Effect of Maunfacturing Methods on Content Uniformity of Tablets (제제방법(製劑方法)이 정제(錠劑)의 함량균일성(含量均一性)에 미치는 영향(影響))

  • Safety, efficacy and reliability are the three basic criteria that define the quality of any well-designed pharmaceutical dosage form. Content uniformity directly bears on each of the three criteria defining the quality of drug product. Effect of manufacturing methods and physicochemical properties of the drug on content uniformity of tablets was investigated. Chlorpheniramine maleate and micronized salicylic acid were used as main ingredients. Three different methods for incorporation of main ingredients with diluents were solvent mixing method, geometric dilution method, and simple mixing method. The solvent mixing method was the best one of the three.

Enhancing Resistance of Red Pepper to Phytophthora Blight Diseases by Seed Treatment with Plant Growth Promoting Rhizobacteria

  • Plant growth promoting rhizobacteria (PGPR) have been shown to suppress phytopthora blight. This suppression has been related to both microbial antagonism and induced resistance. The PGPR isolates were screened by dual culture plate method and most of the isolates were showed varying levels of antagonism. Among the PGPR isolates pyoverdin, pyochelin and salicylic acid producing strains showed the maximum inhibition of mycelial growth of Phytopkhora capsici and increased plant growth promotion in red pepper. PGPR isolates further analysed for its ability to induce production of defence related enzymes and chemicals. The activities such as Phenyle alanin ammonia Iyase (PAL), Peroxidase (PO), Polyphenol oxidase (PPO) and accumulation of phenolics were observed in PGPR pretreated red pepper plants challenged with Phytopkhora capsici. The present study shows that an addition of direct antagonism and plant growth promotion, induction of defense related enzymes involved to enhance resistance against invasion of P. capsici in red pepper.

  • Aromatic hydrocarbon degrading bacteria were isolated from industrial waste by using an agar plate method. The isolate DY-1 was identified as Acinetobacter sp. and found to utilize phenanthrene as tis sole carbon source. THe bacteria were proved to produce salicylic acid as an intermediate from phenanthrene through naphthalene pathway, when the products in the culture were wxamined by thin-layer chromatography. THe $Phn^+$ genes were found to be involved in two plasmids of about 4 and 40kb which were lost and not detected in the DNA samples prepared from the mitomycin C-cured cells by a gel electrophoretic analysis.

  • Genes that are expressed early in specific response to high salinity conditions were isolated from rapeseed plant (Brassica napus L.) using an mRNA differential display method. Five PCR fragments (DD1.5) were isolated that were induced by, but showed different response kinetics to, 200 mM NaCl. Nucleotide sequence analysis and homology search revealed that the deduced amino sequences of three of the five cDNA fragments showed considerable similarity to those of ${\beta}$-mannosidase (DD1), tomato Pti-6 proteins (DD5), and the tobacco harpin-induced protein hin1 (DD4), respectively. In contrast, the remaining clones, DD3 and DD2, did not correspond to any substantial existing annotation. Using the DD3 fragment as a probe, we isolated a full-length cDNA clone from the cDNA library, which we termed BnSWD1 (Brassica napus salt responsive WD40 1). The predicted amino-acid sequence of BnSWD1 contains eight WD40 repeats and is conserved in all eukaryotes. Notably, the BnSWD1 gene is expressed at high levels under salt-stress conditions. Furthermore, we found that BnSWD1 was upregulated after treatment with abscisic acid, salicylic acid, and methyl jasmonate. Our study suggests that BnSWD1, which is a novel WD40 repeat-containing protein, has a function in salt-stress responses in plants, possibly via abscisic acid-dependent and/or -independent signaling pathways.

Phenolic Acid Composition and Antioxidative Activity of Red Ginseng Prepared by High Temperature and High Pressure Process (고온고압 처리 홍삼의 페놀산 조성과 항산화 활성)

  • This study was conducted to develop HTHP ginseng (high temperature and high pressure ginseng) with improved antioxidative activity and phenolic acid composition by high temperature and high pressure process. The HTHP ginseng extract was analyzed for the total phenol content, DPPH radical scavenging activity and phenolic acid composition. The total phenol content was increased in HTHP ginseng (14.76 mg/g) compared to raw ginseng (3.59 mg/g) and red ginseng (3.93 mg/g). DPPH radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 4.8~78.4%, 1~47.4% and 1.8~56.5% at $1{\sim}100mg/m{\ell}$ concentration. Also ABTS radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 8.9~99.8%, 3.4~96% and 1.2~96.5% at $1{\sim}100mg/m{\ell}$ concentration. In HPLC analysis, amounts of measured phenolic acid of HTHP ginseng greatly increased than raw ginseng and red ginseng, but salicylic acid was not detected in HTHP ginseng. In addition, DPPH radical scavenging activity of phenolic acid from HTHP ginseng was increased. Consequently, we believe high temperature and high pressure process is better method than existing method to increase the bioactivity of ginseng.

  • 높은 이온전도도와 충분한 기계적 강도, 전해질 누수가 적은 새로운 형태의 고분자 전해질막(pore-gel SPE)을 연구 개발하였다. 다공성 PVDF-HFP 고분자막의 기공 내에 전해질 용액을 흡수시킨 후 막 내에서 젤화를 진행시켰다. 전해질 용액은 2:2:1의 비를 갖는 PC/EC/DMA에 1M SA(Salicylic acid)를 용해하고 여기에 고분자막을 용해시킬 수 있는 아세톤을 첨가하였다. 초음파를 이용함으로써 고분자막의 용액 흡수율을 증가시키고 또 고분자막에서 젤화를 촉진 시킬 수 있었다. 이렇게 젤화한 막의 이온전도도는 젤화 전 막보다 $1{\sim}2.2$ 배 향상되었고, 인장강도는 gel-type SPE 보다 40 배 증가하였으며, 전해질 누수실험결과 hybrid-type SPE는 13%의 누수를 보였으나 본 연구의 막(pore-gel SPE)은 6%로 감소함을 보였다.

Functional Analysis of PepRSH (Pepper relA/spoT homolog) cloned from Capsicum annuum showing Systemic Acquired Resistance against Phytophthora capsici

  • RSH (relA/spoT homolog) has been known to determine the level of guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp), which are the effector nucleotide of the prokaryotic stringent response and also play a role in antibiotic production and differentiation in Streptomyces species but not a little in eukaryotic organism, especially in plant. Salicylic acid (SA), a critical signal molecule of establishing systemic acquired resistance (SAR), could induce SAR in Pepper (Capcicum annuum) against Phytophthora capsici. And the extent of SAR induction was in proportion to the dosage of SA (or BTH). Suppression subtractive hybridization (SSH), a PCR-based method for cDNA subtraction, was carried out between SA-treated and non-SA-treated pepper leaves to isolate genes which may be responsible for defense signaling against pathogens. Early upregulated gene was selected from reverse northern and kinetics of SSH-genes transcripts in SA-treated pepper leaves upon SA treatment. Full-length cDNA of the gene (PepRSH; Pepper RelA / SpoT homolog) had an open reading frame (ORF) of 2166 bp encoding a protein of 722 amino acids and a significant homology with (p)ppGpp phosphohydrolase or synthetase. Genomic DNA gel blot analysis showed that pepper genome has at least single copy of PepRSH. PepRSH transcripts was very low in untreated pepper leaves but strongly induced by SA and methyljasmonic acid (MeJA), indicating that PepRSH may share common SA and MeJA-mediated signal transduction pathway Functional analysis in E. coli showed PepRSH confers phenotypes associated with (p)ppGpp synthesis through a complementation using active site mutagenesis.

Synthesis and Characterization of Some Transition Metal Complexes of Unsymmetrical Tetradentate Schiff Base Ligand (비대칭 Tetradentate Schiff 염기 리간드의 전이금속 착물에 대한 합성 및 특성)

  • The solid complexes of Cu(II), Ni(II), Co(II), Mn(II) and Fe(III) with 4-hydroxy-3-(1-{2-(2-hydroxybenzylidene)- amino phenylimino}-ethyl)-6-methy-pyran-2-one (H2L) derived from o-phenylenediamine, 3-acetyl- 6-methyl-(2H) pyran, 2,4 (3H)-dione (dehydroacetic acid or DHA) and salicylic aldehyde have been synthesized and characterized by elemental analysis, conductometry, magnetic susceptibility, UV-visible, IR, $^1H$-NMR spectra, X-ray diffraction, thermal analysis, and screened for antimicrobial activity. The IR spectral data suggest that the ligand behaves as a dibasic tetradentate ligand with ONNO donor atoms sequence towards central metal ion. From the microanalytical data, the stoichiometry of the complexes has been found to be 1:1 (metal: ligand). The physico-chemical data suggests square planar geometry for Cu(II) and Ni(II) complexes and octahedral geometry for Co(II), Mn(II) and Fe(III) complexes. The x-ray differaction data suggests orthorhombic crystal system for Cu(II) complex, monoclinic crystal system for Ni(II), Co(II) and Fe(III) and tetragonal crystal system for Mn(II) complex. Thermal behaviour (TG/DTA) of the complexes was studied and kinetic parameters were determined by Coats-Redfern method. The ligand and their metal complexes were screened for antibacterial activity against Staphylococcus aureus and Escherichia coli and fungicidal activity against Aspergillus Niger and Trichoderma.

Efficacy of Three Antiviral Agents and Resistant Cultivars on Tomato Yellow Leaf Curl Virus in Tomato (토마토황화잎말림바이러스병에 대한 저항성 품종과 항바이러스 활성 물질 3종의 효과 검증)

  • Recently, several in vitro studies have reported antiviral activity of agents of systemic acquired resistance against plant virus infection, but the approach has not been applied in a wide range of agricultural fields. The objective of this study was to evaluate the inhibitory effect of the exogenous application of salicylic acid (SA), chitosan (CH), or eugenol (EG) in tomato yellow leaf curl virus (TYLCV) infection of greenhouse-grown tomato plants. In vitro, the initial time of symptom was observed in TYLCV-infected plants (VP) of the resistant cultivar 'Superdotaerang' at 12 days post inoculation (dpi) after application of antiviral agents. At 32 dpi, the disease rate of TYLCV in the CHT+VP (0.1% chitosan and virus infected control) treated plants was 87.5%, lower than that of the other treatment. However, the virus content in the CHT+VP treated plants was higher than those of the other treatments, and SA, EG, and CH did not show significant effect on plant height or shoot and root fresh weight. Our results from summer-cultivated greenhouse-grown tomatoes show that none of the tested agents had an inhibitory activity on viral infection or yield of tomato 'Dotaerangsola'cultivar. In contrast, all treated 'TY Giants' cultivars that possessed TYLCV resistance genes Ty-1 and Ty-3a did not show typical symptoms and the virus content was remarkably lower than those in the TYLCV treated plants in 'Superdotaerang'. The results of this research indicated that the planting of resistant tomato cultivars was effective method instead of using SA, EG, and CH (known as resistance-inducing factors for control) of TYLCV in the field.
