In this study, the identity and distribution of plants and the structure of their associated rhizobacterial communities were examined in an oil-contaminated site. The number of plant species that formed a community or were scattered was 24. The species living in soil highly contaminated with total petroleum hydrocarbon (TPH) (9,000-4,5000 mg/g-soil) were Cynodon dactylon, Persicaria lapathifolia, and Calystegia soldanella (a halophytic species). Among the 24 plant species, the following have been known to be effective for oil removal: C. dactylon, Digitaria sanguinalis, and Cyperus orthostachyus. Denaturing gradient gel electrophoresis (DGGE) profile analysis showed that the following pairs of plant species had highly similar (above 70%) rhizobacterial community structures: Artemisia princeps and Hemistepta lyrata; C. dactylon and P. lapathifolia; Carex kobomugi and Cardamine flexuosa; and Equisetum arvense and D. sanguinalis. The major groups of rhizobacteria were Beta-proteobacteria, Gamma-proteobacteria, Chloroflexi, Actinobacteria, and unknown. Based on DGGE analysis, P. lapathifolia, found for the first time in this study growing in the presence of high TPH, may be a good species for phytoremediation of oil-contaminated soils and in particular, C. soldanella may be useful for soils with high TPH and salt concentrations. Overall, this study suggests that the plant roots, regardless of plant species, may have a similar influence on the bacterial community structure in oil-contaminated soil.
The palmitoyl acyl carrier protein (ACP) specific thioesterase (EC 3.1.2.14) from Iris pseudoacorus was purified and characterized. The thioesterase which was very unstable in relatively high salt concentrations was eluted using a co-gradient of Triton X-100 and low concentration of KCl or Na-phosphate from Q-Sepharose, DEAE-Sepharose, and hydroxyapatite chromatography. SDS-PAGE analysis showed a single band with a molecular weight of 35,000. The native molecular weight of approximately 37,000 was estimated by Sephacryl S-200 chromatography, indicating that the enzyme is a monomer. The thioesterase activity was inhibited about 75% and 50% by N-ethylmaleimide (2 mM) and phenylmethylsulfonyl fluoride (2 mM). respectively. The N-ethylmaleimide-inactivation was protected by sodium palmitate but the inactivation with phenylmethylsulfonyl fluoride was not protected. Oxidation of thiols by 2 mM 5.5'-dithio-bis-(2-nitrobenzoic acid) resulted in 65% inactivation of the enzyme. These results suggest that a cysteinyl residue is essential to the catalytic reaction of the enzyme. The enzyme activity was increased by sodium citrate and also by $Cu^{2+}$

Experimental investigations have been made to study the double-diffusive nature of convection of an initially stratified salt-water solution due to heating from below in a rotating cylindrical cavity. The objective is to examine the flow phenomena and the heat transfer characteristics according to the changes in temperature gradient, concentration gradient and rotating velocity of cavity. Thermal and solutal boundary conditions at side wall are adiabatic and impermeable, respectively. The top and bottom plate are maintained each at constant temperature and concentration. The cavity is put into a state of solid body rotation. Like the stationary case, the types of initially-formed flow pattern are classified into three regimes depending on the effective Rayleigh number and Taylor number; stagnant flow regime, single mixed-layer flow regime and successively formed multi-mixed layer flow regime. At the same effective Rayleigh number, the number of initially-formed mixed layer and its growth rate decrease as the effect of rotation increases. The temperature and concentration profiles are both uniform in each layer due to convective mixing in the layered-flow regime, but look both liner in stagnant flow regime and single mixed-layer flow regime. At the interface between adjacent layers, the temperature changes smoothly but the concentration changes rapidly.
An improved procedure for the rapid purification of glucose-6-phosphate dehydrogenase from extracts of Saccharomyces cerevisiae was developed by using affinity chromatography. Among six affinty media tested, NADP$^{+}$-agarose and Affi-gel Blue were more effective than others (i.e., Affi-gel Red, AMP-agarose, ATP-agarose, and NAD$^{+}$-agarose). Conditions to desorb the enzyme bound to the affinity media were examined to increase the purity as well as yield. The best result was obtained when the column was developed with a linear gradient of KCl (0-1.0M). In case of Affi-gel Blue, introduction of NAD$^{+}$ (15mM) washing step prior to the salt gradient was most effective to remove NAD$^{+}$-binding proteins. For a large scale preparation of G-6-P dehydrogenase higher recovery was obtained by Affi-gel Blue than NADP$^{+}$-agarose, however, the purity of the enzyme was decreased by 10 times if the former was used as the affinity medium. The capacity of Affi-gel Blue for G-6-P dehydrogenase was found to be 5 times higher than that of NADP$^{+}$-agarose. Furthermore Affi-gel Blue could be reused repeatedly and its preparation is relatively easier and less expensive than NADP$^{+}$-agarose.X> +/-agarose.

At the interface between adjacent layers, the temperature changes smoothly but the concentration changes rapidly. The layers expand by diffusion of concentration through the interface along with its random fluctuation.
A new method for the simultaneous determination of heavy metal ions in cigarette material by microwave digestion and reversed-phase high-performance liquid chromatography (RP-HPLC) has been developed. The cigarette material was digested by microwave digestion. Lead, cadmium, mercury, nickel and tin ions in the digested samples were pre-column derivatized with tetra-(2-chlorophenyl)-porphyrin ($T_2$-CPP) to form color chelates, which were then enriched by solid phase extraction with a $C_{18}$ cartridge. The chelates were separated on a Waters Xterra$^{TM}RP_{18}$ column by gradient elution with methanol (containing 0.05 mol/L pyrrolidine-aceticacid buffer salt, pH = 10.0) and acetone (containin0.05 mol/L pyrrolidine-acetic acid buffer salt, pH = 10.0)as mobile phase at a flow rate of 0.5mL/min and analyzed with a photodiode array detector from 350-600 nm. The detection limits of lead, cadmium, mercury, nickel and tin were 4,3,3,8 and 5 ng/L, respectively, in the original samples. This method was afforded good results.
An ion-association chromatography was applied for the separation and determination of individual rare earth elements (REE) contained in mineral monazite. Prior to the determination, the group separation of REE was achieved by a cation exchange column of Dowex 5OW-X8 resin. The quantitative recovery of REE by the resin column, free from coexisting elements in monazite, was confirmed with radioactive tracers as well as with ICP-MS. Individual REE at ppm level was separated on reversed-phase column ($\mu$-Bondapak $C_{18}$) using gradient elution from 0.05 to 0.3 M $\alpha$-hydroxyisobutyric acid at pH 4.6. The individual REE was detected at 546 nm following post-column reaction with PAR (4-(2-pyridylazo)-resorcinol monosodium salt).

Assessing for flora distribution is necessary for land management and environmental research in reclaimed lands. This study was conducted to find out the relationship between vegetation distribution and soil salinity on three reclaimed tidal flats of Kyonggi-bay in the mid-west coast of Korea. We investigated the soil salinity and identified the vegetation at the continuum distribution spots, and describe the characteristics of continuum distribution. On the reclaimed tidal flats, spatial variation of vegetation formed partially, however as the result for connection of each spatial variation along with the soil salinity, continuum distribution formed and it was overlapped edaphic gradient with vegetation distribution, it means that the continuum distribution correspond with soil salinity gradient, as the evidence high salt tolerance species occurred at high saline spots, non salt tolerance species occurred at low saline spots. On the aged reclaimed tidal flats, continuum type was various and also clearly distinguished but it was not clear on the early stage of reclamation. The continuum distribution distinguished sequential and non-sequential type. Sequential type started from high saline zone and connected to low saline zone gradually, on this type, vegetation changed from pioneer halophyte to facultative halophyte and glycophyte along with the salinity gradient. Non-sequential type formed by non-sequential change of soil salinity, on this type, vegetation distribution was non-regular form because it has not changed gradually. In the aged reclaimed land, vegetation wilted zone existed with high salinity, and continuum distribution started from this zone with bare patch.
In the elastic wave equations, both horizontal and vertical displacements are defined. Since we can measure both the horizontal and vertical displacements in field acquisition, these displacements compose a displacement vector. In this study, we propose a frequency-domain elastic waveform inversion technique taking advantage of the magnitudes of displacement vectors to define objective function. When we apply this displacement-vector objective function to the frequency-domain waveform inversion, the inversion process naturally incorporates the back-propagation algorithm. Through the inversion examples with the Marmousi model and the SEG/EAGE salt model, we could note that the RMS error of the solution obtained by our algorithm decreased more stably than that of the conventional method. Particularly, the density of the Marmousi model and the low-velocity sub-salt zone of the SEG/EAGE salt model were successfully recovered. Since the gradient direction obtained from the proposed objective function is numerically unstable, we need additional study to stabilize the gradient direction. In order to perform the waveform inversion using the displacementvector objective function, it is necessary to acquire multi-component data. Hence, more rigorous study should be continued for the multi-component land acquisition or OBC (Ocean Bottom Cable) multi-component survey.

The community dynamics of benthic diatoms in the hypersaline environment are investigated to advance our understanding how salinity impacts marine life. Diatoms were sampled in the two salterns encompassing salt Ponds, ditches, and seawater reservoirs (n = 11), along the salinity gradient (max = 324 psu), and nearby tidal flats (n = 2). The floral assemblages and distributions across sites and stations showed great variations, with a total of 169 identified taxa. First, not surprisingly, higher diversity of benthic diatoms was found at natural tidal flats than salterns. The saltern diatoms generally showed salinity dependent distributions with distinct spatial changes in species composition and dominant taxa. Biota-environment and principal component analysis confirmed that salinity, mud content, and total nitrogen were key factors influencing the overall benthic community structure. Some dominant species, e.g., Nitzschia scalpelliformis and Achnanthes sp. 1, showed salinity tolerance / preference. The number of diatom species at salinity of >100 psu reduced over half and no diatoms were found at maximum salinity of 324 psu. The highest salinity for the observed live diatoms was 205 psu, however, a simple regression indicated a theoretical salinity threshold of ~300 psu on the survival. Finally, the indicator species were identified along the salinity gradient in salterns as well as natural tidal flats. Overall, high species numbers, varying taxa, and euryhaline distributions of saltern diatoms collectively reflected a dynamic saltern ecosystem. The present study would provide backgrounds for biodiversity monitoring of ecologically important microalgal producers in some unique hypersaline environment, and elsewhere.
