This study was conducted to survey the current pest occurrence and grower's pest management practices, and to investigate biodiversity including birds, snakes, rodents, and amphibians in organic pear orchards. From the questionnaires and field survey of 22 organic orchards in 2010, the materials such as lime sulphur, plant extracts and their mixtures with microorganisms, and oils were preferred over insecticides. Scab, mealybugs, woollybugs and barkminers were the major pests for organic pear growers. Environment-friendly fungicides and insecticides were applied 9 to 10 and 5 to 6 times respectively. Incidence and severity of scab on Niitaka pear fruits in organic orchards was higher than in conventional ones. Whereas for mealybugs there was no significant difference between organic and conventional orchards. Pear fruits in organic orchards were significantly smaller than those of non-organic orchards. For the biodiversity of amphibians, snakes, pheasants, and rodents, there was no significant difference between organic and conventional orchards, but Rufous turtle doves were monitored more in organic areas than in conventional ones.

The fungus Venturia nashicola is the causal agent of scab on Asian pears. For the rapid and reliable identification as well as sensitive detection of V. nashicola, a PCR-based technique was developed. DNA fingerprints of three closely related species, V. nashicola, V. pirina, and V. inaequalis, were obtained by random amplified polymorphic DNA (RAPD) analysis. Two RAPD markers specific to V. nashicola were identified by PCR, after which two pairs of sequence characterized amplified region (SCAR) primers were designed from the nucleotide sequences of the markers. The SCAR primer pairs, designated as D12F/D12R and E11F/E11R, amplified 535-bp and 525-bp DNA fragments, respectively, only from genomic DNA of V. nashicola. The specificity of the primer sets was tested on strains representing three species of Venturia and 20 fungal plant pathogens. The nested PCR primer pair specific to V. nashicola was developed based on the sequence of the species-specific 525-bp DNA fragment amplified by primer set E11F/E11R. The internal primer pair Na11F/Na11R amplified a 235-bp fragment from V. nashicola, but not from any other fungal species tested. The nested PCR assay was sensitive enough to detect the specific fragment in 50 fg of V. nashicola DNA.
In 1963, a severe epidemic of cereal scab caused by Gibberella zeae occurred in southern Korea and to a less extent in central and northern Korea. In some areas losses were $80{\sim}100$ percent. The epiphytotic was due to heavy rainfall during the heading and flowering season which provided a favorable environment for this severe epidemic. Yield losses resulted a great social problem because of the resultant food and feed grain shortage, lose of seeds for planting the following crops and mycotoxicoses to man and animals. In the same year, a nationwide research committee was organized including plant pathologists, microbiologists, agronomists and biochemists under the juristiction of the Ministry of Agriculture and Forestry. The committee initiated research on etiology, epidemiology, and control of the disease and on the toxic effect of infected grains to man and animals. The present paper will review some research carried out in Korea on cereal scab with special reference to epidemiology and mycotoxicoses to animals and man. In addition, the present status of research in Korea on aflatoxins in foods and toxic moldy rice will be briefly reviewed.
In the course of our screening for biocontrol agent (BCA) against Streptomyces scabiei and S. turgidiscabies causing potato scab using 5,000 actinomtcete isolates, 9 antagonistic strains were selected as BCA candidates through in vitro and in vivo assay. An antagonistic strain, A020645 was highly resistant to some pesticides and antibiotics such as dazomet and mancozeb and showed high control value in vivo. Two bioactive compounds (compound A, B) were purified by anion exchange chromatography, solid phase (ODS) extraction, TLC and reverse phase HPLC. Their chemical structures are now thought to be nucleoside derivative as determined by $^1H-NMR$ data analysis. Their full chemical structures would be elucidated through $^{13}C-NMR$, HMQC and HMBC analyses. Further studies will be focused on fitness in soil and formulation of the BCA candidates.
Potato common scab disease is caused by pathogens belonging to Streptomyces spp. and results in a serious yield loss worldwide. Despite decades of research aimed at disease management, a definitive control method remains undiscovered. This study aims to explore the correlation between the C/N ratio and urea application with potato common scab pathogen dynamics and disease severity. We applied sawdust with a high C/N ratio and urea into the soil prior to potato cropping, both in pot and field experiments. Disease severity assessments and quantification of the TxtB gene were conducted at the harvest stage. Furthermore, culture experiments were performed to assess the direct impact of urea on the pathogen. Our findings revealed that higher disease severity was correlated with a high C/N ratio application and pathogenic gene quantity. Urea exhibited a direct influence on S. scabiei activity, reducing the disease severity in pot experiments. However, the effects of urea application on disease suppression in the conductive field were inconclusive. Although the results of urea application experiments displayed inconsistencies between pot and field trials, urea worked as the control to suppress S. scabiei activity. Further investigations are needed under various field conditions to confirm these findings.
This experiment was conducted to investigate the changes of rate of infected common scab and yield characteristics of fall potato(Solanum tuberosum L.) where green manual crops had been previously cultivated, crushed and tilled and P\ulcornerE transparent vinyl film had been mulched and tunneled to solar heating of soil from May 21. 1998 to July 28. 1998. The total yields of green manual crops which had been previousely cultivated were as followed order : pioneer 855F(64.3MT/ha), soybean(25.0MT/ha), red clover and orchardgrass. The average below-ground temperature at the depths of 5, 10 and 20cm were 54, 45 and 44$^{\circ}C$ during the mulching period, respectively The rate of infected area per potato tuber of common scab decreased by solar heating the soils with mulching after the soybean and red clover cultivation as with previous croppings. Plant height, SPAD(soil plant analysis development) reading, fresh weight of stems, and fall potato's tuber yields tended to increase by the cultivation of green manual crops and mulching of P\ulcornerE film. Fall potato's tuber yields were remarkably affected in the plot of soybean, red clover and pioneer 855F+cultivation of fall potato. T-N, K and Ca contents of fall potato(stem and tuber) also tended to increase by the cultivation of green manual crops and mulching of P\ulcornerE film. T-N, K and Ca contents of soil tended to increase after the cultivation of green manual crops and mulching of P\ulcornerE film and then reduced to contents of soil before green manual planting after potato harvesting.
In Korea, EBI fungicides which are highly effective for control of pear scab and rust but of high risk for development of resistance have been frequently sprayed by majority of pear growers. To detect any possible resistance or reduced sensitivity in the field strains of scab fungus to five EBIs, difenoconazole, fluquinconazole, flusilazole, fenarimole and hexaconazole, sensitivity tests were conducted with fungal specimens collected in Ulsan and Naju where scab usually occurs and EBIs have been intensively sprayed for many years. As the strains for which $EC_{50}$ values of the EBIs were largely shifted from those of base-line were occasionally found, the resistant or less sensitive strains were supposed to be distributed. In the activity test for the EBIs by artificial inoculation, in which EBI-treated pear leaves on the potted seedlings were inoculated with fungal spores collected in the two regions, development of resistances to EBIs were confirmed. Since the fungal spores collected at 4 and 2 orchards in Naju and Ulsan, respectively, produced much higher disease incidence on the leaves treated with hexaconazole than those on the untreated control, those fungal specimens were determined as resistant to hexaconazole. Similar results were also obtained with two specimens from Ulsan for flusilazole.
