Pinealocytes in the lower vertebrate are known to have photoreceptive function. These photoreceptor cells have been characterized morphologically in various species of lower vertebrates. No such ultrastructural studies, however, were reported in fresh water turtle. The purpose of this study is to characterize the pinealocytes and the phylogenetic evoluton of these cells is discussed in terms of functional analogy. I. Light microscopy: The pineal body was divided into incomplete lobules by connective tissue septa containing blood vessels, and parenchymal cells were arranged as irregular cords or follicular pattern. In the lobules, glandular lumina were present and contained often densely stained materials. II. Electron microscopy: The pineal parenchyma had three categories of cells: photoreceptor cells, supportive cells and nerve cells. The photoreceptor cells had darker cytoplasm compared to the supportive cells, and the enlarged apical cytoplasm(inner segment) containing abundant mitochondria and dense cored vescles protruded into the glandular lumen in which lamellar membrane stacks(outer segment), dense membranous materials, and cilia were present. Some of these lamellated membrane stacks appeared to be dege-nerating while others were apparently newly formed. Constricted neck portion of the photoreceptor cells contained longitudinally arranged abundant microtubules. centrioles and cross-striated rootlets. Cell body had well developed Golgi apparatus, abundant mitochondria, dense granules($0.5{\sim}1{\mu}m$), dense cored vesicles($70{\sim}100nm$), and rough endoplasmic reticulum occasionally with dense material within its cisterna. Basal portion of the photoreceptor cells had basal processes often with synaptic ribbons, which terminate in the complicated zone of cellular and neuronal processes. Synatpic ribbons often made contact with the nerve processes and the cell processes of neighboring cells. In some instances, these ribbons were noted free within the basal process and were also present at the basal cell mem-brane facing the basal lamina. Obvious nerve endings with clear and dense cored vesicles were observed among the parenchymal cells. Photoreceptor cells of the snapping turtle pineal body were generally similar in fine structure to those of other lower verterbrates reported previously, and suggested to have both photoreceptive and secretory functions which were modulated by pinealofugal and pinealopedal nerves. The supportive cells were characterized by having large dense granules($0.3{\sim}1{\mu}m$), abundant ribosomes, well developed Golgi apparatus and rough endoplasmic reticulum. These cells were furnished with microvilli on the luminal cell surfaces, and often had centrioles, striated rootlets, abundant filaments especially around the nucleus, and scattered microtubules. Some supportive cells had cell body close to the lumen and extended a long process reaching to basal lamina, which appeared to be a glial cell. Nerve cells within the parenchyma were difficult to identify, but some large cells located basally were suspected to be nerve cells, since they had synaptic ribbon contact with photoreceptor cells.
The sciatic nerves of anesthetized rabbits were exposed and stimulated by a nerve stimulator in order to observe the myoneural response. These rabbits were divided into three groups and respectively injected with morphine (Group 1), meperidine(Group 2) and pentazocine (Group 3). The sciatic nerves were stimulated periodically and gait changes were observed to see the myoneural activity after the injections. When the distal part of the sciatic nerves were stimulated by the nerve stimulator after the respective drug injections, the normal muscle twitch responses were observed in all the progressional stages of Group 1. However, in Group 2 and 3, the muscle twitch responses decreased gradually, finally disappearing after approximately 10 minutes in these two groups. Complete motor paralysis continued for about 60 minutes. The muscle reactions returned to normal approximately 90 minutes after injection. Specimens drug-injected tissues were severed 4 hours, 24 hours and 1 week after injection respectively. These tissue were investigated under light as well as electron microscopy. The tissue revealed rare to moderate vacuolizations scattered in the axons of the myelinated and unmyelinated nerves of some of the specimens; however, there were no significant pathologic lesions. These results provide evidence that neurophysiologically, meperidine and pentazocine have a local anesthetic-like effect such as motor paralysis, but morphine does not. In addition, the results indicated that neurohistologically, the three narcotics have no significant toxic effects on the nerve tissue.
Some RE (Zr, Nb, REE) ore deposits are located in the middle part of the Korean peninsula. Geotectonically, the RE ore deposits situated on the Kyemyeongsan Formation of northern margin of the Okcheon geosynclinal belt and in the transitional zone between Kyeonggi massif and Okcheon belt. The rare metal deposits distributed in Kyemyeongsan Formation which consists of schist and alkaline granite. The alkali granite has suffered extensive post-magmatic metasomatism and hydrothermal processes. The ore contains mainly Ce-La, Ta-Nb, Y, Y-Nb, Ti-Nb-(U), Nd-Th group minerals. Fergusonite, one of Nb-Y rich REE minerals belonging to the A-B oxides, is most common mineral in the rare metal deposits. The fergusonite bearing rocks may be devided into four types by occurrence features and mineral association, that is, zircon type, allanite vein, feldspar type, and fluorite type. Fergusonites show wide variations in optical properties, due to part of differences in their chemical composition (depending on the types), but also the degree of crystalinity of the individual specimens. Fergusonite metamicts enclosed in biotite are generally surrounded by well developed pleochroic haloes. Usually, fergusonite is accompanied with zircon and other REE-bearing minerals. Petrographical and chemical data are presented for fergusonites which collected different types. $Nb_2O_3$ and $Y_2O_3$ contents range from 48.51 to 53.01 wt.% and 29.18 to 42.02 wt.% respectively. Also, $ThO_2$, (1.83~6.93), $UO_2$, (0.17~2.84), ${\sum}RE_2O_3$ (except to Y) (1.11~8.73), and $TiO_2$, (0.19~1.19 wt.%) contents show variational compositions according to fergusonite types. The ${\sum}RE_2O_3$ of fergusonites are positive relation with $Y_2O_3$ and negative relaton with $ThO_2$ and $({\sum}{RE_2O_3}-{Y_2O_3})$. The $Nb_2O_3$ is sightly negative relation with $Ta_2O_3$. Back-scattered electron microscope images (BEI) of fergusonite show the mineral composition and textural feature is very complicated. The variation of Nb, Th and REE content of fergusonite and the modes of occurrence of mineral, suggests that REE may have been mobilized during the circulation of hydrothermal fluids related to contact metamorphism (metasomatism). The chemical variation of the fergusonites with occurrences and mineral association can be related to metasomatism of alkaline fluid was probably the dominant ore-forming process in Chungju district.
The gemological charateristics of Gwangdong tektites and Baikdusan obsidians were investigated, using microscopy, density and refractive index measurements, X-ray fluorescence spectrometry, Inductively Coupled Plasma Mass spectrometry, X-ray powder diffraction, and electron microprobe analysis. The Gwangdong tektites and Baikdusan obsidians are both black in colour and slightly trans-lucent with various shades of brown when cut into a few mm thick. Both the materials yield conchoidal fracture on broken surface. The tektites occur as tear-drop shapes, ranging from 4 to 10 cm long, and in spheres, from 3 to 5 cm in diameter. On the surface numerous shallow pits up to 3 mm in diameter are present. Mohs' hardness and specific gravity are 5 to 5.5 and 2.66, respectively. The tektites are singly refractive, with an refractive index of 1.51. Numerous spherical air bubbles are randomly scattered throughout the tektites, and silica-rich glass inclusions are occasionally seen. X-ray powder diffraction analysis verifies that they are non-crystalline. The Baikdusan obsidians show very similar properties to those of the Gwangdong tektites, especially in hardness, amorphous nature and fracture. Nevertheless, the Baikdusan obsidian can readily be distinguished from the Gwangdong tektites by refractive index ($1.49{\sim}1.50$), specific gravity (2.67 to 2.68), and inclusions (absence of bubbles and presence of sanidine and magnetite crystals).
Detailed knowledge on maskelynite, a glassy phase of plagioclase found in shocked meteorites and impact craters, is essential to understand a shock metamorphism. Here, we explore an inhomogeneous shock metamorphism in the lunar meteorite Mount DeWitt (DEW) 12007 with an aim to understand the formation mechanism of maskelynite. Most plagioclase grains in the DEW 12007 partially amorphized into maskelynite with a unidirectional orientation. Back-scattered electron (BSE) images of maskelynite show a remnant of planar deformation fracture possibly indicating that the maskelynite would be formed by solid-state transformation(i.e., diaplectic glass). Plagioclase with flow texture is also observed along the rim of maskelynite, which would be a result of recrystallization of melted plagioclase. Results of Raman experiments suggest that shock pressure for plagioclase and maskelynite in the DEW 12007 is approximately 5-32 GPa and 26-45 GPa, respectively. The difference in shock pressures between plagioclase and maskelynite can be originated from 1) external factors such as inhomogeneous shock pressure and/or 2) internal factors such as chemical composition and porosity of rock. Unfortunately, Raman spectroscopy has a limitation in revealing the detailed atomic structure of maskelynite such as development of six- or five-coordinated aluminum atom upon various shock pressure. Further studies using nuclear magnetic resonance spectroscopy are necessary to understand the formation mechanism of maskelynite under high pressure.
This study was undertaken to clarify the microstructure of the different IPS Empress ingots by etching and to observe the change of leucite crystal structure according to subsequent heat treatment and the crystal distribution according to sprue types(${\phi}2.8mm$, single sprue; ${\phi}1.8mm$, double sprue) by scanning electron microscopy. IPS Empress T1, O1 ingots used for staining technique, and Dentin(A2) ingots used for layering technique were selected for this study. To observe the microstructures of these ingots before pressing, the specimens were prepared in splinters($3{\times}3{\times}3mm$) taken from the original ingots. And to estimate crystal distribution and microstructural change by sprue type and subsequent heat treatment. the specimens($3{\times}3{\times}3mm$) were heat-pressed through the two types of sprues with different diameters and numbers, and all specimens were fired according to the recommended firing schedule. The observed surface was ground with waterproof papers($#800{\sim}#1800$) on the grind polisher and was cleaned ultrasonically. All specimen were etched with 0.5% hydrofluoric acid. After etching, the surface was treated by ion sputter coating for SEM observation at an accelerating voltage of 20kV. In all specimens, the central area of ground surface was observed because there was less difference in microstructure between the peripheral area and the central area. The results were as follows ; 1. In the microstructure according to the ingot type, there was a wide difference between the staining (T1,O1) and layering(Dentin A2) ingots, but there was not a considerable difference between the T1 ingot and the O1 ingot for staining technique. 2. In all specimens, the crystal dispersion of IPS Empress ceramic using double sprue was significantly more scattered than that of IPS Empress ceramic using single sprue. The degree of scattering was strongest in the Dentin(A2) specimen and weakest in the O1 ingot. 3. In the microstructural change according to the subsequent heat treatment, all of ingots had some microcracks in the inside of the leucite crystal and the glass matrix after pressing. The inner splinters of the leucite crystal became smaller, and more microcracks occurred in the glass matrix due to increasing heat treatment times. 4. The size of leucite crystals varied from $1{\mu}m\;to\;5{\mu}m$. The mean size of mature crystals was about $5{\mu}m$. The form of the crystal was similar to a circle when it was smaller and similar to an ellipse when it was larger.
The Kusandong Tuff, known as a representative key bed in the Cretaceous Gyeongsang Basin, is a crystal-rich tuff of pyroclastic flow and surge origin. It is 1-4 m thick and laterally extends for more than 200 km intercalated in the upper part of the Hayang Croup. Observations and analyses of the feldspar crystals in the tuff, using polarizing microscopes, EPMA, and BSE images, reveal that the plagioclase crystals in the tuff were completely albitized (>$97\%$ Ab) whereas those in the southernmost localities where the tuff is rich in fine ash matrix are unaltered or partly albitized. K-feldspars are partly albitized at all localities, irrespective of the matrix content of the tuff, Perthitic textures, chessboard twinning, albitization along micro-fractures and cleavages, and the relationship between matrix content and the degree of albitization suggest that feldspars in the Kusandong Tuff were albitized by Na-rich fluid after burial. Albitization is interpreted to start preferentially along micro-fractures and cleavages and be hampered in matrix-rich tuffs with a low permeability. Original composition of the plagioclases in the Kusandong Tuff is also interpreted to have ranged between oligoclase and andesine ($Ab_{62.5}-Ab_{83.3}$) before the albitization.
Light microscope, and scanning and transmission electron microscopy were used to investigate the fine structure of sperm of the Korea squirrel, Tamias sibiricus. The sperm head of T. sibiricus was paddle in shape. The total length of T. sibiricus sperm was 67.8 ${\mu}m$. The length of sperm head was 7.8 ${\mu}m$, and the tail (60.0 ${\mu}m$) was consisted of four major segments: the neck (1.0 ${\mu}m$), middle (8.0 ${\mu}m$), principal (48.5 ${\mu}m$) and end piece (2.5 ${\mu}m$), respectively. Especially, the length of the middle piece is short, and end piece was very shorter than those of other rodents. The post-nuclear cap was occupied about a fifth of nucleus. The equatorial segment is located between the post-nuclear cap segment and acrosomal cap on the nuclear surface. Nine segmented columns were surrounded by the mitochondria, and numbers of gyres of mitochondria were 26. One segmented column was consisted ten to twelve knobs, and each of segmented column in the neck region connected with the nine outer dense fiber in the middle piece. Numerous satellite-like fibers were scattered around the segmented columns. Nos. 1, 5 and 6 of the outer dense fibers in the middle piece were larger than the others. A fibrous sheath and longitudinal column of the principal piece were in evidence, but the fibrous sheath and longitudinal column was not seen at the end piece. In conclusion, the structural features of sperm head and tail may be useful information to patterns of sperm evolution and classification of species.
Morphology and microstructure of the suction trap in aquatic Utricularia japonica were examined using scanning electron microscopy. Branched stems bear numerous suction traps without root formation. The traps are derived axillary from the node, and their antennae and appendages extend in a peculiar fashion. The trap walls are thin, two-celled, parenchyma tissue and simple, small glands are scattered in both internal and external surface of the trap. The entrance of the trap is surrounded by one pair of dorsal antennae and ventral appendages, where the former guides the prey to the entrance. Trap door is situated below the entrance and numerous sessile and stalked capitate trichomes cover the entrance and even on the door surface. The capitate trichomes are secretory, but four trigger hairs formed on the central areas of the door are not. They are believed to function in activating and tripping the trap door. A specialized region of the threshold come in contact with the lower portion of the door upon closing. The secretory capitate trichomes near this region are responsible for producing and secreting a mucilage-like substance which composes the velum. Two-armed bifid glands are located in the interior side of the threshold, while four-armed quadrifid glands are considerably numerous occurring over the entire inner trap wall. Bifid and quadrifid glands develop semi-spherical basal cells that connect them to the inner wall surface. Antennae, trigger hairs, capitate trichomes, bifid and quadrifid glands are more important structures in the carnivory of U. japonica.
In order to investigate the histopathological, mechanism of Rompun-induced shock, the development of mammary carcinoma, the numerical changes and the morphological findings of the mast cells appeared in the carcinoma were microscopically observed in the rat treated with DMBA and each chemical of compound 48/80 and Rompun. Also mast cell degranulation induced by Rompun was observed with electron microscope. The results observed were summarized as follows: Tumor appeared in 100% of the animals. Tumors grew more rapidly to $10{\times}10mm$ in rats depleted of mast cells ($37.7{\pm}4.2$ days) than was observed in the control group ($42.5{\pm}4.7$ days) (p<0.005). The mean number of tumors per rat was $2.8{\pm}1.3$ in the compound 48/80- treated group in contrast to $3.4{\pm}1.3$ in the control group. No significant difference was apparent in the tumor induction time of Rompun treated group compared with the compound 48/80-treated group, but the tumor measuring at least $10{\times}10mm$ appeared more quickly in the Rompun treated group than in the control group (p<0.005). The numbers of mast cells in the control group were inclined to increase significantly according to the mammary tumor development (p<0.005). In contrast, the mast cells were fewer significantly in the compound 48/80-treated group and Rompun-treated group than in the control group (p<0.005). The numbers of mast cells in the compound 48/80-treated group and Rompun-treated group were inclined to reduce significantly according to the stages of the mammary carcinoma growth in contrast to the control group respectively. The ultrastructural morphologies of mast cells at 30 minutes after Rompun injection were appeared many normal granules in the cytoplasm, but many normal and degranulated granules were scattered along the cell membrane. And at 1 hour after Rompun injection mast cell granules were disappeared nearly or rarely seen. many long cytoplasmic projections were folded back to adhere to their own surface membrane. and mast cells resulted in a reduced size of these cells. Otherwise. compound 48/80 caused extensive degranulation of mast cells by disrupting cell membrane. but mast cell degranulation by Rompun was observed exocytosis of granules through a channel. From the above results. it is concluded that the Rompun may give rise to the dealth of animals as a shock caused by mast cell degranulation.
