송화 메탄올 추출물의 항산화적 항돌연변이 효과 (Antimutagenic and Antioxidative Effects of Methanol Extract of Pine Pollen)

  • 직ㆍ간법 변이원과 산화적 돌연변이원에 대한 송화분 추출물의 항돌연변이원성에 대하여 Ames test로 조사하였다. 메탄을 추출물을 이용하여, 10종의 돌연변이에 대한 돌연변이 억제효과를 검색한 결과 S. typhimurium TA98에서는 daunomycin, AFB$_1$과 Trp-P-1의 변이원성에 대해서만 각각 17.8, 82.2, 80.9%의 돌연변이 억제효과를, S. typhimurium TA100에서는 AFB$_1$의 변이원성에 대해서만 72.3%의 돌연변이 억제효과를 나타내었으며, S. typhimurium TA102, 104에서 t-과산화부틸과 $H_2O$$_2$의 돌연변이원성에 대한 메탄을 추출물의 돌연변이 억제 효과는 t-과산화부틸을 돌연변이원으로 한 S. typhimurium TA102에서 16.3%의 억제효과를 선택적으로 나타내었다. 따라서 메탄을 추출물을 클로르포름, 에틸아세테이트, 부탄올, 물 순으로 순차용매 분리한 다음, 이 분획물에 대한 항돌연변이원성을 검색하였다. 그 결과 S. typhimurium TA98에서 daunomycin, AFB$_1$과 Trp-P-1에 대해서 플로로포름 분획물은 각각 55.6, 93.7, 93.5%, 에틸아세테이트 분획물은 각각 11.4, 74.3, 85.2%, 부탄을 분획물에서는 10.9, -5.6, 6.8%, 물 분획물은 각각 -3.4, -3.8, 4.6%의 돌연변이 억제효과를 나타내었다. S. typhimurium TA100에서는 AFB$_1$에 대해 클로로포름 분획물과 에틸아세테이트 분획물이 각각 95.1, 62.5%의 억제효과를 나타내었으며, S. typhimurium TA102에서는 t-과산화부틸에 의해 유도된 산화적 돌연변이에 대해 클로로포름 분획물이 93.6%로 매우 강한 억제효과를 나타내었으며, 기타 분획물에서는 매우 낮은 억제효과를 나타내었다.

  • To evaluate the genotoxicity of CKD-602, an anticancer agent the in viかo reverse mutation assay using Salmonella typhimurium, the Chromosome aberration assay using Chinese hamster lung (CHL) cells and the in vivo micronucleus assay using bone marrow cells of ddY mice were performed. In the reverse mutation assay, CKD-602 did not induced mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535, and TA 1537 strains with and without metabolic activation. In the chromosome aberration test using CHL cells, there was an increased incidence of structural aberrations induced by CKD-602 without metabolic activation during 24 and 48 hours, but CKD-602 did not induce chromosome aberration with metabolic activation. The in vivo induction of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ddY mice. At 24 hours after treatment with CED-602 by i.p. once, there was an increased incidence of micronucleated polychromatic erythrocytes in bone marrow of ddY male mice.

옻나무 추출액(Rhus-II)의 안전성에 관한 유전독성학적 평가 (Genotoxicicological Safety Estimate for the Rhus-II)

  • These observations were performed to investigate the safety of the natural herbs (Rhus-II) in respect of genotoxicity. This substance was examined in two in-vitro tests: (1) Salmonella typhimurium reversion assay (Ames test) in strain TA 98, TA 100, TA 1535 and TA 1537, (2) in vitro chromosome aberration test in cultured Chinese hamster ovary (CHO) cells. In the reverse mutation test, Rhus-II did not induced mutagenicity in Salmonella typhimurium reversion assay(Ames test) with or without metabolic activation. In the chromosome aberration assay using CHO cells, there was no increased incidence of structural and numerical aberrations with or without metabolic activation. These results indicated that, the Rhus-II had no genotoxicity.

  • Hypertension is a serious health problem due to high frequency and concomitant other diseases including cardiovascular and renal dysfunction. Olmesartan cilexetil is a new antihypertensive drug associated with angiotensin II receptor antagonist. This study was conducted to evaluate the mutagenicity of olmesartan cilexetil by bacterial reverse mutation test using Salmonella typhimurium (TA100, TA1535, TA98, and TA1537) and Escherichia coli (WP2 uvrA). At the concentrations of 0, 62, 185, 556, 1667, and 5000 ${\mu}g$/plate, olmesartan cilexetil was negative in both Salmonella typhimurium and Escherichia coli regardless of presence or absence of metabolic activation system (S9 mix). These results demonstrate that olmesartan cilexetil does not induce bacterial reverse mutation.

합성화학물질들의 유전독성평가(Ⅸ) -합성 제초제 Pretilachlor - (Evaluation of the Genetic Toxicity of Synthetic Chemicals (Ⅸ) a Synthetic Selective Herbicide, Pretilachlor-)

  • Pretilachlor [2-chloro-N -(2, 6-diethylphenyl)-N-(2-propoxyethyl) acetamide, $C_{17}$H$_{26}$ClNo$_2$, M.W.=311.9, CAS No.51218-49-6]는 제초제의 일종으로 본 연구에서는 박테리아 복귀 돌연변이 시험과 포유동물 세포를 이용한 염색체 이상 시험 및 마우스를 이용한 in vivo소핵 시험을 수행하여 pretilachlor의 유전독성을 평가하였다. 박테리아 복귀 돌연변이 시험에서 pretilachlor는 Salmonella typhimurium TA98, TA 100, TA1535, TA1537 균주의 대사 활성계 존재 및 부재시 313-5,000$\mu\textrm{g}$/plate의 범위에서 농도 의존적인 돌연변이 율의 증가를 관찰할 수 없었다. 또한 포유동물 세포인 Chinese hamster lung (CHL) fibroblast를 이용한 염색체 이상 시험에서 pretilachlor는 대사 활성계 존재 및 부재시 1.56-6.24$\mu\textrm{g}$/mL의 농도에서 clastogenicity를 보이지 않았고, 137.5-550.1 mg/kg의 pretilachlor를 복강 주사한 마우스의 골수세포를 이용한 in vivo소핵 시험의 결과에서도 통계적으로 유의한 소핵 유발능을 관찰할 수 없었다었다

Host-Mediated Assay를 이용한 감궁탕의 돌연변이원성 평가 (Evaluation of Mutagenicity with Gamgung-tang Using Host-Mediated Assay)

  • Mutagenicity of Gamgung-tang (GGT) was tested using in vitro S-9 mixture in vitro host-mediated assay with Salmonella typhimurium. In the previous reports, GGT was tested for the safety using Ames(-S-9), Bacillus subtilis Rec, and umu gene expression mutagenicity tests. Mutagenic activity in any assays we tested was not found. In this report, we further investigated safety of GGT after metabolic activation in vivo. Ames test with S-9 mixture and host-mediated assay with Salmonella typhimurium TA98 were used to identify metagenic property of GGT. GGT was administered 3 times with i.m. to Balb/c mice did not induced mutagenic effect in Salmonella typhimurium TA98 recovered from the liver after 3.5h with i.p. treatment. Over the entire dose range $(3{\sim}150mg/mouse)$ tested no toxicity was detected to the bacterial cells. These results suggest that there was no DNA damage and mutagenicity by GGT.

Salmonella typhimurium Strain TA98, 100에서 감잎차, 녹차, 우롱차 추출물의 돌연변이 억제 효과 (Antimutagenic Effects of Water Extracts of Persimmon Leaf Tea, Green Tea and Oolong Tea on Reversion and Survival of Selected Salmonella Tester Strains)

  • Water extracts of persimmon leaf tea(PLTE), green tea(GTE) and oolong tea(OTE), at the con centration used for human consumption, were examined for inhibitory effects on the mutagenicity of major classes of dietary and environmental mutagens including indirect acting mutagens, B[ ]P (benzo[ ]pyrene), IQ(2 amino 3 methylimidazo[4,5 f]quinoline), 2 AA(2 aminoanthracene) in the presence of S9 mix and direct acting mutagen, 4 NQO(4 nitroquinoline 1 oxide) without S9 mix, using the modified Ames Salmonella/microsome assay. PLTE, GTE and OTE showed very potent and concentration dependent antimutagenic effects against indirect acting mutagens B[ ]P and IQ. At the maximum concentration(16,200 g/plate) of each tea extract, number of colonies decreased in a dose dependent manner up to 82~100%. Similar inhibition of PLTE, GTE and OTE were seen at higher concentration in the mutagenicity of the 2 AA following an initial increase in the activity at lower concentration. However, the mutagenicity of the direct acting mutagen 4 NQO were not suppressed at lower concentration of the three tea extracts, and higher concentration of the tea extracts enhanced mutagenic activity of the mutagen. There were no differences in the mode of antimutagenesis between PLTE, GTE, and OTE, in both Salmonella typhimurium TA98 and TA100 strains against the same mutagen. In conclusion, the water extracts of persimmon leaf tea, green tea and oolong tea possess marked antimutagenic potential against a variety of important dietary and environmental indirect acting mutagens, but the activity was not observed against the direct acting mutagens. These results suggest that the mode of inhibitory action may not have resulted from direct interaction between tea extracts and the mutagens, but rather from indirect metabolic inactivation of mutagens by tea extracts.

Ames 및 umu assay를 이용한 감궁탕의 안전성평가 (Evaluation of Safety with Gamgung-tang Using Ames and umu Assays)

  • 감궁탕의 돌연변이원성을 유무를 알아보기 위해 Salmonella typhimurium TA 98 및 TA100을 이용한 돌연변이 원성 실험에서도 감궁탕은 어느 균에서도 돌연변이원성을 나타내지 않았으며, 이는 S-9 mixture 의해 감궁탕이 대사가 된 후에도 이와 유사한 경향을 나타내었다. 또한, SOS umu test의 경우에서도 $\beta-galactosidase$활성에는 별다른 영향을 미치지 않는 것으로 보아 감궁탕은 돌연변이원성을 일으키지 않는 것으로 판정되었으며 S-9 mixture처리에 의한 대사 후에도 이와 유사한 실험결과가 나타났다. 따라서 감궁탕은 그 자체 및 대사 후에도 DNA에 별다른 영향을 미치지 못하는 비교적 안전한 생약처방으로 여겨진다.

Mutagenic and Clastogenic Activities of the Browning Reaction Model Systems

  • Two short-term bioassays were employed to asses the mutagenic and clastogenic activities in browning reaction of pentose-creatine, pentose-glycine and pentose-creatine-glycine browning reaction model system. Methylene chloride extract of rhamnose-creatine-glycine browning reaction exhibited the strongest mutagenicity toward Salmonella typhimurium TA98 with S-9. Methylene chloride extract of pentose-creatine and pentose-glycine browning reaction solutions was also tested for mutagenicity, with positive responses. Methylene chloride extract of pentose-creatine-glycine browning reaction solutions induced significant increase in chromosome aberrations in the treated Chinese hamster ovary(CHO) cells. Each of pentose-creatine and pentose-glycine browning reaction solutions induced a relatively low frequency of chromosome aberrations in the treated cells.
