Tocopherols are essential lipophilic antioxidant in human cells, while little is known about its function in plant tissues. To study the impact of composition and content of tocopherols on stress tolerance, tobacco (Nicotiana tabacum) was transformed with a construct containing a cDNA insert encoding $\gamma$-tocopherol methyltransferase ($\gamma$-TMT/VTE4) from perilla under the control of the cauliflower mosaic virus (CaMV) 35S promoter. The transgenic tobacco was confirmed by PCR and RT-PCR. The total content and composition of tocopherols in the transgenic lines were similar with wild type controls. However, chlorophyll-a and carotenoid content in the transgenic lines were increased by up to 45% (P<0.01) and 39% (P<0.02), respectively. Also, the over-expression of $\gamma$-TMT increased the salt stress tolerance in tobacco plants. These results demonstrate that over-expression of $\gamma$-TMT gene in tocopherol bio-synthetic pathway can increase salt stress tolerance and contents of chlorophyll-a and carotenoid in transgenic tobacco plants.

Screening for drought and salinity tolerance was undertaken for selected Philippine upland rice landraces during germinative and seedling stages to identify varieties which can potentially be grown in marginally dry and saline soils. While increasing PEG and NaCl concentrations caused obvious signs of injury to all rice genotypes, considerable varietal differences were noted in the nature of responses providing evidence that these genotypes possess broad intraspecific genetic variations for drought and salt tolerance. Inconsistent responses of these varieties during both growth stages highlight complexities involved in stress responses and underscore the futility of utilizing a single stage in the rice plant's life cycle for physiological screening. Notwithstanding these perplexing responses, G_Katiil and Ml-Pilit Tapul were observed to thrive relatively well despite increased salt and drought stress during early growth stages and may therefore possess genes needed in crop improvement efforts for drought and salinity tolerance. While these results do not reflect the entire spectrum of adaptive expression to drought and salinity stress during the life cycle of the upland rice plant, they nonetheless provide an easy, reliable and reproducible method for preliminary identification of drought and salt tolerant rice varieties.
Barnyard millet (Echinochloa crusgalli var. frumentacea) has been detected as the best fodder crop in Korea for earlier and/or later season crop. The objective of the study was to examine varieal differences among the varieties introduced from ICRISAT and cultivated in Korea for salt tolerance and green fodder and grain yield under the saline and normal upland soil conditions, IEC 514, IEC 515, IEC 518 and IEC 528 introduced from ICRISAT were relatively highly tolerant to saline soil and with higher green fodder and grain yields, and also were very earlier headed and matured as compared with the Korean local varieties and King.
Abiotic stress conditions such as cold, drought, and salinity trigger physiological and morphological changes and yield loss in plants. Hence, plants adapt to adverse environments by developing tolerance through complex regulation of genes related to various metabolic processes. This study was conducted to construct a coexpression network for multidirectional analysis of salt-stress response genes in Brassica rapa (Chinese cabbage). To construct the coexpression network, we collected KBGP-24K microarray data from the B. rapa EST and microarray database (BrEMD) and performed time-based expression analyses of B. rapa plants. The constructed coexpression network model showed 1,853 nodes, 5,740 edges, and 142 connected components (correlation coefficient > 0.85). On the basis of the significantly expressed genes in the network, we concluded that the development of salt tolerance is closely related to the activation of $Na^+$ transport by reactive oxygen species signaling and the accumulation of proline in Chinese cabbage.
Under a typical Mediterranean environment in southern Australia, the evaporation rate increases significantly in hot summers, resulting in highly saline drinking water for grazing animals. Also in the cropping areas, dryland salinity is a problem. Grazing animals under these environments can ingest excessive amount of salt from feed, drinking water and soil, which can lead to a reduction in growth rate. To understand the impact of high salt intake on grazing deer, two experiments were conducted to assess the effect of salt levels in drinking water on feed intake and growth rate of red and fallow weaner deer. The results revealed that fallow deer did not show any abnormal behaviour or sickness when salt level in drinking water was increased from 0% to 2.5%. Feed intake was not affected until the salt content in water exceeded 1.5%. Body weight gain was not affected by 1.2% salt in drinking water, but was reduced as salt content in water increased. Compared with deer on fresh water, the feed intake of red deer on saline water was 11-13% lower when salt level in drinking water was 0.4-0.8%. An increase in salt level in water up to 1% resulted in about a 30% reduction in feed intake (p<0.01). Body weight gain was significantly (p=0.004) reduced when salt level reached 1.2%. The deer on 1% salt tended to have a higher (p=0.052) osmotic pressure in serum. The concentration of P, K, Mg and S in serum was affected when salt level in water was over 1.0%. The results suggested that the salt level in drinking water should be lower than 1.2% for fallow weaner deer and 0.8% for red weaner deer to avoid any reduction in feed intake. Deer farmers need to regularly test the salt levels in drinking water on their farms to ensure that the salt intake of grazing deer is not over the levels that deer can tolerate.
Tests show that the growth of lactic bacteria in kimchi varies according to the type and concentration of salt used. Weissella confusa, the early stage bacteria in kimchi fermentation, increased sharply after 5 hr of induction with 3% light salt and refined salt. However, the induction period lengthened to 12 hr with 3% sea salt and bamboo salt. Lactobacillus delbrueckii ss lactis and L. pentosus which grow in the middle stage of fermentation, and L. hamsteri, which grows at the end stage of fermentation, were found after 12 hr of induction with 3% salt of all kinds. When 5% light salt was added to the culture medium, the induction period of bacteria other than W. confusa lengthens to 12 hr. The trend is similar for sea salt, bamboo salt, and refined salt, with a higher NaCl concentration resulting in less growth. W. confusa showed salt tolerance, but L. hamsteri was affected by the type and concentration of salt. With 5% sea salt, bamboo salt, and refined salt, the growth of bacteria was inhibited by up to 24 hr.
Reclaimed tidal area is a great agricultural resource in the world. Improvement and utilization of reclaimed soil is an important measure for expanding land resource. This study was conducted to evaluate relative salt-tolerance of plants and its effect for improving saline soil. Eighteen tolerance plants were selected from China, Pakistan and Korea the climate of which is different. The emergence of different varieties in reclaimed soil was in order as FL478>Barnyard grass>Pokkali>Atriplex>Sesbania>Rumex>Alfalfa>Tall Fescue>Ryegrass>Sudan grass. Four varieties, Barnyard grass, Sesbania, Atriplex and Limonium were selected for soil improvement in reclaimed land. Cultivation of Sesbania, Barnyard grass and Atriplex were good to soil physico-chemical quality. Also these plants increased soil organic matter contents and reduced soil salt concentration. Organic matter contents of cultivated soils of Sesbania, Barnyard grass and Atriplex were $4.10g\;kg^{-1}$, $4.60g\;kg^{-1}$ and $2.81g\;kg^{-1}$ respectively. On the other hand organic matter content of uncultivated soils was $2.65g\;kg^{-1}$. As Sesbania and Barnyard grass were applied to cultivated soil like green manure, bulk density improved from $1.42Mg\;m^{-3}$ to $1.39Mg\;m^{-3}$.
Recently the increasing of vinyl and green houses and development of reclaimed land including Saemangeum induced the need for breeding salt-tolerant crops which can survive and grow in high salinity soil. So we try to develop salt-tolerant transgenic chrysanthemum (Dendranthema grandiflorum.) lines by using anti-porter gene TANHX and HVNHX. Through marker selection and plant regeneration step, we could get 284 putative transgenic chrysanthemum lines. On selected putative transgenic plants, 40 candidates were used for genetic analysis and 30 lines could be made up of target size band on PCR, so about 75% of marker selected lines were decided as real transgenic lines. Selected 284 transgenic lines were also used for salt-tolerance test as a range of NaCl 0.2 ~ 1.2% (300 mM). As a result of salt-tolerance test, 15 selected transgenic lines could live and grow on the continuous supply of 0.8% (200 mM) NaCl solution and another 7 lines were could survive under 1.2% (300 mM) NaCl solution. This salt-tolerant transgenic lines under salt stress also lead a cell alternation especially a guard cell. A stressed guard cell be swelled and grow larger in proportion to NaCl concentration. TTC test for cell viability on transgenic chrysanthemum lines pointed out that more strong salt-tolerant lines can be live more than another under same salt stress. The numerical value of strong salt-tolerant 7 transgenic lines were 0.206 ~ 0.331 under 1.2% NaCl stress, and then it's value is more larger than middle salinity lines' 0.114 ~ 0.193 and non-transgenic's 0.046. And the proline contents as indicated stress compound also pointed out that HVNHX introduced salt-tolerant transgenic lines were less stressed than other under same salt stress. The contents of strong salt-tolerant transgenic lines were 2.255 ~ 2.638 mg/kg and it is much higher than that of middle salinity lines' 1.496 ~ 2.125.
This study was conducted to obtain the basic information on salt-tolerance of rice cultivars in relations with growth retardation and cation content. As the salt-level increased, less retardation in plant height and dry weight were shown in salt-tolerant than salt-susceptible rice cultivars. Salt tolerant cultivars showed lower N $a^{+}$ and higher $K^{+}$ content and lower N $a^{+}$ / $K^{+}$ ratio at each salt-levels than salt-susceptible ones, while there were no significant differences in $Ca^{++}$ and $Mg^{++}$ content. At the tillering stage, the plant height and dry weight of the salt-treated plots were significantly correlated with N $a^{+}$ and $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio, implying that N $a^{+}$ and $K^{+}$ content could be an indicator of salt-tolerance of a rice cultivar. There were no tiller-depending differences in cation content in all cultivars. N $a^{+}$ content and N $a^{+}$ / $K^{+}$ ratio in leaves were lower at the top and higher at the bottom. In three $F_1$ hybrids between salt tolerant parent Pokkali and three salt susceptible parents, plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio were similar to those of Pokkali parent, while N $a^{+}$ content was intermediate of the parents. So, it seemed that salt tolerance is dominant over salt susceptibility. In $F_2$ of Pokkali/wx817 cross, genetic segregation of plant height, dry weight, $K^{+}$ content and N $a^{+}$ / $K^{+}$ ratio varied continuously and was biased onto Pokkali side, while that of N $a^{+}$ content showed normal distribution with intermediate mode. Broad-sense heritability of the characters ranged from 0. 604 to 0. 811.
