Development of DNA Probe Assay System for Salmonella Species using Glass as substrate

  • We developed a DNA probe analytical system with a patterned array of oligonucleotide molecules immobilized on glass surfaces. The detection capability of the system depended mainly on the way the capture probes were attached to the support as wen as the sequence. We optimized major variables to graft DNA molecules onto a glass support and the DNA probe assay was eventually accomplished without purification of the PCR product.

Sex Reversal Pattern of Wrasse in Jeju Coastal Waters

  • Eleven wrasse species inhabit the coastal waters of Jeju Island, Korea. They are the target of leisure fishing and are considered good eating. We investigated the distribution of standard length (SL) by sex of wrasse in Jeju coastal waters for Halichores poecilopterus, H. tenuispinis, Pseudolabrus japonicus, and Pteragogus flagellifera. A cross-section of the ovary showed the ovarian cavity and ovarian lamellae containing oocytes. A cross-section of the testis showed many lobules containing spermatogonia and spermatocytes. A cross-section of a gonad undergoing sex reversal showed the regression or reduction of oocytes and some spermatocytes located in the ovarian lamellae. A cross-section of a sex-reversed testis showed the primary structure of the ovary, with spermatocytes distributed in the epithelium of the lamellae, and reformed seminiferous ducts in the basement lamellae. CONVIRT: A web-based tool for transcriptional regulatory site identification using a conserved virtual chromosome

  • Techniques for analyzing protein-DNA interactions on a genome-wide scale have recently established regulatory roles for distal enhancers. However, the large sizes of higher eukaryotic genomes have made identification of these elements difficult. Information regarding sequence conservation, exon annotation and repetitive regions can be used to reduce the size of the search region. However, previously developed resources are inadequate for consolidating such information. CONVIRT is a web resource for the identification of transcription factor binding sites and also features comparative genomics. Genomic information on ortholog-independent conserved regions, exons, repeats and sequences is integrated into the virtual chromosome, and statistically over-represented single or combinations of transcription factor binding sites are sought. CONVIRT provides regulatory network analysis for several organisms with long promoter regions and permits inter-species genome alignments.   • Two dimensional aerosol dynamics considering the effects of particle generation, coagulation, thermophoresis, sintering and convection has been studied to obtain the growth of non-spherical silica particles in conjunction with determining flame temperature by performing combustion analysis of premixed flat flame. Heat and mass transfer analysis includes 16 species, 29 chemical reaction steps together with oxidation and hydrolysis of SiCl4. The effect of radiation heat loss has also been included. The predictions of flame temperatures and the evolution of particle size distributions were in a reasonable agreement with the existing experimental data.

  • DAFC(direct alcohol fuel cell) takes the same structure and operational principle with PEMFC(Proton exchange membrane fuel cell). However, DAFC, which uses liquid alcohol instead of hydrogen as fuel, is able to be used as a portable power for small-scaled electronic devices such as MP3, PMP, and mobile phone because alcohol is quite convenient steady-state compound to carry and store it. This paper presents the OCV(open circuit voltage) characteristics of the cases which are alcohol species and different weight rate of ethanol, respectively. The OCV of methanol fuel cell is slightly higher 0.2V than ethanol one, and 8% wt. rate ethanol is rated as the most appropriate fuel for DAFC.

  • A numerical investigation on the flame propagation and extinction in a micro combustor is described. Previous measurements of $H_2-air$ flame propagation in a submilimeter scale combustor exhibited significance of wall effects on burning velocity and extinction. The heat transfer to wall becomes important not only in the cooling of burnt gases but also during the flame ropagation, which has be by and large ignored in macro scale combustor calculations. In order to take the heat loss into account the combustion calculation, we developed a numerical code with a heat transfer model that was determined empirically from measured data. PISO algorithm was used for differencing of conservation equations. $H_2-air$ reaction was modeled with 10 species - 16 steps. Comparison with measured data showed good agreement in flame propagation speed. Also the pressure decrease after flame extinction was accurately predicted by the model. A further study is desirable for a better quenching model that can predict the quenching location.

Functional Implications of the Conserved Action of Regulators of Ribonuclease Activity

  • RNase E (Rne) plays a major role in the decay and processing of numerous RNAs in E. coli, and protein inhibitors of RNase E, RraA and RraB, have recently been discovered. Here, we report that coexpression of RraA or RraB reduces the ribonucleolytic activity in rne-deleted E. coli cells overproducing RNase ES, a Streptomyces coelicolor functional ortholog of RNase E, and consequently rescues these cells from growth arrest. These findings suggest that the regulators of ribonuclease activity have a conserved intrinsic property that effectively acts on an RNase E-like enzyme found in a distantly related bacterial species.

Growth Enhancement of the Microalga Tetraselmis suecica by an Extract of the Green Alga Monostroma nitidum

  • Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.

Comparison of Trace Element, Metal, and Metalloid Contents in North and South Korean Plants

  • When relations improve between North and South Korea, there will be demand for North Korean edible plants because of the low labor cost and similar environmental conditions. However, there is no reliable information about trace elements, metals, and metalloids in edible plants from North Korea. Selenium (Se) and germanium (Ge) have positive effects on basic human health and are therapeutical in diverse illnesses. Metal and metalloid (Cd, Pb) poisoning, on the other hand, can cause many health problems. Plants collected from North Korea had higher selenium content than those from South Korea. Although none of the collected species exceeded the permissible levels of cadmium and lead, their content in plants was significantly higher in North Korea than in South Korea. The high metal contents in plants collected from North Korea may be associated with the soil physicochemical properties as well as the accumulated amounts of elements in the soil.

  • A disposable, electro-chemiluminescent immunosensor utilizing a screen-printed carbon electrode and liposome coupled to antibody as tracer has been constructed. In proportion to the analyte (Legionella species as a model) concentration, the analyte-immunoliposome complexes were transferred by the capillary action through a membrane strip to the electrode, the liposomes were lysed in the presence of detergent, and ruthenium was released for signal generation. Such performance of the immunosensor was appropriate for a point-of-care testing.
