation is very important project not only to solve the labor shortage problem at the so-called labor demand peak seasons of the rice transplanting, but also to reduce the production cost of rice by reducing the labor requirements. For these reasons this study was carried out to find the basic data for encourage the project of mechanization of rice transplanting. 381 sample farms were surveyed with questionaries and interviewed with a considerable number of relative personels about the operation, selecting and ownership trend of the rice transplanter. Collected data was analized by computer of Chungnam National University computer center applied to frequencies, cross- tabulation, $x^2$-test. The analized results of this survey are summarized as follows; 1. About 76.09% of the farmers interviewed was individual ownership of rice transplanter but about 52.27% of the farmers who wanted to purchase it in 2 or 3 years supported the cooperative ownership and utilization. This fact suggested that cooperative system of village level should be thoroghly studied. 2. The 93.33% of respondents gave the answer that the yield of rice was not affected by the planting methods between machine and manual. 3. The farmers who had a rice transplanter owned 4- row type rice transplanter with mat type seedling but the 25% of the farmers wanted to purchase it in 2 or 3 years wanted to own a 4 row type rice transplanter with band type seedling. Therefore the introduction of the 4-row type rice transplanter with band type seedling to rural area should be considered again. 4. The percent of farmers who wanted the cooperative system of village level was 49.57-57.83% of the farmers who had it already and wanted to own it in near future. It was strengthened by this fact that seedling nursely work was technically supported by the governmental level.

Density Effects on the Size of 2-1 Korean Pine and 1-1 Jack Pine Nursery Stock (잣나무 2-1 묘(苗)와 방크스소나무 1-1 묘(苗)의 생장(生長)에 미치는 밀도효과(密度効果))

  • One of the most common needle leaf species used in planting in Korea is korean pine (Pinus koraiensis S. et Z.), and jack pine (Pinus banksiana Lamb.) is one of the test species for suitability. The relation of nursery bed density of 2-1 korean pine and 1-1 jack pine was studied at the Kwang Nung Nursery, Central Branch Station of Forest Research Institute, and about 40km north of Seoul. Nursery bed density of 2-1 korean pine, which ranged from 36 to 324 trees per square meters and of 1-1 jack pine, which ranged from 25 to 169 trees per square meters, had a marked effect on caliper, height, dry weight and percent and amount of plantable stock. The soil physical and chemical properties is silt plus clay, 50.55 percent; organic matter, 2.09 percent; total nitrogen, 0.13 percent; available phosphorus, 253.25 ppm; exchangeable potash, 0.46 m.e/100g; and pH, 5.58. As the density of the nursery seedling stand of 2-1 korean pine increases, the average tree height increases (Fig. 1A), but in 1-1 jack pine density do not affect to increase or decrease the average tree height. As the density of nursery bed increases, the average stem caliper (at 2cm above ground line) and dry weight decrease (Fig. 1B), but the decreasing rate is more seriously in 1-1 jack pine than 2-1 korean pine (Fig.5). As increasing the density of nursery bed, the T/R ratio of trees of the test species increase. Also the dry weight of leaf, stem and root parts are decreasing in proportion to the increase of stand density, but the drop rate of jack pine is more rapid than korean pine (table. 1) The patent facts of difference of growth characteristics between 2-1 korean pine and 1-1 jack pine were studied. These facts should be used to select the scale of stand density at the nursery bed or the plantable site. Korean pine is demanded high density, on the other hand in jack pine low density are more suitable to manage the stand density. Stands of comparatively low density had the greatest percentage of high-quality stock, and the stands of high density had less than the high quality trees of low density. An important criterion of the best density is percent and number of high-quality trees produced per square meter of bed area. Stem caliper and stem height of seedling is used in most public nurseries to sort seedling into plantable grades. The stock grade standard has set at 4.5mm caliper and 16cm height of 2-1 korean pine as the minimum desired stem caliper and height. By the result studies, the plantable stock grade standards of 2-1 korean pine used at stem height 16cm and stem caliper 4.5mm from public nurseries should be reformed to stem height 18cm and stem caliper 4.0mm by the growth characteristics and the tree distribution of stem height and caliper of relation to density. For the 2-1 korean pine, best density should be about 160 to 200 trees per square meter according to soil fertility. For the 1-1 jack pine, the suitable standard of plantable stock should be at stem height 25cm and caliper 6mm (at 2cm above ground line) and best density was about 100 to 120 trees.

Establishment of Days after Anthesis(DAA) and Fruit After-ripening Period(FAP) for High-Quality Seed Production of Watermelon (우량 수박종자 생산을 위한 적정 성숙일수 및 후숙일수 확립)

  • This study was conducted to investigate days after anthesis (DAA) and fruit after-ripening period (FAP) for seed-harvesting of high quality watermelon seeds. Fruit weight and number of seed per fruit increased according to DAA, while those did not significant about FAP. Ratio of cotyledon at whole seed was higher about 2 to 4% compared to seed coat irrespective of DAA and FAP. Germinability of watermelon was not a significant effect by DAA, however, it had differences by FAP. Percent of germination (PB) was below 50%, when 30 days maturated fruits after anthesis was omitted ripening, while PB was increased to 92% by ripening. In addition, seeds at DAA 40 and FAP 20 were higher general seedling vigors (hypocotyl length, diameter etc.) in BP test. Results indicated that considering seed productivity, it had maximized seed viability at DAA 40 and FAP 20.

Study on the grading standard of Panax notoginseng seedlings

  • Background: The quality differences in seedlings of medicinal herbs often affect the quality of medicinal parts. The establishment of the grading standard of Panax notoginseng seedlings is significant for the stable quality of medicinal parts of P. notoginseng. Methods: To establish the grading standard of P. notoginseng seedlings, a total of 36,000 P. notoginseng seedlings were collected from 30 producing areas, of which the fresh weight, root length, root diameter, bud length, bud diameter, and rootlet number were measured. The K-means clustering method was applied to grade seedlings and establish the grading standard. Results: The fresh weight and rootlet number of P. notoginseng seedlings were determined as the final indices of grading. P. notoginseng seedlings from different regions of Yunnan could be preliminarily classified into four grades: the special grade, the premium grade, the standard grade, and culled seedlings. Conclusion: The grading standard was proven to be reasonable according to the agronomic characters, emergence rate, and photosynthetic efficiency of seedlings after transplantation, and the yields and contents of active constituents of the medicinal parts from different grades of seedlings.

  • It has been reported that some plants have the potential to metabolize the 2,4,6-trintroluene (TNT) in contaminated soils, sediments and natural water. In this study, the effects of TNT on germination and early seedling development of Abutilion avicennae was characterized in a germination test. Concentration up to 80 mg/L TNT did not affect germination but root and shoot growth, and fresh biomass decreased as TNT concentration increased. A series of axenic hydroponical batch culture of Abutilion avicennae at various initial TNT concentration was used to determine its transformation kinetics, to identify products formed, and to evaluate phytotoxic effects on the TNT transformation process. At higher initial TNT concentrations, TNT removal rate constant decreased, however, total amount of TNT removed was increased in the culture media. Reductive transformation products of TNT were not detected in the plant culture media but higher concentrations of reduced metabolites were detected in the root and stem extracts of plant material at the 7 days of batch incubation. From these results we concluded that Abutilion avicennae has an intrinsic capacity for taking up and transforming TNT.

  • A new grafted cactus cultivar, called 'Hwangseon', Gymnocalycium mihanovichii, is a hybrid crossed between two orange color cultivars 'Geoseong' and 'Huhwang'. Mating was performed in the greenhouse of National Institute of Horticultural & Herbal Science (NIHHS) in 2005. The harvested seeds were sown at 100 mL volumetric flasks filled with 'Kyoto' medium. Seedling had been grafted into a young rootstock, Hylocereus trigonus, and cultured in vitro in 100 mL test tube for more than six months. After re-grafting and planting in the green house in 2006, their characteristics were evaluated three times until 2009. The new cultivar has a flat round shape and bright yellow skin color. In addition, it has 7 to 9 ribs with gray and straight spines on the body and produces an average of 12.3 offsets.

Studies on Resistance of Rice to the Leaf and Planthoppers (한국 주요 수도품종의 부진자류 저항성에 관하여)

  • The pbjective of this study to clarify the varietal resistance to green leaf-and planthopper and the evaluate the nature of the resistance to the insects in connection with the Antibiosis. this study was investigated the reaction of the varieties to insects with 46 rice varieties including recommended varieties of korea, At seedling stage they were in fested with second of third inster nymphs. the results were as follows. 1. Most of the rice vari eties tested showed highly susceptible reaction to Brown planthopper except Mudgo, KR 108-143- and HR529-41-3-2 as resistance, and Akamoch, Satominori, IR 24 and IR 8 asmoderately suscptible. 2. In the test of varietal resistance to white backed planthopper Mudgo, KR 108-243-1, KR 109-154-2 and HR 529-45-3-2 were resistance, and Suweon 82, Tongil, IR 8 palkeum, Iri 309, Hokwang and Chuhoku #31 were Moderatly susceptible. 3. Most of the varieties tested were observed highly resistance to green leafhopper, but Tongil, Suweon 82, IR 24, Milsung and Chuhoku #31 were Moderatly susceptible. 4. the new bred lines HR 108-243-1 are multiple resistance of Brown planthopper, green leafhopper and white babked planthopper, and some others showed moderatly susceptible as. IR 8: Brown planthopper, white backed planthopper. IR 24: Brown poanthopper, green leafhopper. Tongil: suweon #82, Chukoku #31, whist backed planthopper, Green leafhopper.

Germination Percentages of Different Types of Sweet Corn in Relation to Harvesting Dates

  • Germination of sweet and super sweet corn is lower than normal corn due to the higher sugar and lower starch contents of kernels. Sweet corn seeds are easily deteriorated in the field under the unfavorable condition, therefore it is important to identify the optimal harvesting time for seed production. This trial was conducted to investigate the responses of germination percentage of shrunken-2(sh2), brittle(bt), sugary(su), and sugary enhancer(se) hybrids in relation to harvesting dates. Eight hybrids of four different gene sweet corns were harvested at 15, 20, 25, 30, 35, 40, 45, and 50 days after silking(DAS). Germination test was performed using paper towel method. Mean germination percentages across eight hybrids showed the highest value at 45 DAS. There were significant differences among genes and within gene for germination. Shrunken-2 hybrid Mecca was higher than su hybrids for germination, indicating that sh2 would not be poorer than su Late harvesting beyond the optimal harvesting date might not be desirable because of more lodging and ear rots. Theoretical optimal harvesting date estimated from the regression equation was 40.9 DAS, however, practical date for harvesting would be a few days later than the estimated date if seedling vigor might be considered. Kernel dry weight per ear showed similar response to germination. Regression equation showed the highest kernel dry weight at 40.7 DAS. Significant correlations between kernel dry weight and germination were observed, impling that kernel dry matter accumulation would be an important factor for germination.

Phytotoxic Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Biomaterial, on Rice and Barnyardgrass

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide by the action of the protoporphyrinogen IX oxidase (Protox IX). A study was conducted to determine photodynamic herbicidal effect of ALA on seedling growth of rice (Oryza sativa L.) and barnyard grass (Echinochloa crus-galli Beauv. var. oryzicola Ohwi) under dry and wet conditions. ALA effect on early plant growth of rice and barnyardgrass was greatly concentration dependant, suggesting that it promotes plant growth at very low concentration and inhibits at high concentration. No significant difference in herbicidal activity of biologically and synthetically produced ALAs on plant lengths of test plants was observed ALA exhibited significant photodynamic activity regardless of PSDIP and its duration. Significant shoot growth inhibition by ALA soaking treatment exhibited apparently, indicating that ALA absorbed through root system was translocated into shoot part of plants. ALA reduced plant heights of rice and barnyardgrass seedlings by 6% and 27%, respectively, showing more tolerant to ALA in rice under wet condition. Leaf thickness was reduced markedly by ALA with increasing of ALA concentration, due to mainly membrane destruction and severe loss of turgidity in mesophyll cells, although the epidermal was little affected. It was observed that photodynamic herbicidal activity of ALA applied by pre-and post-emergence application exhibited differently on plant species, and that the activity of ALA against susceptible plants was highly correlated with growing condition.
