Effects of Rearing Condition and Species of Microalgae on Growth and Survival of Larvae of the Sunray Surf Clam, Mactra chinensis (사육환경과 먹이종류에 따른 개량조개, Mactra chinensis 유생의 성장과 생존)

  • Rearing condition and species of microalgae on growth and survival of the sunray surf clam, Mactra chinensis larvae were investigated for artificial seedling production. The larvae of M. chinensis on higher temperature showed high growth and low survival and was grown over $230{\mu}m$ in shell length 14 days after hatching, but low growth as $151.1{\mu}m$ at $18^{\circ}C$. The larvae of M. chinensis on salinity showed highest daily growth and survival as $11.3{\mu}m$ and 65.8% at 30 psu, respectively. The optimum of water temperature and salinity for the larval rearing were $23^{\circ}C$ and 30-35 psu over 25 psu at least. The density of larval rearing was below 10 per 1 ml in rearing seawater for elevating the development rate from D-shaped to settled (metamorphosing) stage. The larvae fed the mixed diet of Isochrysis galbana, Pavlova lutheri, Isochrysis sp. (green), Chlorella ellipsoidea showed highest growth and survival. The larvae fed the single diet of I. galbana, Isochrysis sp. (green) showed high growth and survival. But the larvae fed the single diet of P. lutheri and C. ellipsoidea showed low growth and survival. The optimum diet of larvae of M. chinensis was over two species of microalgae included I. galbana, Isochrysis sp. (green) for elevating the high growth and survival.

Effect of Nurseries on Production of High Quality Seedlings in Panax ginseng C. A. Meyer (우량묘 생산을 위한 삼 묘대에 관한 연구)

  • This study was conducted to produce high quality ginseng seedlings by improvement of the physical properties of nursery soil. In order to select optimum nursery conditions, nursery types were treated with the yangiik nursery and semi-yangiik nursery. Soil conditioners were treated with two different materials of puffed rice husks and perlite. This experiments were tested at Gyunggido Agricultural Research and Extention Services, Hwaseng, from 2003 to 2004. Soil permeability was better at the plot of yangiik nursery than that of semi-yangjik nursery, and was better at plot of puffed rice husks than that of perlite. Soil porosity was improved 2.2% at plot of yangjiik nursery compare at that of semi-yangjik nursery, and was improved 2.0% at plot of puffed rice husks compared to control. There was no difference in the number of ginseng plants per kan among treatments. Root length was longer at plot of yangjik nursery than that of semi-yangjik nursery. More available ginseng plants per kan were obtained from the plot of yangjik nursery by more 252 seedlings than that of semi-yangjik nursery, but there was no significant difference between soil conditioners. The number of available seedlings per kan produced from yangjik nursery was 21.7% higher than those produced by semi-yangjik nursery. The number of available seedlings per kan produced from plot of puffed rice husks was 16.3% higher than that of control.

Critical Saline Concentration of Soil and Water for Rice Cultivation on a Reclaimed Saline Soil (간척지 벼 재배시 토양 및 관개수 염의 안전 한계농도)

  • Reclaimed tidal areas for rice cultivation are irrigated with salt mixed water when there is severe drought. Therefore, we identified the critical concentration of saline water for rice growth on a reclaimed saline soil in Korea. The experiment was conducted at the Kyehwado substation of the National Honam Agricultural Experiment Station (NHAES) during 2001-2002. Two experimental fields with 0.1-0.2% for low soil salinity and 0.3-0.4% for medium soil salinity levels were used. The experiment involved four levels of salt solution mixed with sea water (at 0.1, 0.3, 0.5, 0.7%) compared with a control using tap water in a split-plot design with three replicates. Saline solution was applied only two times at seedling stage (10 DAT and 25 DAT) for 5 days. Gyehwabyeo and dongjinbyeo, japonica rice varieties, were used in this experiment. Plant height and number of tillers sharply decreased in the 0.5% saline water in low soil salinity level and 0.1% in medium soil salinity level. For yield components, panicle number per unit area and percentage of ripened grain dramatically decreased in the 0.5% saline water in low soil salinity and 0.1% in medium soil salinity level. But 1,000-grain weight of brown rice decreased sharply in the 0.5% saline water in low soil salinity and 0.3% in medium soil salinity, indicating that this component was not much affected unlike other yield components. Milled rice yield decreased significantly with saline water level in both low and medium soil salinity. In the 0.7% low saline soil, the yield index was only 36% compared with the control. In medium soil salinity, even the control plot showed only 62% yield index compared with the control in the low soil salinity treatment. Results indicated that the critical concentration of saline water for rice growth in terms of economical income of rice production was 0.5% in low soil salinity and tap water in medium soil salinity.

  • Along with the worldwide rapid increase of the cultivation area and commercial production of genetically modified (GM) crops, the amount of GM grains imported to Korea has also been increasing. Roundup-Ready soybean (RRS) was introduced with 5-enolpyruvyl shikimate-3-photphate synthase (EPSPS) gene derived from Agrobacterium CP4 to confer the resistance to herbicide, glyphosate. In this study, we tried to develop PCR-based analytical method to detection the presence of RRS among non-GM soybeans. In order to detect RRS specifically, oligonucleotide primers were specifically designed based on the nucleotide sequence of EPSPS transgene. Qualitative PCR method was established and its specificity and accuracy were confirmed by analysing the nucleotide sequence of PCR DNA fragments. Bioassay was also conducted by spraying glyphosate at seedling stage. Survived individuals showed obvious resistance to Roundup Ready, however all of non-GM seedlings died in two weeks after spray. Conclusively, the highly selective detection systems for RRS were successfully established by both PCR using specific primers to EPSPS transgene and bioassay using the herbicide resistance of RRS. In addition to, the imported soybean showed to be mixed to several varieties regarding to 100-seed weight and hilum color.

Effects of Highland-Raising Seedlings for Retarding Culture of Cucumber in Lowland (오이의 평난지 억제재배를 위한 고랭지육묘 효과)

  • This study was conducted to investigate the effect of highland (Daekwallyung, altitude of 800m above sea level) production of cucumber (Cucumis sativus L.) seedlings for retarding culture in lowland (Kangnung, altitude of 20m above sea level). Cucumbers (cv. Jangilbanbaek, Jeongsunyeoreum and Ginseollok) were raised during 25 days from 31st July to 25th August, and then planted under the rain-shelter at Kangnung 25th August. Mean and minimum temperature during raising seedlings at Daekwallyung, $20.9^{\circ}C$ and $16.1^{\circ}C$ were $3.7^{\circ}C$ and $5.3^{\circ}C$ lower than those of Kangnung, respectively. T/R ratio of the seedlings produced at Daekwallyung was lower than that of Kangnung, but chlorophyll contents were higher. Daekwallyung seedlings were higher female/male ratio compared with seedlings from Kangnung. Marketable yield from cucumber seedlings (cv. Jeongsunyeoreum) produced at Daekwallyung was 32% higher than that of Kangnung. These results showed that cucumber seedling raising place was Daekwallyung for the retarding culture during summer season, particularly, and the best cultivars was Baekchim (cv. Jeongsunyeoreum).

  • The growth, development, and morphological characteristics of Echinochtoa colons (L.) Link were determined through one life cycle. E. colons emerged 2 to 3 days after seeding (DAS) and reached the two leaf stage by 8 DAS. During the early growth stages, root length was greater than plant height, but the relationship was reversed from 4 weeks after seeding (WAS). Tillering started from the third leaf of the main culm as the sixth leaf on the main calm emerged. The unfolding of the leaves and tillering followed a regular pattern during the vegetative growth period. This resulted in the production of 19 tillers (5 primary, 12 secondary, 1 tertiary, and I nodal) at the 14-leaf stage. Shoot-root weight ratio was highest just before panicle initiation. The second spike from the top of the panicle was the shortest and produced the fewest seeds. Thereafter, spike length and the number of seeds per spike generally increased, the lower the position of the spike on the panicle. Seeds on the lower spikes weighed less and had lower germination ability than those from the upper spikes. Adventitious roots arose from the leaf sheath bases of a flowering stalk. The ability to produce adventitious roots was greater in a younger stalls than in an older stalk.

Mechanization for Labor-Saving in Seeding and Harvesting of Bupleurum falcatum L. (시호(柴胡) 파종(播種) 및 수확(收穫)의 성력기계화(省力機械化))

  • This experiment was conducted to know the labor saving effect and reducing production cost by agricultural mechanization in the cultivation of Bupleurum falcatum. Labor reducing effects of the drilling seeder by hand and the machine attached to two wheel tiller were 97%, but emergency rate was highest in the former. Dry root yield per plant was increased by low amount of seed sowing but that yield per unit area was increased at much seeding amount in the seeder attached to the tiller. The drilling seeder by hand was showed highest standing ratio of seedling and produced yield to 84.1kg of root yield per 10a. Labor saving effect was the best at the multipurposes mechanized harvester and labor saving and famer's income ratio were increased to 69% and 50% respectively. Labor time and cost were reduced to 74% and 69% respectively by mechanization of sowing and harvest cultivation practice on Bupleurum falcatum.

Dietary Value of Three Benthic Diatom Species on Haliotis discus hannai Larvae (북방전복 Haliotis discus hannai 유생에 대한 3종 부착 규조류의 먹이효율)

  • Although the method of seedling production of Haliotis discus hannai is well known, the optimum benthic diatom species as a live food at early larval stage are not fully developed. In this study three Pennales diatom species, Caloneis schroederi, Rhaphoneis sp., and Cocconeis californica were examined on settlement, metamorphosis, survival, and growth of Haliotis discus hannai larvae. The larvae fed Raphoneis sp. or C. californica showed high settlement rate with 80-82% within 48 hrs, which was significantly higher than those fed C. schroederi or mixed diets with three diatom species. The larvae fed the former microalgal species also showed higher metamorphosis rate with 32-34% than the latter species with 10-12% within 4 days. With regard to survival and growth of the larvae, single diet with Rhaphoneis sp. or C. californica had better dietary value than the mixed diets for the early larvae of H. discus hannai.

Dietary Value of Frozen and Freeze-Dried Tetraselmis suecica (냉동.동결건조된 Tetraselmis suecica의 먹이효과)

  • To substitute fromzed and freeze-dried food for live food in the development of the artificial seedling production of bivalve larvae, the dietary value of live T. suecica was compared with those of freeze-dried T. suecica, frozed T. suecica, live T. suecica (50%)+frozen T. suecica (50%), and live T. suecica (50%)+freeze-dried T. suecica (50%) in the rearing of oyster (Crassotrea gigas) and hen clam (Mactra chinensis) larvae, and manila clam (Tapes philipninarum) spats. Oyster larvae fed live T. suecica showed the highest growth (shell hight $231.9^{\mu}$m) and survival rate (72.6%) and those fed freeze-dried T. suecica showed the lowest growth (shell height $168.9^{\mu}$m) and survival rate (35.3%). However, in the hen clam larvae, there were not significantly different among diet group in growth and survival rate. The small spats of manila clam fed live t. suecica or live T. suecica (50%)+freeze-dried T. suecica (50%) showed higher growth and survival rate than those fed other diet group. In the case of large spats of manila clam, live T. suecica and live T. suecica (50%)+frozen T. suecica (50%) showed better growht. But, survival rates were not different among diet groups. Dietary valuse of frozen and dried T. suecica were different on species and growth stage, and frozen and freeze-dried T. suecica can be partially used as substitute food for T. suecica live T. succica in shellfish hatchery.

Natural Spawning and Characteristics of Egg Development of the Indoor Cultured Grunt, Haphlogenys nitens (동갈돗돔, Hapalogenys nitens의 자연산란과 난발생 특성)

  • This study was carried out to obtain the fundamental data for the mass seedling production of grunt, Hapalogenys nitens in terms of the natural spawning and some characteristics of the eggs spawned. The wild grunt were reared at indoor tanks for three years. The adults spawners were 34.0∼44.0 cm (38.6$\pm$4.0 cm, n=7) in total length, 1.00∼2.23 kg (1.62$\pm$0.50 kg, n=7) in body weight. Spawning were observed 9 times from September 22 to October 1, 2000 and 37 times from August 22 to October 3, 2001, with a water temperature range of 19.8$\pm$28.5$^{\circ}C$. The total number of eggs collected was 2.29${\times}$10$^{7}$ (1.7${\times}$10$^{3}$/ml). The relative proportion of floating eggs to total eggs was 41.7%. The fertilization rate of floating eggs was ranged between 85.0 and 99.9% and the hatching rate was ranged between 2.9 and 93.0%. Fertilized eggs were buoyant and spherical in shape, and were 0.85∼0.98 mm in diameter. Each egg contained 1-5 oil globules which were, 0.18∼0.25 mm in diameter. The incubation time from fertilization to blastodisc formation was 10 minutes, to blastula was 3 hours, and to the hatched larvae at 26$^{\circ}C$ was 20 hours 30 minutes. The newly hatched larvae attained total length of 1.81$\pm$0.18 mm. The time required from fertilization to hatching was 31∼34 hours at 23$^{\circ}C$ and 17∼20 hours at 29$^{\circ}C$.
