Compositional Characterization and Colorant Identification of Omija (Schizandra chinensis) Fruit Extract

  • A major polyphenolic compound extracted from omija (Schisandra chinensis) fruit was structurally identified, and its composition of major nutrients was investigated as well in this study. A dominating high performance liquid chromatography (HPLC) peak of water-extracted anthocyanin represented 94.1% of total absorbable compounds at 520 nm, which was further identified with HPLC-mass spectrometry (MS). As a result, mass-to-charge ratio (m/z) of the predominant anthocyanin was determined to be 727, and it was identical to molecular mass of cyanidin-3-xylosylrutinoside (Cya-3-O-xylrut). This is the first report that colorant of omija is predominantly composed of Cya-3-O-xylrut. Omija fruit contained exclusively 3 types of monosaccharide such as glucosc (0.68 g), galactose (0.01 g), and fructose (0.52 g) per 100 g of fruits. Several organic acids, citric (3.29 g), malic (1.4 g), acetic (0.4 g), and succinic acids (0.36 g) per 100 g of fruits, were detected by high performance anion exchange chromatography (HPAEC) analysis. During the compositional analysis of tree amino acid by HPLC, it was noticed that omija fruit contained substantial amount (0.01 g/100 g of fruits) of $\gamma$-amino butyric acid (GABA).

Effect of the extracts from Schisandra chinensis Fruit and Morus alba Leaf on Insulin Secretion in Glucose-induced HIT-T15 Cells (오미자와 뽕잎 추출물이 glucose에 의해 유도된 HIT-T15세포의 인슐린 분비능에 미치는 영향)

  • This study aimed to examine the effect of the Schizandra chinensis fruit and Morus alba leaf on insulin expression in HIT-T15 cells, which is exposed by glucose. The total polyphenol contents of the S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract were $20.11{\pm}0.35$ mg/g and $50.02{\pm}0.62$ mg/mL, respectively. The S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract contained $2.85{\pm}0.15$ and $8.76{\pm}0.43$ mg/g flavonoids, respectively. The antioxidant ability of the M. alba leaf hot-water extract was found to be superior to that of the S. chinensis fruit ethanol extract. Compared to the HIT-T15-treated 10 mM 2-deoxy-D-glucose, the $100{\mu}g/mL$ S. chinensis ethanol extract was found to have a two fold increase in insulin productivity. Moreover, the $100{\mu}g/mL$ M. alba leaf hot-water extract promoted the insulin secretion of high-glucose-damaged HIT-T15 almost ten fold. The above results showed that the S. chinensis fruit ethanol extract and M. alba leaf hot-water extract may improve the insulin productivity of the beta cell with glucose-induced oxidative damage. These data suggest that the S. chinensis fruit ethanol extract and the M. alba leaf hot-water extract can be used as food materials for the regulation of insulin secretion.

Culinary Characteristics of Hwachae in Korean Cookbooks Published in 1600-1940s (1600년대-1940년대 조리서에 등장한 화채류의 종류 및 조리법 특성)

  • Purpose: This study aimed to characterize the recipes of Hwachae in Korean cookbooks published from the 1600's to 1940's. Methods: We conducted a content analysis of thirteen old cookbooks (Eumsikdimibang, Jubangmun, Jeungbosallimgyeongje, Kyugonyoram, Imwonsibyukji, Gyuhapchongseo, Sieuijeonseo, Buinpilji, Chosunyorijebub, Chosunmusangsinsikyorijebub, Ganpyeonchosunyorijebub, Chosunyori, Chosunyoribub). We collected a total of 99 recipes of Hwachae and analyzed the ingredients used and characteristics of the recipes according to five groups of Hwachae: noodle type Hwachae, dumpling type Hwachae, fruit Hwachae, flower Hwachae, and the others. Results: The noodle type Hwachae included Changmeyon, Smyeon, Nanmyeon, and Waemyeon. Mung bean (81.0%) and Schisandra (95.2%) were mostly employed as solid ingredient and as beverage base, respectively. In the noodle type Hwachae, Wonsobyeong and Sudan belonged to the dumpling type Hwachae, in which mung bean (51.7%) was mostly used as solid ingredient and just water without any additive was used as the most popular beverage base (62.1%) unlike in other types of Hwachae. There were various types of fruit Hwachae, which used different fruits as solid ingredient such as pear, peach, cherry, strawberry, etc. Pears (44.4%) have been employed as the most popular ingredient and sweets such as honey and sugar have been used in all the fruit Hwachae. The flower Hwachae included Hwamyeon, Sunchaehwachae, Jangmihwachae, and Bomhwachae. In all the flower Hwachae, schizandra has been used as a beverage base. Conclusion: These findings provide basic information required for developing standard recipes of traditional Hwachae.

  • Research on pesticide residues as endocrine disruptors in natural medicines was initiated by Korea Food & Drug Administration this year. We determined the presence and levels of certain pesticides in selected natural medicines. The natural medicines collected this year are Glycyrrhiza Root, Cinnamon Bark, Pueraria Root, Polygonatum Rhizome, Jujube, Schizandra Fruit, Lycium Fruit, Liriope Tuber, Eucommia Bark, Peony Root, Korean Angelica, Dioscorea Rhizome, Cnidium Rhizome, Cassia Seed, Platycodon Root, Comus Fruit, Mentha Herb, Epimedium Herb, Bupleurum Root, and Ginger, which have no data for pesticide residues and 192 samples of them were circulated in Korea, 28 samples were circulated in China. In order to analyze many pesticides in large number of samples we used simultaneous multi-residue analysis of pesticides by GC-ECD, which was followed by GC-MSD analysis to confirm the identity of the detected pesticide in each sample.

On the Proximate composition, Organic acids and Anthocyanins of Omija, Schizandra chinensis Baillon (오미자(五味子)의 일반성분(一般成分), 유기산(有機酸) 및 Anthocyanin 색소(色素)에 관하여)

  • Omija (Schizandra chinmensis Baillon) grown widely in Korea has been used as a traditional drink and as an ingredient in herbal medicine. A determination of the proximate composition, organic acids and major anthocyanins in Omija was made. 1. The proximate composition of Omija showed water to be 84.2%, protein to be 1.1%, fats to be 0.9%, reducing sugars to be 10.9% and total anthocyanins to be 0.2% in terms of weight percent of fresh, ripe Omija 2. Gas chromatography showed citric acid(3.5%), malic acid(1.4%) and succinic acid(0.2%) to be the major organic acids of the fruit. 3. The anthocyanins, the major pigments present in Omija were separated and isolated into 3 fractions using column chromatograph on insoluble polyvinylpyrrolidone. The main anthocyanin was tentatively identified as peonodin 3-glucoside on the basis of its absorptive properties before and after treatments with acid and $AlCl_3$ in the UV and visible ranges.

Investigation on the Technology Trend in Omija by the Patent Index (특허지표를 통한 오미자 기술 동향 연구)

  • The Omija (Schizandra chinensis Baillon) is a fruit native to northeast Asia that is cultivated in South Korea and China. Using 1,938 valid patents of 6 group countries, we analyzed the patent trend based on year, countries, applicants, and technology. The technologies are categorized the 10 sub-technologies such as medicine, quasi-drugs, food, feed, cosmetics, cultivation, genome, manufacture, preprocessing, and etc. The technology level and competitiveness are analyzed using patent index such as cites per patent, patent impact index, patent family size and technology strength. In Korea, patent number rapidly increasing and individual technical level is lower than other countries. However, overall technical competitiveness is estimated high due to multiple patents. We suggest that cosmetics and cultivation fields are most likely to be developed in future omiza technology development in Korea. Our study will provides to the information of technical trend to support performing of new projects for Omija plant.

Varietal and Yearly Differences of Lignan Contents in Fruits of Collected Lines of Schizandra chinensis Baillon (오미자 수집종별 리그난 함량의 연차간 차이)

  • To know the varietal difference and yearly changes of lignan contents in the fruits of collected lines of Schizandra chinensis Baillon, three lignan compounds, schizandrin, gomisin A, and gomisin N, were quantitatively analyzed using High Performance liquid Chromatography. The average contents of schizandrin, gomisin A, and gomisin N, showed 0.78%, 0.25%, and 0.63% in the 6-year-old fruits of 24 lines harvested in 1996, and 0.72%, 0.22%, and 0.63% in the 7-year-old ones of 59 lines harvested in 1997. There were the wide range of coefficient of variation (CV) values, the significant differences by the lines and harvest years, and the significant interaction between line and year for lignan contents. Schizandrin contents during 3 years, $1995{\sim}1997$, showed relatively higher amounts more than 0.9% in Chungju 143, Chungju 453, and Chungju 532. Among 23 lines analyzed from 1995 to 1997, Chungju 542, Chungju 547, and Chungju 580 contained continuously higher amounts more than 0.8% of schizandrin and had lower CV values of lignan contents by the harvest years than the other lines. There were a highly significant and negative correlation between fruit weight and lignan contents, and a positive correlation among contents of schizandrin, gomisin A, gomisin N, and total lignan.

Suppression of Th2-associated Cytokines Expression by Schizandrin in Basophils and Mast Cells (Schizandrin에 의한 호염구와 비만세포의 Th2 관련 사이토카인 발현 억제)

  • Schizandrin (SZ), was isolated from the fruit of Schizandra chinensis, has been reported to have many biological properties, including anti-inflammatory and antitumor activities. However, its anti-allergic effects are not completely elucidated. We focused on the anti-allergic effects of SZ in PMA/ionomycin (PI)-induced rat basophilic leukemia (RBL-2H3) cells and P815 mast cells. Cytokines (IL-4, IL-13), synthesized by basophils and mast cells, are implicated in pathological conditions such as asthma and allergy. The production of IL-4 and IL-13 was quantified by ELISA and the mRNA expression was detected by using RT-PCR assay. In this study, we found that SZ did not show cytotoxic effect at up to 100 ${\mu}M$ on RBL-2H3 cells and mast cells. In addition, SZ inhibited the production of IL-4 and IL-13 and also decreased the level of mRNA in PI-induced RBL-2H3 cells and mast cells in a dose-dependent manner. Thus, we suggest that SZ may have the effect on preventing allergic disorders by inhibiting IL-4 and IL-13 cytokines.

Evaluation of Anti-oxidant, Anti-microbial and Anti-thrombosis Activities of Fruit, Seed and Pomace of Schizandra chinensis Baillon (오미자 열매, 씨, 착즙 후 박의 항산화, 항균 및 항혈전 활성 평가)

  • In this study, for the efficient use of the byproduct of the omija (Schizandra chinensis Baillon: SC) processing industry, the ethanol extracts of the fruit (F), seed (S), and pomace (P) of SC were prepared, and their useful bioactivities were evaluated. For F-SC, S-SC, and P-SC, the extraction yields were 28.3%, 22.1%, and 7.2%, respectively, and the polyphenol contents were 8.81, 37.22, and 9.20 mg/g, respectively. The total flavonoid content in P-SC (4.31 mg/g) was 3.5-fold higher than that in F-SC (0.76 mg/g). In an antioxidation activity assay, P-SC showed stronger radical scavenging activities against DPPH anion, ABTS cation, and nitrite and stronger reducing power activities than the other extracts. The calculated concentration required for 50% radical scavenging activity, $RC_{50}s$, of P-SC for DPPH anion, ABTS cation, and nitrite was 226.2, 192.5, and $92.5{\mu}g/ml$, respectively. In an antimicrobial activity assay, F-SC, S-SC, and P-SC showed similarly strong growth inhibitions against Bacillus subtilis and P. vulgaris at a concentration of 0.5 mg/disc. F-SC and P-SC showed 15-fold extended time in thrombin, prothrombin, and activated partial thromboplastin time assays at a concentration of 5 mg/ml. The anticoagulation activity of P-SC (2.5 mg/ml) was comparable to that of aspirin (1.5 mg/ml). Furthermore, F-SC and S-SC showed very good platelet aggregation inhibitory activities. F-SC, S-SC, and P-SC did not show significant hemolysis against human red blood cell up to a concentration of 0.5 mg/ml. These results suggest that S-SC and P-SC, both of which are byproducts of the omija processing industry, show strong potential as novel antioxidant, antimicrobial, and antithrombosis agents.

Quality Characteristics of Vinegar Fermented Using Omija (Schizandra chinensis Baillon) (오미자를 이용한 식초발효 및 품질특성)

  • This study was conducted to ferment high quality vinegar by using Omija fruit. Strains of bacteria producing acetic acid were isolated from spoiled Omija wine (OV1 and OV2) and traditional rice vinegar (RA). The bacterium was from optimally-oxidized Omija wine containing 6.0% ethanol and from Omija vinegar with 5.21% acetic acid by 14 days of fermentation. The Omija vinegar got the highest sensory score (5.80) among several commercial vinegars. The Omija vinegar showed a potent antibacterial activity against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, and Pseudomonas fluorescens by the paper disc test, while Omija wine had limited antibacterial activity against S. aureus and P. fluorescens. The diameter of the clear zone formed by the paper disc test using Omija vinegar was not less than 9.5 mm. The DPPH free radical-scavenging activity of Omija wine was higher than that from Omija water extracts. The nitrite scavenging ability of Omija wine (36.98% of all nitrites present) and vinegar (31.14%) was higher than that from Omija water extracts (22.53%). Omija vinegar exhibited strong antibacterial and antioxidant activities. In conclusion, we prepared high quality vinegar from dried Omija.
