Stain Fungi and Discoloration Control on Rubberwood (Hevea brasiliensis Muell. Arg.) by Vacuum-Pressure Treatment with Catechin from Gambir (Uncaria gambir Roxb.)

  • Recently, the morphological and molecular features of five stain fungi infecting rubberwood (Hevea brasiliensis), namely Paecilomyces maximus, Paecilomyces formosus, Penicillium crustosum, Paecilomyces lecythidis and Aspergillus chevalieri, have been studied. Prior to this study, the authors revealed that catechin from gambir (Uncaria gambir) could inhibit the growth of the white-rot fungus Schizophyllum commune, and it was important to determine the bioactivity of the aforementioned agent against A. chevalieri. The efficacy of the biocidal agent was examined using a laboratory wooden block test. Rubberwood blocks, 8 mm in thickness, 20 mm in width, and 30 mm in length, were impregnated with catechin solution at concentrations of 6%, 9%, 12%, and 15% (w/v) using the vacuum-pressure method, and their bioactivity was monitored over three weeks through visual and scanning electron microscope assessment of fungal growth as well as the discoloration intensity of the wood samples. The results showed that catechin treatment increased the resistance of wood samples to A. chevalieri. Overall, the higher the catechin concentration, the lower the fungal growth. The lowest fungal growth was observed in the wood samples treated with 12% and 15% catechin (score of 0), demonstrating no discoloration. In contrast, the fungal growth score of the untreated wood samples reached 4, indicating severe discoloration. Catechins appear to be adequate biofungicides against stain fungi in rubberwood.

목재부후균의(木材腐朽菌)의 기생(寄生)에 의한 한국산(韓國産) 유용목재(有用木材)의 비교내후성(比較耐朽性) 연구(硏究) (Studies on the Relative Durability of Useful Woods Grown in Korea to the Parasitic Wood Destroying Fungi)

  • This experiment was undertaken for the purpose of investigation of relative durability about thirty sorts of the korean native wood and nine sorts of the wood destroying fungi Polystictus versicolor, Irpex consors. Poria vaporaria, Merulius lacrymans, Schizophyllum commune, Trametes pini, Lenzites betulina, Paxillus panuoides by means of a laboratory methods. The results obtained are; (from the table 2 to 10) 1. By measuring the percentage of loss in weight with regard to Polystictus versicolor, the sap wood contained in Junipeus chinesis was calculated at 18.2%, therefore it is much higher durability than any others and the sap wood contained in Pinus densiflora was measured at 44.10%, therefore it is much lower durability than any others. 2. By measuring the percentage of loss in weight with regard to Irpex consors, the heart wood contained in Butula schmidtii was calculated at 1.23%, therefore durability is very high and the heart wood contained in Quercus dentata was calculated at 26.79%, therefore durability is very low. 3. By measuring the percentage of lass in weight with regard to Lenzites betulina, the heart wood contained in Betula schmidtii was calculated at 5.33%, therefore it is much higher durability than any others and the sap wood with regard to Abies holophylla was measured at 45.48%, therefore durability is very low. 4. By measuring the percentage of loss in weight with regard to Poria vaporaria, the sap wood contained in Fraxinus rhynchophylla was calculated at 5.57%, therefore durability is very high and the sap wood contained in Paulownia coreana was calculated at 35.6%, therefore it is much lower durability than any others. 5. By measuring the percentage of loss in weight with regard to Merulius lacrymans, the heart wood contained in Castanea crenata was measured at 0.09% and in the next place, the heart wood contained in Abies holophylla calculated at 0.15% did not decay almost and the sap wood contained in Cornus controversa was measured at 32.88%, therefore it is lower durability than any others. 6. By measuring the percentage of loss in weight with regard to Schizophyllum commune, the sap wood cantained in Junipensis chinensis was calculated at 1.09%, therefore durability is very high and the sap wood contained in Populus deltoides was calculated at 36.87%, it is much lower durability than any others. 7. By measuring the percentage of loss ill weight with regard to Trametes pini, the sap wood contained in Robinia pseudo-acacia was calculated at 1.33%, therefore durability is very high and the sap wood contained in Comus controversa was calculated at 33.19%, it is much lower durability than any others. 8. By measuring the percentage of loss ill weight with regard to Paxillus panuoides. the heart wood contained in Zelkova serrata was calculated at 1.73%, therefore durability is very high and the sap wood contained in Populus deltoides was calculated at 42.30%, therefore durability in very low. 9. By measuring the percentage of loss in weight with regard to Dardalea quercina, the heart wood contained in Castanea crenata was calculated at 0.92%, therefore durability is very high and the sap wood contained in Cornus controversa was calculated at 32.00% therefore durability is very low. The above results are summarized as follows 1. Durability between the heart wood and the soft wood find that it is nonsignificant, while durability is higher heart wood than sap wood. 2. Special sorts including in the element like Castanea cenata, Robinia pseudo-acacia, Juniperus chinensis and heart wood like Betula schmiditii and Styrax obasia are highly durable and soft wood like Cornus controversa, Populus davidiana, Salix koreensis, Celtis jessensis are lowly durable in general. 3. Among the wood destroying fungi Paxillus panuoides is highly durable and as follows. Poria vaporaria, Lenzites betulina and the Daedalea quercina is less in general and Irpex consors, Mersulius lacrymans are a middle degree.

The Mycobiota of Air Inside and Outside the Meju Fermentation Room and the Origin of Meju Fungi

  • The fungi on Meju are known to play an important role as degrader of macromolecule of soybeans. In order to elucidate the origin of fungi on traditional Meju, mycobiota of the air both inside and outside traditional Meju fermentation rooms was examined. From 11 samples of air collected from inside and outside of 7 Meju fermentation rooms, 37 genera and 90 species of fungi were identified. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp., Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, Asp. nidulans, Aspergillus sp., Cla. cladosporioides, Eurotium sp., Penicillium sp., Cla. tenuissimum, Asp. niger, Eur. herbariorum, Asp. sydowii, and Eur. repens were collected with high frequency. The concentrations of the genera Aspergillus, Eurotium, and Penicillium were significantly higher in inside air than outside air. From this result and those of previous reports, the origin of fungi present on Meju was inferred. Of the dominant fungal species present on Meju, Lichtheimia ramosa, Mucor circinelloides, Mucor racemosus, and Scopulariopsis brevicaulis are thought to be originated from outside air, because these species are not or are rarely isolated from rice straw and soybean; however, they were detected outside air of fermentation room and are species commonly found in indoor environments. However, Asp. oryzae, Pen. polonicum, Eur. repens, Pen. solitum, and Eur. chevalieri, which are frequently found on Meju, are common in rice straw and could be transferred from rice straw to Meju. The fungi grow and produce abundant spores during Meju fermentation, and after the spores accumulate in the air of fermentation room, they could influence mycobiota of Meju fermentation in the following year. This could explain why concentrations of the genera Aspergillus, Eurotium, and Penicillium are much higher inside than outside of the fermentation rooms.

국내 참나무시들음병 병원균에 대한 버섯 균주의 길항 특성 (Antagonistic Properties of Mushroom Strains to Korean Oak Wilt Pathogen, Raffaelea quercus-mongolicae)

  • 국내 참나무시들음병 병원균인 Raffaelea quercus-mongolicae에 대한 버섯 균주들의 길항 특성을 대치배양을 통해 조사하였다. 총 158개의 시험 균주 중 33개의 균주는 병원균에 대해 단기간 (6~7일) 및 장기간(27~31일) 길항력이 전혀 없었으며, 96개의 균주는 길항력이 미약하거나 길항력이 장기적으로 지속되지 못했다. 이와 달리 29개의 균주는 단기간 및 장기간 길항력이 각각 $\geq$ R+와 $\geq$ R로 타 시험 균주에 비해 높게 나타났으며, 이들과 같이 R. quercus-mongolicae에 대해 길항력을 갖는 균주들은 활엽수(참나무류 포함)나 침엽수 등 발생 기주가 서로 다른 다양한 서식지에서 살아가는 버섯 균종들의 균주임을 알 수 있었다. 참나무시들음병 피해목을 버섯 재배용 자원으로 활용하기 위해 이에 적합한 7개의 길항 균주를 다음과 같이 선발하였다; 붉은덕다리버섯(KFRI 914), 치마버섯(KFRI 832), 구름버섯(KFRI 100), 덕다리버섯(KFRI 654), 간버섯(KFRI 588), 표고(KFRI 269, 764).

소나무와 잣나무 원목에서 변재변색 발생 (Development of Fungal Sapstain in Logs of Japanese Red Pine and Korean Pine)

  • 본 연구는 동절기(1월 중순)에 벌채된 후 목재집하장으로 운반되어 야적장에 저목 중인 소나무와 잣나무 원목을 대상으로 변재변색의 발생시기와 변색 특성을 조사하기 위하여 수행되었다. 벌채 후 3, 4, 5, 6, 8개월 경과시 변색 평가를 실시하였는데, 매 평가시 원목 3개를 임의로 선정하여 원목당 수축방향에 대해 일정 간격으로 7~9개의 두께 3 cm 원판을 채취하여 변색 원인균을 분리한 후 변재변색의 방사방향 최대 침투깊이와 % 변색율을 측정하였다. 변재변색은 전적으로 충형변색이었으며, 주요 변색원인균인 Ophiostomatoid 균을 매개하는 수피천공충은 소나무좀으로 확인되었다. 소나무와 잣나무 원목은 5월 이전까지는 변재변색의 위험없이 저목할 수 있으나 5월 이후부터는 원목의 변재변색이 급속하게 증가하였다. 변색의 정도는 소나무보다 잣나무에서 심하였으며, 장마철 저목 중에 소나무 원목에서 개떡버섯과 치마버섯의 자실체를 다수 관찰할 수 있었다. 본 연구를 통해 밝혀진 결과들은 앞으로 동절기에 벌채되어 저목 중인 소나무와 잣나무 원목에 발생하는 변재변색을 예방하기 위한 제반 조치를 취하는데 매우 중요한 자료로 사용될 것이다.

팽이버섯 수확후배지를 이용한 곰팡이 균사체 기반 복합소재의 특성 (The characteristics of fungal-mycelium-based composite materials using spent mushroom substrates of Flammulina velutipes)

  • 야생버섯 및 재배버섯들 중에서 생장이 우수한 균주를 선발하기 위하여 PDA 배지 상에서 생육이 빠른 9 균주를 선발하였다. 톱밥배지에서 균주별 균사체의 생장을 알아보기 위하여 참나무 톱밥과 미강 80% : 20% (w/w) 비율로 혼합한 배지에서 선발된 치마버섯(No. 7), 상황버섯(No. 29), 아까시재목버섯(No. 39), 영지(No. 52) 균주를 접종하여 14일간 배양한 결과, 영지(No. 52) 균주의 균사체 생장이 가장 우수한 것으로 나타났으며, 기질 건조 후 취급비율도 영지버섯 균사체를 배양한 배지에서 가장 우수하였다. 수확후배지(SM)와 참나무 톱밥(OS) 혼합비율 50% : 50% (w/w), 60% : 40% (w/w), 80% : 20% (w/w)로 배지를 제조하여 선발 균주인 영지(No. 52) 균주를 접종하여 배양한 결과, 균사체의 직경은 수확후배지(SM) : 참나무 톱밥(OS) = 60% : 40% (w/w) 혼합배지에서 균사체 생육과 취급비율이 우수한 것으로 나타났다. 영지(No. 52) 균주를 접종하여 배양한 균사체 소재 사각시편의 압축강도는 300.7 - 302.4 kgf로 스티로폼에 비하여 약 4배 정도 강도가 우수하였으나 탄력성은 없는 것으로 확인되었다. 위와 같은 결과는 버섯농가의 고민거리인 수확후배지를 활용한 균사체 복합소재 개발을 통하여 수확후배지의 새활용을 위한 높은 가능성을 확인하였으며, 향후 기초적인 자료로 활용도가 높으리라 기대된다.

The Origin of Meju Fungi - Fungal Diversity of Soybean, Rice Straw and Air for Meju Fermentation

  • Meju is a brick of dried fermented soybeans and is the core material for Jang such as Doenjang and Ganjang. Jang is produced by addition of salty water to Meju and is considered the essential sauces of authentic Korean cuisine. Meju is fermented by diverse microorganisms such as bacteria, fungi and yeasts. It is known that fungi play an important role in the Meju fermentation and they degrade macromolecules of the soybeans into small nutrient molecules. In previous study, 26 genera and 0 species were reported as Meju fungi. However, it is not comprehensively examined where the fungi present on the Meju are originated. In order to elucidate the origin of the fungi present on the Meju, the mycobiota of 500 samples soybean kernels, 296 rice straw pieces and air samples of Jang factories was determined in 0, 2 and 7 Jang factories respectively. Forty-one genera covering 86 species were isolated from the soybeans and 33 species were identical with the species from Meju. From sodium hypochlorite untreated soybeans, Eurotium herbariorum, Eurotium repens, Cladosporium tenuissimum, Fusarium fujikuroi, Aspergillus oryzae/flavus and Penicillium steckii were the predominant species. In case of sodium hypochlorite-treated soybeans, Eurotium herbariorum, E. repens and Cladosporium tenuissimum were the predominant species. Of the 4 genera and 86 species isolated from soybeans, 3 genera and 33 species were also found in Meju. Thirty-nine genera and 92 species were isolated from the rice straws and 40 species were identical with the species from Meju. Fusarium asiaticum, Cladosporium cladosporioides, Aspergillus tubingensis, A. oryzae, E. repens and Eurotium chevalieri were frequently isolated from the rice straw obtained from many factories. Twelve genera and 40 species of fungi that were isolated in the rice straw in this study, were also isolated from Meju. Especially, A. oryzae, C. cladosporioides, E. chevalieri, E. repens, F. asiaticum and Penicillium polonicum that are abundant species in Meju, were also isolated frequently from rice straw. C. cladosporioides, F. asiaticum and P. polonicum that are abundant in low temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $5^{\circ}C$ and $25^{\circ}C$, while A. oryzae, E. repens and E. chevalieri that are abundant in high temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $25^{\circ}C$ and $35^{\circ}C$. This suggests that the mycobiota of rice straw have a large influence in mycobiota of Meju. Thirty-nine genera and 92 species were isolated from the air of Jang factories and 34 species were identical with the species from Meju. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp. Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, A. nidulans, Aspergillus sp., C. cladosporioides, Eurotium sp., Penicillium sp., C. tenuissimum, A. niger, E. herbariorum, A. sydowii, and E. repens were collected with high frequency.
