Immunological Studies on the Natural Products I -Production of Antibody Specific to Saikosaponin a-

  • In the course of the immunological studies on the natural products, the antibody specific to saikosaponin a was producted. Saikosaponin a was treated with succinic anhydride to give 6'-O-hemisuccinyl saikosaponin a, which was successively converted to saikosaponin a-BSA conjugate (4. 5 mole saikosaponin a/mole of BSA) by carbodiimide method. The antibody obtained from rabbits immunized with saikosaponin a-BSA conjugate as usual manner reacted with both the conjugate and BSA, while after the absorption with BSA, the antibody reacted with the conjugate alone.

In vitro Degradation of Saikosaponin-a in Physiological Condition

  • In vitro degradation of saikosaponin-a in physiological condition was conducted. Saikosaponin-a in 0.1 N-HCl of 25% 1,4-dioxane was incubated at $37^{\circ}$ and the products were analyzed and the time course of degradation was observed. Saikosaponin-a(Sa) was transformed to $saikosaponin-b_1(Sb_1)$ and saikosaponin-g(Sg) in the course of time. Sa was decreased drastically and not detected after 6 hours. $Sb_1$0 and Sg was increased with the time. After 6 hours the ratio of $Sb_1$ and Sg was about 4:1 and was maintained for 24 hours.

Saikosaponin Contents and Growth Characteristics on Cutting and Flower Removal in Bupleurum falcatum L. (예취 및 적화처리에 따른 시호의 생육특성 및 saikosaponin 함량)

  • This study was conducted to investigate the effect of cultural methods(cutting, flower removal) on growth and quality of B. falcatum L. Jeongseon local cultivar collected in Korea and Mishima local cultivar introduced from Japan were used. Some of the results obtained are as follows; Jeongseon local cultivar showed less stem branches and shoot weight compared to Mishima. However, Jeongseon local cultivar showed tall plant height, high root fresh and dry weight, and high levels of saikosaponin-a and total-saikosaponin, but low saikosaponin-c content than that of Mishima. Both cultivars seeded on March 20 had longer main root, bigger stem diameter, few stem branch, and high saikosaponin-c content compared with those of late seeded one, April 30. Growth characteristics such as plant height, stem diameter, stem branch number, shoot weight, root diameter, root fresh and dry weight, and root branch number were increased in a low planting $density(30{\times}15\;cm)$, but the content of saikosaponin was not affected by planting density. Jeongseon and Mishima cultivars seeded on April 10 with $30{\times}15\;cm$ planting density and April 30 with $30{\times}10\;cm$ planting density contained highest total saikosaponin levels, respectively. However, average root dry weight was not affected by planting time or density in both Bupleurum cultivars. Both cutting and flower removal increased the length of main root, fresh and root weight, and saikosaponin-a, saikosaponin-d and total-saikosaponin in Jeongseon cultivar than that of Mishima. As the harvesting time was delayed, plant height, stem diameter, shoot weight, length of main root, root fresh and dry weight were increased, but content of saikosaponin-a, saikosaponin-d and total-saikosaponin were decreased.

Effects of IAA, IBA, and Media on Growth and Saikosaponin Biosynthesis in Bupleurum falcatum Hairy Root Culture (시호 모상근 배양에서 생장과 시호 사포닌 생합성에 미치는 배지와 IAA 및 IBA의 영향)

  • We examined the effects of various basal media and exogenous auxins on the growth and saikosaponin production in hairy root cultures of Bupleurum falcatum. Of the various media tested, 2RCM medium was superior for growth and saikosaponin content, in which saikosaponin a, c, and d was produced, however MS medium was resulted in extremely low level in saikosaponin production. By the treatment of 0.5 mg/L IBA the growth rate was increased by about 60% in 2RCM medium, whereas both IAA and IBA showed a potent inhibition effect in saikosaponin biosynthesis by the treatment of 0.01 mg/L~5mg/L.

  • This study was carried out to find out favourable soil conditions to give good quality of Bupleuri Radix for Korean medicine. The major ingredients were saikosaponin a and c, which were determined by HPLC. The contents of saikosaponin a and c in the root were determined to be 0.141-0.479% and 0.042-0.094%, respectively. The content of saikosaponin a was negatively correlated with the total nitrogen contents in surface soils, but that of saikosaponin c was independent of physico-chemical properties of the soil. The contents of saikosaponin a and c were decreased in the root with high Ca and Fe.

  • This study was carried out to analyse saikosaponin contents in adventitious root formed from callus of Bupleurum falcatum L. The induction of adventitious root from callus of 4 weeks of culture was most favorable to MS medium supplemented with 0.1 mg/L 2,4-D. The adventitious root formation ratio per callus was highest when the size of callus was 900 to 1,000 $\mu$M The content of saikosaponin a and d was 2089$\pm$124 $\mu$g and 4778 $\pm$ 214 $\mu$g at 150 days of culture respectively on the basis of g dry root wt, whereas that of saikosaponin c was 3492 $\pm$ 123 $\mu$g at 60 days of culture.

  • The optimal condition for the determination of saikosaponin a and d, the major pharmacologically active saponins of the roots of Bupleurum falcatum, was studied with the conversion of these saponins into diene saponins $(saikosaponin\;b_1\;and\;b_2)$. The complete separation and quantitative analysis of these saponins were performed by the method of high performance liquid chromatography using $NH_2$ column. The conversion of saikosaponin a and d into diene saponins under gastric pH was calculated. Thirty-three percent of saikosaponin a was converted to saikosaponin $b_1$ and 63 percent of saikosaponin d was converted to saikosaponin $b_2$.

  • This experiment was conducted to find out the effects of L-phenylalanine on the saikosaponin content of callus induced from Bupleurum falcatum leaf segments. In the fresh and dry weight of callus, the addition of 2,4-D than L-phenylalanine was significantly effective. However, the L-phenylalanine treatment rather than 2,4-D was effective for high saikosaponin accumulation in the callus of Bupleurum falcatum.

  • Saikosaponin은 한방에서 중요하게 사용되고 있는 시호의 항염증작용 성분으로서 주로 염증의 후기 단계에 작용하고 있다. 그러나 현재 시호는 생약자체로만 사용되고 성분이 약물로서 개발되지 못하고 있다. 그것은 Saikosaponin의 용해성이 극히 저조하여 제제화하기 힘든 때문이다. 연자는 시호 Saponin의 용해성을 증가시킬 목적으로 시호 Saponin의 하나인 saikosaponin-a를 Pyridine 중에서 succinic anhydride와 반응시켜 6"-hemisuccinyl saikosaponin-a를 얻고 이를 THF에 용해한 후 KOH와 반응시킨 후 다량의 THF를 추가하여 hemisuccinyl esler의 potassium염을 합성하였다. 이 화합물의 용해도를 원래의 Saponin과 비교시험하였다.

Root Yield and Saikosaponin Content in Local Strains of Bupleurum falcatum L. (시호 수집종의 근 수량 및 Saikosaponin 함량)

  • Six local strains collected in Korea and one cultivar (Samdo) introduced from Japan were applied for comparative cultivation to investigate growth characteristics, root yield potentials and saikosaponin contents of root in Bupleurum falcatum L. The one and two year old plants of the native local strains flowered in August 2~3 and July 16~18, respectively, whereas Samdo was late flowering type, being delayed by 18~32days and sustained the growth of above ground parts to the end of November. All the native local strains have similiar characteristics but Samdo was completely different in plant and root type. Local strain collected from Chunchon showed better in growth and root characteristics, consequently resulting high yield of dry roots and saikosaponin contents in both one and two year old plants. The average yield of two year cultivation system was 2.6 times high as 105Kg /10a, compared with that of one year.
