The Content and Risk Assessment of Heavy Metals in Herbal Pills (유통 환제의 유해 중금속 함량 및 위해도 평가)

  • The objective of this study is investigation of contamination levels and assessment of health risk effects of heavy metals in herbal pills. 31 Items and 93 samples were obtained for this investigation from major herbal medicine producing areas, herbal markets and on-line supermarkets from Jan to Jun in 2010. Inductively coupled plasma mass spectrometer method was conducted for the quantitative analysis of Pb, Cd and As. In addition, the mercury analyzer system was conducted for that of Hg without sample digestion. The average contents of heavy metals in samples were as follows : 0.87 mg/kg for Pb, 0.08 mg/kg for Cd, 2.87 mg/kg for As and 0.16 mg/kg for Hg, respectively. In addition, the average contents of heavy metals in different parts of plants, including cortex, fructus, herba, radix, seed, algae and others were 0.63 mg/kg, 3.94 mg/kg, 1.42 mg/kg, 1.05 mg/kg, 0.16 mg/kg, 22.31 mg/kg and 10.17 mg/kg, respectively. After the estimations of dietary exposure, the acceptable daily intake (ADI), the average daily dose (ADD), the provisional tolerable weekly intake (PTWI) and the relative hazard of heavy metals were evaluated. As the results, the relative hazards compared to PTWI in samples were below the recommended standard of JECFA as Pb 3.1%, Cd 0.9%, Hg 0.5%. Cancer risks through slope factor (SF) by Ministry of Environment Republic Korea and Environmental Protection Agency was $4.24{\times}10^{-7}$ for Pb and $3.38{\times}10^{-4}$ for As (assuming that the total arsenic content was equal to the inorganic arsenic). Based on our results, possible Pb-induced cancer risks in herbal pills according to parts used including cortex, fructus, herba, radix, seed, algae and others were $1.95{\times}10^{-7}$, $1.45{\times}10^{-6}$, $2.14{\times}10^{-7}$, $6.27{\times}10^{-7}$, $1.99{\times}10^{-8}$, $3.61{\times}10^{-7}$ and $9.64{\times}10^{-8}$, respectively. Possible As-induced cancer risks in herbal pills by parts used including cortex, fructus, herba, radix, seed, algae and others were $1.54{\times}10^{-5}$, $7.24{\times}10^{-5}$, $1.23{\times}10^{-4}$, $2.02{\times}10^{-5}$, $3.25{\times}10^{-6}$, $2.18{\times}10^{-3}$ and $5.67{\times}10^{-6}$ respectively. Taken together, these results indicate that the majority of samples except for some samples with relative high contents of heavy metals were safe.

Socheongja and Socheong 2 Extracts Suppress Lipopolysaccharide-induced Inflammation and Oxidative Stress in RAW 264.7 Macrophages through Activating Nrf2/HO-1 Signaling and Suppressing MAPKs Pathway (RAW 264.7 대식세포에서 Nrf2/HO-1 신호 전달계 활성화와 MAPKs 경로 억제를 통한 소청자와 소청2호의 LPS 매개 염증성 및 산화적 스트레스 반응의 억제)

  • Inflammatory response and oxidative stress play critical roles in the development and progression of many human diseases. Therefore, a great deal of attention has been focused on finding functional materials that can control inflammation and oxidative stress simultaneously. The purpose of this study was to investigate the effects of Socheongja and Socheong 2, Korean black seed coat soybean varieties, on the inflammatory and oxidative stress induced by lipopolysaccharide (LPS) in RAW 264.7 macrophages. Our data indicated that the extracts of Socheongja (SCJ) and Socheong 2 (SC2) significantly suppressed LPS-induced production of nitrite oxide (NO) and prostaglandin $E_2$, key pro-inflammatory mediators, by suppressing the expression of inducible NO synthase and cyclooxygenase-2. It was also found that SCJ and SC2 reduced the LPS-induced secretion of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ and $interleukin-1{\beta}$, which was concomitant with a decrease in the protein levels. In addition, SCJ and SC2 markedly diminished LPS-stimulated intracellular reactive oxygen species accumulation, and effectively enhanced nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase (HO)-1 expression. Furthermore, LPS-induced activation of mitogen-activated protein kinases (MAPKs) was abrogated by SCJ and SC2. Taken together, these data suggest that SCJ and SC2 may offer protective roles against LPS-induced inflammatory and oxidative responses in RAW 264.7 macrophages through attenuating MAPKs pathway, and these effects are mediated, at least in part, through activating Nrf2/HO-1 pathway. Given these results, we propose that SCJ and SC2 have therapeutic potential in the treatment of inflammatory and oxidative disorders caused by over-activation of macrophages.

Anti-inflammatory effects of seed ethanolic extracts of the common buckwheat and tartary buckwheat are mediated through the suppression of inducible nitric oxide synthase and pro-inflammatory cytokines in LPS-induced RAW 264.7 macrophage cells (일반메밀과 쓴메밀 종실 추출물의 RAW 264.7 대식세포에서 LPS에 의해 유도되는 iNOS 및 염증성 사이토카인 발현 저해를 통한 항염증 효과 비교)

  • The ethanolic seed extracts of the common buckwheat (CB) and tartary buckwheat (TB) were examined for their anti-oxidant and anti-inflammatory effects on lipopolysaccharide (LPS)-induced RAW 264.7 cells. In this study, it was observed that the rutin content of TB extracts was 65-78 times higher than the CB extracts, while quercetin was only detected in the TB extracts. In addition, TB extracts were observed to have 1.8-2.0 times higher flavonoid and polyphenolic content than the CB extracts. Cytotoxicity was not observed when both the buckwheat extracts were evaluated at concentrations in the range of 6.25-400 ㎍/mL. The treatment with TB extracts significantly suppressed the LPS-induced nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression at the protein and mRNA levels. The TB extracts more potently inhibited the LPS-induced production of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 than the CB extracts. The mRNA levels of TNF-α, IL-1β, and IL-6 were also significantly inhibited both by the TB and CB extracts in a pattern similar to their production.

Synergistic Inhibition of Aronia melanocarpa and Moringa oleifera Seed Extract on Experimental Atopic Dermatitis (아로니아 및 모링가 종자 복합물의 항아토피 상승효과)

  • Atopic dermatitis is a chronic, relapsing inflammatory skin disease. This study aimed to investigate the therapeutic benefits of Aronia melanocarpa (AM) and Moringa oleifera seed extract (MO) on experimental atopic dermatitis. We examined the effects of AM or MO and their combination on 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis in BALB/c mice as well as tumor necrosis factor $(TNF)-{\alpha}$ and interferon $(IFN)-{\gamma}-stimulated$ HaCaT keratinocytes. Mice were orally treated with extract during repeated application of DNCB to shaved dorsal skin. Our results show that treatment with AM and MO in combination reduced histological manifestations such as epidermal hyperplasia and inflammatory cell infiltration. Furthermore, it significantly decreased skin thickness and serum immunoglobulin E (IgE) level compared to the AM or MO alone treated group. Combined extract of AM and MO suppressed expression of $TNF-{\alpha}/IFN-{\gamma}-induced$ T helper 2 (Th2) chemokines such as thymus and activation-regulated chemokine and macrophage-derived chemokine. To sum up, combination of AM and MO suppressed the inflammatory response and serum IgE as an indicator of several allergic diseases in DNCB-induced experimental atopic dermatitis and Th2 chemokine expression in HaCaT cells. This result suggests that combination of AM and MO could be a valuable strategy to improve atopic dermatitis.

STUDIES ON THE DIMORPHISM AND TRANSITION OF BISEXUALITY OF HETEROSTYLOUS POLYGONACEAE (여뀌과 이형경식물의 Dinorphism과 Bisexuality의 변화)

  • The present experiments were designed in order to clarify the differences between the long and short styled plants and the transgressive gradition in the degree of dimorphism among the three heterostylous species of the Polygonus, P. japonica, F. esculentum, and P. senticosa, based on investigations regarding the floral structure, ecological and physiological traits, the results of which are summarized as follows: (1) P. japonica, although it exhibits typical dimorphism, has undergone so high a differentiation between long and short styled that its long styled individuals behave as if they were female; and short styled individuals as if male. In long-styled individuals, filament, anther, and pollen grains show signs of degeneration, most of the pollen being abortive. On the other hand, in short styled individuals, the filament, anther, and pollen grains have attained remarkable development; the pollen grians are large and fertile. In short-plant the fertilized flowers readily drop off in every stage of their embryo development. This species has completely lost the self-fertile property, which is characteristic of the non-dimorphic Polygonum genus. Although this specsei typically exhibits the physiological characteristics of the non-dimorphic Polygonum genus. Although this specisei typically exhibits the physiological characteristics of dimorphism in controlled pollination, the short-styled individuals bear no seed in nature, thus misleading taxonomists to idenfity the short-styled plant as male. 2) The morphological feature of the flower organ of P. senticosa obviously indicates definite dimorphism. Physiologically, however, no differentiation towards dimorphism was observed, the species still retaining, both in long and short-individuals, the self-fertile property common to the Polygonum genus. Elaborate examinations revealed that regardless of the modes of pollination, both fertiization and seed setting flourish, no differentiation betwen legitimate and illegitimate unions being recognizable. This sort of physiological property has not been observed in the investigations of other heterostylous plants. It is assumed that this species is differentiated structurally into dimorphism, but not yet physiologically. In nature, however, this plant would have more opportunities to be cross-pollinated, i.e., legitimately combined, than self-pollinated because of the development of two forms of flowers. 3) In terms of heterostylism, the F. esculentum just occupies the intermediate position between P. japonica and P. senticosa structurally, ecologically, and physiologically. Doescription of some of the physiological behavior of the plant will suffice to demonstrate the above facts. While P. japonica has completely lost its self-fertile property, P. senticosa still retains it wolly. In F. esculentum 2-6% of self-fertility is the result in illegitimate combination. There occur occasionally hereditary self fertile individuals among some of the F. or 20 min. irradiation plot, when they reach any stage of the same bacterial population. In addition to this increase of total population in the plots with the more dose of UV light irradiation, it seems that the more dose of UV light irradiation is the more shortened the generation time of Azotobacter. Therefore, it is clear that variation of reproductive rate must be, mere or less, due to the genetic effects induced by UV light irradiation. On the other hand, the lag phase or logarithmic growth phase in nonirradiated culture is shortened prominently, and this must be due to the difference in bacterial number of the original inoculm. The generation time of Azotobacter is shortened by exogeneous treatment of nuclei acid derivatives, and the degree is greater in case of DNA derivatives than RNA dervatives. W.H. Price reported that the rate of ribose nucleic acid to protein in Staphylococcus muscae is proportional to the generation time: that is the faster the cell can form ribose nucleic acid, the more rapid its growth. This explains the shortening of generation time by exogeneous RNA derivatives in this work reasonably. On the other hand, it is well known that the desoxyribose nuclic acid content per cell is constant and independent of the generation time. A.D. Laren and W.N. Takahashi reported that the infectious RNA from TMV is 6 times as sensitive to inactivation by UV as it is in the form of intact virus, and that inactivation of infectious TMV involves onlu a local change on RNA chain. But, the effect of exogeneous DNA in this work suggests that irradiated living cell which cotain DNA bring about some change on DNA moleculs as well as RNA molecules. And if the mutagenic effects of UV take into consideration, it is very reasonable. Therefore, it is clear that the variation of the generation time by UV irradiation is, more or less, due to the genetic effects. Therefore, it seems that the shortness of the average lifewpan of Azotobacter by UV irradiation is resulted not only from the influence of the environmental conditions, but also from the variation of genetic factor of the individual.

  • This study was carried out to investigate the influence of light intensity on the growth of Korean white pine (Pinus koraiensis) seedlings. The seedlings were grown under four different relative light intensities: 100%, 63%, 37%, and 19% of full sunlight by covering with saran screen on seed bed. The results obtained were summarized as follows; 1. The highest shoot elongation was found at 37% in relative light intensity (RLI) plot, and the best growth in root, seedling length and diameter increment appeared at 100% on RLI plot. Contribution rates of shading to growth in shoot elongation, root growth, seedling length, and diameter increment were 9.3%, 26.2%, 21.0%, and 48.7%, respectively. 2. The greatest number of one-year-old needle fascicle was found at 100% in RLI plot, whereas that of two-year-old needle fascicle appeared at 37% in RLI plot. The contribution rate of the light intensity to number of needle fascicle was 3.9% and factor of leaf age contributed to number of needle fascicle in 27.8%. The length of needles grown under different light intensities varied with needle ages. The longest length of new needles appeared at 37% in RLI plot, but old needles were not clear in influence of light intensity. The contribution rate to needle length by the light intensity was 2.5%. 3. The heaviest value of the fresh and dry weight of seedlings appeared at 100% of RLI plot then decreased with light intensity. The contribution rates to fresh and dry weight by the shading were 38% and 7.6% respectively. 4. The largest value of the dry weight of needles appeared at 100% of RLI plot and then decreased with light intensity. The contribution rates to dry weight of needles by the light intensity was 13.18%. 5. The values of T/R ratio increased with decreased light intensity and the contribution rate was 7.0%. 6. Positive correlation and linear regressions were recognized between dry weight of leaf and other factors (dry weight of shoot, root, seedling and diameter increment).

Vulnerability Assessment on Spring Drought in the Field of Agriculture (농업지대 봄 가뭄에 대한 취약성 평가)

  • Seasons in Korea have very distinguishable features. Due to continental high pressure, spring in Korea is dry and has low precipitation. Due to climate change derived from the increase of greenhouse gases, climate variability had increased and it became harder to predict. This caused the spring drought harsher than usual. Since 1990s, numbers of chronic drought from winter to spring increased in southern regions of Korea. Such drought in the spring damages the growth and development of the crops sown in the spring and decreases its quantity. For stable agricultural production in the future, it is necessary to assess vulnerability of the relationship between spring drought and agricultural production as well as to establish appropriate measures accordingly. This research used CCGIS program to perform vulnerability assessment on spring drought based on climate change scenario SRES A1B, A1FI, A1T, A2, B1, B2 and RCP 8.5 in 232 regions in Korea. As a result, Every scenario showed that vulnerability of spring drought decreased from 2000s to 2050s. Ratio of decrease was 37% under SRES scenario but, 3% under RCP 8.5 scenario. Also, for 2050 prediction, every scenario predicted the highest vulnerability in Chungcheongnam-do. However, RCP-8.5 predicted higher vulnerability in Gyeonggi-do than SRES scenario. The reason for overall decrease in vulnerability of agriculture for future spring drought is because the increase of precipitation was predicted. The assessment of vulnerability by different regions showed that choosing suitable scenario is very important factor.

Correlation of Lignan Contents with Protein and Oil Contents in the Seeds of Sesamum indicum L.

  • The contents of lignan, protein, and oil of nine Sesamum indicum cultivars were analyzed. To determine the lignan contents, sesamin 1 and sesamolin 2 were isolated from the ethylacetate extracts of the Suwon cultivar, and the structures were fully characterized by spectral and physical methods. The seeds of nine cultivars were screened for two lignans, which were determined by HPLC using a $C_{18}$ reversed phase column coupled with a photodiode array detector. Suwon cultivar showed the highest concentration (1: $6.24{\pm}0.04$ and 2: $3.58{\pm}0.01\;mg/g$), whereas Soonheuk displayed the lowest (1: $0.91{\pm}0.01$ and 2: $0.73{\pm}0.01\;mg/g$). The average content ratio of sesamin 1 (3.64 mg/g) was significantly higher than that of sesamolin 2 (2.57 mg/g). The protein content ranged from $21.52{\pm}0.35$ to $31.22{\pm}0.25%$, Suwon containing the highest level and Soonheuk had the lowest. Kwangbaek showed the highest oil level ($49.84{\pm}0.40%$), while Soonheuk exhibited the lowest ($42.52{\pm}0.05%$). Sesamin 1 exhibited a stronger radical-scavenging activity in the ABTS ($IC_{50}:\;63.2{\pm}2.4\;{\mu}M$) than its DPPH radical-scavenging activity ($IC_{50}>200\;{\mu}M$). These results lead to the conclusion that lignan content is affected by protein and oil contents. Concentration of the chemical components in the cultivars could be a key factor in the selection process of a high quality species.

  • Venture companies, which can commercialize various kinds of new ideas and creating added value, are amid of great attention. Since most of venture companies in initial development stages cannot finance its capital sufficiently all by their own, investments from outside investors are key factor to their survival. However, they have lots of difficulties in financing from outside investors because of their typical uncertain but favorable investment characteristics. Moreover, even though they had successfully financed from outsiders, problems related to their stakeholders, such as interrupting in management and types of investments, cause contrary results to firms. Therefore there are various kinds of systems for startups like angels, venture capitals and governmental supports. Even so, investments and supports for venture companies are still not sufficient. 'Accelerator', which is a brand-new investment type started from Silicon Valley in United States during the mid 2000, is growing attention these-days. It mainly supports startups financially in the initial development stages, however, it also supports the firms by providing mentoring, education and networking services. On the other hand, difference between existing investment types and performance of the investment is still in controversy. Therefore this study compared investment behavior between accelerator and venture capital. As a result, we found that the difference in investment behavior of accelerators and venture capital.

Influence of N-alkoxy groups on the activity of photodynamic herbicidal 6-Benzofuryl-2-[1-(alkoxyimino)alkyl]-3-hydroxycyclohex-2-en-1-one derivatives (광역동 6-Benzofuryl-2-[1-(alkoxyimino)alkyl]-3-hydroxycyclohex-2-en-1-one 유도체의 제초활성에 미치는 N-alkoxy기의 영향)

  • New photodynamic herbicidal 2,3-dihydro-2,2,4,5,6-pentamethylbenzofuran-5-yl, (I) and 2,3-dihydro-2-ethyl-2,4,5,6-tetramethylbenzofuran-5-yl, (II) substituents in 6-benzofury-2-[1-( alkoxyimino )alkyl]-3 hydroxycyclohex-2-en-1-one derivatives were synthesized and their herbicidal activities against rice plant (Oryza sativa L.), barnyard grass (Echinochloa crus-galli) and pickerel weed (Monochoria vaginalis presl.) were measured under submerged pre and post-emergence conditions. Particularly, a series of (I) compounds showed good selective herbicidal activities between the 3 leaf stage of rice plant and barnyard grass at rates of $0.25{\sim}0.0007kg/ha$. The structure activity relationships (SAR) on the herbicidal activity with changing N-alkoxy groups were discussed quantitatively. According to the SAR results, it was expected that the selectivity factor between seed of rice plant and barnyard grass should be rely on the N-alkoxy groups with bigger dipole moment and bigger (or smaller) $B_3$ constant than optimal value $(B_3)_{opt.}=4.41{\AA}$. Compared with (I), the (II) substituents showed more superior herbicidal activities.
