Dynamic Relative Displacement of Geosynthetic-Soil Interface Considering Chemical Effect (화학적 영향을 고려한 토목섬유-지반 접촉면의 동적상대변위)

  • Recently, the construction of onshore waste landfill sites has been studied due to the increase of waste and geosynthetics are widely utilized to enforce and protect waste landfill. Geosynthetics comprises the interface with soil and the seismic behavior and stability mostly depend on the dynamic shear behavior of the geosynthetic-soil interface. Therefore, the understanding of dynamic shear behavior and dynamic relative displacement of the interface is critical. The dynamic shear behavior of the interface is affected by surrounding conditions and loading and shows very complicated response, and, it is difficult to study theoretically. In this study, laboratory test to investigate dynamic relative displacement is performed under chemical condition. Dynamic interface apparatus is utilized and cyclic simple shear tests are conducted under short term (60 days of submerging period) and long term (840 days of submerging period) conditions. Consequently, relative displacement of the interface shows the largest values under acid condition, which means more severe damage of the interface.

Usefulness of Chlorine Dioxide to Airborne Bacteria at a Hospital Using Biological Information (생물학적 정보를 활용한 병원에서 존재하는 공기중 부유 세균에 대한 이산화염소의 유용성)

  • In the present study, using biological information of bacteria and biochemical information of chlorine dioxide gas, Gram-positive bacteria, e.g., Alloiococcus otitis, Erysipelothrix rhusiopathiae, Staphylococcus caprae, Staphylococcus lentus, and gram-negative bacteria, e.g., Acinetobacter baumannii complex, Aeromonas salmonicida, Brucella melitensis, Oligella ureolytica were used whether a plastic kit to release ClO2 gas could inhibit their growth. Overall, chlorine dioxide gas showed about 99% inhibition of bacterial growth, with less than 10 CFU. However, it was found that Gram positive Alloiococcus otitis and Gram negative Aeromonas salmonicida had more than about 50 CFU. When comparing the results of experiments with several bacteria, it suggested that the concentration of chlorine dioxide gas would be at least 10 ppm to 400 ppm for the bacterial inhibition. The results of this study could be used as basic data to evaluate the clinical usefulness of chlorine dioxide gas. If this study helps with prior knowledge to help clinicians to recognize and prevent the presence of micro-organisms that cause infections in hospitals, it would be helpful for activities such as patient care as a convergence field. In the future, it is considered that the research results will be the basis for rapidly inhibiting the microbes infected with patients by utilizing data of the information of the microbes that are inhibited for chlorine dioxide gas.

Synthesis of Low Concentration of NaOH Solution using $Na^+$ ion in the Concentrated Water from Membrane Separation Process (분리막 농축수에 포함된 Na를 이용한 저농도 NaOH 용액의 합성)

  • Concentrated water discharged from seawater desalination process contains a high concentration of $Na^+$ ion. Electrolysis was applied to synthesize NaOH solution from the highly concentrated NaCl solution. The effect of various operating parameters of composited laboratory-scale chlor-alkali (CA) membrane cell was investigated. The operating parameters such as membrane types (CIMS and Nafion membranes), pretreatment of the membrane, flow rate (73 mL/min~200 mL/min), initial $Na^+$ ion concentration (1.5 M, 3M and 5 M) and current (1.5A and 2A) were evaluated. It was observed that synthesis efficiency of NaOH solution with CIMS membrane was higher than that with Nafion membrane, but the durability of CIMS membrane on $Cl_2$ gas was poor. The synthesis efficiency of NaOH solution increased with increasing initial $Na^+$ ion concentration and current, while the efficiency decreased with increasing flow rate using Nafion membrane.

Wastewater Treatment and Microbial Structure Analysis by Fluorescence In Situ Hydridizationin a Biofilm Reactor (생물막 반응키에서의 폐수 처리 및 Fluorescence In Situ Hybridization에 의한 복합 미생물계 구조 해석)

  • Laboratory scale aerobicfanaerobic biofilm reactor was used for simultaneous and stable removal of organics, N and P components to investigate optimum design and operation parameters and to analyze the microbial distribution and consortium structure of nitrification and denitrification bacteria in aerobic and anaerobic biofilm systems. The biofilm reactor was successfully operated for 143 days to show $COD_{cr},\;BOD_5$, SS removal efficiencies of 88, 88, and 97%, respectively. During the experiment period, almost complete nitrification efficiency of 96% was achieved. Denitrification efficiency was about 45% without addition of any external carbon sources. In case of total phosphorus removal, 74% of the inlet phosphorus was removed. Fluorescence in situ hybridization (FISH) results showed that most of the ammonia oxidizing bacteria in the aerobic nitrification zone was found to be Nitrosomonas species while Nitrospira was the representative nitrite oxidizing bacteria. For the denitrification, Rhodobacter, Rhodovulum, Roseebacter and Paracouus were the dominant denitrification bacteria which was 10 to 20% of the total bacteria in numbers.

Effects of pH and Temperature on the Adsorption of Cationic Dyes from Aqueous Suspension by Maghnia Montmorillonite (수용액으로부터 양이온 염료 흡수에 대한 pH 및 온도 효과)

  • The effects of pH and temperature on the removal of two dyes (neutral red; NR and malachite green oxalates; MG) from aqueous effluents using Maghnia montmorillonite clay in a batch adsorption process were investigated. The results showed the stability of the optical properties of MG in aqueous solution and adsorbed onto clay under wide range of pH 3-9. However, the interaction of NR dye with clay is accompanied by a red shift of the main absorption bands of monomer cations under pH range of 3-5, whereas, those of neutral form remains nearly constant over the pH range of 8-12. The optimal pH for favorable adsorption of the dyes, i.e. ${\geq}$90% has been achieved in aqueous solutions at 6 and 7 for NR and VM respectively. The most suitable adsorption temperatures were 298 and 318 K with maximum adsorption capacities of 465.13mg/g for NR and 459.89 mg/g for MG. The adsorption equilibrium results for both dyes follow Langmuir, Freundlich isotherms. The numerical values of the mean free energy $E_a$ of 4.472-5.559 kj/mol and 2.000-2.886 kj/mol for NR and MG respectively indicated physical adsorption. Various thermodynamic parameters, such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$, ${\Delta}G^{\circ}$ and Ea have been calculated. The data showed that the adsorption process is spontaneous and endothermic. The sticking probability model was further used to assess the potential feasibility of the clay mineral as an alternative adsorbent for organic ion pollutants in aqueous solution.

Antimicrobial Effect of Garlic Extract against Pathogenic Bacteria (병원성 세균에 대한 마늘추출물의 항균효과)

  • The purpose of this study is to verify the antimicrobial effect of garlic and black garlic against pathogenic bacteria. For the comparative analysis of antibacterial effects of garlic, Ampicillin $10{\mu}g$ (BBL) was used as control antibiotics. Research experiments were conducted on each of November 2013 and January 2014. Susceptibility to the antimicrobial effect was measured through Kirby-Bauer disc diffusion method and verified according to the standard proposed by the CLSI. Antimicrobial effect of fresh garlic was higher regardless of the method to extract than Ampicillin $10{\mu}g$. In contrast, the manufacturing methods of the black garlic had no effective differentiations. In antimicrobial susceptibility test, black garlic showed resistance to all of 4 strains. However, in the ethanol-extract of fermented black garlic(natural aging of 15 days.) was found the small changes of the growth-inhibition-zone against S. aureus (8 mm)and E. coli(7 mm). This study proposes a variety attempts about the extraction methods of black garlic for the possibility of food preservation.

Pathogenicity of Entomopathogenic Nematode, Steinernema carpocapsae GSN1 Strain (Rhabditida: Steinernematidae) against Tebenna issikii (Lepidoptera: Choreutidae) (우엉뭉뚝날개나방(Tebenna issikii)에 대한 Steinernema carpocapsae GSN1 계통의 병원성)

  • Entomopathogenic nematode, Steinernema carpocapsae GSN1 strain was evaluated for the environmentally sound control of Tebenna issikii (Lepidoptera: Choreutidae) in the laboratory. The corrected mortality of Tebenna issikii larvae was 100% at the 40 infective juveniles (Ijs)/larva 3 days after treatment with S. carpocapsae GSN1 strain in Petri dish. $LC_{50}$ value of S. carpocapsae GSN1 strain against Tebenna issikii was 5.7 Ijs. The mean penetration numbers of Ijs of S. carpocapsae GSN1 strain at the 5, 10, 20, 40 and 80 Ijs/larva in a Tebenna issikii larva were 1.4, 1.4, 3.2, 5.6 and 11.9 Ijs/larva, respectively. However penetration rate of Ijs of S. carpocapsae GSN1 strain at 5 Ijs/larva was the highest among other nematode concentrations. Progeny of S. carpocapsae GSN1 strain in a Tebenna issikii larva was higher with increasing nematode concentration.

Epidemiological Studies on the Methicillin Resistant Staphylococcus aureus Isolated from Clinical Samples (임상가검물에서 분리한 Methicillin내성 Staphylococcus aureus의 분자역학적 연구)

  • A total of 45 Staphylococcus aureus strains from clinical samples were tested for the biochemical test and antibiotic susceptibility test. Forty-five S. aureus strains were subjected to the molecular epidemiological study by susceptiblity test, antibiogram, bacteriophage typing, polymerase chain reaction and mec-associated hypervariable region gene in order to detect of mecA gene which was one of the structural gene related to antibiotic resistant expression factors. Three of 15 mecA-negative S. aureus isolates were classified as oxacillin resistant despite borderline minimal inhibitory concentration values. Methicillin susceptiblities were completely consistent with PCR results for these strains. On the other hand, 4 of 30 mecA-positive isolates yielded results in the oxacillin and methicillin susceptibility tests which were discrepant from those of PCR analysis. Except for SA6, the methicillin resistant S. aureus strains tested were highly resistant to penicillin, oxacillin, gentamicin, and chloramphenicol. In the phage typing, 27 strains were typable. The Iytic group III was as many as 12 strains, and 7 of 12 were 75/83A/84 type. In the PCR of specific mecA gene probe with chromosomal DNA of 30 methicillin resistant S. aureus, the amplified DNA band of 533 bp was confirmed in 30 strains and not in methicillin sensitive S. aureus. The single amplified band of hypervariable region related to mec was investigated in all of 30 methicillin resistant S. aureus, but in methicillin sensitive S. aureus it was amplified. The size of PCR products was between 200 bp and 600 Up. Four units was directly repeated.

A Comparative Analysis on The Efficiency of Various Clinical Methods for Diagnosis of Tuberculosis (결핵 진단을 위한 검사 방법간의 효율성에 관한 비교 분석)

  • In recent years continuously increasing number of tuberculosis (TB) cases due to the emergence of strains with multidrug resistance and AIDS is a significant global health problem. Therefore, more rapid and reliable diagnosis of TB may be one of the most urgent needs in efforts to eradicate the disease. The present study was designed to compare and assess the diagnostic values and efficiencies between the conventional methods (X-ray, AFB stain and culture) and PCR for pulmonary TB on 171 cases. Chest X-ray finding and clinical features revealed that 39 (22.8%) of 171 sputum specimens were pulmonary TB cases. The statistical data were taken on the basis of the definitive diagnosis: In X-ray, overall sensitivity, specificity, efficiency and false positive and false negative incidence was respectively 69.2%, 87.1%, 83.0%, 12.9%, and 30.8%; 79.5%, 95.5%, 91.8%,4.6% and 20.5% in AFB-stain; 56.4%, 99.2%,89.5%, 0.8% and 43.6% in culture; 82.1%, 96.2%, 93.0%, 3.8% and 17.9% in PCR. PCR got a highest sensitivity and efficiency as well as a lowest false negative incidence. Culture had a highest specificity with a lowest false positive incidence. These results imply that PCR assay is fast, sensitive and efficient method for diagnosis of pulmonary TB. However, combined use of the conventional methods with thorough quality control may offer more opportunities for detecting Mycobacterium tuberculosis and diagnosting TB although they have some limits.

Effect of Aging on the Liver Damage in Bromobenzene-pretreated Rats (연령이 다른 흰쥐에 Bromobenzene 미치는 영향)

  • To evaluate an effect of growth periods on the bromobenzene-induced liver damage, bromobenzene was administrated to 5-week-old rats and 10-week-old rats pretreated with bromobenzene 5 times every other day for 10 days and then the animals were sacrificed. The results were obtained as follows; The increasing rate of serum levels of alanine aminotransferase, xanthine oxidase activity, hepatic lipid peroxide contents, liver weight per body weight (%) and decreasing rate of hepatic contents of protein to each control group were higher in 10-week-old rats than 5-week-old rats by the pretreatment of bromobenzene. According to the above results, 10-week-old rats indicated more severe liver injury than 5-week-old those in case of bromobenzene pretreatment. On the other hand, hepatic aniline hydroxylase activity was more increased in 10-week-old rats than 5-week-old rats both in control and bromobnezene pretreated rats where as the reverse in hepatic glutathione S-transferase. In case of hepatic GSH determination at the intervals of 2, 4, 8, 24 hours throughout 24 hr after administration of single dose of bromobenzene to 5-week-old and 10-week-old rats both in control and bromobnezene pretreated, the rate of GSH utilization was lower in 10-week-old rats than 5-week-old rats. In conclusion, from the above experimental, it is deduce that the 10-week-old rats showed more severe liver injury than 5-week-old rats by the bromobenzene treatment because the disposal ability of bromobenzene in liver was lower in 10-week-old rats than 5-week-old rats.
