Studies on Erythropoietic Action by the Administration of Pilose Antler Extract in SAM P6.

  • In previous studies we reported that the levels of RBC, hemoglobulin and hematocrit in SAM Rl and SAM P6 were increased significantly from 7 day after oral administration of the pilose antler extract, 5g/kg/day, and were lasted during the study. Therefore, this study was performed to elucidate mechanism of erythropoietic action by the extract administration. SAM Rl and SAM P6 were chosen as experimental animals. At age of 12 weeks, pilose antler extract were given 0.3 and 5 g/kg/day (p.o.) each for 0, 7 and 14 days in both animals. Complete blood cells (CBC) such as WBC, lymphocytes, monocytes, granulocytes, RBC, hemoglobulin, and hematocrit were counted. And plasma concentration of erythropoietin (EPO) which is the major regulator of erythropoiesis was measured using $\^$125/I-antierythropoietin IgG. Ferritin concentration in plasma was also analyzed.

노화 방지를 위한 한약재의 효능 연구 (I) (Age-related Changes in blood Chemistry and Thyroid hormones in Senescence Accelerated Mice(SAM R1 and SAM P6))

  • Aging process can be explained by many factors. In this study, we counted complete Blood Cells (CBC) such as WBC, Lymphocytes, Monocytes, Granulocytes, RBC, HGB, and HCT of both SAM P6 and SAM P1 during the aging process. Plasma albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea concentrations were also measured at the same time. In addition to these, plasma concentrations of cortisol, total T3, and total T4 were analyzed by Chemiluminescent Immunoassay. There were no change in CBC counts of SAM R1 and SAM P6 during this study. Plasma concentratins of albumin and glucose decreased significantly in SAM R1. However, plasma alkaline phosphatases and creatinine concentration in SAM P6 decreased significantly at 16 week after birth comparing to the control. Total T4 levels were siginificantly increased although cortisol and total T3 concentrations were the same in SAM R1 groups. Especially, the after birth of creatinine, alkaline phosphatase, T4 of SAM P6 at 16 week were significantly different form those of SAM R1. At 12 week after birth, pilose antler extract was given 5g/kg/day p.o. for 0, 7, 14, 21, and 30 days each in both SAM R1 and SAM P6. The RBC, HGB, and HCT levels started to increase significantly from 7 days after the dose at SAM P6 only. Total T4 conectrations were elevated gradually during the study although the antler extract administration did not prevent or inhibit the increase in total T4 concentration during the study. Therefore, the elevation of erythrocytes after administration of the extract needs to be studied in future.

ITO 표면의 자기 조립 단일막 형성에 의한 유기 발광 소자의 특성

  • 본 연구에서는 ITO 표면 개질에 의한 유기 발광 소자의 특성 변화에 대해서 연구하였다. ITO 전극은 발광 소자의 투명 전극으로 널리 사용되고 있으며 이러한 발광 소자의 특성은 ITO의 표면 상태에 따라 민감하게 반응한다. ITO 표면 개질은 ITO와 유기물 사이의 쇼트기 장벽을 감소시키며, 전극과 유기물의 점착을 향상시켜 준다. 본 실험에서는 습식 처리 방식으로 self-assembled monolayer(SAM)을 사용하였다. 유기 발광 소자의 특성은 SAM 처리에 의해 향상 되었다. 유기 발광 소자는 ITO/SAM/TPD(50nm)/$Alq_3$(70nm)/LiF(0.5nm)/Al(100nm)의 구조로 제작하였으며, ITO의 표면 특성은 일반적인 특성 기술에 의해 연구되었다. SAM 처리된 소자는 SAM 처리하지 않은 소자에 비해 구동 전압, 발광 세기, 외부 양자 효율 등이 향상되었다. ITO의 SAM 처리 시간을 0/10/15/20/25분으로 하여 소자를 제작하였다. 15분간 SAM 처리한 소자는 SAM 처리하지 않은 소자에 비해 외부 양자 효율과 전류 효율이 2.6배 상승하였다. 본 실험을 통하여 ITO 표면 위에 SAM층을 삽입한 걸과, 구동 전압, 발광 세기, 효율 등이 향상됨을 알 수 있었다.

Selection and Characteristics of Fermented Salted Seafood (jeotgal)-Originated Strains with Excellent S-adenosyl-L-methionine (SAM) Production and Probiotics Efficacy

  • This study is executed to develop probiotics which produce S-adenosyl-L-methionine (SAM), a methyl group donor of the 5-methyltetrahydrofolate methylation reaction within the animal cell. SAM is an essential substance for the synthesis, activation, and metabolism of hormones, neurotransmitters, nucleic acids, phospholipids, and cell membranes of animals. The SAM is also known as a nutritional supplement to improve brain functions of the human. In this study, the SAM-producing strains are identified in 18 types of salted fish, and then, the strains with excellent SAM productions are being identified, with 1 strain in the Enterococcus genus and 9 strains in the Bacillus genus. Strains with a large amount of SAM production include the lactic acid bacteria such as En. faecium and En. durans, En. sanguinicola, as well as various strains in the Bacillus genus. The SAM-overproducing strains show antibacterial activities with certain harmful microbes in addition to the weak acid resistances and strong bile resistances, indicating characteristics of probiotics. It is possible that the jeotgal-originated beneficial strains with overproducing SAM can be commercially utilized in order to manufacture SAM enriched foods.

Effect of S-Adenosylmethionine on Hepatic Injury from Sequential Cold and Warm Ischemia

  • We investigated whether S-adenosylmethionine (SAM) treatment improved ischemic injury using perfused rat liver after sequential periods of 24 h cold and 20 min re-warming ischemia. SAM (100 $\mu\textrm{mol/L}$) was added to University of Wisconsin (UW) solution and Ringers lactate solution. After cold and sequential warm ischemia, releases of lactate dehydrogenase (LDH) and purine nucleoside phosphorylase (PNP) markedly increased during repefusion. The increase in PNP was significantly reduced by SAM treatment. While the concentration of reduced glutathione (GSH) in ischemic livers significantly decreased, the concentration of glutathione disulfide (GSSG) increased. This decrease in GSH and increase in GSSG were suppressed by SAM treatment. Lipid peroxidation was elevated in cold and warm ischemic and reperfused livers, but this elevation was also prevented by SAM treatment. Hepatic ATP levels were decreased in the ischemic and reperfused livers to 42% of the control levels. However, treatment with SAM resulted in significantly higher ATP levels and preserved the concentration of AMP in ischemic livers. Our findings suggest that SAM prevents oxidative stress and lipid peroxidation and helps preserve hepatic energy metabolism.

김치의 재료별 첨가량에 따른 SAM 함량의 변화 (Change of S-Adenosyl-L-Methionine(SAM) Content in Material by Addition of the Kimchi)

  • 본 연구에서는 발효기간 중 SAM 생성량 증진김치의 최적조건을 개발하기 위해 배추김치의 재료별 및 제조법에 따른 발효기간 중의 SAM 함량 변화를 측정하였다. 발효가 진행됨에 따라 모든 김치군의 pH는 발효초기에는 급격히 감소하다가 후기에는 pH $4.2{\sim}4.3$으로 비슷하게 나타났다. 산도는 pH와 비슷한 경향을 보이면서 발효초기에는 급격히 감소하다가 후기에는 서서히 증가하는 경향을 보였다. SAM 함량은 가장 기본적인 재료로만 제조한 표준(A)김치는 가장 낮게 나타났고, SAM이 검출되는 원 부재료에 SAM 함량이 많은 김치가 SAM 함량도 높게 나타났다. 모든 김치군 중 I김치가 가장 높은 SAM 함량을 나타냈으며, 김치별 SAM 함량이 가장 높은 발효시점은 A, K김치는 pH $4.6{\sim}4.9$, 산도 $0.38{\sim}0.57$0.57, D, H김치는 pH 6.3, 산도 $0.24{\sim}0.27$, E, G김치는 pH $6.1{\sim}6.6$, 산도 $0.05{\sim}0.14$0.14, B, C, F, I, J김치는 pH $4.3{\sim}5.3$, 산도 $0.45{\sim}1.02$에서 나타났다. 따라서 대부분의 김치는 적숙기시점이 되었을 때 B, E, G, H김치는 초기발효 때부터 SAM 함량이 높게 나타났다. 또한 발효가 진행됨에 따라 SAM 함량은 감소하는 경향을 보였다. 따라서 연구결과 주재료인 채소류와 젓갈의 함량비를 적절히 제조하여 최적의 SAM 생산능력의 발효시점과 김치맛에 영향을 끼치지 않는 관능적으로 적합하고 SAM 생성이 보다 효과적으로 유도되는 최적의 조건에 대한 실험들이 추가적으로 필요하다고 판단된다.

초소수성 코팅 튜브에서의 비응축가스 영향에 대한 응축 열전달 연구 (The effects of non-condensable gas on condensation heat transfer on a super-hydrophobic surface tube)

  • 본 연구의 목표는 수평 관외 초소수성 튜브에서 비응축가스(NCG)의 영향을 관찰하고, 이를 일반 알루미늄 튜브의 응축 현상과 비교하는 것이다. 초소수성 튜브 제작을 위해 Self-Assembled Monolayer(SAM) 코팅으로 알루미늄 튜브의 외부를 표면개질 했다. 응축 성능을 나타내기 위해 총합 열전달계수를 사용하였고, 이 값으로 응축 성능을 비교 검토하였다. 본 연구의 주요 변수는 비응축가스 질량 분율로, 0.08에서 0.45의 범위에서 실험을 진행하였다. 응축 실험을 통해 비응축가스 질량 분율이 낮아질수록 응축 성능이 SAM 튜브와 일반 튜브에서 모두 향상되는 것을 확인했다. SAM 튜브에서 적응축 열전달 성능은 일반 튜브 대비하여 평균 약 1.9배에서 2.5배 정도 큰 것을 관찰하였다. SAM 튜브에서 비응축가스 질량 분율이 낮아지면서 응축 성능 상승폭이 감소하게 되는데, 이는 Flooded 응축 현상이 발현되었기 때문이다. 응축이 더 활발하게 진행되면 SAM 튜브에서 막응축이 일어나는 것을 관찰하였고, 이 때 성능은 일반 알루미늄 튜브보다도 저하된 성능을 보였다. SAM 알루미늄 튜브에서 Flooded 응축과 막응축이 일어나는 원인으로 표면에서의 Pinning 효과를 이용하여 설명하였다. 결론적으로, SAM 튜브를 실제 응축기에 적용해 표면개질로 인한 응축 성능 개선 효과를 얻기 위해서는 적응축 또는 Flooded 응축이 일어나는 조건으로 응축기 내 환경 조성을 해야 한다.

Development of S-Adenosyl-L-methionine (SAM)-reinforced Probiotic Yogurt Using Bifidobacterium bifidum BGN4

  • S-Adenosyl-L-methionine (SAM) exerts several beneficial effects on depression, chronic diseases, and cognitive impairments. Bifidobactrium bifidum BGN4 reportedly produces higher amounts of SAM than any other lactic acid bacterium used in yogurt. The aim of this study was to develop a SAM-reinforced probiotic yogurt using Bifidobacterium. The sensory aspects of the yogurt via response surface methodology (RSM) and the texture and SAM content of the yogurt were assessed. Based on the sensory assessments for sweetness, sourness, and thickness evaluated by 48 panelists, the optimized conditions for preparation of SAM-reinforced yogurt were 4.0-4.4%(w/w) sugar, 3.2-3.5%(w/w) skim milk, and a pH of 4.7-4.8. The SAM content of the developed yogurt was 0.05 nmol/mL. In conclusion, SAM-reinforced probiotic yogurt may provide a vehicle for the potential exploitation of the benefits of increased dietary SAM.

S-Adenosylmethionine (SAM) Regulates Antibiotic Biosynthesis in Streptomyces spp. in a Mode Independent of Its Role as a Methyl Donor

  • S-Adenosylmethionine (SAM) is a ubiquitous biomolecule serving mainly as a methyl donor. Our recent studies revealed that SAM controls antibiotic production in Streptomyces. In this study, the functional mode of SAM was studied in S. coelicolor and S. antibioticus ATCC11891, employing S-adenosylhomocysteine (SAH), a methylation reaction product of SAM. Actinorhodin biosynthesis did not require SAM as a methyl donor, whereas SAH enhanced the actinorhodin biosynthesis up to the level comparable to SAM, and the most effective concentration of SAH was higher than that of SAM. In the case of oleandomycin that requires SAM for its biosynthesis, both SAM and SAH at the concentration as low as 100 mM showed comparable efficacy in enhancing the production; SAM at 1 mM concentration additionally stimulated to give a 5-fold enhancement of oleandomycin production. In vitro autophosphorylation of protein kinase AfsK was found to be activated by both SAM and SAH, as well as other structurally related compounds. Our studies demonstrate that SAM regulates antibiotic biosynthesis in a mode independent of its role as a methyl donor and suggest that SAM acts directly as an intracellular signaling molecule for Streptomyces.

마이크로몰딩의 이형성 향상을 위한 소수성 Self-assembled Monolayer(SAM) 코팅 (Hydrophobic Self-assembled Monolayer(SAM) Coating for Enhanced Demolding Performance in Micromolding)

  • In this paper, the surface modification effect of self-assembled monolayer(SAM) of 1-dodecanethiol [$CH_3$($CH_2$)$_{11}$SH] used as an anti-adhesive film in micromolding process was studied. Monolayers of 1-dodecanethiol[$CH_3$(CH$_2$)$_{11}$SH] were obtained by immersing a metal place in pure 1-dodecanethiol. SAM film on the nickel plate has been examined by using X-ray photoelectron spectroscopy(XPS). The focus has been placed on S-Ni bonding. From the XPS analysis, sulfur atoms were detected from the SAM film as a chemical composition of S-Ni. In order to measure an adhesion force of the SAM-coated nickel surface, atomic force microscopy(AFM) was used in force-distance mode, which whows the micro-adhesive force on solid surface. It was shown that adhesion forces measured from the SAM-coated nickel surface and the Ni surface without SAM coating were 3.52nN and 5.32nN, respectively. In order to investigate the effect of SAM coating on the surface foughness the replica in demolding process, hot embossing experiments were performed using a SAM-coated nickel master and a nickel master without SAM coating. Surface roughness of replica from the SAM-coated master showed 25nm and that of replica from master without SAM coating was 35nm. The smoother surface roughness of the replica from the SAM-coated, master is believed to result from reduction in the adhesion forces.ces.
