Growth responses of New Zealand Spinach [Tetragonia tetragonoides (Pall.) Kuntze] to different soil texture and salinity (신규 채소작물용 번행초의 토성 및 염도에 대한 생육 반응)

  • This research was conducted to investigate potential use of New Zealand spinach (Tetragonia tetragonoides) as a new vegetable crop which will be cultivating in salt-affected soils including reclaimed land. Traditionally New Zealand spinach has been studied to explore functional compound or salt removing potential. To cultivate the crop species in the salt-affected soil widely, it is essential to obtain salt and soil texture responses under the controlled environment. Fifty nine New Zealand spinach ecotypes native to Korean peninsula first collected over seashore areas, and primitive habitat soil environment was evaluated by analyzing soil chemical properties from 32 locations. Different textures of sandy, silt loam, and sandy loam soils were prepared from nearby sources of sea shore, upland and paddy soils, respectively. Target salinity levels of 16.0 dS/m, 27.5 dS/m, 39.9 dS/m, and 52.4 dS/m in electrical conductivity (ECw) were achieved by diluting of 25, 50, 75, 100% (v/v) sea water to tap water (control, 0.6 dS/m), respectively. Various measurements responding to soil texture and irrigation salinity included plant height, root length, fresh weight (FW), dry weight (DW), leaf parameters (leaf number, leaf length, leaf width), lateral branching, and inorganic ion content. was found to adapt to diverse habitats ranging various soil chemical properties including soil pH, organic matter, exchangeable bases, EC, and cation exchange capacity (CEC) in Korea. Responding to soil texture, New Zealand spinach grew better in silt loam and sandy loam soil than in sandy soil. Higher yield (FW and DW) seemed to be associated with branch number (r=0.99 and 0.99, respectively), followed by plant height (r=0.94 and 0.97, respectively) and leaf number (r=0.89 and 0.84, respectively). Plant height, FW, and DW of the New Zealand spinach accessions were decreased with increasing irrigation salinity, while root length was not significantly different compared to control. Based on previous report, more narrow spectrum of salinity range (up to 16 dS/m) needs to be further studied in order to obtain more accurate salinity responses of the plant. As expected, leaf Na content was increased significantly with increasing salinity, while K and Ca contents decreased. Growth responses to soil texture and irrigation salinity implied the potential use of New Zealand spinach as a leafy vegetable in salt-affected soil constructed with silt loam or sandy loam soils.

The Effect of Passing Aged Years to the Polarization Characteristics of Embedded Steel Bar of Mortar Specimen(W/C:0.4) (몰타르 시험편(W/C:0.4) 내부철근의 분극특성에 미치는 재령년수의 영향)

  • The structures of reinforced concrete has been extensively increased with rapid development of industrial society. Futhermore, these reinforced concretes are easy to expose to severe corrosive environments such as sea water, contaminated water, acid rain and seashore etc.. Thus, corrosion problem of inner steel bar embedded in concrete is very important in terms of safety and economical point of view. In this study, multiple mortar test specimen(W/C:0.4) with six types having different cover thickness each other was prepared and was immerged in seawater solution for five years to evaluate the effect of cover thickness and immersion years to corrosion property of embedded steel bar. And the polarization characteristics of these embedded steel bars was investigated using electrochemical methods such as measuring corrosion potential, cathodic polarization curve, and cyclic voltammogram. At the beginning of immersion, the corrosion potentials exhibited increasingly nobler values with increasing cover thickness. However, after immersed for 5 years, the thicker cover of thickness, the corrosion potentials shifted in the negative direction, and the relationship between corrosion potential and cover thickness was not in good agreement with each other. Therefore, it is considered that the thinner cover of thickness, corrosive products deposited on the surface of the embedded steel bar plays the role as a resistance polarization which is resulted in decreasing the corrosion rate as well as shifting the corrosion potential in the positive direction. As a result, it seemed that the evaluation which corrosion possibility of the reinforced steel would be estimated by only measuring the corrosion potential may not be a completely desirable method. Therefore, it is suggested that we should take into account various parameters, including cover thickness, passed aged years as well as corrosion potential for more accurate assessment of corrosion possibility of reinforced steel which is exposed to partially or fully in marine environment for long years.

A Study on the Application of the Solar Energy Seasonal Storage System Using Sea water Heat Source in the Buildings (해수냉열원을 이용한 태양열계간축열시스템의 건물냉방 적용에 관한 연구)

  • Paradigm depending only on fossil fuel for building heat source is rapidly changing. Accelerating the change, as it has been known, is obligation for reducing green house gas coming from use of fossil fuel, i.e. reaction to United Nations Framework Convention on Climate Change. In addition, factors such as high oil price, unstable supply, weapon of petroleum and oil peak, by replacing fossil fuel, contributes to advance of environmental friendly renewable energy which can be continuously reusable. Therefore, current new energy policies, beyond enhancing effectiveness of heat using equipments, are to make best efforts for national competitiveness. Our country supports 11 areas for new renewable energy including sun light, solar heat and wind power. Among those areas, ocean thermal energy specifies tidal power generation using tide of sea, wave and temperature differences, wave power generation and thermal power generation. But heat use of heat source from sea water itself has been excluded as non-utilized energy. In the future, sea water heat source which has not been used so far will be required to be specified as new renewable energy. This research is to survey local heating system in Europe using sea water, central solar heating plants, seasonal thermal energy store and to analyze large scale central solar heating plants in German. Seasonal thermal energy store necessarily need to be equipped with large scale thermal energy store. Currently operating central solar heating system is a effective method which significantly enhances sharing rate of solar heat in a way that stores excessive heat generating in summer and then replenish insufficient heat for winter. Construction cost for this system is primarily dependent on large scale seasonal heat store and this high priced heat store merely plays its role once per year. Since our country is faced with 3 directional sea, active research and development for using sea water heat as cooling and heating heat source is required for seashore villages and building units. This research suggests how to utilize new energy in a way that stores cooling heat of sea water into seasonal thermal energy store when temperature of sea water is its lowest temperature in February based on West Sea and then uses it as cooling heat source when cooling is necessary. Since this method utilizes seasonal thermal energy store from existing central solar heating plant for heating and cooling purpose respectively twice per year maximizing energy efficiency by achieving 2 seasonal thermal energy store, active research and development is necessarily required for the future.

The activity of antioxidants and suppression of cancer cell proliferation in extracts of Orostachys japonicus A. Berger (바위솔 추출물의 항산화활성 및 암세포 증식억제)

  • The antioxidative activities and biological properties in the EtOAc extracts and purified extracts of Orostachys japonicus were measured by assay of DPPH, xanthine/xanthine oxidase and mammalian cells(2-12). Scavenging of DPPH radical and inhibition of xanthine/xanthine oxidase of EtOAc extracts were showed the highest activity in the arable land and in September. The S-4 fraction showed the highest activity among the silica-gel column chromatography fractions. LH-4 fraction showed higher activity than the other fractionsins in assay of DPPH and xanthine/xanthine oxidase. Fatty acids and phenolic compounds were identified by GC/MS and main compounds were 1,2,3-benzenetriol, alpha-androsta-7,14-diene in LH-4 fraction. The activities of POD and SOD in samples havested on different habitats were high such as arable land> intermountain> seashore. That of POD and SOD in crude extracts of late stage were higher than early stage. Isozyme bands of crude extracts samplinged in all habitats and all growing stages showed two bands and the signal intensity showed strongly according to passage of growing stage. The purified extracts of LH-4 fraction showed excellent inhibition effect in proliferation of HL-60 cells and markedly suppressed colony formation in mouse fibroblast cells. Dose response between partially purified extracts(400ppm) and negative control did not produced statistically significant reduction in colony formation.

Clean-up of the Crude Oil Contaminated Marine Sediments Through Biocarrier-Mediated Bioaugmentation (생물담체 활용 생물접종에 의한 원유로 오염된 해양토양의 정화)

  • This study was carried out to develop an effective biocarrier-mediated bioaugmentation technology which will be useful for remediation of the crude oil-contaminated marine sediments. Enrichment of several microbial communities was made from several oil-polluted seashore sites and the two distinctively functional consortia have been successfully selected. These two consortia were grown together and used to manufacture the microbial agents for bioaugmentation of marine sediments polluted with crude oil. The most dominant species in the mixed culture was identified as Alcanivorax borkumensis based on pure culture and DGGE analysis. Bioaugmentation of oil-polluted marine sediments with the microbial agent MA-2 formulated using the mixed culture and biocarriers (activated carbon and minerals) was more effective, especially in combination with an oxygen producing (releasing) compound (ORC). Ninty percent of TPH was removed in the presence of ORC in 35 days while 74% in the absence of ORC. This indicated that the indigenous consortial degraders could be immobilized on the active carbon as a biocarrier to manufacture microbial agents and then effectively bioaugmented for remediation of the oil-polluted sediments.

Phylogenetic and Morphological Comparison between Thamnaconus septentrionalis and T. modestus Collected in Southwest Seashore (서남해에서 채집된 말쥐치 (Thamnaconus modestus)와 유사종 (T. septentrionalis)의 형태 및 계통유전학적 비교)

  • Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.

The Mask-Dance Performances in the Shaman Rituals: and (굿 속의 탈놀이:<영산 할아?.할?굿>과 <탈굿>)

  • The Korean Mask-Dance Theatre has been developed closely related to Korean Shaman rituals. As many scholars noticed, the performers of the Mask-Dance Theatre were closely related to the Shaman family. In addition, there are mask-dance performances in actual shaman rituals. and are the representative performances among them. This essay intends to compare these two mask-dance performances in the shaman rituals to the similar performance of Old Grandpa and Grandma episode in the regional Mask-Dance Theatre. This study would bring us further in proving the close relation between the shaman ritual and the Mask-Dance Theatre. is one episode, Keori, in the shaman ritual of 'Baeyeonsin-kut' and 'Taedong-kut' in the mid-west seashore area. 'Taedong-kut' is the village shaman ritual for fertility and prosperity, while 'Baeyeonsin-kut' is a private shaman ritual for a large catch of the ship. is held in the later part of the whole shaman ritual since the later part tend to be more for entertainment than actual ritual. The story of is very similar that of in Pongsan Mask-Dance Theatre of the mid-west region. In addition, some of their dialogues are very similar. Only the later part is different. These similarities indicate that the Mask-Dance Theatre, which came into being in later period than the shaman ritual, has likely taken the story motif of the shaman ritual. is also a performance in the shaman ritual of east coasts. is more elaborate and recreational than of the west coasts. is also performed near the end of the ritual, and sometimes it is not performed at all. This indicates that has little ritual meaning left. When we compare it with the regional Mask-Dance Theatres such as Keosung Okwangdae, Tongyong Okwangdae, and Suyong Yaryu, the structure and the story lines are also very similar. It is a question why only the motif of the Grandpa and Grandma isfound both in the shaman ritual and the Mask-Dance Theatre. Many other motifs of other episodes in the Mask-Dance Theatre are not found in the shaman rituals. It seems that the Grandpa and Grandma motif is related to the ur-belief in fertility. In other words, this motif seems to be originated from the old belief in the fertility couple of Chonha Taechanggun and Jiha Yeochanggun. The shaman ritual for fertility first picked up this motif, and then the mask-dance theatre also adapted this motif for its recreational purpose. When we compare with , still has more aspects of fertility ritual, while lost its ritualistic meaning and its main purpose is to develop dramatic needs. and are invaluable existent performances to prove theatre's origin in ritual. The existence of mask-dance performances in the shaman rituals shows us the transit performance between theatre and ritual.

Reconsideration and Conservational Scientific Diagnosis of Silla Stone Monument in Bongpyeong-ri, Uljin (울진 봉평리 신라비의 재판독과 보존과학적 진단)

  • This study was focused on the recognition of historical values and the establishment of conservation schemes for a Silla stone monument in Bongpyeong-ri, Uljin by combining the humanistic investigation of inscription reinterpretation and the scientific conservational diagnosis of deterioration. According to the investigation of inscription, a total of 13 letters were reconsidered compared to the preceding researches. Thus, the meaning and interpretation of previous inscription was partially changed. This monument is composed of gneissose leucogranite and the most suitable site as provenance of the stone would be the Jukbyeon seashore (2.1km). The site shows similar color, size and composition of minerals, gneissose structure and magnetic susceptibility as the Bongpyeong stone monument. This monument developed a structural crack (crack index 0.4) and a microcrack (crack index 2.0) along the gneissose structure. The horizontal strength is weaker than the vertical strength. Therefore, the cracks should be reinforced and treated. However, consolidating is not urgently needed because the total weathering grade by ultrasonic velocity shows the stage of moderate weathering(3,403m/s, 0.32). Also, the major problems of chemical deterioration are blackening (85.2%) with soil, iron oxide,rubbing mark, and salt crystals (17.3%) from the sea. Therefore, the contaminants and the salt crystals should be removed using pressure spray and pulp paper, while the application of poultice should be examined through clinical tests.

Isolation of Simiduia sp. SH-2 and Characterization of Its β-Agarase (한천분해세균 Simiduia sp. SH-2 균주의 분리 및 β-agarase의 특성조사)

  • This study isolated a new agarase-producing bacterium and characterized its agarase. A new agar-degrading strain was isolated from the seashore of Namhae in Gyeongnam province, Korea, and was purely cultured using the Marine Agar 2216 media. The isolated bacterium was identified as Simiduia sp. SH-2 after 16S rRNA gene sequencing. The crude agarase was obtained from the culture medium of the Simiduia sp. SH-2 strain, and the agar-degrading activity was measured. The highest level of activity of the Simiduia sp. SH-2-derived agar-degrading enzyme was 625 U/l. Agar degradation activity was most significant at 40℃ and pH 7.0. Compared to the activity at 40℃, the relative activity was 31% at 20℃ and 71% at 30℃. Compared to the activity at pH 7.0, the relative activity was 94% and 89% at pH 6.0 and pH 8.0, respectively. Residual activity was greater than 96% after exposure to 20℃ and 30℃ for 2 hr and more than 49% after exposure to 40℃ for 2 hr. Simiduia sp. SH-2 was identified as a strain producing β-agarase that creates neoagarooligosaccharides, such as neoagarotetraose and neoagarohexaose. Therefore, the Simiduia sp. SH-2 strain and its β-agarase are expected to be useful functional material producers in the food, cosmetic, and pharmaceutical industries.

Regeneration of adventitious root from Calystegia soldanella L. in Jeju island and mass proliferation method using bioreactor system (제주지역 갯메꽃(Calystegia soldanella L.) 유래 부정근 재분화 및 생물반응기 시스템 이용 대량증식법)

  • Calystegia soldanella L. is a perennial herbaceous halophyte belonging to the convolvulaceae family, which mainly grows in coastal sand dunes in Korea. Shoots and rhizomes are edible, and roots called 'Hyoseon Chogeun' are known to have medicinal effects such as antipyretic, sterilization, and diuretic. In addition, physiological activities of antioxidant, anti-inflammatory, antiviral, antifungal and PTP-1B (protein tyrosine phosphate-1B) inhibition have been reported. In this study, in vitro induction cell lines of C. soldanella L. collected from the coastal sand dunes in Jeju island was redifferentiated into adventitious roots that can be used as medicinal resources. Also the biomass of mass-proliferated adventitious roots using a bioreactor were evaluated. Plants of C. soldanella L. were collected from the crevice of the seashore in the coastal area of Taeheung 2-ri, Namwon-eup, Seogwipo-si. Then, it was separated into leaves, stems, rhizomes, and roots, and surface sterilized with 70% ethyl alcohol and 2% NaOCl (sodium hypochlorite). After washing with sterilized water, each organ section was cultured in Hormone-free MS medium (Murashige & Skoog Medium). As a result, the induction response rates were evaluated at 85% and 55%, respectively, in terms of callus formation and shoot generation in the rhizome segment. In the case of the adventitious roots morphological characteristics induced by single-use treatment of auxin-based plant growth regulators IBA and NAA from redifferentiated shoots were compared. Most efficient adventitious root culture method as a rooting rate, number, length, and biomass proliferation in the bioreactor system was confirmed when treated by culturing in MS salts, Sucrose 30 g·L-1, and IBA 1mg·L-1 for 4 weeks. In this study, the medium composition and culture period were confirmed using a bioreactor system to mass-proliferate adventitious roots derived from C. soldanella L. in Jeju island. Also this adventitious root line developed a new medicinal material could increase value of the bio-industry ingredient through quantitative and qualitative screening of phyto-bioactive compounds.
