Defining the N-Linked Glycosylation Site of Hantaan Virus Envelope Glycoproteins Essential for Cell Fusion

  • The Hantaan virus (HTNV) is an enveloped virus that is capable of inducing low pH-dependent cell fusion. We molecularly cloned the viral glycoprotein (GP) and nucleocapsid (NP) cDNA of HTNV and expressed them in Vero E6 cells under the control of a CMV promoter. The viral gene expression was assessed using an indirect immunofluorescence assay and immunoprecipitation. The transfected Vero E6 cells expressing GPs, but not those expressing NP, fused and formed a syncytium following exposure to a low pH. Monoclonal antibodies (MAbs) against envelope GPs inhibited cell fusion, whereas MAbs against NP did not. We also investigated the N-linked glycosylation of HTNV GPs and its role in cell fusion. The envelope GPs of HTNV are modified by N-linked glycosylation at five sites: four sites on G1 (N134, N235, N347, and N399) and one site on G2 (N928). Site-directed mutagenesis was used to construct eight GP gene mutants, including five single N-glycosylation site mutants and three double-site mutants, which were then expressed in Vero E6 cells. The oligosaccharide chain on residue N928 of G2 was found to be crucial for cell fusion after exposure to a low pH. These results suggest that G2 is likely to be the fusion protein of HTNV.

A Study on the Decrease of Platelet Count, Yield, and Efficiency after Plateletpheresis (Haemonetics MCS 3p에 의한 혈소판 성분채혈 후 혈소판 감소율, 회수량 및 효율에 대한 연구)

  • The purposes of this study were to evaluate the changes in hematologic indices after plateletpheresis and to identify the preapheresis platelet count and clinical factors (age, gender, height, and weight) that showed some influence on the percentage of platelet decrement, yield and efficiency. Plateletpheresis was performed on 101 healthy donors in Bundang CHA general hospital. The data was analyzed using the SAS program with t-test, ANOVA test and Multiple regression. The mean percentage decrease after plateletpheresis was 2.0% in hemoglobin, 1.8% in hematocrit, and 29.7% in the platelet count, while a WBC count showed an increase of 2.6%. The mean percentage decrease of hemoglobin and hematocrit were 1.7% and 1.4%, in males and 3.6% and 3.7% in females, respectively. Particularly the percentage decrease of platelet count was significantly higher in females (40.0%) than in males (27.2%). The platelet decrementage and yield were significantly higher in females, but the efficiency did not differ significantly between males and females. The yield showed the lowest levels in subjects who were 40 years old or over but the platelet decrement and efficiency did not change according to age. The platelet decrement increased as height and weight increased. Also, the platelet decrement and yield increased as the initial platelet counts increased, but the efficiency did not. From multiple regression analysis, the platelet decrement was associated with gender, weight, and initial platelet count. The yield was related to the initial platelet count, but the efficiency was not related to gender, age, weight, height or initial platelet counts. This study has a limitation of the generality of the study results since this study was conducted only in a single university hospital. Further study would be necessary to find out a subpopulation that is sensitive to the hematologic change after plateletpheresis, and to determine the standard criteria for blood donation based on the subpopulation.

  • 본 연구는 대학생의 성격유형에 따른 섭식태도 및 비만스트레스를 알아보기 위해 2009년 11월 2일~11월 19일까지 대전지역 일개대학 대학생을 대상으로 설문지를 배포하여 총 285부의 유효부수를 조사에 활용하였다. 각 문항에 대한 빈도분석과 백분율, 교차분석을 이용하였고, 집단간 차이에 대한 유의성 검증을 위해 카이제곱과 로지스틱 분석을 실시하였다. 본 연구의 주요결과는 대학생의 성격유형(A형,B형)에 따른 섭식태도는 카이제곱 분석을 한 결과 조사대상자의 일반적 특성 통계적으로 유의하지 않아서, 성격유형에 따른 섭식태도의 차이를 나타내지 않았다. 대학생의 성격유형(A형,B형)에 따른 비만 관련 스트레스는 카이제곱 분석을 한 결과 조대상자의 일반적 특성 통계적으로 유의하지 않아서, 성격유형에 따른 비만스트레스는 차이를 나타내지 않았으며, 섭식태도와 비만스트레스에 영향을 미치는 요인을 로지스틱 분석한 결과 각각에 성격유형에 따른 차이를 나타내지 않았다. 본 논문은 대학생의 성격유형에 따른 섭식태도, 비만스트레스에 차이가 있을 것으로 예상하였으나, 연구결과 별다른 차이를 보이지 않아 섭식태도, 비만스트레스는 성격유형과 무관한 것으로 나타났다. 이상의 결과에서 볼 때 대학생의 성격유형과 섭식태도, 비만스트레스는 무관한 것으로 판단되어 진다. 선천적인 성격유형과 비만과의 관계에 대한 다각적인 선행연구가 전무한 실정이므로 앞으로 보다 많은 연구를 통해 비만관련 영양 교육 및 보건교육, 나아가 비만 관련 치료 시 성격유형에 대한 고려가 이루어지기를 기대한다.

  • 병원 근로자의 VDT 자각 증상에 대한 선행연구를 보면 대부분이 간호사 관련 직종으로 한정되어 있고, 방사선사에 대한 연구는 수행되지 않았다. 본 연구는 2009년 10월 15일부터 11월 2일까지 대전광역시에 소재하고 있는 종합병원에 VDT 작업을 시행하는 보험심사, 원무, 의무기록, 전산, 행정부서 근무자와 방사선사를 대상으로 구조화된 설문지를 활용하여 분석하였다. VDT 증후군 자각 증상 점수를 종속변수로 한 다중선형회귀 분석 결과 관련요인으로 안관련 증상에 영향을 미치는 요인을 보면 성별, 건강염려, 일상생활 방해이며, 전신증상은 운동, 스트레스, 건강상태 인지 관련, 근골격 증상에서는 임상경력 6-10년, 대인관계, 건강상태 인지 관련 건강염려와 일상생활 방해로 나타났으며, 통계적으로 유의한 차이를 보였다. 피부 증상으로는 연령 20-29세, 결혼여부, 식습관, 건강상태 인지 관련 소화 정도와 일상생활 방해로 나타났으며, 심리 증상으로는 성별, 결혼 여부, 스트레스, 소화 정도, 일상생활 방해로 나타났으며, 통계적으로 유의한 차이를 보였다. 타부서 근로자와 방사선사의 VDT 증후군 자각점수 차이가 크게 나타나지 않았으며, 이는 방사선사의 업무가 VDT 환경으로 변함으로써 VDT 증후군 자각 증상이 일어날 수 있다는 것을 나타내는 것이다. 이는 정기적인 순환근무를 시행함으로써 VDT 환경에 지속적인 노출을 피하는 것이 예방 대책이라 할 수 있다.

A study on factors relevant to the self-perception of halitosis and the correlation between halitosis and obsessive-compulsive disorders (구취의 자각요인과 강박증에 관한 연구)

  • Objectives : The purpose of this study to provide preliminary data to effective halitosis control. Methods : A survey was carried out the self-perception of causative factors of halitosis in 98 female college students in Chungnam province. In addition, the levels of oral gas and breath gas were measured by use of the B&B checker, and an analysis was made of the correlation between halitosis and obsessive-compulsive disorder (OCD). Results : 1. Subjective halitosis awareness degree different degree of dryness, 'None', 'some', 'little' people who feel as the 'little fly' (68%), 78.3%, 70% was higher (p<0.05), halitosis interest 'very worried', 'worry', 'normal' person with a' not at all 'to 58.3% higher (p<0.01). 2. In regard to OCD severity, 49.0%, 34.7% and 16.3% were found to be mild, moderate and severe respectively. Halitosis tended to be severe in proportion to OCD severity, which was statistically significant (p<0.05). 3. Oral gas had a positively correlation with breath gas (r=0.329, p<0.01), but had a negative one with OCD severity (r=0.204, p<0.05). Conclusions : Symptoms of oral dryness and halitosis interest were associated with self-perception of halitosis. The levels of oral gas and breath gas were in inverse proportion to OCD severity. Hereat, it needs to analyze psychogenic factors properly as regards the diagnosis or treatment of halitosis.

The Effect of a Community-Based Nutrition Intervention Program on Dietary Behavior and Nutritional Status of Low-Income Elderly Women in Gwangju City (광주지역 일부 저소득층 여자노인에서 지역사회 영양중재 프로그램 적용에 따른 식행동 변화 및 영양상태 개선 효과)

  • This study was conducted to measure the effect of a community-based nutritional intervention program on dietary behaviors and nutrient intakes of low-income elderly women in Korea. The subject of the study was a group of 88 elderly women using the free meal service in Dong-gu, Gwangju city in Korea. The study was carried out from April to July 2009, and the pretest, application of nutrition education, and post-test were applied in each stage. This community-based nutrition intervention program consisted of snack supplement and nutrition education provided three times a week and 36 times in total. The nutrition education was led by the nutritionists and professionals using various educational media. Snack supplement included were milk, soymilk, carrot juice, bread, yogurt, and bananas. The result of pretest showed that the nutrient intakes of the subjects were much below the Recommended Nutrient Intake (RNI) level of Dietary Reference Intake for Koreans. Comparison of the dietary behavior score and nutrient intakes before and after the nutritional intervention program indicated that the food behavior score increased from 3.1 to 5.6 (P<0.001) and intake of energy, protein, carbohydrate, thiamin, niacin, vitamin C, iron, and potassium of the subjects all increased significantly (P<0.05). Mean Adequacy Ratio (MAR) of their nutrient intakes was also improved from 0.53 to 0.64 (P<0.01). These results indicated that the nutrition intervention program conducted in this study was effective in improving dietary behavior and nutrient intakes of elderly women in local community.

The Effect of Sexual Rehabilitation Education on the Knowledge and Adjustment of Sexual of Spinal Cord Injury Clients (성 재활 교육이 척수장애인의 성 지식과 성 적응에 미치는 효과)

  • The purpose of this study is to identify the effect of sexual rehabilitation education on the knowledge and adjustment of sexual of spinal cord injury clients by the method of nonequivalant control group pretest-posttest group. The data were collected from Jan. 19 though Feb. 16, 2000. The experimental group was 26 spinal cord injury client from one rehabilitation center, and the control group was 26 spinal cord injury client from two rehabilitation center. Sexual rehabilitation education was done with manuals, videotapes and chart. Education time was 120 minutes a week for 2 weeks. The study tool was about Sexual Knowledge, designed Song, Chung Sook(1990) and SIAC(Sexual, Interest, Activity, & Satisfaction) designed Kruter, Sullivan, Siosteen(1994) translated by Kang, Hyun Sook, Koh, Jung Eun, Suh, Yeon Ok, Yee, Oon Hee(1999). The collected data were analyzed SPSS program. The Data were analyzed descriptive statistics and $X^2$ test, Pearson's Correlation Coefficient, t-test, F-test (ANOVA), Duncan's multiple comparison test. The result were as follows. 1. The first hypothesis was accepted : The group educated about sexual rehabilitation has more sexual knowledge than those of the uneducated group(t = 7.412, p = .001). 2. The second hypothesis was accepted : The group educated about the sexual rehabilitation has more adjustment of sexual than those of the uneducated group(t = 5.524, p= .001). 3. The third hypothesis was accepted: The higher sexual knowledge state, the higher sexual adjustment(r= .449, p= .003). According to results, sexual rehabilitation education helps Spinal cord Injury Clients to gain knowledge and adjustment of sexual.

Asymptomatic Human Paragonimiasis among the Karen People in Tak Province, Thailand: A Case Report

  • During the mobile clinic activities in Tak Province, Thailand, Paragonimus sp. eggs were found in a fecal sample of a 72-year-old Karen resident. Paragonimus DNA was amplified from the stool sample and identified to P. heterotremus. The patient did not have any symptoms. Apparent pulmonary lesion was not found on the chest X-ray. The patient admitted habitual consumption of semi-cooked or roasted waterfall crabs for several years. The waterfall crabs collected from stream near the village were found negative for Paragonimus metacercariae. In northern Thailand, paragonimiasis remains as one of the public health concerns and should be ruled out for asymptomatic pulmonary patients.

Development of a Social Contact Survey Instrument Relevant to the Spread of Infectious Disease and Its Application in a Pilot Study Among Korean Adults

  • Objectives: This study aimed to develop a valid social contact survey instrument and to verify its feasibility for use among Korean adults. Methods: The Delphi technique was used to develop an instrument to assess social contacts, which was then applied in a cross-sectional pilot study. A panel of 15 medical professionals reviewed the feasibility and validity of each item. The minimum content validity ratio was 0.49. Thirty participants used the developed measure to record contacts during a 24-hour period. Results: After a systematic review, the survey instrument (parts I and II) was developed. Part I assessed social contact patterns over a 24-hour period, and part II assessed perceptions of contacts in daily life and preventive behaviors (hand hygiene and coughing etiquette). High validity and feasibility were found. In the pilot study, the 30 participants had a combined total of 198 contacts (mean, 6.6 daily contacts per person). The participants' age (p=0.012), occupation (p<0.001), household size (p<0.001), education (p<0.001), personal income (p=0.003), and household income (p<0.001) were significantly associated with the number of contacts. Contacts at home, of long duration, and of daily frequency were relatively likely to be physical. Assortative mixing was observed between individuals in their 20s and 50s. Contact type differed by location, duration, and frequency (p<0.001). Conclusions: The developed social contact survey instrument demonstrated high validity and feasibility, suggesting that it is viable for implementation.
