Characterization of a Gamma Radiation-Induced Salt-Tolerant Silage Maize Mutant (방사선 유도 내염성 증진 사료용 옥수수 돌연변이체 특성 분석)

  • Salt stress is a significant factor limiting growth and productivity in crops. However, little is known about the response and resistance mechanism to salt stress in maize. The objective of this research was to develop an enhanced salt-tolerant silage maize by mutagenesis with gamma radiation. To generate gamma radiation-induced salt-tolerant silage maize, we irradiated a KS140 inbred line with 100 Gy gamma rays. Salt tolerance was determined by evaluating plant growth, morphological changes, and gene expression under NaCl stress. We screened 10 salt-tolerant maize inbred lines from 2,248 M2 mutant populations and selected a line showing better growth under salt stress conditions. The selected 140RS516 mutant exhibited improved seed germination and plant growth when compared with the wild-type under salt stress conditions. Enhanced salt tolerance of the 140RS516 mutant was attributed to higher stomatal conductance and proline content. Using whole-genome re-sequencing analysis, a total of 328 single nucleotide polymorphisms and insertions or deletions were identified in the 140RS516 mutant. We found that the expression of the genes involved in salt stress tolerance, ABP9, CIPK21, and CIPK31, was increased by salt stress in the 140RS516 mutant. Our results suggest that the 140RS516 mutant induced by gamma rays could be a good material for developing cultivars with salt tolerance in maize.

A Narrative Literature Review on the Neural Substrates of Cognitive Reserve: Focusing on the Resting-state Functional Magnetic Resonance Imaging Studies (인지예비능의 신경적 기질에 대한 서술적 문헌고찰 연구 : 휴지기 기능적 자기공명영상 연구를 중심으로)

  • Cognitive reserve (CR) is a concept that can explain the discrepancies between the pathologic burden of the disease and clinical manifestations. It refers to the individual susceptibility to age-related brain changes and pathologies related to Alzheimer's disease, thus recognized as a factor affecting the trajectories of the disease. The purpose of this study was to explore the current states of clinical studies on neural substrates of CR in Alzheimer's disease using functional magnetic resonance imaging. We searched for clinical studies on CR using fMRI in the Pubmed, Cochrane library, RISS, KISS and ScienceON on August 14, 2023. Once the online search was finished, studies were selected manually by the inclusion criteria. Finally, we analyzed the characteristics of selected articles and reviewed the neural substrates of CR. Total thirty-four studies were included in this study. As surrogate markers of CR, not only education and occupational complexity, but also composite score and questionnaire-based method, which cover various areas of life, were mainly used. The most utilized methods in resting-state fMRI were independent component analysis, seed-based analysis, and graph theory analysis. Through the analysis, we demonstrated that neuroimaging techniques could capture the neural substrates associated with cognitive reserve. Moreover, functional connectivity of brain regions centered on prefrontal and parietal cortex and network areas such as default mode network showed a significant correlation with CR, which indicated a significant association with cognitive performance. CR may induce differential effects according to the disease status. We hope that this perspective on cognitive reserve would be helpful when conducting clinical researches on the mechanisms of traditional Korean medicine for Alzheimer's disease in the future.

The Influential Factor Analysis in the Technology Valuation of The Agri-Food Industry and the Simulation-Based Valuation Analysis (농식품 산업의 기술평가 영향요인 분석과 시뮬레이션 기반 기술평가 비교)

  • Since 2011, DCF(Discounted Cash Flow) method has been used initiatively for valuating R&D technology assets in the agricultural food industry and recently technology valuation based on royalties comparison among technology transfer transactions has been also carried out in parallel when evaluating the technology assets such as new seed development technologies. Since the DCF method which has been known until now has many input variables to be estimated, sophisticated estimation has been demanded at the time of technology valuation. In addition, considering more similar trading cases when applying sales transaction comparison or industry norm method based on information of technology transfer royalty, it is an important issue that should be taken into account in the same way in the Agri-Food industry. The main input variables used for technology valuation in the Agri-Food industry are life cycle of technology asset, the financial information related to the Agri-Food industry, discount rate, and technology contribution rate. The latest infrastructure building and data updating related to technology valuation has been carried out on a regular basis in the evaluation organization of the Agri-Food segment. This study verifies the key variables that give the most important impact on the results for the existing technology valuation in the Agri-Food industry and clarifies the difference between the existing valuation result and the outcome by referring the support information that is derived through the latest input information applied in DCF method. In addition, while presenting the scheme to complement fragment information which the latest input data just influence result of technology valuation, we tried to perform comparative analysis between the existing valuation results and the evaluated outcome after the latest of reference data for making a decision the input values to be estimated in DCF. To perform these analyzes, it was first selected the representative cases evaluated past in the Agri-Food industry, applied a sensitivity analysis for input variables based on these selected cases, and then executed a simulation analysis utilizing the key input variables derived from sensitivity analysis. The results of this study is to provide the information which there are the need for modernization of the data related to the input variables that are utilized during valuating technology assets in the Agri-Food sector and for building the infrastructure of the key input variables in DCF. Therefore it is expected to provide more fruitful information about the results of valuation.

  • I. Project Title A Study of Brachytherapy for intraocular tumor II. Objective and Importance of the project The eye enucleation or external-beam radiation therapy that has been commonly used for the treatment of intraocular tumor have demerits of visual loss and in deficiency of effective tumor dose. Recently, brachytherapy using the plaques containing radioisotope-now treatment method that decrease the demerits of the above mentioned treatment methods and increase the treatment effect-is introduced and performed in the countries, Our purpose of this research is to design suitable shape of plaque for the ophthalmic brachytherapy, and to measure absorbed doses of Ir-192 ophthalmic plaque and thereby calculate the exact radiation dose of tumor and it's adjacent normal tissue. III. Scope and Contents of the project In order to brachytherapy for intraocular tumor, 1. to determine the eye model and selected suitable radioisotope 2. to design the suitable shape of plaque 3. to measure transmission factor and dose distribution for custom made plaques 4. to compare with the these data and results of computer dose calculation models IV. Results and Proposal for Applications The result were as followed. 1. Eye model was determined as a 25mm diameter sphere, Ir-192 was considered the most appropriate as radioisotope for brachytherapy, because of the size, half, energy and availability. 2. Considering the biological response with human tissue and protection of exposed dose, we made the plaques with gold, of which size were 15mm, 17mm and 20mm in diameter, and 1.5mm in thickness. 3. Transmission factor of plaques are all 0.71 with TLD and film dosimetry at the surface of plaques and 0.45, 0.49 at 1.5mm distance of surface, respectively. 4. As compared the measured data for the plaque with Ir-192 seeds to results of computer dose calculation model by Gary Luxton et al. and CAP-PLAN (Radiation Treatment Planning System), absorbed doses are within ${\pm}10\%$ and distance deviations are within 0.4mm Maximum error is $-11.3\%$ and 0.8mm, respectively. As a result of it, we can treat the intraocular tumor more effectively by using custom made gold plaque and Ir-192 seeds.

Effects of Unripe Rubus coreanus Miquel Extract on Improvement of Lipid Metabolism in C57BL/6 Mice Fed a High-Cholesterol Diet (미숙과 복분자 열매 추출물이 고콜레스테롤 식이를 섭취시킨 C57BL/6 Mice의 지질대사 개선 효과)

  • Hypercholesterolemia is the presence of high levels of cholesterol in the blood, and it is regarded as a risk factor for cardiovascular disease due to the effects of cholesterol. The objective of this study was to investigate the effects of unripe Rubus coreanus Miquel (uRC) extract on lipid metabolism in hypercholesterolemic mice fed a high cholesterol diet (HC). uRC 50 (unripe R. coreanus 5% ethanol extract 50 mg/kg/day), uRC 100 (unripe R. coreanus 5% ethanol extract 100 mg/kg/day), uRC 300 (unripe R. coreanus 5% ethanol extract 300 mg/kg/day), and BO (borage seed oil containing minimum of 20% ${\gamma}$-linolenic acid 30 mg/kg/day) were orally administered for 60 days after HC. Oral administration of uRC 50, uRC 100, uRC 300, and BO significantly reduced serum total-cholesterol, triglyceride, LDL-cholesterol, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, atherogenic index, and cardiac risk factor levels. Similarly, uRC treatment elevated serum HDL-cholesterol levels. These results suggest that unripe R. coreanus extract could be established as a functional food for the improvement of lipid metabolism.

Characterization of typical Aeromonas salmonicida isolated from Sea-Chum Salmon (Oncorhynchus keta) (해수에 순치된 첨연어(Oncorhynchus keta)에서 분리된 정형 에로모나스 살모니시다(Aeromonas salmonicida)에 대한 특성 분석)

  • Chum salmon (Oncorhynchus keta) is a species which returns to Korea for spawning and was produced as seed production at the Fisheries Resources Agency located in Uljin-gun, Gyeongsangbuk-do to preserve the species. However, farmed chum salmon showed symptoms of bacterial infection. Therefore, in this study, bacteria were isolated to identify the causative agent from chum salmon in October 2021. The isolated bacteria were identified based on the sequences of 16S rDNA, rpoD (RNA polymerase sigma factor σ70), and vapA (A-layer) genes. Also, salinity-growth curve, biochemical characterization, antibiotic susceptibility test, and pathogenicity analysis were performed in four strains. As a result, four isolated strains were identified as Aeromonas salmonicida subsp. salmonicida. Additionally, the bacterial strains showed a decrease in growth as the salt concentration increased in the medium. All of the isolated strains exhibited γ-hemolysis, and the same biochemical properties. In the antimicrobial susceptibility test, all strains showed an inhibition zone of 40 to 44 mm for oxolinic acid, flumequine, and florfenicol. Pathogenic factors were assessed by RT-PCR at the mRNA level, and found that the four strains expresses the outer membrane ring of T3SS (ascV), inner membrane ring of T3SS (ascC), vapA, enterotoxin (act), and lipase (lip) genes which are well known to significantly contribute to the pathogenicity of A. salmonicida. The results of this study can be used as basic data to prevent A. salmonicida subsp. salmonicida occurring in sea-chum salmon in the future.

Studies on a Factor Affecting Composts Maturity During Composting of SWine Manure (돈분 퇴비화 중 부숙도에 미치는 영향인자 구명)

  • This study was conducted to investigate indices affecting composts maturity for swine manure compost produced in a commercial composting facility with air-forced from the bottom. The composting was made of swine manure mixed with puffing rice hull(6: 4) and turned by escalating agitator twice a day. Composting samples were collected periodically during a 45-d composting cycle at that system, showing that indices of Ammonium-N to Nitrate-N ratio were sensitive indicators of composting quality. Pile temperature maintained more than 62$^{\circ}C$ and water contents decreased about 20% for 25days of composting. A great variety and high numbers of aerobic thermophilic heterotropic microbes playing critical roles in stability of composts have been examined in the final composts, sbowing that they were detected $10^8$ to $10^{10}$ $CFUg^{-1}$ in mesophilic bacteria, $10^3$ - $10^4$ in fungi and $10^6$ - $10^8$ in actinomycetes, respectively. The results of this study for detennining a factor affecting compost stability evaluations based on composting steps were as follows; 1. Ammonium-N concentrations were highest at the beginning of composting, reaching approximately 421mg/kg. However Ammonium-N concentrations were lower during curing, reaching approximately l04mg/kg just after 45 day. The ratio between $NH_4-N$ and $NO_3-N$ was above II at the beginning of composting and less than 2 at the final step(45 day). 2. Seed germination Index was dependent upon the compost phytotoxicity and its nutrition. The phytotocity caused the GI to low during the period of active composting(till 25 days of composting time) depending on the value of the undiluted. After 25 days of composting time, the GI was dependent upon compost nutrition. The Gennination index of the final step was calculated at over 80 without regard to treatments. 3. E4: E6 ratio in humic acid of composts was correlatively decreased from 8.86 to 6.76 during the period of active composting. After 25 days of composting time, the E4: E6 was consistently decreased from 6.76 to 4.67($r^2$ of total composting period was 0.95). 4. Water soluble carbon had a tendency to increase from 0.54% to 0.78%during the period of active composting. After 25 days of composting time, it was consistently decreased from 0.78% to 0.42%. Water soluble nitrogen increased from 0.22% to 0.32% during the period of 15 days after initial composting while decreased from 0.32% to 0.21% after 15days of composting. In consequence, the correlation coefficient($r^2$) between water soluble carbon and water soluble nitrogen was 0.12 during the period of active composting mule was 0.50 after 25 days of composting time

A New Detailed Assessment for Liquefaction Potential Based on the Liquefaction Driving Effect of the Real Earthquake Motion (실지진하중의 액상화 발생특성에 기초한 액상화 상세평가법)

  • The conventional method for assessment of liquefaction potential proposed by Seed and Idriss has been widely used in most countries because of simplicity of tests. Even though various data such as stress, strain, stress path, and excess pore water pressure can be obtained from the dynamic test, especially, two simple experimental data such as the maximum deviatoric stress and the number of cycles at liquefaction have been used in the conventional assessment. In this study, a new detailed assessment for liquefaction potential to reflect both characteristics of real earthquake motion and dynamic soil resistance is proposed and verified. In the assessment, the safety factor of the liquefaction potential at a given depth of a site can be obtained by the ratio of a resistible cumulative plastic shear strain determined through the performance of the conventional cyclic test and a driving cumulative plastic shear strain calculated from the shear strain time history through the ground response analysis. The last point to cumulate the driving plastic shear strain to initiate soil liquefaction is important for this assessment. From the result of cyclic triaxial test using real earthquake motions, it was concluded that liquefaction under the impact-type earthquake loads would initiate as soon as a peak loading signal was reached. The driving cumulative plastic shear strain, therefore, can be determined by adding all plastic shear strains obtained from the ground response analysis up to the peak point. Through the verification of the proposed assessment, it can be concluded that the proposed assessment for liquefaction potential can be a progressive method to reflect both characteristics of the unique soil resistance and earthquake parameters such as peak earthquake signal, significant duration time, earthquake loading type, and magnitude.

  • Fertilized egg, by successive cell divisions, differentiates into different tissues and organs with various structures and functions. Different cells and tissues contain different proteins, products of selective gene expression. Not all the genes in any genomes are equally active, temporal and spatial gene expression being the general rule. Present paper attempts to review the tanscriptional mechanisms or the initiations of transcription from several angles. In some of the organisms the genes in the process of transcription or the genes in the inactive state can be seen under the light microscope. Some bands of Drosophila polytene chromosomes may exhibit a swollen or puff appearance under certain conditions. A puff, unfolded or decondensed form of chromomere, represents sets of intense transcriptional activity or RNA synthesis. The heterochromatic X chromosome whose genes remain inactive in the female mammals can be visualized as a dark staining structure called Barr body, Configuration of chromatin differs between transcribed and nontranscribed chromatin. Modification to the chromatin facilitates RNA synthesis. The movement of large polymerase molecule along the DNA would probably be facilitated if some modifications of the chromatin configuration is effected. Methylation of cytosines in CG sequences is associated with inactive genes. Methylation can play a role in determination of mammalian cells during embryogenesis. Demethylation is necessary for the gene to be expressed during development A histone modification that is also known to be correlated with transcriptional capacity of chromatin is acetylation of the lysine residues of the core histones. Chromatin containing a high level of histone acetylation is very sensitive to DNase 1. For the transcription to occur TBP must first bind to the TATA box. Another TF, TF IIB, then binds to the promoter-TBP complex, facilitating the access of RNA polymerase to the transcription initiation site. As recently as eight years ago researchers assumed that histones were irrelevant to the regulation of gene expression. Histones combine with the DNA to form nucleosome of the chromatin. Histones are vital participant in gene regulation. Histone and basal factors compete for access to TATA box. When DNA is exposed to basal factors before histones are introduced, the basal factors assemble on TATA boxes preventing the access of histones, allowing transcription to occur, for transcription to begin, activator protein at the upstream activation sequence or enhancer must interact with the tail of histone H4 at TATA box and cause the histone role particle to dissociate from the TATA box leading to partial breakup of the histone core particle and allowing the basal factors to bind to the TATA box. New concept of genomic flux in contrast to the old concept of static genome has been developed based on the powerful new molecular techniques. Genomic changes such as repetitive DNAs and transposable elements, it is assumed but not yet proved, may affect some of the developmental patterns that characterize particular cells, tissues, organs, and organisms. In the last decade or so remarkable achievement have been made in the researches of the structures and functions of TFs and the specific target sequences located in promoters or enhancers where these TFs bind. TFs have independent domains that bind DNA and that activate transcription. DNA binding domain of TFs serves to bring the protein into the right location. There are many types of DNA binding domains. Common types of motifs can be found that are responsible for binding to DNA. The motifs are usually quite short and comprise only a small part of the protein structure. Steroid receptors have domains for hormone binding, DNA binding, and activating transcription. The zinc finger motif comprises a DNA binding domain. Leucine zipper consist of a stretch of amino acids with a leucine residue in every seventh position Two proteins form a dimer because they interact by means of leucine zippers on similar α-helical domain. This positions their DNA binding basic domains for interaction with the two halves of a DNA sequence with dyad symmetry of TGACTCA, ACTGAGT.

Comparison of nutritional characteristics between wild and cultured juvenile black rockfish, Sebastes schlegeli (자연산 및 양식산 조피볼락 치어의 영양학적 특성 비교)

  • Cultured juvenile black rockfish, Sebastes schiegeli as a seed for sen ranching project were compared with the similar size of wild fish. Wild fish were analyzed and compared with cultured fish in body indices, and moisture, crude protein, lipid, ash, many kinds of amino acids and various minerals in the whole-body of juvenile rockfish from three different populations. There were significant (P<0.05) differences in the moisture, crude lipid, and ash in the whole-body of fish among three different populations; the moisture content of wild fish was higher than that of both cultured fish. The lipid content in wild and tank cultured fish was lowest and highest, respectively; The lipid content of fish cultured in embanked system was intermediate values. The condition factor (CE), hepatosomatic index (HSI), and intraperitoneal fat (IPE) of fish cultured in tank system and embanked system were significantly (P<0.05) higher than those of wild fish. But fish cultured in tank system produced similar intestinosomatic index (ISI) and muscle ratio (MR) values as the wild fish. There were significant differences in asparatic acid, glutamic acid, isoleucine, leucine, lysine, phenylalanine, threonine, valine of whole-body fish cultured by different methods; the wild fish was highest in these amino acids, followed by fish cultured in embanked system and then fish cultured in tank system. There were also significant differences in Ca, P, Mg, Mn, Zn of whole-body fish cultured by different methods: wild fish and fish cultured in tank system produced highest in Ca, P, Mg, Zn and lowest in Mn, respectively. There were significant differences in moisture, crude lipid, ash, many kinds of amino acids, several minerals, CE, HSI, UE, and ISI among the three juvenile populations; whole-body of wild fish showed higher in moisture, ash, asparatic acid, glutamic acid, isoleucine, leucine, Iysine, phenylalanine, threonine, valine, Ca, P, Mg, and Zn than fish cultured in embanked system, while they showed lower in the rest of the nutritious parameters studied.
