Guava (Psidium guajava L.) contains high amount of vitamins and minerals, and its leaves have been reported to be very effective on reducing blood pressure. In this study, antimicrobial characteristics of extracts from four different parts of guava (fruit, branch, leaf, and seed) with four different solvents (methanol, ethanol, acetone, and water) were evaluated. Four targeted food-borne microorganisms were selected; two Gram negatives (Escherichia coli O157:H7 and Salmonella Typhimurium) and two Gram positives (Staphylococcus aureus and Listeria monocytogenes). By the paper disc method, guava extracts showed stronger clear hollow zone against Gram positive bacteria than Gram negatives in nutrient broth agar plate. Especially, extracts of branches and leaves showed significant antibacterial activity. Guava extracts also showed significant inhibition activity on the growth of Gram positive food-borne bacteria in nutrient broth. For example, S. aureus did not grow at all at 200 ppm of acetone extracts of guava branch and leaf. In the case of L. monocytogenes, the same concentration of acetone extracts of guava branch and leaf inhibited the growth 33.5% and 55.4%, respectively, at 32 hrs of incubation time. The results indicated that extracts of guava branch and leaf showed significant antibacterial activities against food-borne Gram positive microorganisms, and that guava branches, the byproducts of guava, might be a valuable resource for antibacterial materials.
The aim of this research was to develop specific and sensitive PCR-based procedures for simultaneous detection of economically important plant pathogenic bacteria and seed borne virus in commercial Brassicaceae crop seeds, Xanthomonns campestris pv. campestris (Xcc) and Lettuce Mosaic Virus (LMV). Bacterial and virus diseases of Brassicaceae leaves are responsible for heavy losses. PCR with arbitral primers: selection of specific primers, performance of PCR with specific primers and determination of the threshold level for pathogens detection. To detect simultaneously the Xcc and LMV in commercial Brassicaceae crop seeds (lettuce, kohlrabi, radish, chinese cabbage and cabbage), two pairs of specific primer (LMV-F/R, Xcc-F/R) were synthesized by using primer-blast program (http://www.ncbi.nlm.nih.gov/tools/primer-blast/). The multiplex PCR for the two pathogens in Brassicaceae crop seeds could detect specifically without interference among primers and/or cDNA of other plant pathogens. The pathogen detection limit was determined at 1 ng of RNA extracted from pathogens. In the total PCR results for pathogen detection using commercial kohlrabi (10 varieties), lettuce (50 varieties), radish (20 varieties), chinese cabbage (20 varieties) and cabbage (20 varieties), LMV and Xcc were detected from 39 and 2 varieties, respectively. In the PCR result of lettuce, LMV and Xcc were simultaneously detected in 8 varieties.
Rice bakanae is a typical seed-borne and seed-transmitted disease caused by infection by Fusarium fujikuroi. Seed disinfection using chemical fungicides (such as benomyl and prochloraz) is most effective in controlling the disease, but the emergence of fungicide-resistant strains has recently been increasing. Slightly acidic hypochlorous acid water (SAHW) is a safe and environmentally friendly disinfectant that has a potent and broad spectrum of antimicrobial activity against viruses, bacteria, and fungi. In this study, we aimed to investigate the effectiveness of SAHW against F. fujikuroi strains, including chemical fungicide-resistant strains, as an alternative to conventional chemical fungicides in the management of bakanae disease. SAHW showed strong but similar levels of antifungal activity among the F. fujikuroi strains with a minimum inhibitory concentration (MIC90) of 5±2.5 ppm of free available chlorine (FAC). In addition, F. fujikuroi cells lost viability completely within 5 min of SAHW treatment due to the lethal damage to cell integrity. When the rice seeds infected by F. fujikuroi were treated with SAHW containing 20±10 ppm of FAC for 12 hr, the efficiencies of seed disinfection and disease control were 95-98% and 90.1-92.6%, respectively. Altogether, our data suggest that SAHW is an effective compound for controlling rice bakanae disease.
Jatropha curcas L. (Physic nut) is a commercially important non-edible oil seed crop known for its use as an alternate source of biodiesel. In order to investigate the morphogenic potential of immature embryo, explants from four developmental stages were cultured on medium supplemented with combinations of auxins and cytokinins. It was found that the size of embryo is critical for the establishment of callus. Immature embryos (1.1-1.5 cm) obtained from the fruits 6 weeks after pollination showed a good response of morphogenic callus induction (85.7%) and subsequent plant regeneration (70%) with the maximum number of plantlets (4.7/explant) on Murashige and Skoog's (MS) medium supplemented with IBA (0.5 $mg\;l^{-1}$) and BA (1.0 $mg\;l^{-1}$). The above medium when supplemented with growth adjuvants such as 100 $mg\;l^{-1}$ casein hydrolysate + 200 $mg\;l^{-1}$ L-glutamine + 8.0 $mg\;l^{-1}$$CuSO_4$ resulted in an even higher frequency of callus induction (100%). Plant regeneration (90%) with the maximum number of plantlets (10/explant) was achieved on MS medium supplemented with 500 $mg\;l^{-1}$ polyvinyl pyrrolidone + 30 $mg\;l^{-1}$ citric acid + 1 $mg\;l^{-1}$ BA + 0.5 $mg\;l^{-1}$ Kn + 0.25 $mg\;l^{-1}$ IBA. It was observed that plantlet regeneration could occur either through organogenesis of morphogenic callus or via multiplication of pre-existing meristem in immature embryos. The age of immature embryos and addition of a combination of growth adjuvants to the culture medium appear to be critical for obtaining high regeneration rates. Well-developed shoots rooted on half-halfstrength MS medium supplemented with 0.5 $mg\;l^{-1}$ IBA and 342 $mg\;l^{-1}$ trehalose. The rooted plants after acclimatization were successfully transferred to the field in different agro-climatic zones in India. This protocol has been successfully evaluated on five elite lines of J. curcas.
Growth rates of the low temperature growing isolates, Pseudomonas fluorescens M45 and MC07, reached maximum stationary phase within 50 hrs at the low temperature, 4$^{\circ}C$. But an ordinary biocontrol agent P. putida Pf3 did not reach logarithmic growth phase until 80 hrs. The culture filtrates of M45 and MC07 significantly inhibited the mycelial growths of Pythium ultimum, Rhizoctonia solani and Phytophthora capsici. Detached cotyledons of cucumber grown on Murashige and Skoog agar medium were much enhanced in their growth, compared to those without the filtrates. Population densities of M45 and MC07 in the rhizosphere at 14$^{\circ}C$ were more stable than at 27$^{\circ}C$. When M45 and MC07 were treated into soil, the population density of MC07 continuously increased up to 9 days after treatment, and sustained the initial inoculum density up to 60 days. Cucumber damping-offs caused by P. ultimum and R. solani were significantly reduced by applying M45 as seed-inoculant and by soil treatment with MC07. The combined treatment of M45 and MC07 provided greater effect in reducing the disease incidence than that obtained by single treatments.
In order to find out the effect of medicinal plant extracts on germination of zoosporan-gium and mycelium growth of Pythium ultimum, this study was carried out. Among 28 species in 16 families of plants tested, plant extracts from 9 species were strongly inhibitory to zoosporan-gium germination of P. ultimum. Plant extracts from 3 species were strongly inhibitory to mycellium growth of P. ultimum. Especially, Paeonia suffruticosa was strongly inhibitory. P. suffruticosa was shown a strong control effect on damping-off of sesame by P. ultimum sesame, but no effect on cucumber. Seed germination of sesame and cucumber was shown phyto-alexin by extract of Phytolacca esculenta.

Fusarium spp. were isolated from field grown rice, wheat and barley in 1976. The pathogens isolated included Fusarium (Calonectria) nivale, F. (Gibberella) moniliforme and F. (Gibberella) roseum 'Graminearum' and 'Avenaceum'. Among the saprophytes F. (Nectria) episphaeria was isolated. In each of these isolated both the Fusarium and perfect stages were found. F. nivale, and F. episphaeria with there Calonectria, and Nectria stages do not seem to have been recorded previously in Korea. Of the Fusaria isolated, $66.3\%$ from rice were F. moniliforme, and $68.2\%$ from wheat and barley were F. roseum 'Graminearum'. Perithecia also were produced under laboratory conditions. F. moniiforme was recovered wheat heads and also from barley seed.

Burkholderia glumae (bacterial grain rot), Xanthomonas oryzae pv. oryzae (bacterial leaf blight), and Acidovorax avenae subsp. avenae (bacterial brown stripe) are major seedborne pathogens of rice. Based on the 16S and 23S rDNA sequences for A. avenae subsp. avenae and B. glumae, and transposase A gene sequence for X. oryzae pv. oryzae, three sets of primers had been designed to produce 402 bp for B. glumae, 490 bp for X. oryzae, and 290 bp for A. avenae subsp. avenae with the $63^{\circ}C$ as an optimum annealing temperature. Samples collected from naturally infected fields were detected with two bacteria, B. glumae and A. avenae subsp. avenae but X. oryzae pv. oryzae was not detected. This assay can be used to identify pathogens directly from infected seeds, and will be an effective tool for the identification of the three pathogens in rice plants.
Acidovorax citrulli causes bacterial fruit blotch in Cucurbitaceae, including watermelon. Although A. citrulli is a seed-borne pathogen, it can cause diverse symptoms in other plant organs like leaves, stems and fruits. To determine the infection routes of A. citrulli, we examined the virulence of six isolates (Ac0, Ac1, Ac2, Ac4, Ac8, and Ac11) on watermelon using several inoculation methods. Among six isolates, DNA polymorphism reveals that three isolates Ac0, Ac1, and Ac4 belong to Clonal Complex (CC) group II and the others do CC group I. Ac0, Ac4, and Ac8 isolates efficiently infected seeds during germination in soil, and Ac0 and Ac4 also infected the roots of watermelon seedlings wounded prior to inoculation. Infection through leaves was successful only by three isolates belonging to CC group II, and two of these also infected the mature watermelon fruits. Ac2 did not cause the disease in all assays. Interestingly, three putative type III effectors (Aave_2166, Aave_2708, and Aave_3062) with intact forms were only found in CC group II. Overall, our results indicate that A. citrulli can infect watermelons through diverse routes, and the CC grouping of A. citrulli was only correlated with virulence in leaf infection assays.

Seven seed samples of corn obtained from Kangweon Provincial Office of Rural Development, Kerea were tested for seed-borne fungi, and found that all the samples tested were infected with Fusarium moniliforme to an extent of $6.0{\sim}79.5%$. Severely infected seed samples showed poor germination on blotter. Seed component plating showed that the fungus present not only in tip caps, pericarps and endosperms, but also in embryos. Heavy infection of the fungus caused severe seed rot and seedling blight in soil, but the damage was not severe and many plants grew without any symptoms when the seeds with light infection were sown in soil. However the fungus was frequently detected from inside of the stems of healthy looking seedlings. The results indicate that the fungus transmit from seed to plant systemically. In inoculation experiments, the fungus produced stem rots on corn plants of 110 days old. The cultivar of Hwangok 3 was revealed more susceptible to the fungus than that of Suweon 19.
