비타민나무의 약리 효과 및 구성 성분 (Phamalogical effect and component of sea buckthorn(Hippophae rhamnoides L.))

  • Sea buckthorn (Hippophae rhamnoides L.) is deciduous shrubs in the genus Hippophae, mainly cultivated in Europe and Asia. Sea buckthorn berries have a high vitamin C, vitamin E, carotenoids, carbohydrates, protein, organic acids, dietary minerals, triterpenoids, polyphenolic acids and amino acids. Extracts of sea buckthorn berries have anti- obesity, anti-oxidantive, anti-microbial, anti-ulcerogenic, anti-diabetic and nutritional effects. Sea buckthorn used as a traditional medicine for the treatment of cough, aid digestion, invigorate blood circulation and alleviate pain. Extracts of sea buckthorn branches and leaves was administered to humans and animals to treat gastrointestinal distress in Mongolia. This paper briefly reviews the most relevant experimental data on the pharmacological effects and isolated component of sea buckthorn. And, we also describe the importance of sea buckthorn as the environmental-friendly crops.

  • Sea-buckthorn fruits (Hippophae rhamnoides L.) are used in Mongolia as traditional medicine due to the health-benefits associated with its bioactive components. The purpose of this study was to investigate the chemical composition of Mongolian sea-buckthorn fruits. In terms of proximate composition, crude fat content was the highest, whereas its crude ash content was the lowest. In organic acid contents, malic acid content ($6.30{\pm}0.005$ mg/g) was the highest. Free sugars were composed of sucrose, xylose, glucose and mannitol. Vitamin C and carotenoid content were 320 mg/100 g and 715.25 mg/100 g, respectively. The major fatty acids were palmitic (C16:0), palmitoleic (C16:1), caprylic (C8:0) and linoleic (C18:2) acid.

비타민나무열매가루 첨가 식빵의 품질 특성 및 항산화 활성 (Quality Characteristics and Antioxidant Properties of White Pan Bread Added with Sea Buckthorn (Hippophae rhamnoids L.) Berry Powder)

  • The purpose of this study was to prepare bread added with sea buckthorn (Hippophae rhamnoides L.) berry powder (SBBP) (1, 3, and 5%) using a straight dough method. The quality and antioxidant characteristics of the bread were analyzed. The results indicated that the pH value and dough raising power of the dough decreased and the moisture content of the bread added with SBBP increased compared to that without the SBBP. The crust color of the bread did not change significantly. However, the crumb lightness value decreased and the redness and yellowness value increased respectively to the concentration of the SBBP. The texture measurement indicated that the hardness, gumminess, and chewiness of the bread increased with the addition of the SBBP. The total polyphenol content of the bread supplemented with the SBBP was increased (4.60~16.14 mg GAE/g) compare to the control (1.67 mg GAE/g). Dose-dependent, significant-high DPPH (26.86%) and ABTS (42.52%) radical scavenging activities were observed in the bread with the addition of the SBBP up to 5%. Based on these results, the optimum amount of the SBBP to add for baking bread would be 3% and the SBBP could be considered a functional agent.

Sea Buckthorn (Hippophae rhamnoides L.) Leaf Extracts Protect Neuronal PC-12 Cells from Oxidative Stress

  • The present study was carried out to investigate the antioxidative and neuroprotective effects of sea buckthorn (Hippophae rhamnoides L.) leaves (SBL) harvested at different times. Reversed-phase high-performance liquid chromatography analysis revealed five major phenolic compounds: ellagic acid, gallic acid, isorhamnetin, kaempferol, and quercetin. SBL harvested in August had the highest total phenolic and flavonoid contents and antioxidant capacity. Treatment of neuronal PC-12 cells with the ethyl acetate fraction of SBL harvested in August increased their viability and membrane integrity and reduced intracellular oxidative stress in a dose-dependent manner. The relative populations of both early and late apoptotic PC-12 cells were decreased by treatment with the SBL ethyl acetate fraction, based on flow cytometry analysis using annexin V-FITC/PI staining. These findings suggest that SBL can serve as a good source of antioxidants and medicinal agents that attenuate oxidative stress.

비타민나무(Sea Buckthorn, Hippophae rhamnoides) 추출물의 이화학적 성분 분석과 항산화 활성효과 (Analysis of Chemical Composition and in vitro Anti-oxidant Properties of Extracts from Sea Buckthorn (Hippophae rhamnoides))

  • 본 연구에서는 비타민나무의 기능성 소재 개발을 위한 기초 자료로 활용하기 위해 일반 성분, 이화학적특성 중 총 페놀함량과 DPPH free radical 소거활성, SOD 유사활성, Hydroxy radical scavenging activity를 이용한 항산화활성을 측정하였다. 비타민나무는 잎, 잔가지, 굵은 가지, 뿌리의 4가지 부위를 물과 에탄올을 용매로 추출하여 실험에 사용한 결과, 일반 성분의 경우 비타민 나무의 모든 부위에서 조회분과 조단백질의 함량이 에탄올 추출물보다 물 추출물에서 더 높은 것으로 확인되었다. 총 폴리페놀함량은 잎과 뿌리 부위해서 높은 결과를 보였으며, 물 추출물에 비해서 에탄올 추출물의 함량이 더 높은 것으로 확인되었다. DPPH radical 제거활성은 잔가지를 제외한 부위에서 에탄올 추출물보다 물 추출물이 높은 활성을 보였고, 특이적으로 에탄올을 용매로 한 잔가지 추출물의 $IC_{50}$이 23.58${\pm}$0.84%로 가장 높은 활성을 나타내었다. SOD 유사활성능은 물 추출물보다 에탄올 추출물이 높았으며, 그 중 잔가지의 에탄올 추출물이 35.03${\pm}$2.33%의 가장 높은 활성을 나타내었다. Hydroxy radical scavenging 활성은 에탄올 추출물이 물 추출물에 비해 2배 이상의 높은 활성을 보였고, 그 중 뿌리 부위의 에탄올 추출물이 66.12${\pm}$8.73%의 높은 활성을 보였다. 결과적으로 본 연구에서는 비타민나무 추출물의 영양학적 가치와 항산화활성을 확인하였고, 그로 인한 기능성소재로써의 개발이 가능할 수 있음을 확인하였다.

비타민나무 잎 첨가 현미설기떡 제조 및 항산화 활성 (Antioxidative Activity of Sea Buckthorn and Quality Characteristics of Brown Rice Sulgidduk)

  • This study was performed to investigate the antioxidative properties of sea buckthorn (Hippophae rhamnoides L.; leaf, fruit and stem) and the quality characteristics of brown rice sulgidduk prepared using a powder of sea buckthorn leaves. First, the antioxidative activities of sea buckthorn were measured to choose the most effective part of this plant. By analyzing the measured values, we concluded that the effective part of sea buckthorn was its leaves. The $IC_{50}$ value of the DPPH radical scavenging activity and the ABTS radical scavenging activity in sea buckthorn leaves were $7.78{\mu}g/mL$ and $264.04{\mu}g/mL$, respectively. The total polyphenol and total flavonoid contents of sea buckthorn leaves were 3.80 mg/mL and .19 mg/mL, respectively. Therefore, the brown rice sulgidduk was prepared using a powder of sea buckthorn leaves in the weight ratio of 0%, .5%, 1.0%, 1.5%, 2.0% and 2.5%. For analyzing the quality characteristics of the prepared sulgidduk, proximate compositions, color and texture profiles were measured and a sensory evaluation was conducted. With an increase in the added content of the sea buckthorn leaf powder (SBLP), the L-value significantly decreased while the a-value and the b-value increased. In the case of texture profiles, the control group (control A) had a higher score for hardness than the case groups in which the SBLP was added. However, springiness, chewiness and adhesiveness were not significantly different among the groups. In the sensory evaluation, the sample containing 1.5% SBLP yielded the best results. Therefore, we suggest that the powder of sea buckthorn leaves is a good ingredient for increasing the consumer acceptability and functionality of sulgidduk.

The Effect of Extract from Sea Buckthorn on DNCB-induced Atopic Dermatitis in NC/Nga Mice

  • Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-${\kappa}B$ signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-${\kappa}B$ pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p<0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-${\alpha}$, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

UV조사에 의해 유도된 피부섬유아세포의 세포사에 미치는 Sea Buckthorn (Hippophae rhamnoides L.) 열매추출물의 영향 (Effects of Sea Buckthorn (Hippophae rhamnoides L.) Fruit Extract on Ultraviolet-induced Apoptosis of Skin Fibroblasts)

  • Sea buckthorn (Hippophae rhamnoides L.)은 flavonoid, carotenoids, steroids, vitamin, tannins, oleic acid 등의 생물학적으로 활성이 높은 성분을 풍부하게 함유하고 있어 오랫동안 피부질환(skin diseases), 위궤양(gastric ulcers), 천식(asthma), 폐질환 등에 사용되어 왔다. 본 연구에서는 아시아와 유럽의 고지대에서 야생으로 자라는 Sea buckthorn의 열매추출물(SBFE)이 UV조사 후 유도되는 세포사와 연관성을 규명하기 위하여, 세포사와 관련된 다양한 인자의 변화를 관찰하였다. 그 결과, 세포사를 유발할 수 있는 UV조사농도는 400 mJ로 설정하였고, 세포생존율은 UV + LoC, UV + MeC, UV + HiC 처리군에서는 유의적으로 증가하였다. 또한 FACS와 DAPI염색의 결과에서 PI로 염색된 세포수는 UV + vehicle 처리군에서 급격히 증가한 이후 UV + SBFE 처리군에서 유의적으로 감소하였고, Annexin V가 염색된 세포 수는 고농도의 SFBE을 처리한 UV + HiC 처리군에서 유의적으로 감소하였다. 그러나 PI와 Annexin V가 동시에 염색된 세포의 수는 UV + LoC 처리군에서 오히려 증가하였고 나머지 그룹에서는 유의적인 변화가 관찰되지 않았다. 한편, 세포사와 관련된 단백질의 변화에 대한 연구에서 caspase-3는 pro 형태와 active 형태 모두에서 유의적인 차이를 나타내지 않았으며, Bax와 Bcl-2 모두 UV 무처리군에 비하여 UV + Vehicle 처리군에서 감소하였으나 Bax의 발현양은 UV + SBFE 처리군에서 농도의존적으로 증가한 반면, Bcl-2의 발현양은 UV + SBFE 처리군에서 추가적으로 더욱 감소하였다. 따라서 이상의 결과들은 SBFE가 UV조사에 의해 유도된 세포사를 억제하거나 회복시키는 기능을 가지고 있음을 제시하고 있어 향후 UV조사에 의해 유발되는 다양한 상해에 대한 치료제로서의 가능성을 제시하고 있다.
