Identification of PM10 Chemical Characteristics and Sources and Estimation of their Contributions in a Seoul Metropolitan Subway Station (서울시 지하역사에서 PM10의 화학적 특성과 오염원의 확인 및 기여도 추정)

  • Since the underground transportation system is a closed environment, indoor air quality problems may seriously affect many passengers' health. The purpose of this study was to understand $PM_{10}$ characteristics in the underground air environment and further to quantitatively estimate $PM_{10}$ source contributions in a Seoul Metropolitan subway station. The $PM_{10}$ was intensively collected on various filters with $PM_{10}$ aerosol samplers to obtain sufficient samples for its chemical analysis. Sampling was carried out in the M station on the Line-4 from April 21 to 28, July 13 to 21, and October 11 to 19 in the year of 2010 and January 11 to 17 in the year of 2011. The aerosol filter samples were then analyzed for metals, water soluble ions, and carbon components. The 29 chemical species (OC1, OC2, OC3, OC4, CC, PC, EC, Ag, Al, Ba, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Si, Ti, V, Zn, $Cl^-$, $NO_3{^-}$, $SO_4{^{2-}}$, $Na^+$, $NH_4{^+}$, $K^+$, $Mg^{2+}$, $Ca^{2+}$) were analyzed by using ICP-AES, IC, and TOR after proper pretreatments of each sample filter. Based on the chemical information, positive matrix factorization (PMF) model was applied to identify the $PM_{10}$ sources and then six sources such as biomass burning, outdoor, vehicle, soil and road dust, secondary aerosol, ferrous, and brakewear related source were classified. The contributions rate of their sources in tunnel are 4.0%, 5.8%, 1.6%, 17.9%, 13.8% and 56.9% in order.

Preparation and Properties of Moisture-absorbing Film Impregnated with Polyacrylic Acid Partial Sodium Salt Material (폴리아크릴산 나트륨 염이 함침된 흡수성 고분자 복합 필름의 제조 및 특성 연구)

  • Moisture is a major factor causing the deteriorative physical change, microbial growth, and chemical reaction of the products. In this study, the moisture absorbing composite films have been prepared with moisture absorbing material of polyacrylic acid partial sodium salt (PAPSS) impregnated on LLDPE polymer for the functional packaging applications. The results showed that PAPSS impregnated film illustrated uniformly dispersed PAPSS particles in the LLDPE polymer matrix. The transparency of the PAPSS impregnated film decreased slightly at higher PAPSS concentrations. An increase in the PAPSS content for moisture-absorbing films showed a similar decrease in tensile strength, percent elongation at break, and tear strength. Their values of films impregnated with PAPSS of 0.5, 1, and 2% showed no significant difference. Meanwhile, 4% PAPSS films significantly decreased the values of mechanical properties compared to the films impregnated with different PAPSS levels. Values of the oxygen permeability and water vapor permeability for PAPSS impregnated films decreased significantly with greater PAPSS. The results indicate that 4% PAPSS impregnated in LLDPE films had high affinity of moisture absorbencies compared to the other films. The mathematical equation that best described the moisture sorption isotherm of each film sample was the GAB equation at $25^{\circ}C$. The crystallization and melting temperatures of PAPSS films were influenced by the addition of PAPSS material, but showed good thermal stability.

Inspection System using CIELAB Color Space for the PCB Ball Pad with OSP Surface Finish (OSP 표면처리된 PCB 볼 패드용 CIELAB 색좌표 기반 검사 시스템)

  • We demonstrated an inspection system for detecting discoloration of PCB Cu ball pad with an OSP surface finish. Though the OSP surface finish has many advantages such as eco-friendly and low cost, however, it often shows a discoloration phenomenon due to a heating process. In this study, the discoloration was analyzed with device-independent CIELAB color space. First of all, the PCB samples were inspected with standard lamps and CCD camera. The measured data was processed with Labview program for detecting discoloration of Cu ball pad. From the original PCB sample image, the localized Cu ball pad image was selected to reduce the image size by the binarization and edge detection processes and it was also converted to device-independent CIELAB color space using $3{\times}3$ conversion matrix. Both acquisition time and false acceptance rate were significantly reduced with this proposed inspection system. In addition, $L^*$ and $b^*$ values of CIELAB color space were suitable for inspection of discoloration of Cu ball pad.

Chemometric Approach to Fatty Acid Profiles in Soybean Cultivars by Principal Component Analysis (PCA)

  • The purpose of this study was to investigate the fatty acid profiles in 18 soybean cultivars grown in Korea. A total of eleven fatty acids were identified in the sample set, which was comprised of myristic (C14:0), palmitic (C16:0), palmitoleic (C16:1, ${\omega}7$), stearic (C18:0), oleic (C18:1, ${\omega}9$), linoleic (C18:2, ${\omega}6$), linolenic (C18:3, ${\omega}3$), arachidic (C20:0), gondoic (C20:1, ${\omega}9$), behenic (C22:0), and lignoceric (C24:0) acids by gas-liquid chromatography with flame ionization detector (GC-FID). Based on their color, yellow-, black-, brown-, and green-colored cultivars were denoted. Correlation coefficients (r) between the nine major fatty acids identified (two trace fatty acids, myristic and palmitoleic, were not included in the study) were generated and revealed an inverse association between oleic and linoleic acids (r=-0.94, p<0.05), while stearic acid was positively correlated to arachidic acid (r=0.72, p<0.05). Principal component analysis (PCA) of the fatty acid data yielded four significant principal components (PCs; i.e., eigenvalues>1), which together account for 81.49% of the total variance in the data set; with PC1 contributing 28.16% of the total. Eigen analysis of the correlation matrix loadings of the four significant PCs revealed that PC1 was mainly contributed to by oleic, linoleic, and gondoic acids, PC2 by stearic, linolenic and arachidic acids, PC3 by behenic and lignoceric acids, and PC4 by palmitic acid. The score plots generated between PC1-PC2 and PC3-PC4 segregated soybean cultivars based on fatty acid composition.

The Exposure Status and Biomarkers of Polycyclic Aromatic Hydrocarbons in Shipyard Workers

  • Because shipyard workers are involved with various manufacturing process in shipyard industry, and they are exposed to many kinds of hazardous materials. Especially, painting workers were exposed polycyclic aromatic hydrocarbons (PAH). This study was conducted to assess the exposure status of PAH based on job-exposure matrix. We investigated the effect of genetic polymorphism of xenobiotic metabolism enzymes involved in PAH metabolism on levels of urinary metabolite. A total of 93 shipbuilding workers were recruited in this study. Questionnaire variables were age, sex, use of personal protective equipment, smoking, drinking, and work duration. The urinary metabolite was collected in the afternoon and corrected by urinary creatinine concentration. The genotypes of CYP1A1, CYP2E1, GSTM1, GSTT1 and UGT1A6 were investigated by using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods with DNA extracted from venous blood. Urinary 1-OHP levels were significantly higher in direct exposured group (spray and touch-up) than indirect exposed group. Urinary 1-OHP, concentration of the high exposure with wild type of UGT1A6 was significantlyhigher than that of the high exposure with other UGT1A6 genotype. In multiple regression analysis of urinary 1-OHP, the regression coefficient of job grade was statistically significant (p<0.05) and UGT1A6 was not significant but a trend (p<0.1). The grade of exposure affected urinary PAH concentration was statistically significant. But genetic polymorphism of xenobiotics metabolism enzymes was not statistically significant. Further investigation of genetic polymorphism with large sample size is needed.

Verification of Microstructure Qualities of ACR-Approved Mammography Phantoms by Refraction-Enhanced Synchrotron Radiation Imaging

  • Images of microcalcification specks showed large variation in conventional radiographs of phantoms which are approved for mammography image quality standard by the American College of Radiology (ACR). This kind of variation is not appropriate for image quality standards because the number of specks are visually counted in images and that number is important in image quality evaluation. Our study using synchrotron radiation (SR) imaging revealed the overlapping of micro-sized air bubble(s) to some specks, and also the structural deformation or crackings. Eight phantoms approved by ACR from two different makers and an air-bubble phantom were examined. SR imaging was performed at a synchrotron radiation facility, SPring-8, in Japan. The image-detector was a fluorescent-screen optical-lens coupling system using a CCD camera with a spatial resolution of 6 $\square$m. Objects when imaged with longer sample-to-detector distance show edge enhancement due to a difference in refraction indices, that is refraction enhancement. Refraction-enhanced SR images revealed that some of specks carried foreign objects, which were proven to be air. In phantoms provided by one maker, attaching/overlapping airs were observed for 62 out of 150 specks (41%) , with a higher incidence for the smallest specks. A speck becomes hardly visible in a conventional radiograph when air(s) overlaps the majority part of a speck, though depending on the size of the air-inclusion and on its configuration. Those airs might have been adsorbed on a speck surface before being embedded and then introduced into the matrix together with specks. Our study using SR imaging has clearly shown the nature of defects in some mammography phantoms which seriously degrade the quality as an image standard.

Simultaneous Determination of Tetracycline Antibiotics by 3-Phase Hollow Fiber-Liquid Phase Microextraction (HF-LPME) and HPLC-UV/Vis (3-상 속빈 섬유-액체상 미량추출법(HF-LPME)과 HPLC-UV/Vis을 이용한 Tetracycline류 항생제 동시분석)

  • A simple and efficient preconcentration method was developed using three-phase liquid phase microextraction prior to HPLC-UV for simultaneous extraction and determination of tetracycline antibiotics (tetracycline, oxytetracycline, and chlortetracycline). The tetracycline antibiotics were separated simultaneously on a column ($C_8$, $3.0{\times}150mm$, $3{\mu}m$) with high selectivity and sensitivity using gradient elution. Under optimized conditions (extraction solvent, heptanal; pH of donor, 9.0; pH of acceptor, 1.0; stirring speed, 700 rpm; NaCl salt, 0%; and extraction time, 60 min), enrichment factors (EF) were between 5.6 and 22.3. The limit of detection (LOD) and limit of quantitation (LOQ) in the spiked urine matrix were in the concentration range of $0.08{\sim}0.8{\mu}g/mL$ and $0.4{\sim}1.6{\mu}g/mL$, respectively. The calibration curves were linear within the range of $0.1{\sim}32{\mu}g/mL$ with the square of the correlation coefficient being more than 0.995. The precision (as a relative standard deviation, RSD) and accuracy (as a relative recovery) within working range were 1.3~9.1% and 84~118%, respectively.

Effect of storage time and temperature on levels of phthalate metabolites and bisphenol A in urine

  • Urine is a widely used matrix in biomonitoring studies on the assessment of human exposure to environmental chemicals such as phthalate esters and bisphenol A (BPA). In addition to the need to apply valid analytical techniques, assurance of specimen integrity during collection and storage is an important prerequisite for the presentation of accurate and precise analytical data. One of the common issues encountered in the analysis of non-persistent contaminants is whether shipping and storage temperature and time since collection have an effect on sample integrity. In this study, we investigated the stability of phthalate metabolites and BPA in spiked and unspiked urine samples stored at room temperature ($20^{\circ}C$) or at $-80^{\circ}C$ for up to 8 weeks. Concentrations of phthalate metabolites declined, on average, by 3% to 15%, depending on the compounds, and BPA declined by ~30% after 4 weeks of storage of spiked urine samples at $20^{\circ}C$. In a test of 30 unspiked urine samples stored at $20^{\circ}C$ and at $-80^{\circ}C$ for 8 weeks, the concentrations of phthalate metabolites and BPA decreased by up to 15% to 44%, depending on the compound and on the samples. It was found that the small reduction in phthalate concentrations observed in urine, varied depending on the samples. In a few urine samples, concentrations of phthalate metabolites and BPA did not decline even after storage at $20^{\circ}C$ for 8 weeks. We found a significant relationship between concentrations of target analytes in urine stored at $20^{\circ}C$ and at $-80^{\circ}C$ for 8 weeks. We estimated the half-lives of phthalate metabolites and BPA in urine stored at $20^{\circ}C$. The estimated half-life of monoethyl phthalate (mEP) and mono (2-ethyl-5-carboxyphentyl) phthalate (mECPP) in urine stored at $20^{\circ}C$ was over two years, of mono (2-ethyl-5-oxohexyl) phthalate (mEOHP) and monobenzyl phthalate (mBzP) was approximately one year, and of other phthalate metabolites was approximately 6 months. The estimated half-life of BPA in urine stored at $20^{\circ}C$ was approximately 3 months, which is much longer than that reported for aquatic ecosystems.

Measuring the Professional Development Needs for Technical and Vocational Education and Training Staff in Nepal: A Look at Teachers' Perceptions (네팔 직업기술교육훈련 교사들의 교육훈련 프로그램 요구조사)

  • Promoting job growth and quality has been central to developing countries to reduce poverty for decades. In this regard, Nepal is not unique in its need to develop effective TVET programme as it is considered showing positive association with workforce development and productivity. While the copious academic literature has highlighted upon staff training programme in general education, it encounters lack of adequate attention in the field of TVET education. The purpose of the study is to identity and prioritise the professional development needs for the teachers of TVET sector in Nepal. A sample(N=317) of Nepalese teachers from seven provinces were asked to answer the scale. The Borich needs assessment model was employed to assess the perceived level of importance and present preferred training modules among 29 teaching subjects. Moreover, a quadrant analysis using 2×2 matrix was used as a diagnostic tool to denote overriding training topics. The result shows that following competence skills constructs were perceived to be in high need of enhancement as well as in existence of discrepancies: 'occupational characteristics of TVET trainer', 'training needs analysis and curriculum development', 'curriculum development based on national competence standard(NCS)', and 'Institution management evaluation'.

Development of mass spectrometric analysis of $\alpha_1$(I) and $\alpha_2$(I) chain Collagen ($\alpha_1$(I)및 $\alpha_2$(I)사슬 콜라겐의 질량분석법 개발 연구)

  • Collagen is the important structural proteins in mammals with various peptide composition and cross-linkings. The direct analysis of collagen protein was not suitable because of its structural complexity and diversity. In this study, we suggest the simple way of collagen analysis by introducing matrix-assisted laser desorption/ionization time-of flight mass spectrometry (MALDI-TOF MS) to identify the collagen and its trypsin-digested fragments, and by subsequent time-of-flight tandem mass spectrometry(Q-TOF MS/MS) to analyze the amino acid sequences of identified fragments. Using the collagen samples extracted from the tail of mouse, 10 separated bands were found in SDS-PAGE, and the masses of most bands could be more finely determined by MALDI-TOF MS. When each 10 separated proteins was tryptic digested and introduced to MALDI-TOF, the Gly1056-Arg1073 fragment from $\alpha_1$-chain was identified in four bands, and the Gly1056-Arg1073 fragment from $\alpha_2$-chain was identified in five bands, both in type I collagen. Although few fragments were found because of the cross-linkings left in digested collagen sample, it could be determined that the type I collagen existed at least in 7 separated bands. When the amino acid sequences of two identified fragments were analyzed by Q-TOF MS/MS, both sequences were identical with those determined by MALDI-TOF MS. It suggested that the two peaks in MALDI-TOF MS caused by the fragments identified in this work could be used as the fingerprint to simply identify type I collagen in protein samples.
