Anti-Tumor Effect of Cyclophosphamide, TUbercin-3 , and Picibanil on Sarcoma-180 Bearing Mice (Sarcoma-180 유발 생쥐에 대한 Cyclophosphamide, Picibanil 및 Tubercin-3의 투여효과)

  • This study was carried out to detemrine the efficacy of combined treatment of cyclophosphamide with tubericin-3 and or picibanil. One hundred sixty sarcoma-180 bearing mice were divided eight groups. Each group received saline, tubercin-3, picibanil , and cyclophosphamide along and/or received cyclophosphamide with tubercin-3 , with picibanil or with both tubercin-3 and picinanil, respectively.Average surviving time of each group of animals was as follows ; control was 10.9days, tubercin-3 was 15.1 days. picibanil was 12.6 days, and cylophosphamide was 17.9 days, In combined therapyy that cylophosphamide injected with tubercin-3 , the surviving time was 26.8 days an din the case of other therapy that cyclophosphamide injected with tubercin-3, the surviving time was 26.8 days an din the case of other therapy that cyclophosphamide injected with picibanil, the surviving time was 21.9 day and cyclophosphamide treated with both turbercin03 and picibanil, the surviving time was found to be 18.2 days, conclusively , the therapeutic potentiation seemed to be extended when combined tretment of the chemotherapeutics cyclophosphamide with either one of immunotherapeutics tubericin-3 or picibanil was tried, Combinatin of tubercin-3 and picibanil showed to be atagonistic each other. Yield of ascites fluid were determined 7 days after injectino of sarcoma-180 ascites tumor cells. Adminitration of cyclophosphamide, tubercin-3 , and picibanil alone and their various combinations reduced the yield of ascites fluid except for picibanil group.

Suppressed CD31 Expression in Sarcoma-180 Tumors after Injection with Toxoplasma gondii Lysate Antigen in BALB/c Mice

  • The anti-tumorigenic effects of Toxoplasma gondii (RH) antigens were studied in a murine sarcoma-180 tumor model. To determine the anti-tumor effects, the reduction in tumor size and expression of CD31 (an angiogenesis marker in the tumor tissue) were examined after injection of BALB/c mice with T. gondii lysate antigen (TLA) or formalin-fixed, proliferation-inhibited, T. gondii tachyzoites. Tumors were successfully produced by an intradermal injection of sarcoma-180 cells with plain Matrigel in the mid-backs of mice. After injection with TLA or formalin-fixed T. gondii tachyzoites, the increase in tumor size and weight nearly stopped while tumor growth continued in control mice that were injected with PBS. CD31 expression in TLA-treated or formalin-fixed T. gondii-injected mice was lower than the control mice. Accordingly, the present study shows that the treatment of mice with formalin-fixed T. gondii or TLA in the murine sarcoma-180 tumor model results in a decrease of both tumor size and CD31 expression.

Antitumor Activity of Seaweeds toward Sarcoma-180 (해조류의 Sarcoma-180에 대한 항암효과)

  • Protein-polysaccharides(PP) extracted from four species of gulfweed, fusiforme, sea-mustard and sea-tangle were examined for antitumor activity toward sarcoma-180 implanted in mice, ICR. Polysaccharides in protein-polysaccharides extracted from gulfweed, fusiforme, sea-mustard and sea-tangle were 61.48, 55.61, 34.06 and 30.28%, respectively. Monosaccharides of four seaweeds consisted of glucose, galactose, mannose, fructose and xylose, and major amino acids consisted of glutamic acid, aspartic acid, cystein, valine and glycine. Antitumor activity of the protein-polysaccharides extracted from sea-mustard showed the highest inhibition ratio of 69.76% when PP was injected to the mice at the dose of 100mg/Kg/day. PP of sea-tangle showed the maximum survival ratio of 25.22% when injected at the dose of 100mg/Kg/day.

  • Sarcodon aspratus (Thelephoraceae), a native mushroom distributed in Korea and Japan has been widely used as a traditional food and folk medicines. Two proteoglycan polysaccharides, named SAP-A and SAP-B were isolated from the fruitbody of S. aspratus. Their molecular weight were determined as 6184.9 D, and 25749.2 D, respectively, by maldi-tof ms. The composition of sugar and amino acid of these compounds was also determined. The SAP-A showed potent antitumor activity. The $ID_{50}$ value of SAP-A was 8.2 mg/kg/day for ICR mice transplanted with solid sarcoma-180, and the elongation of lifespan effect was 167.4% for ICR mice transplanted with ascitic sarcoma-180. Moreover, the elongation life-span effect of SAP-A increased dose-dependently.

Effect of Korean Red Ginseng Polysaccharide on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (고려홍삼 다당체 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향)

  • This study was devised to observe the inhibitory effects of ethanol treatment precipitate (crude acidic polysaccharide) from Korean red ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been inoculated with sarcoma-180. The yields of crude acidic polysaccharide from Korean red ginseng was 63.5%. In vitro, at the concentration of 500rg /ml, the inhibition rate of lipolysis by the crude acidic polysaccharide of Korean red ginseng was 38.8% and the total inhibitory activity per gram of ginseng material was 4,928 nit. In vivo, the red ginseng polysaccharide(40mg/ml in saline soon.) 16ul/g of body weight was injected to the sarcoma-180 bearing mice once In 3 days until death. The effects against the extension of life span was little but body weight gain of sarcoma-180 bearing mice decreased significantly by administration of Korean red ginseng polysaccharide compared to those of the control group.

Antitumor Activities of the Proteoglycans from the Mycelium of Ganoderma lucidum IY009 (영지버섯 균사체 (Ganoderma lucidum IY009)로부터 추출한 단백다당체의 항암촬성)

  • In this study, the antitumor activities were investigated using $\beta$-lmmunan, a proteoglycan obtained from cultured mycelia of the IY009 strain of Ganoderma lucidum belonging to basidiomycetes. The result showed the significant effect of cytotoxicity test against murine sarcoma 180 and murine lymphocytic leukemia L1210 using immunized macrophage cultures by $\beta$-lmmunan. When intraperitoneally injected at 40 mg/kg/day daily for 10 days, $\beta$-lmmunan inhibited the growth of sarcoma 180 solid tumor in ICR mice by 88.8% (p<0.05). It was also observed that $\beta$-lmmunan increased life span by 85.2% (p<0.01) after treatment of 100 mg/kg/day in BDF1 mice bearing lymphocytic leukemia L1210. And combination therapy with cisplatin (dosage: 4 mg/kg) increased life span by 140.4% (p<0.05) after treatment of 100 mg/kg/day daily in BDF1 mice bearing lymphocytic leukemia L1210.

  • To find antitumor components with low toxicity in natural products of Korean Basidiomycete, the carpophores of Flammulina velutipes (Fr.) Sing. were extracted with hot water for eight hours. The extract was purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained as pale brownish amorphous powder after it was freeze-dried. The fraction was examined for antitumor activity against sarcoma 180 implanted subcutaneously in the left groins of I.C.R. mice. The inhibition ratio of this fraction of against the tumor was 62.3% at the dose of 10 mg/kg/day for the period of ten days. The tumors in three of the ten treated mice were completely regressed.

  • This study was designed to observe the anticancer activity of propolis on human rectal (HRT-18) and human colon (HCT-48) cancer cell lines in vitro, and on sarcoma-180 cells in vitro. The proliferation of HRT-18 and HCT-48 cancer cell lines was potently inhibited in proportion to the concentration of propolis. The survival time of the mice inoculated with sarcoma-180 cells was increased modestly by the administration of propolis compared to the control. Those observations suggest that propolis has anticancer effects against some of the cancer cell lines in vitro and in in vitro.

Effects of Polyacetylenes from Panax ginseng on Some Microsomal and Mitochondrial Enzymes

  • Effects of panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on some enzyme activities were determined. Activities of ATPase, membrane-bound enzyme from Sarcoma 180 and rat liver were slightly inhibited by panaxydol. Activities of 5'-nucleotidase, membrane-bound enzyme and succinate cytochrome c reductase in mitochonidria from sarcoma 180 and rat livers were significantly inhibited in a dose-dependent manner by panaxynol. The inhibitory effects of panaxydol and panaxynol on succinate cytochrome c reductase activities were more potent than those on 5'-nucleotidase activities and panaxynol was found to be a very potent inhibitor of succinate cytochrome c reductase. Activities of glucose-6-phosphatase in endoplasmic reticulum from Sarcoma 180 and rat livers were not affected by all three polyacetylenes. These results suggested that the inhibitory effects of panaxydol and panaxynol on enzyme activities might contribute to their biological activities.

  • Antitumor effect of LC43, a protein-bound ploysaccharide (M.W. 43 kDa) that was purified from intergeneric protoplast fusant of Lentinus edodes and Coriolus versicolor, was elucidated against mouse sarcoma 180 cell in vitro and in vivo. By injecting LC43 into ICR mice bearing solid or ascitic sarcoma 180, tumor regression and survival rates were investigated. To examine the effects of LC43 on immunopotentiation activity. immunoorgan weight, B cell differentiation, T cell activity and macrophage activation were determined. LC43 showed antitumor effects against both solid tumor and ascitic tumor of sarcoma 180. It did not change significantly the immunoorgan weight but potentiated immune responses such as B cell differentiation and the release of superoxide anion from macrophages. These results suggest that the protein-bound polysaccharide of LC43 exhibited antitumor activities through the activation of immune-related cells and acted as an immunmodulator.
