Seed Morphological Characteristics and Dormancy type of Eranthis stellata Maxim., Korea Rare Plant. (희귀식물 너도바람꽃(Eranthis stellata Maxim.) 종자의 형태특성 및 휴면유형 분석)

  • Eranthis stellata Maxim. is a perennial plant that grows around the valley. E. stellata is concerned about the decline in natural habitats due to climate change in KOREA, continues to be observed and protected as an endangered species (Least Concrned, LC). Nevertheless, studies on the characteristics of the seeds of E. stellata are insufficient. So, this study analyzed the morphological characteristics and dormancy types of seeds. Seeds of E. stellata was collected in April at Gyeongsangbuk-do Arboretum and kept at 5 ℃ until using. To investigate the morphology of seeds, an optical microscope and a scanning electron microscope (SEM) were used. GA3 treated or untreated seeds (4 replicates of 25 seeds each) were observed germination and embryo growth for 1 month at 5 ℃ and 25/15 ℃ (12h day/12h night). The seed surface of E. stellata, light brown, was observed as a common characteristic of Eranthis genus, reticulate. The short axis of seeds was 1.11~1.77mm (average 1.44mm), and the long axis was 1.27~1.91mm (average 1.63mm), which was investigated in a slightly round shape (subglose). While no germination was observed at all conditions, Embryo growth was observed at 5 ℃ both in the control group and with GA3treated groups. Thus, seeds of E. stellata are classified as morphological physiological dormancy (MDP), which requires embryonic development and dormant break at the same time. These results can be useful information for determining morphological physiological seed dormancy and germination, and will be an important basic data for seed propagation of E. stellata as a resource.

Development of RAPD-SCAR Molecular Marker Related to Seed-hair Characteristic in Carrot (당근(Daucus carota var. sativa) 종자모 형질 관련 RAPD-SCAR 분자표지 개발)

  • Mechanical hair removal of carrot seed causes seed injuries and suppresses the germination in carrot cultivation. This study was performed to develop molecular markers for breeding high quality cultivars with short-hair seed. To meet this objective, random amplified polymorphic DNA (RAPD)-sequence characterized amplified region (SCAR) markers specifically linked to seed-hair characteristic were identified using CT-SMR 616 OP 389-1 line with short-haired seed and CT-SMR 616 OP 616-33 line with long-haired seed, bred by self-pollination for 6 years from 2008 to 2013, as parents. After seed hair lengths of these lines were analyzed using microscope, next generations were advanced and compared with the molecular markers polymorphism. From RAPD analysis using fixed lines in 2011, twelve RAPD primers showing polymorphic bands specific between the two lines were identified from 80 random primers. To develop RAPD-SACR marker, SCAR primers were designed based on sequence analysis of these specific RAPD bands and more than three combinations of primers were tested. As a result, it was found that the $SCA2_{1.2}$ amplified single polymorphic band from short-haired seed line. To confirm this result, $SCA2_{1.2}$ marker was retested by applying to the 2012 and 2013 progenies. Finally, it was concluded that the developed $SCA2_{1.2}$ marker distinguished short-haired line from long-haired seed line. Therefore, SCAR marker, $SCA2_{1.2}$ is expected to be utilized for breeding of the short-haired seed cultivars.

Studies on Seed Germination Characteristics and Patterns of Protein Expression of Achyranthes japonica by Treating Plant Growth Regulators and Seed Primings (생장조절제와 프라이밍 처리에 의한 우슬종자의 발아특성과 단백질 발현 양상에 관한연구)

  • This study was conducted to improve the postharvest storage techniques of managing and storing seeds, to test qualities and viabilities of the seeds and to examine the germination rate and the protein expression of Achyranthes japonica Nakai. The seeds collected from different areas of Je-Cheon and Gwang-Ju were stored with different temperatures and durations. Two plant growth regulators and two seed priming were treated to investigate their effect on the germination rates and the days required for germination. The weight of one hundred seed collected in Gwang-Ju was heavier than those in Je-Cheon. Seed length collected in Gwang-Ju was also longer about 5.12 mm than those in Je-Cheon about 4.90 mm and seed width was longer in Gwang-Ju than those in Je-Cheon. The rates of seed germination in two different collection areas were higher about 2.9 to 13.0% in Gwang-Ju compared to those in Je-Cheon. Comparing its rates with the storing temperatures and durations, they were not clearly different in between $4^{\circ}C$ and $25^{\circ}C$ and they also were gradually decreased with getting longer storing durations. The germination rates treated by plant growth regulators were higher with $GA_3$ than those with Kinetin. The highest seed germination rate was appeared at 50 ppm of $GA_3$. Comparing its rates with different seed priming, they were relatively higher with $KNO_3$ than those with PEG6000. In protein expression patterns between before the germinating and after the germinating of seeds, more and clear bands were appeared in the seed after the germination compared to those before the germination of seeds, especially 10 ~ 20 kDa. These results showing more and clear bands were more clearly appeared in Gwang-Ju compared to Je-Cheon. Comparing the protein expression with plant growth regulators and seed primings, $GA_3$ was better expression than those with Kinetin and $KNO_3$ was better than those with PEG6000. More and clear bands were closely related to the germination rates of seeds and more detailed studies would be required.

Studies on the Variation of Physico-Chemical Characteristics During Storage and Frying (고추씨 기름의 저장 및 가열에 따른 이화학적 변화에 관한 연구)

  • To observe the propability as the frying oil and oxidative stability of red pepper seed oil, some physico-chemical tests of the oil were examined during 21 days storage period at 5$^{\circ}C$, 15$^{\circ}C$ and 30$^{\circ}C$, and AOM value determined and after heating the oil at 180${\pm}$5$^{\circ}C$ for 40 hours with or without antioxidants. The analysis storage stability of red pepper seed oil showed that the quality of storage group at 5$^{\circ}C$ was almost as good as fresh oil, and the storage group at 30$^{\circ}C$ showed certain degree of rancidity. Change of physico-chemical characteristic during storage were so small so that storage stability of red pepper seed oil was found to be good. AOM stability of red pepper seed oil was 7 hours which is lower than other vegetable oil, but the degree of stability grew greatly after adding phenolic antioxidants, such on TBHQ or PG. As for the chemical change after heating continuously for 40 hours, acid value, peroxide value and refractive index increased, but iodine value decreased as the heating processed. The fatty acid composition also showed the remarkable reduction of linoleic acid. The addition of antioxidants resulted in the delay of oxidation, the degree of which was greater in TBHQ than in PG.

Distribution of Chitinases in Rice (Oryza sativa L)Seed and Characterization of a Hull-Specific Chitinase

  • The uneven distribution of acidic and basic chitinases in different parts of rice seed, and also the characterization of hull-specific chitinases, are reported here. After extraction of chitinases from polished rice, bran, and rice hulls, the chitinases were separated into acidic and basic fractions, according to their behavior on an anion exchanger column. Both fractions from different parts of rice seed showed characteristic activity bands on SDS-PAGE that contained 0.01% glycol chitin. The basic chitinases from rice hulls were further purified using chitin affinity chromatography. The chitinase, specific to rice hulls (RHBC), was 88-fold purified with a 1.3% yield. RHBC has an apparent molecular weight of 22.2 kDa on SDS-PAGE. The optimal pH and temperature were 4.0 and $35^{\circ}C$, respectively. With [$^3H$]chitin as a substrate, RHBC has $V_{max}$ of 13.51 mg/mg protein/hr and $K_m$ of 1.36 mg/ml. This enzyme was an endochitinase devoid of ${\beta}$-1,3-glucanase, lysozyme, and chitosanase activities.

Morphological Characteristics of Endocarp in Relation to Seed Dormancy of 18 Rubus Species in Korea

  • The microstructure observation of seed surface structure is needed for protocols of breaking dormancy of seeds with physical dormancy. The seeds of Rubus species are surrounded by a thick, hard endocarp; together, the seed and endocarp make up the stone. We evaluate stone characteristics of 18 species of Rubus through optical microscopic observation, and correlate different stone characteristics with endocarp thickness. As a result of stone size comparison, Rubus species were classified as big stones group including R. parvifolius and R. idaeus, small stones group including R. longisepalus var. longisepalus, R. corchorifolius and R. hirsutus, and middle stones group including rest of the species. The result of this study revealed that stone size and the endocarp thickness in Rubus species was various characteristics in each species. Furthermore stone size and stone weight were also well correlated endocarp thickness and result indicated that heavy stones had harder endocarp than lighter one. Thus from the result of this study it can be presumed that only one stone characteristic approach may be sufficient to estimate other characteristics in Rubus.

A Study on the Resistve Switching Characteristic of Parallel Memristive Circuit of Lithium Ion Based Memristor and Capacitor (리튬 이온 기반 멤리스터 커패시터 병렬 구조의 저항변화 특성 연구)

  • In this study, in order to secure the high reliability of the memristor, we adopted a patterned lithium filament seed layer as the main agent for resistive switching (RS) characteristic on the 30 nm thick ZrO2 thin film at the device manufacturing stage. Lithium filament seed layer with a thickness of 5 nm and an area of 5 ㎛ × 5 ㎛ were formed on the ZrO2 thin film, and various electrode areas were applied to investigate the effect of capacitance on filament type memristive behavior in the parallel memristive circuit of memristor and capacitor. The RS characteristics were measured in the samples before and after 250℃ post-annealing for lithium metal diffusion. In the case of conductive filaments formed by thermal diffusion (post-annealed sample), it was not available to control the filament by applying voltage, and the other hand, the as-deposited sample showed the reversible RS characteristics by the formation and rupture of filaments. Finally, via the comparison of the RS characteristics according to the electrode area, it was confirmed that capacitance is an important factor for the formation and rupture of filaments.

  • Fast and dynamic biochemical, enzymatic and morphological changes occur during the so-called generative development and during the vegetative processes in seeds. The most characteristic biochemical and compositional changes of this period are the formation and decline of storage components or their precursors, the change of their degree in polymerization and an extensive change in water content. The aim of the present study was to detect the maturation processes in seed nondestructively and to verify the applicability of near infrared spectroscopic methods in the measurement of physiological, chemical and biochemical changes in wheat seed. The amount and variation of different water “species” has been changed intensively during maturation. Characteristic changes of three water absorption bands (1920, 1420 and 1150 nm) during maturation were analysed. It was concluded that the free/bound transition of water molecules could be followed sensitively in different region of NIR spectra. Kinetic changes of carbohydrate reserves were characteristic during maturation. An intensive formation and decline of carbohydrate reserves were observed during early stage of maturation (0 -13 days, high energy demand). An accelerated formation of storage carbohydrates (starch) was detected in the second phase of maturation. Five characteristic absorption bands were analysed which were sensitive indicators the changes of carbohydrates occurred during maturation. Precursors of protein synthesis and the synthesis of reserve proteins and their kinetic changes during maturation were followed from NIR spectra qualitative and qualitatively. Dynamic formation of amino acids and the changes of N forms were detected by spectroscopic, chromatographic and by capillary electrophoresis methods. Calibration equations were developed and validated in order to measure the optimal maturation time protein and moisture content of developing wheat seeds. The spectroscopic methods are offering chance and measurement potential in order to detect fine details of physiological processes. The spectra have many hidden details, which can help to understand the biochemical background of processes.

A Study on the Standardization and Cooking Properties of Imjasootang (임자수탕(荏子水湯) 조리법의 표준화와 품질특성에 관한 연구)

  • The objective of this study was to standardized the cooking method of Imjasootang, a traditional health food in summer. By mixing chicken soup with 20 g (I$_1$), 30 g (I$_2$), 40 g (I$_3$), 50 g (I$_4$) of seasam seeds, the distinctive characteristic and level of preference of Imjasootang were examined through the analyses of the proximate composition, fatty acid contents, color index, turbidity, brix$^{\circ}$, and sensory evaluation. 1. The proximate composition of Imjasootang was 90.2∼95.1% of total moisture, 0.985∼1.271% of crude protein, 0.90∼0.98% of crude fat, 1.592∼2.456% of nonfiber, 0.003∼0.004% of fiber, and 0.77∼0.97% of crude ash. Fatty acid were composed of 15.15∼17.36% of saturated fatty acid and 79.73∼82.54% of unsaturated fatty acid. Imjasootang contains high level of oleic acid and linoleic acid which were dependant on the content of seasame seeds. 2. For color index of samples, the lightness ranged 47.2∼56.9, “a”values 1.82∼2.15%, and “b”values 15.5∼16.5%. The turbidity of samples ranged 39∼61%, in which I$_4$ (chicken stock 100 g+sesame seed 50 g+water 25 g) was the most turbid. Solid content was also the highest in I$_4$, ranging 1.4∼5.1%. 3. In sensory evaluation, all sesame seed Imjasootangs had no significance differences in appearance. The higher the level of sesame seed in Imjasootangs, the more Imjasootang was preferred in terms of nutty aroma, color nutty taste. and viscosity. I$_4$ had the strongest nutty aroma. I$_2$ (chicken stock 100 g+sesame seed 30 g+water 15 g) and I$_3$ (chicken stock 100 g+sesame seed 40 g+water 20 g) were the best in the overall preference.

In Vitro Antioxidant Activities and Antimicrobial Activity of Lotus (Leaf, Stem, and Seed Pod) Extracts (연잎, 연 줄기 및 연자방 추출물의 In Vitro 항산화 활성과 항균 활성에 관한 연구)

  • The purpose of this study was to investigate and analyze the total phenolic content (TPC), antioxidant activities (FRAP, ABTS, and DPPH), and antibacterial properties of lotus (Nelumbo nucifera) extracts. Lotus leaves, stems, and seed pods were extracted with deionized water at $95^{\circ}C$, and with 70.5% ethanol at $85^{\circ}C$. The TPC ranged from 8.12 to 215.12 GAE mg/g. The ethanol extract of the seed pod had the highest TPC, and the TPC of the corresponding deionized water extract was 161.45 mg/g. FRAP values ranged from 104.03 to $3,546.39TEAC\;{\mu}mol/g$, ABTS radical cation scavenging activities ranged from 105.11 to $3,956.94TEAC\;{\mu}mol/g$, and DPPH radical scavenging activities ranged from 37.29 to $2,549.46TEAC\;{\mu}mol/g$. $EC_{50}$ values ranged from 0.26 to 9.63 mg/mL, and 0.31 to 21.21 mg/mL for ABTS and DPPH, respectively. The ethanol and deionized water extracts of the seed pod showed higher TPC and stronger antioxidant properties (FRAP, ABTS, and DPPH) than those of characteristic of the leaf extracts. The ethanol and deionized water extracts of the seed pod showed antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, and Pseudomonas aeruginosa with inhibition zones of 9.0 to 14.0 mm, and the ethanol extract of the leaf showed antimicrobial activity against B. subtilis and S. aureus with inhibition zones of 9.0 and 10.0 mm, respectively. Thus, the lotus seed pod could be used to produce novel teas, and could be a potential source of therapeutic ingredients for food and medicine.
