Microscopy of Microbial Gas Vesicles

  • Gas vesicles are intracellular gas-filled protein-shelled nanocompartments. The structures are spindle or cylinder-shaped, and typically $0.1{\sim}2{\mu}m$ in length and 45~250 nm in width. A variety of prokaryotes including photosynthetic bacteria and halophilic archaea form gas vesicles in their cytoplasm. Gas vesicles provide cell buoyancy as flotation devices in aqueous habitats. They are used as nanoscale molecular reporters for ultrasound imaging for biomedical purposes. The structures in halophilic archaea are poorly resolved due to the low signal-to-noise ratio from the high salt concentration in the medium. Such a limitation can be overcome using focused ion beam-thinning or inelastically scattered electrons. As the concentric bodies (~200 nm in diameter) in fungi possess gas-filled cores, it is possible that the concept of gas vesicles could be applied to eukaryotic microbes beyond prokaryotes.

  • Recently developed new step wedge has a small size as $10{\times}10\;cm^2$. It can use central beam, and is affected little by scattered rays emmitted from the neighboring step wedges. It's characteristics are same as the intensity scale method, and are excellent compared with conventional boot strap methods. Moreover, the method using new step wedge is simple in operation, and the characteristics are excellent. Now, the above method using new step wedge can supercede the distance method.

  • The flow and heat transfer characteristics behind a backward facing step located in a vertical channel has been studied. In this study, the numerical prediction has been performed by solving the Navier-Stokes equation and energy equation simultaneously with the SIMPLE algorithm embedied in TEACH code. Local heat flux was measured by using Schlieren Interferometer. The flow visualization was performed using the cylindrical lens and the laser beam that is scattered by the supplied glycerine particles. The velocity and temperature distributions, recirculation region, reattachment length, and local heat flux are obtained under the various parameters to investigate the buoyancy effect on the flow and heat transfer characteristics behind the step.

  • A pulsed ultrasonic doppler flowmeter for mesurements of velocity profils in man is described. The device projects a beam of ultrasound in burst of 570 ns duration at 3.5 MHz. The back-scattered signals are processed to produce a signal oxrresponding to the mean velocity over a small region of the flowing stream. The observation range of 112mm is divided into 128 depth channels. The size of this sample volume determines the flowmeter sensitivity and accuracy. The device uses a quadrature detector to detect the direction of the moving target(hemoglobin). The main feature of the novel instrumnet is its simple hardware structure due to sequential signal processing.

  • The high coherence of laser beam has made it possible to observe interference effects even in the light scattered from rough surfaces. That's why, when object with a scattering surface is illuminated with laser light, we do see a speckled appearance due to random interference. This sort of unique property of laser speckle has bruht into existence the new noncontaciting techniques such as speckle metrology method of measuring deformation, displacement, and vibration etc of objects with high optical sensitivity. The measurable range of speckle metrology especially used to measure in -plane information, however, is limited by some factors, the so-called strain, rotation tilt of surface and out of displacement perpendicular to the plane of analysis This restrictions severly limits the measurable range of speckle metrology by causing the decorrelation of speckle patterns. It is the purpose of this paper to give a survey on the measurable limitation of speckle photography method that is one of speckle metrology. Namely we will discuss the mutual relationships and problems of each limitations adding the restriction on the largest and smallest displacement measurable with speckle methods.

  • Three-dimension paraffin compensator was designed to construct the tissue equivalent compensator for irregular body contours and obiliques beam incidence. The ratio of compensator thickness to tissue deficit was depended on field size, depth and air gap because the scattered dose loss. The ratio of compensator-tissue was optimized 0.79, 0.73, 0.61 and 0.56 in 6MV x-rays as function of field size $4{\times}4$, $10{\times}10$, $20{\times}20$ and $30{\times}30cm^2$ respectively. in our study. Using this tissue equivalent compensator, it can be got 2% difference of dose at same mid-plane in phantom study.

  • Collective Thomson scattering (CTS) system is being developed in KAERI based on high power gyrotrons. CTS is a promising diagnostic method to measure fast ion distributions and potentially the fusion product alpha particles in magnetically confined plasmas. By utilizing millimeter-waves from high power gyrotrons as a probing beam, spatially and temporally resolved 1-D ion velocity distributions can be obtained from the scattered radiation with less scattering geometrical constraints. The pulse modulation of gyrotrons enables to separate scattering signal from ECE background noises. The feasibility was assessed with the calculation of spectral density functions under the condition of KSTAR plasmas. Further CTS system requirements are also discussed.

Textures of Claystones and Their Genetic Significances in the Cheonunsan Formation of the Hwasoon Area (화순지역 천운산층 중의 점토암의 조직과 그 성인적 의미)

  • Textures of claystones of the Cheonunsan Formation in the Hwasoon area have been studied using optical microscope and electron microprobe. Microscopic images were observed under the optical microscope using the transmitted polarizing light from thin sections and under the electron microprobe using the back-scattered electron beam from the polished sections. Identification of minerals were made using X-ray diffraction analysis and chemical analysis by electon microprobe. Textural analyses show that the original sedimentary claystones rich in aluminium were subjected to metamorphism by which they changed to the metamorphosed claystone consisting mainly of chloritoid, quartz, andalusite and illite. Later intensive hydrothermal kaolinization of this metamorhosed claystones resulted in the formation of high-aluminous claystones rich in kaolinite exhibiting various complicated replacement textures.

A Study on the NC Embedding of Vision System for Tool Breakage Detection (공구파손감지용 비젼시스템의 NC실장에 관한 연구)

  • In this research, a vision system for detecting tool breakage which is hardly detected by such indirect in-process measurement method as acoustic emission, cutting torque and motor current was developed and embedded into a PC-NC system. The vision system consists of CMOS image sensors, a slit beam laser generator and an image grabber board. Slit beam laser was emitted on the tool surface to separate the tool geometry well from the various obstacles surrounding the tool. An image of tool is captured through two steps of signal processing, that is, median filtering and thresholding and then the tool is estimated normal or broken by use of change of the centroid of the captured image. An air curtain made by the jetting high-pressure air in front of the lens was devised to prevent the vision system from being contaminated by scattered coolant, cutting chips in cutting process. To embed the vision system to a Siemens PC-NC controller 840D NC, an HMI(Human Machine Interface) program was developed under the Windows 95 operating system of MMC103. The developed HMI is placed in a sub window of the main window of 840D and this program can be activated or deactivated either by a soft key on the operating panel or M codes in the NC part program. As the tool breakage is detected, the HMI program emit a command for automatic tool change or send alarm to the NC kernel. Evaluation test in a high speed tapping center showed the developed system was successful in detection of the small-radius tool breakage.

Accuracy and reliability of stitched cone-beam computed tomography images

  • Purpose: This study was performed to evaluate the linear distance accuracy and reliability of stitched small field of view (FOV) cone-beam computed tomography (CBCT) reconstructed images for the fabrication of implant surgical guides. Material and Methods: Three gutta percha points were fixed on the inferior border of a cadaveric mandible to serve as control reference points. Ten additional gutta percha points, representing fiduciary markers, were scattered on the buccal and lingual cortices at the level of the proposed complete denture flange. A digital caliper was used to measure the distance between the reference points and fiduciary markers, which represented the anatomic linear dimension. The mandible was scanned using small FOV CBCT, and the images were then reconstructed and stitched using the manufacturer's imaging software. The same measurements were then taken with the CBCT software. Results: The anatomic linear dimension measurements and stitched small FOV CBCT measurements were statistically evaluated for linear accuracy. The mean difference between the anatomic linear dimension measurements and the stitched small FOV CBCT measurements was found to be 0.34 mm with a 95% confidence interval of +0.24 - +0.44 mm and a mean standard deviation of 0.30 mm. The difference between the control and the stitched small FOV CBCT measurements was insignificant within the parameters defined by this study. Conclusion: The proven accuracy of stitched small FOV CBCT data sets may allow image-guided fabrication of implant surgical stents from such data sets.
