  • Population ecological parameters and stock biomass of the purplish washington clam, Saxidomus purpuratus (Sowerby), in the southern coastal areas of Korea were determined based on the fishery data from the Divers Fisheries Cooperative and other available biological data. Instantaneous coefficient of total mortality (Z) of purplish Washington clam was estimated to be 0.7479/year. The estimated instantaneous coefficient of natural mortality (M) was 0.2012/year. From the values of Z and M, the instantaneous coefficient of fishing mortality coefficient (F) for the recent years was calculated to be 0.4578/year. The age of purplish Washington clam at its first capture ($t_c$) was 2.7506 year. Yield-per-recruit and spawning biomass-per-recruit were estimated under harvest strategies that bases on $F_{max}$, $F_{0.1}$, $F_{35%}$ and $F_{40%}$ were shown as 81.60 g, 61.68 g, 115.07 g and 131.51 g, respectively. The acceptable biological catch (ABC) was estimated to be about 1,404 metric tons.

  • Reproductive biology of the purplish Washington clam, Saxidomus purpuratus was investigated based on the samples captured in Jinhae Bay, the East China Sea from January to December 2002. The gonad index (GI) began to increase in January, reached the maximum value in March. The reproductive cycle of this species can be divided into five successive stages: the early active stage (from November to January), the late active stage (from December to February), the ripe stage (from February to May and October), the spawned stage (from May to December), and inactive stage (from November to December). The spawning period was from April to December, and the main spawning occurred between June and August. The shell length at 50% group maturity was estimated to be 71.85 mm. The Sex ratio of this species was not significantly different a 1:1 sex ratio (P > 0.05).

  • Partial sequences of the mitochondrial cytochrome oxidase subunit Ⅰ(mtCOI) gene of veneroid clams were obtained to eluciadate molecular phylogeny. A total of nine veneroids and one corbiculid were collected from southern and western sea of Korea. The mtCOI sequences of the clams obtained from the present study and three veneroids sequences from GenBank were analyzed by maximum parsimony and neighbor-joining methods. The subfamilies Samarangiinae, Dorsiniinae, Cyclinae, Meretricinae and Chioninae were monophyletic, but Pitarinae was paraphyletic. One transitions and two transversions among two samples of Ruditapes philippinarum and a R. variegata were observed.

  • Carbonic anhydrase (CA), which is involved in shell formation processes in bivalves, is one of the major biocatalysts for carbon capture and storage. In this study we investigated CA activity in the total hemocytic proteins of five bivalves. The highest CA activity was observed in Scapharca broughtonii, which had more than twice the activity found in Crassostrea gigas. No CA activity was observed among the total hemocytic proteins of Pinctada fucata and Saxidomus purpuratus. The results suggest that marine invertebrates may provide a better source of CA, as an alternative to mammalian sources.

  • To facilitate the effective use of butter clam shell as a natural calcium resource, we determined the optimal conditions for calcium lactate (BCCL) preparation with high solubility using response surface methodology (RSM). The polynomial models developed by RSM for pH, solubility and yield were highly effective in describing the relationships between factors (P<0.05). Increased molar ratios of calcined powder (BCCP) from butter clam shell led to reduced solubility, yield, color values and overall quality. The critical values of multiple response optimization to independent variables were 1.75 M and 0.94 M for lactic acid and BCCP, respectively. The actual values (pH 7.23, 97.42% for solubility and 423.22% for yield) under optimization conditions were similar to the predicted values. White indices of BCCLs were in the range of 86.70–90.86. Therefore, organic acid treatment improved color value. The buffering capacity of BCCLs was strong, at pH 2.82 to 3.80, upon the addition of less than 2 mL of 1 N HCl. The calcium content and solubility of BCCLs were 6.2–16.7 g/100 g and 93.6-98.5%, respectively. Fourier transform analysis of infrared spectroscopy data identified BCCL as calcium lactate pentahydrate, and the analysis of microstructure by field emission scanning electron microscopy revealed an irregular form.

  • For effective utilization of butter clam shell as a natural calcium resource, the optimal conditions for preparation of calcium acetate (BCCA) with high solubility were determined using response surface methodology (RSM). The polynomial models developed by RSM for pH, solubility, and yield were highly effective in describing the relationships between factors (P<0.05). Increased molar ratio of calcined powder (BCCP) from butter clam shell led to reduction of solubility, yield, color values, and overall quality. Critical values of multiple response optimization to independent variables were 2.70 M and 1.05 M for acetic acid and BCCP, respectively. The actual values (pH 7.04, 93.0% for solubility and 267.5% for yield) under optimization conditions were similar to predicted values. White indices of BCCAs were in the range of 89.7~93.3. Therefore, color value was improved by calcination and organic acid treatment. Buffering capacity of BCCAs was strong at pH 4.88 to 4.92 upon addition of ~2 mL of 1 N HCl. Calcium content and solubility of BCCAs were 20.7~22.8 g/100 g and 97.2~99.6%, respectively. The patterns of fourier transform infrared spectrometer and X-ray diffractometer analyses from BCCA were identified as calcium acetate monohydrate, and microstructure by field emission scanning electron microscope showed an irregular form.

  • Amino acid composition and relationship of the commercially valuable five Korean Veneridae, Protothaca jedoensis, Ruditapes philippinarum, Meretrix lusoria, Saxidomus purpuratus and Cyclina sinensis were compared. The major amino acids of five species in Veveridae clams were ureanine, taurine, proline, glycine, alanine and arginine. A similarity of amino acid between C. sinensis and M. lusoria was highest (0.94) and lowest (0.52) for between C. sinensis and P. jedoensis, respectively.

  • A survey was performed to know the recent infection status of digenetic trematode metacercariae in clams and oysters from 4 sites in western coastal regions of the Republic of Korea (=Korea). Four species of clams (Mactra veneriformis, Ruditapes philippinarum, Cyclina sinensis, and Saxidomus purpuratus) were collected from Taean-gun, Chungcheongnam-do (Province), Buan-gun (County) and Gochang-gun, Jeollabuk-do, and oysters, Crassostrea gigas, from Shinan-gun, Jeollanam-do were transferred to our laboratory on ice and examined by the artificial digestion method. The metacercariae of Himasthla alincia were detected in 3 species of clams, M. veneriformis, R. philippinarum, and C. sinensis from the 3 surveyed areas. The positive rate and the mean density per clam infected were 98.9% (30.8 metacercariae) in M. veneriformis, 60.0% (5.0) in R. philippinarum, and 96.0% (28.4) in C. sinensis. The positive rate (mean density) of Acanthoparyphium tyosenense metacercariae in M. veneriformis was 50.0% (2.1) from Taean-gun and 70.0% (2.8) from Gochang-gun. The metacercariae of Parvatrema spp. were detected in M. veneriformis and R. philippinarum from Taean-gun and Gochang-gun; the positive rate (mean density) was 63.3% (4,123) and 50.0% (19) in M. veneriformis, and 6.7% (126) and 100% (238) in R. philippinarum from the 2 regions, respectively. The metacercariae of Gymnophalloides seoi were detected in all 30 oysters from Shinan-gun, and their average density per oyster was 646. From the above results, it has been confirmed that more than 3 species of metacercariae are prevalent in clams from the western coastal regions, and G. seoi metacercariae are still prevalent in oysters from Shinan-gun, Jeollanam-do, Korea.
