Neuroprotective Effects of Medicinal Herbs in Organotypic Hippocampal Slice Cultures (뇌해마의 장기양 조직배양을 이용한 한약물의 뇌신경세포손상 보호효능 연구)

  • Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. Effect of Salviae Radix on Impairment of Membrane Transport Function in Rabbits with Myoglobinuric Acute Renal Failure (마이오글로빈뇨성 급성 신부전 토끼에서 신장 세포막 수송 기능 장애에 대한 단삼의 효과)

  • This study was carried out to determine if Salviae Radix extract (SRE) exerts protective effect against alterations in membrane transport function in rabbits with rhabdomyo lysis-induced acute renal failure. Acute renal failure was induced by intramuscular administration of glycerol (50%, 10 ml/kg). GFR in the glycerol-injected animals was reduced to 11% of the basal value and the fractional $Na^{+}$ excretion was increased to 7.8-fold, indicating generation of acute renal failure. When animals received SRE pretreatment for 7 days prior to glycerol injection, such changes were significantly attenuated. The fractional excretion of glucose and phosphate was increased more than 43-fold and 27-fold, respectively, in rabbits treated with glycerol alone. However, they were increased to 17-and 4.3-fold, respectively, in SRE-pretreated rabbits, and these values were significantly lower than those in rabbits treated with glycerol alone. Uptakes of glucose and phosphate in purified isolated brush-border membrane, the $Na^{+}-K^{+}-ATPase$ activity in microsomal fraction, and cellular ATP levels all were reduced in rabbits treated with glycerol alone. Such changes were prevented by SRE pretreatment. Uptakes of organic ions, PAH and TEA, in renal cortical slices were inhibited by the administration of glycerol, which was prevented by SRE pretreatment. Pretreatment of an antioxidant DPPD significantly attenuated the increase in the fractional excretion of glucose and phosphate induced by rhabdomyolysis. These results indicate that rhabdomyolysis causesimpairment inreabsorption of solutes in the proximal tubule via the generation of reactive oxygen species, and SRE pretreatment may provide the protection against the rhabdomyolysis-induced impairment by its antioxidant action.

The Effect of Salviae Radix on Oxidat-Inhibition of Phosphate Uptake in Renal Proximal Tubular Cells (단삼약침액(丹蔘藥鍼液)이 신장(腎臟) 근위세뇨관세포(近位細尿管細胞)에서 산화제(酸化劑)에 의한 인산(燐酸)의 이동억제(移動抑制)에 미치는 영향(影響))

  • This study was undertaken to determine if Salviae Radix (SR) exerts protective effect against oxidant-induced inhibition of phosphate uptake in renal proximal tubular cells. Membrane transport function and cell death were evaluated by measuring phosphate uptake and trypan blue exclusion, respectively, in opossum kidney (OK) cells, an established proximal tubular cell line. $H_2O_2$ was used as a model oxidant. $H_2O_2$ inhibited the phosphate uptake in a dose-dependent manner over the concentration range of 0.1-0.5 mM. Similar fashion was observed in cell death. However, the phosphate uptake was more vulnerable to $H_2O_2$ than cell death, suggesting that $H_2O_2$-induced inhibition of phosphate uptake is not totally attributed to cell death. Decreasedphosphate uptake was associated with ATP depletion and inhibition of $Na^+$-pump activity as determined by direct inhibition of $N^+-K^+$-ATPase activity. When cells were treated with $H_2O_2$ in the presence of 0.05% SR, the inhibition of phosphate uptake and cell death induced by $H_2O_2$ was significantly attenuated. SR restored ATP depletion and decreased $Na^+-K^+$-ATPase activity, and this is likely responsible for the protective effect of SR on decreased phosphate uptake. The protective effect of SR was similar to the $H_2O_2$ scavenger catalase. SR reacts directly with $H_2O_2$ to reduce the effective concentration of the oxidant. The iron chelator deferoxamine prevented the inhibition of phosphate uptake and cell death induced by $H_2O_2$, suggesting that $H_2O_2$-induced cell injury is resulted from an iron-dependent mechanism. These results indicate that SR exerts the protective effect against $H_2O_2$-induced inhibition of phosphate uptake by reacting directly with $H_2O_2$ like the $H_2O_2$scavenger enzyme catalase, in OK cells. However, the underlying mechanism remains to be explored.

Promoting Effect of a Mixture of 8 Herbal Extracts (SPELA 707) on Hair Growth

  • In Korean folk medicine, several herbs, Glycyrrhizae Radix, Persicae Semen, Salviae Radix, Angelicae Gigantis Radix, Zanthoxyli Fructus, Ginseng Radix Alba, Cnidii Rhizoma, and Carthami Flos, are known to enhance blood circulation and have wound healing or anti-inflammatory effects. These pharmacological actions prompted us to investigate whether these herbs might stimulate hair growth. Thus, using a mixture of their extracts called SPELA 707, we investigated their effects and found that SPELA 707 possessed significant hair cycle converting activity from the telogen phase to the anagen phase in C3H mice. Furthermore, we found that SPELA 707 enhanced the hair density in subjects with hair loss and also promoted the conversion of hair into the anagen phase in subjects with androgenetic alopecia. In addition, hair growth promotion effect of SPELA 707 occurred through inhibition of steroid $5{\alpha}$-reductase activity, which is known to block hair growth. Taken together, these results suggest that SPELA 707 has a potential to be used for the treatment of hair loss.

Antimetastatic effect of several crude drugs with antitumor activity on B16-Fo and A549 cells( II ) (항암활성 수종생약의 B16-Fo와 A549 암세포에 대한 항전이 효과( II ))

  • Ethyl ether fractions of Ferula Resina(EFR) and Lithospermi Radix(ELR) and hexane fraction of Salviae miltiorrhizae Radix(HSR) exerted an inhibitory effect on cell adhesion of A549 to extracelluar matrix most effectively in vitro cell adhesion assay. Thus, with above fractions for the evaluation of antitumor activity, T/C% was monitored in ICR bearing S-180 and for antimetastatic effect, pulmonary colonization assay, weight of organs, changes of WBC and platelet were studied. In pulmonary colonization assay incidence rate to control was 73 % 42 %, 14 % in ELR, HSR and EFR-treated groups repsectively. Gain of lung weight was significantly inhibited in all groups while spleen weight was significantly reduced only in SMR group, but no changes in kidney and liver as compared with control. Number of platelet was significantly increased in all groups to normal range as compared with thrombocytopenic contol. WBC was significantly reduced only in LR group. These results suggest that ethyl ether fraction of Ferula Resina has more effective antimetastatic activity.

A Case Report of Obesity treated with Soshiho-tang gagambang based on Shanghanlun provisions (『상한론(傷寒論)』 변병진단체계(辨病診斷體系)에 근거하여 소시호탕(小柴胡湯) 가감방(加減方)을 투여한 비만 증례 1례)

  • Objective : The purpose of this paper is to report the effect of Sosiho-tang gagambang on Obesity based on Shanghanlun provisions Methods : According to a disease pattern identification diagnostic system based on Shanghanlun provisions, the patient was diagnosed as Yangmyung-byung, and treated for 90 days with Soshiho-tang gagambang that Scutellariae Radix was substituted with Salviae Miltiorrhizae Radix. The result of treatment was evaluated by Body Mass Index(BMI). Results : After taking Soshiho-tang gagambang for 90 days, the patient's BMI was decreased, and the patient is cured of Obesity. Conclusions : This study suggests that the treatment based on Shanghanlun provision has efficacy on Obesity. In addition, further studies are necessary in order to discover the roles of flavor of Shanghanlun herbal decoctions, and ensure the safety of it.

Beneficial Effect of Salviae Miltiorrhizae Radix(SR) on $H_2O_2$-induced Cell Death in Intestinal Epithelial Cells (단삼추출액이$H_2O_2$에 의해 유발된 인간의 장관상피세포의 손상에 미치는 영향)

  • 목적:반응성산소기들은 장관에서 여러 종류의 질병의 발생과 관련을 가지고 있는 것으로 알려져 있어, 이들에 의한 세포손상을 방지하는 약물의 개발은 시급한 실정이다. 본 연구에서는 항산화작용을 가진 약재로 보고 된 단삼추출액이 장관상피세포에서 $H_2O_2$에 의한 세포손상을 방지할 수 있는 지를 조사하고자 하였다. 방법:장관상피세포로는 사람의 소장상피세포에서 유래한 배양세포주인 Caco-2세포를 이용하였고, 세포손상 정도는 trypan blue exclusion assay를 통해 평가하였고, 지질의 과산화는 그 산물인 malondialdehyde의 량을 측정하여 산정하였다. 결과: $H_2O_2$는 처리 시간 및 농도에 비례하여 세포손상을 유발하였으며, 이러한 효과는 단삼추출액에 의해 농도의존적으로 방지되었다. $H_2O_2$에 의한 세포소상은 $H_2O_2$제거제인 catalase와 철착염제인 deferoxamine에 의해 방지되었으나 항산화제인 N,N-diphenyl-p-phenylenamine(DPPD)에 의해 영향을 받지 않았다. $H_2O_2$는 지질의 과산화를 증가시켰으며, 이러한 효과는 단삼추출액과 DPPD에 의해 억제되었다. 단삼추출액은 $H_2O_2$에 의한 세포내 ATP 고갈을 방지하였다. $H_2O_2$는 DNA 손상을 일으켰으며, 이러한 효과는 단삼추출액, catalase 및 deferoxamine에 의해 방지되었으나, DPPD에 의해서는 변화되지 않았다. 결론 : 이상의 결과를 종합하면 단삼추출액은 장관상피세포에서 $H_2O_2$에 의한 세포손상을 방지하며, 이러한 효과는 항산화작용이 아닌 다른 작용기전에 기인할 것으로 생각된다. 또한 본 연구의 결과는 $H_2O_2$가 장관상피세포에서 지질의 과산화를 유발하여 세포손상을 일으키지 않음을 가리킨다.

Effects of Salviae Miltiorrhizae Radix, Rhei Rhizoma and Carthami Flos combined with Samgijiwhang-Tang on Streptozotocin-induced Diabetic Nephropathic Rats (삼기지황탕가단삼(蔘?地黃湯加丹蔘), 대황(大黃), 홍화(紅花)가 Streptozotocin으로 유발된 생쥐의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향)

  • This study was carried out to investigate the preventive effect of Salviae Miltiorrhizae Radix, Rhei Rhizoma and Carthami Flos combined with Samgijiwhang-Tang(SJTSRC) on streptozotocin(STZ)-induced diabetic nephropathy. Rats were divided into a control group of rats with STZ-induced diabetic nephropathy, a sample group of those given SJTSRC, and a normal group. In the experiment diabetic nephropathy was induced by giving STZ(60mg/kg) to rats via the peritoneum, and effects were assessed with measures of serum creatinine, serum BUN, secretion content of albumin and glucose content of urine, malondialdehyde(MDA) and glutathione(GSH) content in cortex of kidney. When STZ was injected into sample rat, the value of creatinine and BUN increased validly and STZ did damage to the kidney. When applying SJTSRC to sample rats, the value of serum creatinine decreased validly but the value of serum BUN decreased invalidity. It was confirmed that SJTSRC had an effect on recovery after kidney damage and secretion content of albumin increasedafter administration of SJTSRC but there was no change in glucose content of urine compared with the control group. The decrease of secretion of albumin after injection of STZ was taken to mean progressive diabetic nephropathy, and that reversal of that trend after SJTSRC administration showed that kidney function had improved, not through decreasing blood sugar, but through other factors. Results suggest that diabetic nephropathy was induced by STZ, and SJTSRC was effective in restricting the extent of damage to the kidney and halting the progression of diabetic nephropathy with improvement in levels of serum creatinine and albumin secretion. More study is needed, particularity pertaining to anti-oxidative effects in the kidney cortex.

Effect of Salviae Radix on renal tubular reabsorption in rabbits with mercury-induced acute renal failure (수은으로 유발된 토끼의 신장 기능 손상에 대한 단삼(丹參)의 효과)

  • 독성약물에 의한 급성신부전시 세뇨관세포의 물질 재흡수 장애에 대한 단삼(丹參) 추출액의 효과를 조사하였다. 토끼에 수은(HgCl2)을 10 mg/kg되게 피하 주사하여 급성신부전을 유발하였고, 단삼(丹參) 추출액의 효과는 수은을 주사하기 전 7일 동안 0.05% 액(液) 0.3 g/kg 용량을 경구 투여하여 관찰하였다. 수은을 주사하기 전 24시간 동안 요와 혈액을 채취하여 신장기능을 측정하여 대조기간(basal period)의 값으로 하였고, 수은을 주사한 후 24시간 동안 요와 혈액을 얻어 수은에 의한 신장기능 변화를 평가하였다. 수은을 처리한 후 사구체여과율이 대조값에 비해 감소하였고, 혈청내 creatinine 농도가 증가하였다. 이러한 결과들은 수은이 급성신부전을 유발하였음을 가리킨다. 수은을 처리한 동물에서 포도당 및 인산의 배설분율이 증가하였고, 이러한 변화는 brush-border membrane에서 물질의 이동장애와 Na-pump 활성의 감소에 기인하였다. 수은을 주사한 동물의 신장피질 절편에서 유기이온인 PAH와 TEA 이동이 억제되었다. 토끼의 신장조직에서 지질의 과산화가 수은을 주사한 후 증가하였다. 단삼(丹參) 추출액을 전 처리한 후 수은을 주사한 경우 수은에 의해 유발된 사구체여과율의 감소와 혈청내 creatinine 농도 증가 현상이 유의하게 완화되었다. 수은에 의한 세뇨관에서 물질의 재흡수 장애가 단삼(丹參) 추출액의 전처리에 의해 방지되었다. 단삼(丹參) 추출액은 수은에 의한 지질의 과산화를 억제하였다. 수은에 의한 급성신부전은 항산화제로 잘 알려진 DPPD에 의해 방지되었다. 이상의 결과를 종합하면 생체실험결과 수은에 의한 급성신부전의 유발과정에 지질의 과산화가 중요한 역할을 할 가능성을 보이고 있고, 단삼(丹參) 추출액은 수은에 의한 급성신부전을 방지하는 효과를 가지고 있으며, 그 효과는 단삼(丹參)의 항산화작용에 기인(起因)할 가능성이 많다.

  • This study was done to investigate effects of SR, CF and AR to angiogenesis of cerebral tissues, protecting damage of cerebral neurons and activating them in cerebral contusion-induced rats. I observed these conclusions as follows ; 1. Observation of VEGF-immunoreactive cells : Groups of administered AR were not meaningful in increasing VEGF-immunoreactive cells for 3 days and 7 days, groups of administered SR meaningfully increased them to control groups in all groups, and groups of administered CF meaningfully increased them to control groups in all groups, too, interestingly, increased double to control group for 7 days. 2. Observation on cerebral neurons by Cresyl violet stain : Dendrites and axons of groups of administered SR, AR for 3 days were clearly observed to control group. Cerebral neurons of groups of administered CF for 3days and 7 days were increased a little, but were not meaningful. In conclusion, AR will be careful of being used in cerebral contusion. CF and SR were effective to activating cerebral hemokinesis by inducing angiogenesis in trauma of tissue, but weakly to protecting trauma of cerebral neurons and activating them. I think more studies will be done in these facts.
