Changes of physicochemical properties of seed longevity from a cross between japonica and weed rice

  • In previously study, we evaluated seed germination for longevity derived from a cross between 'Ilmi' and 'Dharial', a weed rice collected in Bangladesh. The strong germination trait originated from 'Dharial' was incorporated into 'Ilmi', through backcross method. The germination ratio was evaluated after two years of room temperature storage conditions. A high germination ratio of 80.5% in donor plant of 'Dharial' and 77.3% in an introgression lines was observed based on the two years of storage while the recurrent japonica cultivars, 'Ilmi' was failed in germination. In this study, we investigate changes in physicochemical properties of 'Ilmi' and introgression lines (ILs) stored at room temperature. We analyzed germination rate, texture of cooked rice, toyo glossiness value, pasting properties, amylose content, protein content and ${\alpha}-amylase$ content of 'Ilmi' and 5 introgressions every 4 months on the room temperature condition. Seed germinations were decreased by storage periods. Three ILs germination rate was slowly decreased more than 'Ilmi' and 2 another ILs after 4 months. Toyo glossiness value of 'Ilmi' and three ILs were no difference, but, 2 ILs gradually decreased every 4 months at storage periods. Pasting properties were affected by storage temperatures and periods of 'Ilmi' and ILs. The increase at breakdown was observed but setback was decreased by storage periods. Amylose content and protein content were no significant difference at storage periods, respectively. The ${\alpha}-amylase$ content was gradually increased during the storage periods. The introgression line could be useful to increase longevity and maintain quality during storage of japonica rice seed.

대두유전자원의 안전보존을 위한 계통간 발아수명검정 (Storability of Soybean Lines for Maintanance of Germplasm)

  • 대두유전자원의 안전보존을 위해서 50계통을 선정하고 온도 23$^{\circ}C$와 습도 60%가 넘지 않도록 유지시킬 수 있는 본 연구소의 저장고에 종자를 저장하면서 계속 일정 간격으로 발아력을 조사하는 한편 동일 계통의 종자를 온도 4$0^{\circ}C$와 습도 90%에서 인공적으로 노화촉진시켜 저장고에서의 발아력감퇴와 비교하였다. 저장초기의 평균발아율이 97%이었는데 반해 저장 42개월에는 62%로 저하되었는데 비교적 본 저장고에서 오랫동안 발아력이 유지된 셈이다. 종자의 크기와 종피색에 따라 발아력 유지의 정도에 차이가 있는 것으로 나타났는데 대립보다 소립 또한 갈색이나 녹색보다 검정색이나 황색 계통에서 발아력 유지가 오래 지속되는 경향을 보였다. 인공노화촉진처리한 후의 계통간 발아력감퇴차이는 저장고에서의 것과 일치하는 것으로 나타났고, 특히 저장전에 계통간 종자 수명을 파악코저 할 때, 온도 4$0^{\circ}C$와 습도 90%에서 7일 처리가 가장 알맞는 것으로 사료된다.

The Effects of Fatty Acid Composition and Storage Conditions on the Oxidative Stability of Various Vegetable Seed Oils

  • Vegetable seed oils (VSOs) have been extracted and used not only as ingredients in food and as sources of dietary lipids, but also as sources of nutraceuticals used to overcome the various oxidative stresses that contribute to the development of diseases, including cancer and other chronic conditions. The chemical compositions and oxidative stabilities of various VSOs were therefore investigated; samples were stored for 35 d, with each oil having been tested under $O_2$ exposure, sealed from $O_2$ exposure and sealed from $O_2$ exposure while containing $O_2$ scavengers. Oxidative stability was evaluated by peroxide value (POV), p-anisidine value (p-AnV), iodine value (IV), and thiobarbituric acid (TBA) value. Perilla seed and flaxseed oil were mostly composed of linolenic acid (45.5% and 59.7%, respectively), whereas pine seed oil was mostly composed of linoleic acid (48.3%). Meanwhile, camellia seed and olive oils contained 80% oleic acid, which correlated strongly with oxidative stability. The POV, p-AnV, and TBA values were the highest under $O_2$ exposure, and the lowest in the presence of $O_2$ scavengers. These results indicate that VOS oxidative stability depends not only on storage conditions, but on unsaturated fatty acid profiles as well.

자동제어식 파종조절장치 개발 (Development of Automatic Seed Metering Device)

  • Planting, transplanting, and harvesting are important processes for the successful production of farm products in Korea because those require the high labor intensity during limitted period. Recently, many researches of using automatic control with a microcomputer are carried in the agricultural field, but are not much spread to the seeder development. Automatic sowing technology would be much attractive if there was a way to assure that each seed was count accurately in the seed metering device. Thus, an automatic seed metering device was designed and constructed to be controlled by microcomputer. This device could be improved in not only counting the number of seeds in but also sowing seeds between row spacings. Automatic seed metering device consisted of conveyor belt and temporary storage device. Performance of seed metering device depends on the apparatus including sensor, stepping motor and DC-solenoid. Research contents and results are summarized as follows. 1. The seed metering device involving seed hopper, sorter and temporary storage device was designed and constructed. 2. A seed counting system with six photo electric sensors, designed and built for this project, was adequate for tranferring and counting seeds accurately. 3. Operating algorithm for stepping motor and photo electric DC-solenoid was developed. The Seed metering device proved to be a smooth and accurate operating device using the algorithm. 4. The performance of second prototype metering device was examined with five kinds of seeds ; mung beans, red beans, white beans, black beans and corn to transfer and count the seeds. The error ratio of seed metering was less than 3.5%.

쥐오줌풀 개화·결실 특성과 적정 채종방법 및 육묘이식재배에 관한 연구 (Study on Flowering, Bearing Fruit, Seed Harvesting and Seedling Transplanting Cultivation of Valeriana fauriei Briquet)

  • This study was carried out to know the characteristics of flowering and bearing fruit, the optimum period, regions and methods for seed harvesting, the optimum temperatures for seed storage and germination, and the optimum period for sowing at nursery bed and seedling transplanting of Valeriana fauriei Briquet. The flowering and bearing fruit of Valeriana fauriei was developed from the before-year root. Optimum period for seed harvest of Valeriana fauriei was from late July to middle August, and optimum areas were the high elevated areas over 500 m above the sea level as Jinbu-myeon, Pyeongchang-gun, Gangwon-do. Using of net-bag for seed harvesting was the effective method to gather the full ripe seed, and bagging of net-bag was necessary from the season of middle May that was the flowering middle-stage. Germination rates don't show the difference among the different temperatures of storage as approximately 41% at $-20^{\circ}C$, $2^{\circ}C$ or $20^{\circ}C$ of seed storage temperatures. The optimum temperature range was in $15{\sim}30^{\circ}C$ for seed germination at nursery bed. The optimum period for seed sowing at nursery bed was the late February, and the optimum period for seedling transplanting was the middle April.

건조 및 초저온 처리에 의한 뽕나무 종자의 장기 보존 (Cryopreservation of Mulberry(Morus) Seeds in Liquid Nitrogen(L$N_2$))

  • To investigate the possibility of crypreservation of mulberry seeds in liquid nitrogen(LN$_2$), characteristics of the seeds were examined after picking mulberry syncarps and drying-heat treatment. Storage in LN$_2$has the potential of providing indifinite preservation of valuable seed germplasm. Determining the tolerance of seeds among given cultivars to LN$_2$cooling and subsequent rewarming is the first step to establishing the feasibility of LN$_2$storage. Seeds of 4 mulberry varities were treated to LN$_2$(-196$\^{C}$) for 24 hours after drying heat treatment. Seed moisture content of Daeryukppong was the highest. As moisture content of mulberry seed was below 1%, storage in LN$_2$was safe. And drying heat treatment for 60 minutes was suitable to prevent decreased germination rate and germination vigor of seeds. The seeds of Cheongilppong were unsuibable to cryopreserve in LN$_2$for longterm storage.

종자 혼입율과 입도를 달리한 고추가루의 저장 중 색과 매운맛 성분의 변화 (Changes of Colors and Pungent Principles of Red Pepper Powder with Different Seed Contents and Particle Sizes During Storage)

  • Red pepper powders with variuos seed contents (0%, 10%, 20%) and particle sizes were stored at 0$^{\circ}C$, 20$^{\circ}C$, and 30$^{\circ}C$ for 90 days, and the changes in their characteristics were monitored. The initial moisture content was about 13%, and the powders were packed in 0.1 mm-thick polypropylene (PP) bags. Moisture content of each power remained constantly, so PP bags appeared to be effective to prevent moisture transmission. All the values of L*, a* and b* were higher in fine particles than in coarse particles. Storage at 0$^{\circ}C$ did not change any color values, however, overall color values devreased apparently when stored at 30$^{\circ}C$ for 90 days. Capsanthin contents decreased gradually at all storage temperatures and more rapidly at higher temperature. The contents of capsaicinoids in fine particles (49∼59 mg%) were twic as much as those in coarse particles (15∼36 mg%), and they did not change throughout the storage.

Seed Deterioration Response of Different Genes of Sweet Corn during Long-tenn Storage

  • Sweet com seeds deteriorate faster due to low starch content than field com seeds when stored for a long tenn. This study had been conducted to observe the seed deterioration of four different sweet corns in a long tenn storage conditions in room temperature. Four kinds of sweet com genes (sh2, bt, su, and se) were harvested from 15 days to 50 days after silking with 5-day intervals. These seeds were stored in the room temperature and tested for germination percentages from 3 months to 18 months period with 3-month interval. su seeds germinated better than other types of gene. Hybrid Mecca which is sh2 gene germinated better when stored for 3 months to 18 months. For all genes, mean regression equations in relation to storage periods showed linear responses. For regression equation, the slope of sh2 gene was lower than that of su gene. The highest slope value was observed in bt gene showing faster deterioration rate. The rate at which seed deteriorates seems to be affected by the date at which it was harvested. The seeds that were harvested at the optimum time deteriorated more slowly than those which were not.

벼 생태형별 저장기간에 따른 종자 발아율 및 이화학적 특성변화 (Changes of Seed Viability and Physico-Chemical Properties of Milled Rice with Different Ecotypes and Storage Duration)

  • 저장기간이 벼 생태형에 따라 미립 특성에 영향하는 정도를 검토하기 위하여 자포니카형, 통일형 및 통일형에서 유래된 1대잡종형의 정조를 $4{\sim}40$개월간 외부환경이 제어되지 않은 종자 저장실에 저장하여 저장기간에 따른 물리화학적 특성 변화에 대하여 조사한 결과는 다음과 같다. 1. 종자 발아율은 저장기간이 길어짐에 따라 낮아졌는데 자포니카형은 16개월 저장시 66.7%, 28개월 저장시 2.5%로 크게 떨어졌고 40개월 후에는 전혀 발아되지 않았으며, 통일형과 1대잡종형은 28개월까지는 뚜렷한 차이가 없었으나 40개월에는 각각 54.0% 및 67.9%로 발아율의 차이를 보였다. 2. 현미 경도는 통일형, 1대잡종형, 자포니카형의 순서로 높았으며 저장기간에 따른 유의한 차이는 없었다. 3. 알카리 붕괴도는 저장기간이 길어짐에 따라 증가하는 경향이었으며, 단백질 함량은 완만하게 감소하였고, 아밀로즈 함량은 일정한 경향이 없었다. 4. 전당 함량과 환원당 함량은 저장기간이 경과함에 따라 증가하였는데 저장 28 개월 이내에서 증가폭이 컷고 그 이후는 완만한 증가를 보였다. 5. 전당 함량에 대한 환원당 함량의 비율은 저장기간이 길수록 전당 함량보다는 환원당 함량의 증가가 많아서 비율이 높아졌다.

Effect of Various Phyto-extracts on Physico-chemical, Colour, and Oxidative Stability of Pork Frankfurters

  • Lipid oxidation, colour stability and physico-chemical quality of pork frankfurters with the incorporation of 0.30% sea buckthorn (SBT), 0.10% grape seed (GSE), 0.03% green tea (GTE), 0.12% fenugreek seed (FSE) and 0.10% Acacia catechu (ACE) were studied during 20 days of refrigerated aerobic storage. The SBT and ACE were identified as being the most effective antioxidants to retard lipid oxidation with the potency decreasing in the following order: SBT>ACE>GSE>GTE>FSE based on thiobarbituric acid reacting substances, peroxide value and free fatty acids. In all samples pH and $a_w$ decreased during storage period. The $L^*$ value of treated as well as control samples decreased over time while SBT and ACE exhibited an increased redness producing higher $a^*$ values than other treatments. However, GTE was more effective in increasing $b^*$ values than other treatments at the end of storage. The results suggest that functional plant-derived extracts can be valuable to the modification of frankfurter formulations for improved oxidative stability as well as quality characteristics.
