Development of High Intensity Focused Ultrasound (HIFU) Mediated AuNP-liposomal Nanomedicine and Evaluation with PET Imaging

  • Liposomes as drug delivery system have proved useful carrier for various disease, including cancer. In addition, perfluorocarbon cored microbubbles are utilized in conjunction with high-intensity focused-ultrasound (HIFU) to enable simultaneous diagnosis and treatment. However, microbubbles generally exhibit lower drug loading efficiency, so the need for the development of a novel liposome-based drug delivery material that can efficiently load and deliver drugs to targeted areas via HIFU. This study aims to develop a liposome-based drug delivery material by introducing a substance that can burst liposomes using ultrasound energy and confirm the ability to target tumors using PET imaging. Liposomes (Lipo-DOX, Lipo-DOX-Au, Lipo-DOX-Au-RGD) were synthesized with gold nanoparticles using an avidin-biotin bond, and doxorubicin was mounted inside by pH gradient method. The size distribution was measured by DLS, and encapsulation efficiency of doxorubicin was analyzed by UV-vis spectrometer. The target specificity and cytotoxicity of liposomes were assessed in vitro by glioblastoma U87mg cells to HIFU treatment and analyzed using CCK-8 assay, and fluorescence microscopy at 6-hour intervals for up to 24 hours. For the in vivo study, U87mg model mouse were injected intravenously with 1.48 MBq of 64Cu-labeled Lipo-DOX-Au and Lipo-DOX-Au-RGD, and PET images were taken at 0, 2, 4, 8, and 24 hours. As a result, the size of liposomes was 108.3 ± 5.0 nm at Lipo-DOX-Au and 94.1 ± 12.2 nm at Lipo-DOX-Au-RGD, and it was observed that doxorubicin was mounted inside the liposome up to 52%. After 6 hours of HIFU treatment, the viability of U87mg cells treated with Lipo-DOX-Au decreased by around 20% compared to Lipo-DOX, and Lipo-DOX-Au-RGD had a higher uptake rate than Lipo-DOX. In vivo study using PET images, it was confirmed that 64Cu-Lipo-DOX-Au-RGD was taken up into the tumor immediately after injection and maintained for up to 4 hours. In this study, drugs released from liposomes-gold nanoparticles via ultrasound and RGD targeting were confirmed by non-invasive imaging. In cell-level experiments, HIFU treatment of gold nanoparticle-coupled liposomes significantly decreased tumor survival, while RGD-liposomes exhibited high tumor targeting and rapid release in vivo imaging. It is expected that the combination of these models with ultrasound is served as an effective drug delivery material with therapeutic outcomes.

Developing Content System for Home Economics Curriculum in Connection with Education for Sustainable Development(ESD): Focusing on the 'Life Environment and Sustainable Choice' Area (지속가능발전교육(ESD)을 연계한 가정과 교육과정의 내용체계 개발: '생활환경과 지속가능한 선택' 영역)

  • The purpose of this study is to develop a content system for the home economics curriculum that integrates Education for Sustainable Development(ESD) and provides basic material for ESD implementation in schools. In view of this, the content elements of the revised home economics curriculum for 2022 were analyzed, and a content system for the home economics curriculum, linked to ESD, was proposed based on the implications drawn from the analysis. The results are as follows. First, the three components of competencies, namely knowledge, values, and skills, were organized equally as a whole. However, the association between the content elements and key competencies in sustainability was found to be insufficient. Consequently, it is proposed that key competencies in sustainability should be cultivated integrally. Second, no content element was identified that can promote social participation. Therefore, it is proposed that solutions should be dealt with at the level of social participation. Third, the connection with Sustainable Development Goals(SDGs) was observed in only six of the 28 content elements. Wherever relevant, it is proposed to incorporate key issues related to SDGs. Fourth, the analysis confirmed that only the environmental dimension of sustainable development was considered. Therefore, it is proposed to pursue coexistence based on temporal and spatial relationship and consider the dimensions of environment, society, and economy in an integrated manner.

Cultural Diversity and Repression in Communities: A Study on China and Latin America (공동체에서의 문화 다양성과 억압 -중국과 라틴아메리카를 중심으로-)

  • In this study, discussions of the suppression of cultural diversity in communities was conducted. First, based on the studies conducted so far and recent changes, the oppression that exists between the Chinese government and ethnic minorities was considered. The visible suppression mentioned was the expansion of Han Chinese Mandarin language education, sanctions on minority languages, and the expansion of higher education at the exclusion of minority identities. In terms of 'invisible' oppression, urbanization, urban development with modernization at the forefront, and the use of officials from minority ethnic groups educated by the central government were items that were discussed. Next, the case of Latin America was examined. In particular, attention was paid to the theory of resistance against Europeans and European culture. Based off of the worries and experiences of Latin American intellectuals who have underwent oppression as individuals from culturally diverse backgrounds, a mature theory was formulated that could be used to defend Chinese minorities in the future. There is a specificity to the problem of Chinese minority communities. However, from a large perspective, experience and self-critical exploration in Latin America serve as an opportunity to expand the specificity of Chinese minority communities. Their situation resembles previous situations in Latin America when native cultures were being culturally eroded by Europe. Thus, as Latin American scholars argue, a shift in perception is necessary. In addition to this, in the text, it is likewise necessary to reflect on diversity, freedom, and mutualistic respect. There are proposals advocating for the realization of Heyibutong (和而不同 harmony but not through sameness) based on the situation in China. In the process of this consideration, much thought was given about what the observed communities are like and what a hypothetically desirable community would be like. This extends not only to Chinese minority communities and native residents of Latin America, but also to Asians in the United States and foreigners in Korea. Through this, it is hoped that desirable communities characterized by cultural diversity can be skillfully pursued.

A Study on the Classification of Rural Tourism Resources through a Card Sorting Test -Focused on Rural Amenity Resources Database- (카드분류법을 통한 농촌관광자원 유형 분류 -농촌어메니티자원 DB를 중심으로-)

  • As the interest in rural tourism has been increasing since the late 1990s, the research associated with rural tourism has increased, including research on the classification of rural tourism resources. The research classifying these resources has proved useful to many other studies. Although such studies have been conducted for a long time, they have expressed only experts' perspectives and been supported by statistics, without reflecting on users' opinions. Given this background, this study aims to classify rural tourism resources by focusing on the rural activities for tourists who use those tourism resources. To achieve this, each study participant proceeded to collect tourism resources by using a rural amenity resources database, and a card sorting test was conducted. Thirty-two people who had previously gone sightseeing in the rural areas were chosen as participants in the card sorting test. After the card sorting test was complete, the results were reviewed by experts. These results yielded six categories: doing nature activities, eating and cooking local dishes, putting up (overnight stays), going sightseeing/appreciating the landscape, enjoying leisure activities, and doing artistic activities. In the doing nature activities category, there were four subcategories: experiencing local resources, experiencing nature, experiencing tradition, and harvesting. This study was conducted to improve the satisfaction and understanding of the tourists who visit rural areas. Thus, the classification of rural tourism resources developed by this study will be widely used to establish the framework or contents of websites, applications, and so on, for promoting rural tourism resources and local resources.

Project of Improving Good Agriculture Practice and Income by Intergrated Agricultural Farming (미얀마 우수농산물 재배기술 전수사업)

  • The objectives of the project are to increase farmers' income through GAP and to reduce the loss of agricultural produce, for which the Korean partner takes a role of transferring needed technologies to the project site. To accomplish the project plan, it is set to implement the project with six components: construction of buildings, installation of agricultural facilities, establishment of demonstration farms, dispatching experts, conducting training program in Korea and provision of equipments. The Project Management Committee and the Project Implementation Team are consisted of Korean experts and senior officials from Department of Agriculture, Myanmar that managed the project systematically to ensure the success of the project. The process of the project are; the ceremony of laying the foundation and commencing the construction of training center in April, 2012. The Ribbon Cutting Ceremony for the completion of GAP Training Center was successfully held under PMC (MOAI, GAPI/ARDC) arrangement in SAl, Naypyitaw on June 17, 2012. The Chairman of GAPI, Dr. Sang Mu Lee, Director General U Kyaw Win of DOA, officials and staff members from Korea and Myanmar, teachers and students from SAl attended the ceremony. The team carried out an inspection and fixing donors' plates on donated project machineries, agro-equipments, vehicles, computers and printer, furniture, tools and so forth. Demonstration farm for paddy rice, fruits and vegetables was laid out in April, 2012. Twenty nine Korean rice varieties and many Korean vegetable varieties were introduced into GAP Project farm to check the suitability of the varieties under Myanmar growing conditions. Paddy was cultivated three times in DAR and twice in SAl. In June 2012, vinyl houses were started to be constructed for raising seedlings and finished in December 2012. Fruit orchard for mango, longan and dragon fruit was established in June, 2012. Vegetables were grown until successful harvest and the harvested produce was used for panel testing and distribution in January 2013. Machineries for postharvest handling systems were imported in November 2012. Setting the washing line for vegetables were finished and the system as run for testing in June 2013. New water tanks, pine lines, pump house and electricity were set up in October 2013.

Fish Community Structure and Biodiversity of the Korean Peninsula Estuaries (한반도 하구의 어류군집 구조 및 다양성)

  • Fish assemblage of total 325 of Korean peninsula estuaries were surveyed to analyze the characteristics of community structure and diversity by sea areas for three years from 2016 to 2018. The scale (stream width) of Korean estuaries were various (14~3,356 m), and 68.9% of all estuaries showed salinity of less than 2 psu. Total 149 species classified into 52 families of fish were identified, and the dominant and sub-dominant species were Tribolodon hakonensis (relative abundance, RA, 12.5%) and Mugil cephalus (RA, 9.5%), respectively. The estuary of the Korean Peninsula had different physical and chemical habitat environments depending on the sea area, and accordingly, fish community structure also showed statistically significant differences (PERMANOVA, Pseudo-F=26.69, P=0.001). In addition, the NMDS (nonmetric multidimensional scaling) results showed the patterns that indicating fish community difference by sea areas, even though low community similarity within sea area (SIMPER, 21.79~26.39%). The estuaries of east sea areas were distinguished from the others in the aspects of which, the higher importance of migratory fishes and endangered species, and that of brackish species were characterized at south sea estuaries. However, the estuaries of west sea showed higher importance of species that have a relation with freshwater (primary freshwater species, exotic species), which is the result that associating with the lower salinity of west sea estuaries because of the high ratio of closed estuaries(78.2%). The SIMPER analysis, scoring the contribution rates of species to community similarity, also showed results corresponding to the tendency of different fish community structures according to each sea area. So far, In Korea, most studies on fish communities in estuaries have been conducted in a single estuary unit, which made it difficult to understand the characteristics of estuaries at the national level, which are prerequisite for policy establishment. In present study, we are providing fish community structure characteristics of Korean estuaries in a national scale, including diversity index, habitat salinity ranges of major species, distribution of migratory species. We are expecting that our results could be utilized as baseline information for establishing management policies or further study of Korean estuaries.

Benthic Macroinvertebrates Inhabiting Estuaries in Sea Area and Relationship with Major Drivers of Change in Estuaries (해역별 하구에 서식하는 저서성 대형무척추동물 현황과 하구 서식지 주요 변화 동인과의 관계)

  • This study analyzed the relationship between the community structure of benthic macroinvertebrates and habitat changes in open estuaries among the sites included in the national estuary monitoring program. The estuary survey was conducted under the "Guidelines for Investigation and Evaluation of Biometric Networks" and classified by sea area, 80 places in the East Sea, 102 places in the South Sea, and 19 places in the West Sea were investigated. In a total of 201 open estuaries, benthic macroinvertebrates were identified with 4 phyla, 9 classes, 41 orders, 139 families, 269 species and 196 species in the East Sea, 182 species in the South Sea, and 90 species in the West Sea. The highest population densities were Insecta in the East Sea, the Malacostraca in the South Sea, and the Annelida in the West Sea. Through SIMPER analysis, species contributing to the similarity of benthic macroinvertebrates communities in each sea area were identified. Some species greatly influenced the similarity of clusters. The benthic community in the East Sea was affected by the salinity, so the contribution rate of freshwater species was high. On the other hand, the benthic communities of the South and West Seas showed species compositions are influenced by the substrate composition. As results, the benthic macroinvertebrate community in Korean estuaries was impacted by salinity and substrate simultaneously, and the close relationship with geographical distance was not observed. The result of this study is expected to be used to respond to environmental changes by identifying and predicting changes in the diversity and distribution of benthic macroinvertebrates in Korea estuaries.

Brittle rock property and damage index assessment for predicting brittle failure in underground opening (지하공동의 취성파괴 예측을 위한 암석물성 및 손상지수 평가)

  • Laboratory tests are performed in this paper to investigate the brittle failure characteristics of over-stressed rocks taken in deep depth. Also, numerical simulation performed using that the so-called CWFS(Cohesion Weakening Frictional Strengthening) model is known to predict brittle failure phenomenon reasonably well. The most typical rock types of Korean peninsula - granite and gneiss - were used for testing. Results of uniaxial compression tests showed that the crack initiation stress was about 41 % to 42% of the uniaxial compressive strength regardless of rock types, where as, the crack damage stress of granite was about 75%, and that of gneiss was about 97%. Through the damage-controlled test, strength parameters of each rock were obtained as a function of damage degree. After the peak, the crack damage stress and the maximum stress were decreased, The cohesion was decreased and the friction angle was increased with increase of rock damage. Before reaching the peak, the elastic modulus was slightly increased, while decreased after the peak. Poisson's ratio was increased as the damage of rock proceeds. Comparison of uniaxial compression tests and damage-controlled tests shows the crack initiation stress estimated from the damage-controlled test fluctuated within the range of crack initiation stress obtained from the uniaxial compression test; the crack damage stress was less than that estimated from the uniaxial compression test. In order to predict the critical depth that brittle failure occurs, numerical simulations using the CWFS model were performed for an example site. Material parameters obtained from the laboratory tests mentioned above were used for CWFS simulation. Comparison between the critical depth predicted from the numerical simulation using the CWFS model and that predicted by using the damage index proposed by Martin et al.(l999), showed that critical depth cannot be reasonably predicted by the currently used damage index except for circular tunnels. A modified damage index was proposed by the author which takes the shape of tunnels other than circular into account.

A Study on the Application of IUCN Global Ecosystem Typology Using Land Cover Map in Korea (토지피복지도를 활용한 IUCN 생태계유형분류 국내 적용)

  • Over the past few centuries, widespread changes to natural ecosystems caused by human activities have severely threatened biodiversity worldwide. Understanding changes in ecosystems is essential to identifying and managing threats to biodiversity. In line with this need, the IUCN Council formed the IUCN Global Ecosystem Typology (GET) in 2019, taking into account the functions and types of ecosystems. The IUCN provides maps of 10 ecosystem groups and 108 ecological functional groups (EFGs) on a global scale. According to the type classification of IUCN GET ecosystems, Korea's ecosystem is classified into 8 types of Realm (level 1), 18 types of Biome (level 2), and 41 types of Group (level 3). GETs provided by IUCN have low resolution and often do not match the actual land status because it was produced globally. This study aimed to increase the accuracy of Korean IUCN GET type classification by using land cover maps and producing maps that reflected the actual situation. To this end, we ① reviewed the Korean GET data system provided by IUCN GET and ② compared and analyzed it with the current situation in Korea. We evaluated the limitations and usability of the GET through the process and then ③ classified Korea's new Get type reflecting the current situation in Korea by using the national data as much as possible. This study classified Korean GETs into 25 types by using land cover maps and existing national data (Territorial realm: 9, Freshwater: 9, Marine-territorial: 5, Terrestrial-freshwater: 1, and Marine-freshwater-territorial: 1). Compared to the existing map, "F3.2 Constructed lacustrine wetlands", "F3.3 Rice paddies", "F3.4 Freshwater aquafarms", and "T7.3 Plantations" showed the largest area reduction in the modified Korean GET. The area of "T2.2 Temperate Forests" showed the largest area increase, and the "MFT1.3 Coastal saltmarshes and reedbeds" and "F2.2 Small permanent freshwater lakes" types also showed an increase in GET area after modification. Through this process, the existing map, in which the sum of all EFGs in the existing GET accounted for 8.33 times the national area, was modified so that the total sum becomes 1.22 times the national area using the land cover map. This study confirmed that the existing EFG, which had small differences by type and low accuracy, was improved and corrected. This study is significant in that it produced a GET map of Korea that met the GET standard using data reflecting the field conditions. 

Korean nation-centralism and Confucianism(I) - the reflection of controlled modern rationality (한국의 국가 중심주의와 유교(I) - 통제적 근대 합리성에 관한 성찰 -)

  • This thesis is to check whether the modernization, promoted mainly in 1960-1970 of Korea may have the relations with Confucian values or have a gap between this and that, and have the question about the idea of the appearance of modernity under Park Jung-hee's government, which is based on the viewpoint that Confucianism would have been made ill use of or have contribution to the nation-directing modernity, especially to the modernization of people. In a sense this thesis demands the overcome of the modern ills such as social dichotomy, leaving out matters of locals, and neglecting the diversities and singularities of creatures, resulted from efficiencies and uniformity caused by nation-centralism. At first Confucianism have represented humanism with a view to finding the mean between the two of locals, affairs, and men. As such it has seek to find centrality, which means my real mind for meeting outward things or the optimum as the mean, the best state between of the two. The political doctrines modified from Confucian learnings including chung, hyo, samgang, and oryun worked as the mechanism for finding nation-directing modernity. As a result we have lived in the modernity, strengthened by nation-centralism. And the leading concepts in related with Neo-Confucianism had people lose their spaces of desire for their own future or got them to be narrow. Accordingly the modernization of Korea means not an integral space in which we can achieve what we want in various aspects, but a deficient space to be complemented, resulted from the centralization of all conditions of life, dichotomous way of approaching matters by nation-centralism, far from being the essence of Confucianism. In the end the rapid modernization by the leaders in Korea has given rise to the concentration politics, economy, and so forth on Seoul as the center. Then we should deeply reflect the deficiency state of centralism like this and how Confucianism would have been responsible for it and will give how to relieve the unequal centralism of nation. Now for this matter we would like to expect our study in the future.v
