만성 역류성 식도염에서 황련과 오수유 혼합물이 식도 점막에 미치는 영향 (Effect of Rhei Rhizoma and Scutellariae Radix Mixture in Chronic Acid Reflux Esophagitis Rats)

  • Gastroesophageal reflux disease (GERD) is a gastrointestinal disorder in which stomach contents reflux into the esophagus, causing complications such as mucosal damage. The purpose of this study was to determine the effect of Rhei Rhizoma and Scutellariae Radix mixture (RS) in chronic acid reflux esophagitis (CARE), one of the GERD. After inducing reflux esophagitis through surgery, the group was separated and the drug was administered for 2 weeks; Normal rats (Normal, n=8), chronic acid reflux esophagitis rats (Control, n=8), tocopherol 30 mg/kg-treated chronic acid reflux esophagitis rats (Toco, n=8), Rhei Rhizoma and Scutellariae Radix mixture 100 mg/kg-treated chronic acid reflux esophagitis rats (RSL, n=8), Rhei Rhizoma and Scutellariae Radix mixture 200 mg/kg-treated chronic acid reflux esophagitis rats (RSH, n=8). Gross lesion of esophageal mucosa after RS treatment showed a superior enhancement compared with that of Control group. Additionally, RS significantly decreased the levels of MPO and MDA, effectively inhibited NADPH oxidase, and regulated the expression of the AMPK/LKB1/NF-κB pathway. Moreover, it significantly increased the expression of tight junction proteins. Taken together, RS not only alleviates inflammation of the esophageal mucosa via the AMPK/LKB1/NF-κB pathway by reducing oxidative stress, but also improves esophageal function by modulating tight junction proteins.

황금(黃芩) 발효 추출물이 LPS로 유발된 마우스대식세포에서 염증인자증가에 미치는 영향 (Effect of Fermented Scutellariae Radix Extract on Production of Inflammatory Mediator in LPS-stimulated Mouse Macrophages)

  • Objectives : This study aims at examining the immuno-modulating activity in the fermentative extract of the root of Scutellaria baicalensis Georgi (Scutellariae Radix) on the production of inflammatory mediator in LPS-stimulated RAW264.7 mouse macrophages. Method : Measurements were done for the influences on the cell viability, generation of hydrogen peroxide in cells and nitric oxide (NO) generation using the macrophage of mouse with the specimen SBS as the fermentative extract of Scutellariae Radix (SBS) with Saccharomyces cerevisiae STV89. Result : As a result of carrying out MTT assay to check the cellular toxicity of the fermentative extract of Scutellariae Radix, any excessive toxicity to the macrophage did not occur from treatments by concentration for SBS. SBS increased the generation of hydrogen peroxide in the macrophage. SBS suppressed the NO generated in macrophages and SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the increased NO generated in LPS-stimulated macrophages. SBS concentration higher than $25{\mu}g/mL$ significantly suppressed the generation of IL-6, IL-10, IL-12p40 and MCP-1 in LPS-stimulated macrophages. Conclusion : Our findings indicate that SBS has an immuno-modulating activity in macrophage activation through suppressing the generation of inflammatory substances, NO, IL-6, IL-10, IL-12p40 and MCP-1.

청열약(淸熱藥)의 질내(膣內) 미생물(微生物)에 대(對) 항균효과(抗菌效果) (Antibiotic Effect of Heat-Clearing Medicinals on Vaginal Microorganisms)

  • Purpose : This study was conducted to investigate the in vitro inhibitory effects of heat-clearing medicinal on common bacterias in gynecology. Methods : The heat-clearing medicinals ( Trichosanthis Radix, Sophorae Fructus, Phragmitis Rhizoma, Buddleiae Flos, Bambusae Folium, Anemarrhenae Rhizoma, Celosiae Semen, Gardeniae Fructus, Prunellae Spica, Sophorae Radix, Dictamni Radicis Cortex, Coptidis Rhizoma, Gentianae Scabrae Radix, Scutellariae Radix, Phellodendri Cortex) were used in this study. Staphylococcus aureus, MRSA, Candida albicans and Gardnerella vaginalis were used for vaginal pathogenic microorganisms. Streptococcus spp., Escherichia coli HB101, Lactobacillus gasseri were used for normal vaginal florae. We evaluated antibiotic effect by the optical density and the colony test. Results : The optical density and colony test showed that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex of herbs among heat-clearing medicinal had antimircobial effect. Celosiae Semen and Prunellae Spica had antimicrobial susceptibility and selective toxicity in MRSA. Scutellariae Radix and Phellodendri Cortex had antimicrobial susceptibility and selective toxicity in Gardnerella vaginalis. Conclusion : According to the above results, we could suggest that Celosiae Semen, Prunellae Spica, Scutellariae Radix and Phellodendri Cortex among heat-clearing medicinal be available to antimicrobial agent of vaginal pathogenic microbial species in vitro.

  • Seven flavonoid components were isolated from fr. C of Scutellariae radix which showed antagonistic effects against bradykinin(BK). The results indicated that two oxygen functions (either -OH or $-OCH_3$) at 2'- and 6'-positions and/or an oxygen function at 6-position of flavone seemed to be favored for the BK inhibitory activities. Skullcapflavone-II(IV) which contains $6-OCH_3$, 2'-OH and $6-OCH_3$ in the structure was the most active among the flavones tested.

  • The O/W and W/O emulsions containing Scutellariae Radix extract(SRE) which is very slightly soluble in oil phases and sparingly soluble in water phases, were prepared by homogenizing water and oil phases with emulsifier. The diameters of emulsion were ranged from 100 to $300\;{\mu}m$. The viscosity of W/O emulsion was higher than that of O/W emulsion. W/O emulsion was more stable than O/W emulsion which was gradually degraded when tested by centrifuge method and temperature tolerance method at $50^{\circ}C$. The antibacterial activity of two emulsions was not significantly different from that of aqueous solution of SRE, and showed similar MIC and bacterial growth inhibition rate.

황금이 천식모릴 생쥐의 혈청 사이토카인에 미치는 영향 (Effect of Scutellariae Radix Extract on Serum Cytokines in OVA-induced Asthmatic Mice)

  • The purpose of this research is to examine the effects of Scutellariae Radix(SR) extract on cytokines in ovalbumin (OVA)-induced asthmatic mice. In vivo, C57BL/6 mice were sensitized and handicapped by OVA for 12 weeks. During this experiment, the one group was then treated with SR extract for the later 8 weeks (3 times per week) and analyzed by ELISA. There were significant decreases in IL-4(p<0.05), IL-5(p<0.05), IL-13(p<0.01), histamine(p<0.05) in serum of SR group. IgE also decreased, but was not significant compared with that of control group. The results of this study support a role for SR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions.

황금(黃芩)의 항산화 효과 (Anti-Oxidative Effects of Scutellariae Radix)

  • Objectives : This study was carried out to determine if Scutellariae Radix have protective effect against cell injury induced by various toxic agents in rat kidney slices. Methods : Water(SWe) and methanol(SMe) extracts were prepared for this experiment. Cell injury was estimated by measuring lactate dehydrogenase(LDH). Lipid peroxidation was examined by measuring malondialdehyde. Results : SMe prevented the LDH release by CCl4, menadione, tert-butyl hydroperoxide and mercury treatment in vitro in kidney slices, but SWe prevented the LDH release by CCl4 and mercury. SMe also prevented reduction in GSH by CCl4 and lipid peroxidation induced by mercury. Conclusions : Thus, SMe may have more powerful efficacy on anti-oxidative effects when compared with SWe. And further studies have to be followed concerned with procedure of extraction of SMe and its change of effects.

천연염재의 자외선 차단성능 연구 (A Study on Ulraviolet-cut Ability of Silk Fabric Dyed with Natural Dyestuffs)

  • This study was carried out on the process of natural dye substances which can flock ultraviolet rays without producing harmful substances by pollution, germs, etc. While making human body safe in contact, extracting Gardenia, Sappan wood, Radix scutellariae, Cromwell, Mugwort, Gallnut, Lac. Those are used for curing diseases, dyeing silk cloth with those extracts, and examining the color fastness, Ultraviolet protection. From this study, the following conclusion was produced. Brightness was lower in pre-mordanting than in post. And color difference showed that Radix scutellariae was the highest. Aluminum-mordanting showed a higher color difference than Fe. Colorfastness to washing was high in all samples, and the degree of color-changing by washing was the highest in the sample dyed by gardenia, pre-mordanted gallnut showed the highest degree. Colorfastness to perspiration was the highest in gardenia. UV-A test showed that all samples a high rate of 85% or higher compared with test white cloth. Especially, post-mordanted radix scutellariae, pre-mordanted gallnut and post-mordanted sappanwood showed a superior blocking rate.

황금약침(黃芩藥鍼)이 Type 1 Hypersensitivity에 미치는 영향 (Effect of scutellariae radix pharmacopuncture on the type 1 hypersensitivity)

  • Objectives : We studied the effects of Scutellariae Radix pharmacopuncture solution (SRHAS) on the type 1 hypersensitivity. Methods : In vivo, we measured compound 48/80-induced active systemic anaphylactic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rats. In vitro, we showed effects on cytotoxicity and ${\beta}-hexosaminidase$ release from RBL-2H3 cells. Results : In vivo, SRHAS pretreatments (100% or 50%) at BL13 inhibited active systemic anaphylactic shock induced by compound 48/80. PCA was only inhibited by pretreatments of SRHAS at optional points. In vitro, $0.1{\sim}2%$ SRHAS treatments did not affect cell viability while ${\beta}$-hexosaminidase release was significantly inhibited. Conclusions : These results suggest that SRHAS may be beneficial in the inhibition of type I hypersensitive inflammatory response.

황금 추출물의 항균효과 및 열안정성 (Antimicrobial Effect of Scutellariae Radix and Its Thermal Stability)

  • The present study investigated the antimicrobial properties of medicinal herbs including Scutellariae Radix (SR: dried root of Scutellariae bicalensis Georgi). Among hot-water extracts of medicinal herbs tested in this study, SR extract showed the most potent antimicrobial activity with minimum inhibitory concentration (MIC) of 0.625 mg/mL. In particular, synergistic effects of antimicrobial activity were observed upon combined application of SR and chitooligosaccharide as indicated by MIC of 0.125 mg/mL and FIC (fractional inhibitory concentration) index of 0.45. Thermal stability analysis indicated that the components responsible for antimicrobial activity was stable for 8 months at $45^{\circ}C$. Antimicrobial activity was proven to be effective in foods as well as in cosmetics as comparable to that of the chemical preservatives.
