A study on the removal of mercury and lead by microorganisms, Saccharomyces cerevisiae and Aureobasidium pullulans, was performed, in which the comparison of adsorption model between these two heavy metals was done. The amounts of mercury removed were more than those of lead in both microorganisms. In case of mercury, the adsorption isotherm of S. cerevisiae was accorded with Langmuir model but A. pullulans was followed to Freundlich model. In the case of lead, however, the adsorption isotherm had opposite results. The adsorption rate of mercury to S. cerevisiae was faster than that of A. pullulans, but in the case of lead, it revealed contrary results. It seems, therefore, that the type of microorganisms used as biosorbents should be selected differently with the type of heavy metals removed for applying these to real adsorption process.

Eukaryotic translation initiation factor 5B (eIF5B) plays a role in recognition of the AUG codon in conjunction with translation factor eIF2, and promotes joining of the 60S ribosomal subunit. To see whether the eIF5B proteins of other organisms function in Saccharomyces cerevisiae, we cloned the corresponding genes from Oryza sativa, Arabidopsis thaliana, Aspergillus nidulans and Candida albican and expressed them under the control of the galactose-inducible GAL promoter in the $fun12{\Delta}$ strain of Saccharomyces cerevisiae. Expression of Candida albicans eIF5B complemented the slow-growth phenotype of the $fun12{\Delta}$ strain, but that of Aspergillus nidulance did not, despite the fact that its protein was expressed better than that of Candida albicans. The Arabidopsis thaliana protein was also not functional in Saccharomyces. These results reveal that the eIF5B in Candida albicans has a close functional relationship with that of Sacharomyces cerevisiae, as also shown by a phylogenetic analysis based on the amino acid sequences of the eIF5Bs.
Cellobiase ($\beta$-glucosidase) is an enzyme of the cellulase system in cellulolytic microor-ganisms. The chromosomal DNA fragment which include cellobiase gene of Cellulomonas biazotea was cloned in Eschericia coli via plasmid pBR 322 vector. Restriction enzyme Sal I was used to obtain adequate size of fragments from C. biazotea. chromosomal DNA. The transformant of E. coli HB101 with recombinant plasmid pBG101 showed cellobiase activity, which is not ordinary in E. coli HB101. The enzyme activity of the transformant was as of 20% lower than that of C. biazotea.

유산균발효에 이용하는 Streptococcus속과 Lactobacillus속 균주를 Sacharomyces cerevisiae KFCC 32017과 각각 대두유상에서 혼합배양하여 산생산이 좋은 균주 S. lactis KFCC 32406을 선별하였다. 혼합배양시 산생산성은 20시간 후 거의 정상치에 도달하였고 배양온도는 34$^{\circ}C$ 부근에서 가장 좋은 것으로 나타났다. Sucrose와 skim milk의 영향을 조사하기 위해 0.5 - 3%까지 각각 첨가한 결과 sucrose 1.5% 농도에서 2배 정도로 산도가 높아졌고 skim milk의 경우는 산도에 별다른 영향을 끼치지 않는 것으로 밝혀졌다. 또한 soywhey에서는 뚜렷한 차이를 보이지 않았지만 2.0-3.0% 부근의 skim milk 농도에서 산도의 증가를 나타내었다.
To investigate the promoter effect on heterologous gene expression in S. cerevisiae, the recombinant plasmids pYI11, pYI12, pYI10-2, and pYIGP were constructed to contain the inulinase gene (INUI) as a reporter under the control of GAL10, GAL7, GAL1, and GAP promoters, respectively. When the yeasts transformants were cultivated on galactose-containing rich media, the cell growth reached to 36-39 OD600 at 72 hours of cultivation. The specific growth rates of the cells harboring the four different plasmids decreased similarly : they dropped from $0.24 h^{-1}$ during the glucose-consuming period to 0.04 -$0.10 h^{-1}$ during the galactose-consuming period (gene expression phase for GAL promoter system). After the depletion of glucose, the expression of inulinase gene was started and reached to maximal levels of 4.3(GAL1 promoter), 4.0(GAL10 promoter), 3.8(GAL7 promoter), and 1.6(GAP promoter) unit/mL at 72 hours of cultivation. Based on the maximal expression level and activity staining on the plate, the promoter strength was in the order of GAL1, GAL10, GAL7 and GAP promoter. While the GAL-promoter systems showed a high plasmid stabilities of more than 78%, the GAP-promoter plasmid revealed a lower plasmid stability of 55%. Most of inulinase activity (98%) was found in the extracellular medium, indicating that the secretion efficiency of inulinase is independent on the type of promoter.

HSF1 is the heat shock transcription factor in Saccharomyces cerevisiae. S. cerevisiae KNU5377 can ferment at high temperature such as $40^{\b{o}}C$. We have been the subjects of intense study because Hsf1p mediates gene expression not only to heat shock, but to a variety of cellular and environmental stress challenges. Basing these facts, we firstly tried to construct the hsf1 gene-deleted mutant. PCR-method for fast production of gene disruption cassette was introduced in a thermotolerant yeast S. cerevisiae KNU5377, which allowed the addition of short flanking homology region as short as 45 bp suffice to mediate homologous recombination to kanMX module. Such a cassette is composed of linking genomic DNA of target gene to the selectable marker kanMX4 that confers geneticin (G418) resistance in yeast. That module is extensively used for PCR-based gene replacement of target gene in the laboratory strains. We describe here the generation of hsf1 gene disruption construction using PCR product of selectable marker with primers that provide homology to the hsf1 gene following separation of haploid strain in wild type yeast S. cerevisiae KNU5377. Yeast deletion overview containing replace cassette module, deletion mutant construction and strain confirmation in this study used Saccharomyces Genome Deletion Project (http:://www-sequence.standard.edu/group/yeast_deletion_project). This mutant by genetic manipulation of wild type yeast KNU5377 strain will provide a good system for analyzing the research of the molecular biology underlying their physiology and metabolic process under fermentation and improvement of their fermentative properties.
The fusants which contain starch utilizing ability and alcohol fermenting ability were developed by protoplast fusion of Saccharomyces cerevisiae KOY-1 and Saccharomyces diastaticus KCTC 1804. Sacharomyces cerevisiae KH-12 was obtained by haploid induction from Saccharomyces cerevisiae KOY-1. The auxotropic mutants of yeast were obtained by using an ethylmethane sulfonate (EMS). The frequency of protoplast formation in Saccharomyces cerevisiae KOY-1 $(Met^-)$ and Saccharomyces diastaticus KCTC 1804 $(Trp^-)$ were 90.5% and 97.7%, respectively. The frequency of fusant formation was $1.79{\times}10^{-4 }$ for the regenerated protoplast and the 1,000 fusants were obtained. Fusant FA 776 was selected as a potential yeast which contain an alcohol fermenting ability in the starch medium. The genetic stability was 4.64% for 10 passages of generation. Fusant FA 776 produced 13mg/ml of alcohol in 24% starch medium and showed 1.86-fold higher alcohol fermenting ability than Saccharomyces diastaticus KCTC 1804.

[ ${\beta}-Glucan$ ], one of the cell wall components, is most plentiful polysaccharides in cell wall and has several advantages in immune system. In yeast ${\beta}-glucan$ is mainly contained in the yeast cell wall, and thus it is important to produce high levels of cell mass for the mass production of yeast ${\beta}-glucan$. The best carbon and nitrogen sources on cell mass production were high fructose syrup and yeast extract. Response surface methodology (RSM) was very potential tool for the optimization of process factor and medium component. It was applied to estimate the effects of medium components on the production of cell mass. Optimal concentrations of high fructose syrup and yeast extract by response surface methodology were 8.0% (v/v) and 5.2% (w/v), respectively and the cell mass predicted was $17.0\;g/{\ell}$ at 20 h of cultivation.
In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.
The functional relationship between the initial cell concentration and the ethanol productivity was investigated in the repeated batch fermentation of Sacharomyces cerevisiae ATCC 24858. The repeated batch fermentations were performed in the range of 60 to $150g/\ell$ of initial sugar concentration and 17.5g/$\ell$ to $53.1g/\ell$ of initial cell concentration. The time of one batch fermentation was 1 or 2 hours and the batch fermentation was repeated ten times in every repeated formentaction. The functional relationship showed that the productivity increased non-linearly according to the increase of initial cell concentration regardless of initial sugar concentration. When the initial concentration of sugar was $60g/\ell$ and that of biomass was $34.5g/\ell$, the fermentation was completed within one hour and its ethanol productivity was $26.7g/\ell$.hr, the latter including the times of cell separation, pouring the new substrate into a flask and sampling. When the initial sugar concentration was $120g/\ell$ and the initial cell concentration $50.3g/\ell$, the fermentation was also finished within one hour and its productivity was $45.8g/\ell$$.$hr, The maximum ethanol productivity for eight different repealed fermentations in this work was $53g/\ell$.hr.
