A comparative study of the prevalence of Helicobacter pylori in the oral biofilms of a group of dental and non-dental undergraduates from Sri Lanka

  • Dental health care workers (DHCW) are at a risk of occupational exposure to Helicobacter pylori from the aerosolized oral biofilms and saliva of patients. We designed this study to investigate the prevalence of H. pylori in the oral biofilms of a group of dental and non-dental undergraduates from Sri Lanka. After obtaining informed consent, oral biofilms were collected from 38 dental undergraduates (19 males and 19 females) undergoing clinical training and 33 non-dental undergraduates (14 males and 19 females). The participants were in the age range of 22-27 years and had healthy periodontium. Total DNA from the oral biofilms were extracted, and H. pylori DNA was detected using polymerase chain reaction (PCR) amplification of 16S rRNA gene of H. pylori using JW22-JW23 primers, and the results were confirmed using PCR amplification of H. pylori-urease specific HPU1-HPU2 primers. Out of 71 participants, 11 (28.95%) dental and 3 (9.09%) non-dental undergraduates had H. pylori in their oral biofilms indicating an overall prevalence rate of 19.72% (14/71). Thus, the prevalence of H. pylori in oral biofilms was significantly higher in dental undergraduates than in non-dental undergraduates (p < 0.05). An odds ratio of 4.07 indicated that dental undergraduates were four times more likely to harbor H. pylori in their oral biofilms than non-dental undergraduates. Foregoing data support the fact that there may be greater occupational risk of exposure to H. pylori for dental undergraduates during clinical training than that for non-dental undergraduates, warranting meticulous infection control practices during clinical dentistry.

Ruminal microbial responses in fermentation characteristics and dry matter degradability to TDN level of total mixed ration

  • An in vitro trial was conducted to examine the effects of total mixed rations (TMR) on fermentation characteristics and effective degradability (ED) by rumen microbes. Three TMR diets were growing period TMR (GR-TMR, 67% TDN), early fattening period TMR (EF-TMR, 75.4% TDN) and late fattening TMR (LF-TMR, 80% TDN). Three TMR diets (3 g of TMRs in each incubation bottles) was added to the mixed culture solution of stained rumen fluid with artificial saliva (1:1, v/v) and incubated anaerobically for 48 hours at $39^{\circ}C$. The pH in all incubation solutions tended to decrease up to 48h, but the opposite results were found in concentration of total gas production, ammonia-N and total VFA in all incubations.The total gas production (p<0.05) in LF-TMR was highest compared with those of other diets. Also, concentration of total VFA was tended to increase in LF-TMR compared with other TMR diets in all incubations. The EDDM in both EF-TMR and LF-TMR was tended to high compared with GR-TMR (p=0.100). In this in vitro trials, concentration of propionate in all incubation solution was not affected by increased concentration of TDN. The results of the present in vitro study indicate that TMR may provide more favorable condition for nutrient digestion both in the rumen.

  • Salivary proteins which are produced in the saliary acinar cells have been known to be involved in the Calcium and phosphate metabolism. The acquired pellicle resulting from such metabolism is considered as a secondary defence membrane against tooth caries. In this respect, some proteins included in saliva probably play an important role in the prevention of demineralization in enamel. On the other hand, fluoride has long been known to prevent the demineralization of enamel by the inhibition of the growth of Streptococcus mutans(S. mutans) and by the chemical reaction with calcium and phosphate, Therefore, I have examined the roles of amylase and albumin in the demineralization of enamel and compared these preteins with fluoride in terms of anticariogenic effect. 1. The demineralization caused by S. mutans occurred slowly and progressively for the first 60 min, then the rate of demineralization was accelerated afterwards. 2. pH decreased continuously during the entire period of each experiment. 3. The demineralization was significantly inhibited by the preteatment of amylase and fluoride but albumin had little effect on it. 4. An addition of 0.1 mM lactic acid (final concentration 0.1 ${\mu}M$) caused a rapid increase in calcium concentration reaching a maximum within 10 min. 5. pH decreased rapidly by the addition of 0.1 mM lactic acid and reached a minimum within a few seconds followed by an increase in pH. pH reaced a plateu with 10 min. 6. Fluoride, amylase and albumin played little role in the 0.1 mM lactic acid-induced demineralization. 7. A slow infusion of 0.1 M lactic acid at a rate of 5 ${\mu}l/min$ caused a slower increase in calcium concentration compared with the bolus addition of lactic acid. 8. Fluoride had an inhibitory effect on the calcium release caused by slow infusion of lactic acid while amylase and albumin had no effect on it. These results suggest that fluoride inhibits demineralization by protecting the HA from the acid attack whereas amylase has a direct effect on S. mutans to prevent demineralization.

국내 시판 우유와 저지방 우유의 Streptococcus mutans 세균막에 대한 우식원성 비교 (Comparison of Cariogenicity of Bovine Milk and Low-fat Milk on Streptococcus mutans Biofilm)

  • 본 연구의 목적은 국내 상품화된 일반 우유와 저지방 우유의 우식원성을 CDC Biofilm Reactor를 사용한 세균막 모델을 통해 비교하는 것이다. 소의 법랑질 시편에 Streptococcus mutans ATCC 25175 세균막을 형성하였다. 하루에 3번 일반 우유, 저지방 우유와 0.9% 생리식염수를 세균막에 노출시켰다. 시간의 흐름에 따른 배지의 pH 변화를 측정하였다. 실험 5일째 시편에서 세균막을 분리하여 세균의 집락 형성단위를 측정하였다. 세균막의 두께는 공초점현미경으로 관찰하였다. 실험 전, 후의 표면미세경도를 측정하여 시편의 미세경도변화율을 평가하였다. 세 군 간의 pH 변화 양상과 세균막 두께는 유사하였으며 미세경도변화율과 세균의 집락형성단위는 유의한 차이를 보이지 않았다(p > 0.05). 본 연구 결과, 일반 우유와 저지방 우유의 우식원성은 차이를 보이지 않았으며, 우유는 우식에 대해 안전한 식품임을 확인하였다.

A Study of a Sodium Bicarbonate Cotransporter Functionally Expressed in Xenopus laevis Oocytes

  • Purpose: $HCO_3{^-}$ is the most important ion to buffer the acidity of saliva. The transport of $HCO_3{^-}$ is mediated by electrogenic $Na^+/HCO_3{^-}$ cotransporter 1 (NBCe1), which expressed in various tissues including salivary glands, kidney and pancreas, etc. This experiment was performed to investigate regulatory site of NBCe1involved in the pH regulation using various mutants of NBCe1. Materials and Methods: Human parotid gland NBCe1 (hpNBCe1) and mutants by deletion of 1~285 bp and 1~1,035 bp were prepared. After microinjection of each cRNA to oocytes of Xenopus laevis, they were incubated for 2~3 days. The function of each protein was tested by electrophysiological method. Results: When oocytes were exposed to the $HCO_3{^-}$ buffered solution, 1~285 bp deleted mutant hpNBCe1 evoked a marked hyperpolarization ranging from -90 mV to -160 mV (average: -134 mV; n=12) compared to the full length of hpNBCe1. Although 1~1,035 bp deleted mutant hpNBCe1 was also expressed in the plasma membrane, but it did not show any changes of membrane potentials. Conclusion: Our deletion mutant study demonstrated that 1~285 bp of the NBCe1 is the major domain to determine $HCO_3{^-}$ transport ratio.

Peroxida System을 함유한 세치제가 생체에서 전타액에 미치는 영향에 대한 연구 (Effects of a Peroxidase-System-Containing Toothpaste on Whole Saliva in Vivo)

  • Peroxidase system은 구강내 고유의 항균체계로서 항세균물질인 $HOSCN/OSCN^-$을 생산하여 다양한 그램 양성 그리고 그램 음성 세균에 대해 살균 및 정균작용을 나타낸다. 본 연구는 peroxidase system을 함유한 치약을 사용한 후 건강한 대상자의 자극성 전타액내 생성되는 $HOSCN/OSCN^-$의 양을 측정하여 최대 생성시간을 결정하였다. 그 후 건강한 대상자에게 임의로 실험치약과 대조치약을 할당하여 2주간 사용하도록 한 후 $HOSCN/OSCN^-$ 최대 생성 시간에 타액을 채취하여 치약 사용전과 2주후의 타액분비량, pH, 점조도 그리고 S. mutans와 lactobacilli에 대한 항균효과를 비교하였다. 구강건조증 환자를 대상으로 한 두 번째 연구에서도 peroxidase system을 함유한 세치제를 사용한 후 생성되는 $HOSCN/OSCN^-$의 최대생성시간을 측정하였으며, peroxidase system을 함유한 세치제가 S. mutans와 lactobacilli에 미치는 영향을 양치전과 양치후 $HOSCN/OSCN^-$의 최대생성시간에 타액을 채취하여 비교하였다. 이와같은 실험을 통해 다음과 같은 결론을 얻었다. 1. 건강한 대상자의 자극성 전타액내의 $HOSCN/OSCN^-$의 농도는 양치후 30분 후 대조군에 비해 peroxidase systems을 함유한 세치제를 사용한 군에서 유의성있게 높게 나타났다 (p < 0.05). 2. 건강한 대상자의 자극성 전타액의 타액분비량, pH, 타액점조도는 peroxidase system을 함유한 세치제를 2주간 사용한 후에도 유의한 변화를 나타내지 않았다. 3. 2주간 실험치약을 사용한 후 건강한 대상자의 자극성 전타액내 S. mutans와 lactobacilli의 양은 peroxidase system을 함유한 세치제를 사용한 군에서 상대적으로 낮게 나타났으나 통계학적 유의성은 없었다(p = 0.206 : lactobacilli, p = 0.944 : S. mutans). 4. 구강건조증 환자의 자극성 전타액내의 $HOSCN/OSCN^-$의 농도는 양치후 30분(p < 0.01)과 60분(p < 0.05)에서 대조군에 비해 peroxidase system을 함유한 세치제를 사용한 군에서 유의성있게 높았다. 5. 구강건조증 환자의 양치 후 30분에 채취한 자극성 전타액내의 S. mutans와 lactobacilli의 양은 peroxidase system을 함유하거나(p < 0.01) 함유하지 않은(p < 0.05) 치약을 사용한 군 모두에서 양치전에 비해 유의성있게 감소하였다. 6. 양치후 30분에 채취한 구강건조증 환자의 자극성 전타액내 S. mutans 양은 peroxida system을 함유한 세치제를 사용한 군에서 대조군에 비해 유의성있게 낮았다(p < 0.05). 7. 양치후 30분에 채취한 구강건조증 환자의 자극성 전타액내 lactobacilli양은 peroxidase system을 함유한 세치제를 사용한 군에서 대조군에 비해 상대적으로 낮게 나타났으나(p = 0.067) 통계학적 유의성은 없었다.

Deprivation of Esophageal Boluses and Dry Forage Intake in Large-type Goats

  • In goats fed on dry forage twice a day, an esophageal fistula was used to investigate the physiological factors present in the marked suppression of dry forage intake that occurs after 40 min of feeding. The animals used in this study were five large-type male esophageal- and ruminal-fistulated goats. Roughly crushed alfalfa hay cubes with any large remaining chunks removed were used as feed for this research. The study was conducted under both normal feeding conditions (NFC) and sham feeding conditions (SFC). In the NFC control, the esophageal fistulae were closed by plugs and the animals ate dry forage in the normal manner. In the SFC treatment, before starting the experiment the plugs for closing the esophageal fistula were removed and the cannulae for collecting boluses were fitted into the fistulae. Therefore, the esophageal boluses were removed via an esophageal fistula before they entered the rumen. In the NFC control, eating rates sharply decreased in the first 40 min of feeding and were subsequently maintained at low levels. However, eating rates in the SFC treatment remained high after 40 min of the feeding period had elapsed and the goats ate continuously during the 2 h feeding period. In comparison with the NFC control ($1,794{\pm}203.80\;g$/2 h), cumulative dry forage intake in the SFC treatment ($3,182{\pm}381.69\;g$/2 h) was 77.4% greater (p<0.05) upon conclusion of the 2 h feeding period. In the SFC treatment, cumulative bolus output ($6,804{\pm}469.92\;g$/2 h) was about twofold the cumulative dry forage intake due to cumulative salivary secretion volume ($3,622{\pm}104.13\;g$/2 h) upon conclusion of the 2 h feeding period. The result indicates that large amounts of secreted saliva during dry forage feeding act in conjunction with consumed feed to form the ruminal load responsible for ruminal distension. The increased plasma total protein concentrations were higher in the SFC treatment than in the NFC control. However, plasma and ruminal fluid osmolalities increased in the NFC control during and after feeding but were mostly unchanged in the SFC treatment. In comparison with the NFC control ($3,440{\pm}548.04\;g$/30 min), thirst level in the SFC treatment ($1,360{\pm}467.02\;g$/30 min) was 60.5% significantly less (p<0.05) upon conclusion of the 30 min drinking period. The results of the present study indicate that In the second hour of the 2 h feeding period, dry forage intake is regulated by factors produced when boluses enter the rumen.

일부 산(Acid)취급 근로자의 치아산식증 (Dental Erosion in two Factories using Acids)

  • Acid erosion of teeth was studied in two factories. The A was the textile factory possessing dyeing process using acids. The B was the metal product manufactoring factory possessing electroplating process. The control group was selected at the same factory not exposed to acids. The results were summarized as below. 1. The pH of saliva was the range of $6.6{\sim}6.8$ in both factories. 2. The prevalence rate of erosion of teeth was higher in case group at the B. It was statistically significant. 3. The rate of erosion of incisor were the range of $12%{\sim}16%$(case group), $7%{\sim}21%$(control group) at the A and $10%{\sim}20%$ (case group), $2%{\sim}15%$ (control group) at the B. 4. The rate of erosion of degree 3 which is necessary for conforming, owing to occupational induction, were 8.9% (incisor: A) and 6.8% (incisor: B).

유산균 발효유 섭취가 법랑질 표면에 미치는 영향 (The effects of fermented milk intake on the enamel surface)

  • 연구목적: 본 연구는 액상발효유, 스터드 타입 및 드링크 타입 농후발효유가 정상법랑질 표면에 미치는 영향을 알아보고자 하였다. 연구방법: 3종의 유산균 발효유와 대조군인 제주삼다수에 우치시편을 8일 동안 매일 10분씩 4회 처리하고 그 외의 시간에는 인공타액에 침지하는 pH 순환처리를 시행한 후 표면미세경도 측정과 주사전자현미경을 이용하여 표면 형태를 관찰하였다. 연구결과: 3종의 유산균 발효유군 모두 대조군에 비해 표면경도차(ΔVHN)가 크게 나타났다. 이 중 액상발효유군은 표면경도차(ΔVHN)가 가장 크게 나타났으며, 주사전자현미경에 의한 법랑질 표면 형태 관찰에서도 가장 심한 표면 손상이 확인되었다. 결론: 유산균 발효유 종류에 따른 물리적 성상과 pH가 치아표면에 영향을 미치는 것으로 나타났다. 따라서 유산균 발효유의 치아부식증 발생 가능성을 고위험군에게 알림으로써 섭취 시 이를 고려하도록 하며, 치아부식증을 줄일 수 있는 예방적 연구가 계속적으로 필요하리라 생각된다.

클러스터 초상자성체 산화철 나노입자를 이용한 색채학적 해석 기반 당 측정 (Colorimetric Based Analysis Using Clustered Superparamagnetic Iron Oxide Nanoparticles for Glucose Detection)

  • Superparamagnetic iron oxide nanoparticles (SPIONs) are approved by the Food and Drug Administration (FDA) in the United States. SPIONs are used in magnetic resonance imaging (MRI) as contrast agents and targeted delivery in nanomedicine using external magnet sources. SPIONs act as an artificial peroxidase (i.e., nanozyme), and these reactions were highly stable in various pH conditions and temperatures. In this study, we report a nanozyme ability of the clustered SPIONs (CSPIONs) synthesized by the oil-in-water (O/W) method and coated with biocompatible poly(lactic-co-glycolic acid) (PLGA). We hypothesize that the CSPIONs can have high sensitivity toward H2O2 derived from the reaction between a fixed amount of glucose and glucose oxidase (GOX). As a result, CSPIONs oxidized a 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS) commonly used as a substrate for hydrogen peroxidase in the presence of H2O2, leading to a change in the color of the substrate. We also utilized a colorimetric assay at 417 nm using various glucose concentrations from 5 mM to 1.25 μM to validate β-D-glucose detection. This study demonstrated that the absorbance value increases along with increasing the glucose level. The results were highly repeated at concentrations below 5 mM (all standard deviations < 0.03). Moreover, the sensitivity and limit of detection were 1.50 and 5.44 μM, respectively, in which CSPIONs are more responsive to glucose than SPIONs. In conclusion, this study suggests that CSPIONs have the potential to be used for glucose detection in diabetic patients using a physiological fluid such as ocular, saliva, and urine.
