Impact of Topsoil Stockpiling Methods on the Viability of Seed Banks

  • The aim of this study was to determine the appropriate stockpiling methods for revegetation by comparing the germination status of seed banks before and after preservation for 2 years. Soil temperature in stockpiled topsoil was higher in open treatment and at 1.5 m, whereas soil water content was maintained at lower levels (14.06-19.08%), than those in the control group. The seed banks in stockpiled topsoil had 48 species and 1,559 individuals, among which perennials showed the highest number in terms of life forms, whereas Compositae and Gramineae were dominant in terms of families. Based on seed bank type, persistent seed banks had the highest number of species, while transient seed banks had the highest number of individuals. By stockpiling period, the number of species in the seed bank started to increase after 24 months, while the number of individuals began increasing after 12 months and exceeded that of the control group after 24 months. Regarding the treatment of stockpiling methods, the number of species and individuals in open treatment were closer to those of the control group. When analyzed by height, the number of species and individuals were higher at 0 m, but still lower than those of the control group. A multivariate analysis of variance (MANOVA ) showed that the optimal combination was obtained in open treatment and the number of individuals increased with the lengthening of the stockpiling period.

Effects of Osmotic and Non-Osmotic Priming Methods for Enhancing Germination of Pepper Seed

  • This study was conducted to investigate the effects of osmotic priming, liquid smoke (LS), sonication and modified drum priming treatments to improve the germination speed and uniformity of pepper seeds for high quality export seeds. Seeds were treated in 0.5, 1.0, 5.0, 10% LS solution only or with 100mM $K_2SO_4$ solution for 6days at $15^{\circ}C$. Sonication treatment was performed for 5, 10, and 20 minutes at an intensity of 5.2, 10.4, 15.7, 21.0, 26.1kHz in water at $15^{\circ}C$. After sonication treatment, seeds were primed with water or 100mM $K_2SO_4$ for 4days. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 84, 96 and 108h in a container with a relative humidity of 99% at 26rpm for a modified drum priming treatments. 0.5% LS treatment with water or 100mM $K_2SO_4$ priming showed significant effects with 71% GP (9.0days MGT) and 66% GP (7.4days MGT), respective, while untreated seeds resulted in only 61% GP and 10days MGT. The healthy seed % was increased by 24% by $K_2SO_4$ priming with LS treatment compared to untreated seeds. However, sonication treatment showed less germination or no difference compared to untreated seed, regardless of intensity and treatment time. The modified drum priming treatment (108h incubation after 50% SMC hydration) significantly improved the GP (on the $4^{th}$days after sowing), germination rate (GR), and MGT to 65%, $28%{\cdot}day^{-1}$, and 3.6days, whereas untreated seed resulted only 7% GP, $17%{\cdot}day^{-1}$ GR and 5.9days MGT. Thus, modified drum priming treatment effectively improved seed germination and is considered as an industrially promising treatment methods considering the shortening of the treatment period and environment-friendly aspects.

Phytotoxic effects of mercury on seed germination and seedling growth of Albizia lebbeck (L.) Benth. (Leguminosae)

  • A study was conducted to determine the phytotoxic effect of mercury on seed germination and seedling growth of an important arid legume tree Albizia lebbeck. The seeds germination and seedling growth performance of A. lebbeck responded differently to mercuric chloride treatment (1 mM, 3 mM, 5 mM and 7 mM) as compared to control. Seed germination of A. lebbeck was significantly (p < 0.05) affected by mercury treatment at 1 mM. Root growth of A. lebbeck was not significantly affected by mercury treatment at 1 mM, and 3 mM. Shoot and root length of A. lebbeck were significantly (p < 0.05) affected by 5 mM concentration of mercury treatment. Increase in concentration of mercury treatment at 5 mM and 7 mM significantly (p < 0.05) reduced seedling dry weight of A. lebbeck. The treatment of mercury at 1 mM decreased high percentage of seed germination (22%), seedling length (10%), root length (21.85%) and seedling dry weight (9%). Highest decrease in seed germination (51%), seedling (34%), root length (48%) and seedling dry weight (41%) of A. lebbeck occurred at 7 mM mercury treatment. A. lebbeck showed high percentage of tolerance (78.14%) to mercury at 1 mM. However, 7 mM concentration of mercury produced lowest percentage of tolerance (51.65%) in A. lebbeck. The seed germination potential and seedling vigor index (SVI) clearly decreased with the higher level of mercury. Plantation of A. lebbeck in mercury-polluted area will help in reducing the burden of mercury pollution. A. lebbeck can serve better in coordinating in land management programs in metal contaminated areas. The identification of the toxic concentration of metals and tolerance indices of A. lebbeck would also be helpful for the establishment of air quality standard.

  • Thin films of $SBT(SrBi_2Ta_2O_9)$ having $Pt/SBT/Seed/Pt/Ti/SiO_2/Si$ structure were fabricated using self-seed layer method by R.F. Magnetron sputter. Self-seed layers were deposited at room temperature and $600^{\circ}C$, which had 30 nm thickness. To investigate crystallization of self-seed layer we characterized by XRD after various heat treatment. And we characterized the crystallinity and electrical properties of SBT on self-seed layer after various heat treatment.

Enhanced Germination of Echinacea angustifolia Seed with Ethephon and Sodium Hypochlortie Treatment

  • The influence of sodium hypochlorite and ethephon on the germination of narrow-leaved purple coneflower (Echinacea angustifolia) was investigated. Treatment of seeds with ethephon (1 mM) for 2 h followed by soaking in 0.525% sodium hypochlorite greatly increased germination (>90%). The treatment of seed with a combination of sodium hypochlorite and ethephon shortened number of days required to reach 50% of the final germination ($T_{50}$) from 4.2 days in control to 1.1 days. Seedlings grown from ethephon-treated seeds had shorter and thicker hypocotyls and roots with higher dry weights compared to the control. The same trend was observed during seedling emergence from soil. Ethephon treatment longer than 2 h resulted in weaker seedlings, probably due to leaf senescence. Anthocyanin content in seedling leaves increased linearly from 0.04 mg/g fresh weight in control to 3.72 mg/g fresh weight in 24 h treatment as the time of seed exposure to ethephon increased. Seed treatment with bleach and ethephon may well be practiced to facilitate the establishment of E. angustifolia in the field.

The Water Content and Treatment Period of SMP for Enhanced Seedling Emergence in Tobacco Seeds. (담배 종자의 입묘율 향상을 위한 SMP 수분함량과 처리기간)

  • The purposes of this study are to investigate water content and treatment period in Solid Matrix Priming(SMP; method for enhanced rapid and uniform seedling emergence in tobacco seeds). The higher water content made the faster imbibition speed of tobacco seed. But the radicle emergence shorten the period of water equilibrium. The range of water content in SMP treatment within the limits of no radicle emergence of tobacco seed was 28∼32% to dry weight of tobacco seed. The rate of radicle emergence and seedling emergence was higher in water content 30% and 40% treatment plot than that in 50% and 60% treatment plot. But it was not different between SMP treatment and non-SMP treatment. In water content 40% during 9 days treatment, germinative energy was highest, and T50 and mean germination time(MGT) were the shortest. The germination speed was higher in SMP treatment than that in non-SMP treatment. It was not different on the germination rate between non-SMP treatment and SMP treatment.

Effect of Seeds Treatment on Germinablity of Tetragonia Tetragonides Seeds (번행초의 대량번식을 위한 종자처리가 발아력에 미치는 영향)

  • Tetragonia tetragonides is a medicinal plant native to ocean sand soil of southern provinces and has significant effects on the prevention and curing of gastroenteric disorders. Despite of its popularity, supply of the plant has never met the level of demand because of the absence of an adequate culturing method. The present study, thereby, was conducted for classifying the plants with geographically different characteristics, studying growth habits, developing a new culturing method and establishing a large scale propagation system of selected superior individual plants. The study was also aimed for revealing optimum conditions for seed treatment, fertilization, and efficient culturing system and thereby, for utilizing the plant as a new income source for rural communities. The seed was elongated with size of 2.6 mm (width) ${\times}$ 1.8 mm (length). No difference in seed size was observed depending on different inhabitate. Each flower produced about 4.5~4.8 seeds. Germination rate was high for seeds matured for 40 days after fertilization, but deceased to 50% for seeds matured only for 20 or 30 days. Seed dormancy lasted 6 months and seed storage at humid $5^{\circ}C$ facilitated germination. Mechanical obstruct of seed germination was due to seed coat and removal of seed coat enhanced the germination rate. Optimum temp. for seed storage was $5^{\circ}C$, and high germination rate was maintained for 350 days. However, for stratification condition or at room temperature, germination was significantly reduced as storage time increased Optimum treatment of plant growth regulators was soaking in $GA_3$ 250 mg/L for 1 hr. The priming treatment with 50 mM $Ca(NO_3)_2$ at $20^{\circ}C$ for two days improved the seed germination with 10% compared to non-treated control. The treatment of 20% NaOCl for 3 hr. improved the seed germination rate up to 10% and 1 day ahead.

The Effect of Ethephon, ABA, BA and NAA Soaking Treatment of Seed Germination of Cool Season Turfgrass -es (한지형 잔디의 종자발아에 미치는 Ethephon,ABA,BA,NAA 침지처리 효과)

  • The study is aimed to understand effects of ethephon(lmg/1), ABA(0.lrng/l), BA(0.lmg/l). and NAA (0.lmg /1) soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw'. These experiments are carried in vitro and their results are summarized as the followings ; In case of ethephon (lmg /1) soaking treatment of seed, the early germination of tall fescue 'Arid' and perennial ryegrass 'Dandy' was good without the soaking times. creeping bentgrass 'Penlinks' and kentucky bluegrass 'Nasaw' obtained the best results at 12 hr. soking treatment, and the longer the soaking time, the higher the germination percentage. The early germination percentage of tall fescue 'Arid' and perennial ryegrass 'Dandy' increased more highly at ABA(0. tmg /1) soaking treatment than at the control. The germination percent-age of creeping loentgrass 'Penlink' and kentucky bluegrass 'Nasaw' had respectively the best results at 6hr. and l2hr. soaking treatment. The early germination percentage of tall fescue 'Arid' and kentucky bluegrass 'Nasaw'increased more highly at BA (0.lmg /1) soaking treatment than at the control, and especially had the best results in 6hr. soaking treatment. perennial ryegrass 'Dandy' had the best results at l0min. soaking treatment, and the longer the soaking time, the lesser the germination percentage. The early germination of perennial ryegrass 'Dandy'. creeping bentgrass 'Penlinks' and tall fes- cue 'Arid' increased more highly at NAA (0.lmg /1) soaking treatment than at the control, and especially had the best results at l2hr. soaking treatment. Studies on the Promoting of Seed Germination of Adenophora triphylla var. Japanica $H_{ARA}$ (잔대종자 발아촉진에 관한 연구)

  • The objective of this study is to promote germination for mass cultivation of Adenophora triphylla. After the seed soaking for I day in BA and $GA_3$ solution, seed germination was effectively promoted in the treatment of 500mg/L $GA_3$ and the highest germination rate was 94%.The average germination days were 1.5days earlier than those of non-treated seeds. In a chemicals treatment, seed germination rate was 54% in 1% $KNO_3$ treatment but KOH treatment was no significantly effected. The durations of chilling treatment for breaking of dormancy were longer than 2weeks. The optimal temperature for germination was $25^{\circ}C$. The promotion of seed germination is presumed to be due to the breaking of dormancy by $GA_3$, chilling treatment rather than seed coat maceration by KOH or $KNO_3$ treatments.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.
