In Vitro Selection of RNA Aptamer Specific to Salmonella Typhimurium

  • Salmonella is a major foodborne pathogen that causes a variety of human diseases. Development of ligands directly and specifically binding to the Salmonella will be crucial for the rapid detection of, and thus for efficient protection from, the virulent bacteria. In this study, we identified a RNA aptamer-based ligand that can specifically recognize Salmonella Typhimurium through SELEX technology. To this end, we isolated and characterized an RNase-resistant RNA aptamer that bound to the OmpC protein of Salmonella Typhimurium with high specificity and affinity ($K_d$ ~ 20 nM). Of note, the selected aptamer was found to specifically bind to Salmonella Typhimurium, but neither to Gram-positive bacteria (Staphylococcus aureus) nor to other Gram-negative bacteria (Escherichia coli O157:H7). This was evinced by aptamer-immobilized ELISA and aptamer-linked precipitation experiments. This Salmonella species-specific aptamer could be useful as a diagnostic ligand against pathogen-caused foodborne sickness.

  • 우유내 Salmonella를 검출하기 위한 Sandwich ELISA의 특이성을 조사하였다. Sandwich ELISA에 사용한 항체들은 OMP 분획을 닭에 면역주사하여 얻은 IgY와 OMP 분획을 gel filtration하여 얻은 분자량 40,000의 OMP를 토끼에 면역 주사하여 얻은 토끼 IgG를 사용하였다. Immnoblot assay에서 IgY는 분자량 6,000의 OMP에 강하게 반응하였으며 토끼 IgG는 분자량 40,000, 35,000과 6,000의 OMP들에 강하게 반응하였다. IgY와 토끼 IgG는 Salmonella typhimurium의 다른 단백질에도 반응하였다. Competitive ELISA에서 IgY가 Salmonella의 두 개 균주에 대해 특이성을 나타냈으며 Eshcherchia coli와 Yersinia enterocolitica에 의하여서는 반응을 나타내지 않았다. 우유에 세균을 첨가하여 실시한 sandwich ELISA에서 Salmonella typhimurium 2균주가 가장 높은 흡광도를 보였다. Salmonella cholerasuis 균주들은 상대적으로 흡광도가 낮았으며 이루 일부 Salmonella cholerasuis 균주들은 비 Salmonella 균주들과 차이가 없었다.

Protection Against Salmonella Typhimurium, Salmonella Gallinarum, and Salmonella Enteritidis Infection in Layer Chickens Conferred by a Live Attenuated Salmonella Typhimurium Strain

  • In the present study, we investigated the protection conferred by a live attenuated Salmonella enterica serovar Typhimurium (ST) strain against Salmonella Typhimurium, Salmonella Gallinarum (SG), and Salmonella Enteritidis (SE) infection in layer chickens. Birds were orally primed with the attenuated ST strain at 7 days of age and then boosted at 4 weeks post prime immunization (PPI). Sequential monitoring of plasma IgG and mucosal secretory IgA (sIgA) levels revealed that inoculation with ST induced a significant antibody response to antigens against ST, SE, and SG. Moreover, significant lymphoproliferative responses to the 3 Salmonella serovars were observed in the immunized group. We also investigated protection against virulent ST, SE, and SG strain challenge. Upon virulent SG challenge, the immunized group showed significantly reduced mortality compared to the non-immunized group. The reduced persistence of the virulent ST and SE challenge strains in the liver, spleen, and cecal tissues of the immunized group suggests that immunization with the attenuated ST strain may not only protect against ST infection but can also confer cross protection against SE and SG infection.

  • 식중독 미생물 Salmonella typhimurium ATCC 14028, Staphylococcus aureus KCTC 2199, Vibrio parahaemolyticus ATCC 17802에 대한 은 이온의 항균력을 PAper Disk 방법과 액체 배양법에서 조사하였다. S. typhimurium와 V. parahaemolyticus의 경우 각각 2,10ppm의 은 이온 농도에서 생육저해환이 발견되었으며 농도가 증가함에 따라 생육 저해의 정도가 증가하였다. 그러나 S.aureus는 20 ppm의 은 이온 농도에서도 생육 저해환이 관찰되지 않았다. 액체 배양에서 은 이온의 항균 능력은 S. typhimurium, V.parahaemolyticus, S.aureus에 대해서 각각 03 ppm, 0.5 ppm, 5 ppm 이상에서 관찰되었고, 은 이온의 농도가 증가함에 따라 생육 저해는 크게 나타났다.

  • The result of studying the pathogenicity of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province were summarized as follows. In Congo-red binding test, S typhimurium had much more rough types than S enteritidis. In colicin production test, 4 strains of S typhimurium were positive but all of S enteritidis were negative. In hemolysin production test, all of S typhimurium and S enteritidis were negative. In Guinea pig serum resistant test, all of S typhimurium and S enteritidis were positive. As a result of pathogenicity test to mice, 54.4% of mice were died. Therefore, S typhimurium and S enteritidis were considered as highly pathogenic. S typhimurium DT104 and S enteritidis PT4 were more pathogenic to mice than other phage types of same serovar. S typhimurium and S enteritidis were considered not so pathogenic for 6-day-old chickens. The recovery rates of Salmonella stains from mice and chickens inoculated were 96.8%, and 54%, respectively. In chickens, proportional to the time from 2 weeks after challenge inoculation, the recovery rates were noticeably decreased.

초생추 유래 Salmonella속균의 생물학적 특성 (Studies on Salmonella isolated from chicks)

  • This paper deals with the distribution, reservoir and mode of spread of infection on 6 hatcheries in Taegu, Kyungpook and 5 broiler farms in Kyungpook during the period from June 1991 to June 1992. Isolated Salmonella were examined for serotypes, biotyping of Salmonella(S) typhimurtum, antibiotic susceptibility and some biochemical characteristics. Forty two Salmonella strains were isolated from 42(2.7% of 1,577 caecal samples of chicks, and their serotypes were S typhimurium 10, S typhimurium var Copenhagen 5, S infantis 4, S thompson 3, and untypable 20. The isolation rate of Salmonella varied from 0 to 5.1% in 6 hatcheries and that of Salmonella from 5 broiler farms was 10. 5%. Biotypes of 10 S typhimurium and 5 S typhimurium var Copenhagen strains isolated from chicks of hatcheries and broiler farms were biotype 2(86.6%), 8(6.7%), and 10a(6.7%), and 26i(6.7%) according to Duguid's scheme. Antibiotic susceptibility test of Salmonella isolated were performed by agar dilution method, using 9 antibiotics as follows: ampicillin(Am), chloramphenicol(Cm), gentamicin(Gm), kanamycin(Km), nalidixic acid(Na), rifampicin(Rf), streptomycin(Sm), sulfadimethoxine(Su), and tetracycline(Tc), All the strains were sensitive to RF. But 8 strans(23.8%) were resistant to one or more drugs and the most common resistance patterns of transferred R plasmids were SmSuGm and SmSu. Among 42 isolates, one had transferable citrate utilizing plasmid. S typhimurium and S typhimurium var copenhagen strains were resistant to killing by 90% normal guinea pig serum.

  • This study was performed to predict the multiplication growth patterns of Salmonella typhimurium according to the holding methods and times in cooked foods served at foodservice institutions. When simmered pork in soy sauce and ham and cucumber salad were inoculated with S. typhimurium (7.00 logCFU/g) and tested according to holding methods and times, room temperature holding showed a continuous increase in S. typhimurium count as time passed, whereas 80$^{\circ}C$ heating table sharply reduced the count to 4.00 (logCFU/g) after 2hrs of holding and to zero after 6hrs, which suggested that all S. typhimurium were destroyed. However, 5$^{\circ}C$ refrigerator holding showed a count increase, although it wasis still lower than that at room temperature and at 10$^{\circ}C$ cold table holding, which suggested that S. typhimurium is comparatively resistive to low temperature.

오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향 (Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours. Mousse cake와 Tiramisu에 인위접종된 Salmonella Typhimurium의 식품공전 분리배지, Real-time PCR과 Loop-mediated isothermal amplification-bioluminescence의 검출 특성 비교 (Comparison of Isolation Agar Method, Real-Time PCR and Loop-Mediated Isothermal Amplification-Bioluminescence for the Detection of Salmonella Typhimurium in Mousse Cake and Tiramisu)

  • 최근 한국에서 발생한 Salmonella로 인한 식중독 사고는 2018년 9월 학교급식에서 제공된 초콜릿 무스 케이크가 원인이 되었다. 이 연구의 목적은 Salmonella Typhimurium이 인위적으로 접종된 무스케이크와 티라미수에서 3M Molecular Detection Assay 2 - Salmonella와 식품공전에 등재된 방법인 분리배지와 real-time PCR을 비교하는 것이었다. 무스케이크 2종과 티라미수 2종 25 g에 225 mL BPW를 넣고 $37^{\circ}C$에서 24시간 동안 증균 배양하였다. 배양 후, 3M Molecular Detection Assay 2 - Salmonella, 분리배지 그리고 real-time PCR로 분석하였다. 초콜릿 무스 케이크를 제외하고 3가지 방법은 유사한 결과를 보였다. 초콜릿 무스 케이크에서 분리배지와 3M Molecular Detection Assay 2 - Salmonella는 모든 접종수준에서 동일한 결과를 나타낸 반면 real-time PCR은 $10^4CFU/25g$ 수준에서 1번의 양성결과를 제외하고 모두 검출되지 않았다. 초콜릿 무스에 S. Typhimurium을 $10^2CFU/25g$ 수준으로 접종하였을때, real-time PCR를 이용한 검출은 15%에서는 부분적인 음성을 나타냈고, 20-100% 함량의 초콜릿 무스에서는 모두 음성이었다. Real-time PCR로는 chocolate이 15% 이상 함유된 식품에서의 Salmonella균 검출이 불가능하였지만, LMAP 기반의 3M Molecular Detection Assay 2으로는 chocolate 농도에 관계없이 검출이 가능하였다.

사멸 살모넬라와 뱅코마이신 첨가 사료가 Salmonella typhimurium 인공 감염 육계 병아리의 생산성과 면역 반응에 미치는 영향

  • 사멸 살모넬라와 뱅코마이신 첨가 사료가 Salmonella typhimurium을 감염시킨 육계 병아리의 생산성과 면역계 및 항산화계에 미치는 영향을 조사하였다. 살모넬라 인공 감염전(27일)에는 육계 병아리의 생산성은 사료에 따른 유의한 영향이 없었다. 항산화계에서 사멸 살모넬라와 뱅코마이신 첨가 사료가 기초사료에 비하여 적혈구 세포액내 MnSOD 활성을 낮추었다. 살모넬라 인공 감염 7일후 육계 병아리의 생산성은 사멸 살모넬라와 뱅코마이신 첨가 사료가 대조감염구에 비하여 높은 일당증체량과 사료효율을 나타냈다. 살모넬라 인공 감염 7일후 및 15일후 사멸 살모넬라와 뱅코마이신 첨가 사료가 대조감염구에 비하여 적혈구 세포내 과산화물 분해효소의 활성을 낮춘 반면, LPS 자극에 의한 PBMC 증식도와 PBMC 증식에 분비된 IL-1수준을 높였다. 본 성적은 사료중 사멸 살모넬라와 뱅코마이신 첨가가 살모넬라 감염 육계 병아리 생산성 증가에 영향을 미쳤으며, 이는 항산화 효소계와 PBMC 증식도, IL-1 분비 및 혈장 TNF-a농도 등 타고난 면역계에 의한 것으로 생각된다.
