A study on the removal of mercury and lead by microorganisms, Saccharomyces cerevisiae and Aureobasidium pullulans, was performed, in which the comparison of adsorption model between these two heavy metals was done. The amounts of mercury removed were more than those of lead in both microorganisms. In case of mercury, the adsorption isotherm of S. cerevisiae was accorded with Langmuir model but A. pullulans was followed to Freundlich model. In the case of lead, however, the adsorption isotherm had opposite results. The adsorption rate of mercury to S. cerevisiae was faster than that of A. pullulans, but in the case of lead, it revealed contrary results. It seems, therefore, that the type of microorganisms used as biosorbents should be selected differently with the type of heavy metals removed for applying these to real adsorption process.

Eukaryotic translation initiation factor 5B (eIF5B) plays a role in recognition of the AUG codon in conjunction with translation factor eIF2, and promotes joining of the 60S ribosomal subunit. To see whether the eIF5B proteins of other organisms function in Saccharomyces cerevisiae, we cloned the corresponding genes from Oryza sativa, Arabidopsis thaliana, Aspergillus nidulans and Candida albican and expressed them under the control of the galactose-inducible GAL promoter in the $fun12{\Delta}$ strain of Saccharomyces cerevisiae. Expression of Candida albicans eIF5B complemented the slow-growth phenotype of the $fun12{\Delta}$ strain, but that of Aspergillus nidulance did not, despite the fact that its protein was expressed better than that of Candida albicans. The Arabidopsis thaliana protein was also not functional in Saccharomyces. These results reveal that the eIF5B in Candida albicans has a close functional relationship with that of Sacharomyces cerevisiae, as also shown by a phylogenetic analysis based on the amino acid sequences of the eIF5Bs.
Cellobiase ($\beta$-glucosidase) is an enzyme of the cellulase system in cellulolytic microor-ganisms. The chromosomal DNA fragment which include cellobiase gene of Cellulomonas biazotea was cloned in Eschericia coli via plasmid pBR 322 vector. Restriction enzyme Sal I was used to obtain adequate size of fragments from C. biazotea. chromosomal DNA. The transformant of E. coli HB101 with recombinant plasmid pBG101 showed cellobiase activity, which is not ordinary in E. coli HB101. The enzyme activity of the transformant was as of 20% lower than that of C. biazotea.
유산균발효에 이용하는 Streptococcus속과 Lactobacillus속 균주를 Sacharomyces cerevisiae KFCC 32017과 각각 대두유상에서 혼합배양하여 산생산이 좋은 균주 S. lactis KFCC 32406을 선별하였다. 혼합배양시 산생산성은 20시간 후 거의 정상치에 도달하였고 배양온도는 34$^{\circ}C$ 부근에서 가장 좋은 것으로 나타났다. Sucrose와 skim milk의 영향을 조사하기 위해 0.5 - 3%까지 각각 첨가한 결과 sucrose 1.5% 농도에서 2배 정도로 산도가 높아졌고 skim milk의 경우는 산도에 별다른 영향을 끼치지 않는 것으로 밝혀졌다. 또한 soywhey에서는 뚜렷한 차이를 보이지 않았지만 2.0-3.0% 부근의 skim milk 농도에서 산도의 증가를 나타내었다.
본 연구에서는 GALl, GALl, GALlO 및 GAP promoter 하류에 reporter 유전자인 K. marxianus의 inulinase 유전자(lNUl)를 연결하여 각각의 재조합 plasmid들을 구축하고, 이들로 형질전환된 S. cerevrswe를 회분배양(YPOG 배지 )하여 외래 유전자 발현에 미치는 promoter의 영향을 비교.검토하 였다. 재조합 효모의 최종 균체농도는 36-39 00600 값을 보여 promoter에 따른 큰 차이를 보이지 않았으나, 포도당 소모기간 동안 비증식속도는 평균 $0.24 h^{-1}$로 유지되다가 galactose 소모기간 동안에 GAL promoter 함유 효모배양의 경우 $0.04-0.06 h^{-1}$, pYIGP 함유 재조합 효모배양은 $0.10 h^{-1}$로 감소하였다. 포도당 고갈 후 inulinase 발현은 시작되었고 균체외 inulinase의 발현 수준은 배양 72시간에 4.3 (GALl promoter), 4.0 (GAL7 promoter), 3.8 (GAL10 promoter) 및 1.6 (GAP promoter) unit/mL에 도달하였다. 평판배지상에서의 활성염색과 회분배양의 결과(최종발현양 및 초기 inulinase 말현속도), inulinase 발현에 미치는 promoter 세기 는 GALl > GALlO > GAL7 > GAP 순임을 알 수 있었다. GAL promoter가 배양말기까지 78 % 이상의 높은 plasmid 안정성을 보인 반면에, GAP promoter의 경우 55%의 낮은 plasmid 안정성을 보였다. 또한, 재조합 inulinase는 promoter 종류에 상관없이 98% 이상 배양액으로 분비되였다.
출아효모인 Sacharomyces cerevisiae S288C균주를 이용한 효모의 게놈이 완성된 후 S. cerevisiae는 다양한 연구 모델로 이용되어져 왔다. 현재까지 효모를 이용한 기능 유전체학 측면에서의 연구는 laboratory strainin인 S288C 균주 또는 그 유래의 균주들이다. 그러나 자연에서 분리된 효모 또는 산업적으로 이용되어지고 있는 S. cerevisiae의 유전학 측면에서의 연구는 낮은 포자형성률 및 형질전환률, 그리고 S288C 균주와의 게놈상의 상이성 때문에 거의 이루어지지 않고 있다. 여기서 우리 연구진은 자연에서 분리된 Saccharomyces cerevisiae KNU5377 균주를 이용하여 random spore analysis를 통해 MATa 및 $MAT{\alpha}$ 타입의 각각의 haploid cell을 분리 후 이미 보고된 KanMX module를 가지고 round PCR기법에 의한 short flanking homology 기법을 이용하여 전사조절인자인 HSF1 유전자가 치환된 변이주를 구축할 수 있었다. 덧붙여, 모든 유전자에 이 기법을 적용할 수는 없다는 것을 확인하였다. 앞으로 이 변이주를 통해 기능 유전체학적인 측면에서 이 유전자의 스트레스와의 관련성을 연구하고자 한다.
Saccharomyces diastaticus KCTC 1804 균주와 Saccharomyces cerevisiae KOY-1 균주를 이용하여 전분분해활성과 알코올 발효능을 동시에 가지는 효모융합체를 개발하였다. Saccharomyces cerevisiae KOY-1 균주의 단수체 유도를 통하여 Saccharomyces cerevisiae KH-12 균주를 획득하였다. EMS를 사용한 돌연변이 유발을 통하여 Saccharomyces cerevisiae KH-12 균주의 Met 영양요구성 변이주를 획득하였다. Lyticase 처리로 Saccharomyces cerevisiae KOY-1 $(Met^-)$, Saccharomyces diastaticus KCTC 1804$(Trp^-)$ 균주의 원형질체 형성률은 각각 90.5%, 97.7%로 관찰되었다. 원형질체 융합은 polyethylene glycol 4,000을 이용하여 실시하였고, 이때 원형질체 융합율은 $1.79{\times}10^{-4}$으로 관찰되었다. 원형질체융합과정을 통해 총 1,000주의 융합체를 획득하였고, 획득한 융합체의 전분분해능과 알코올발효능을 비교하여 최종적으로 전분배지 YPS24에서 알코올생성능이 가장 우수한 효모융합체인 FA 776을 최종 선발하였다 최종 선발된 효모융합체 FA 776은 10세대 계대배양 후의 revertent 복귀율이 4.64%로 유전적 안정성을 확인하였으며, 효모융합체의 total DNA 함량을 비교한 결과, Saccharomyces cerevisiae 균주로부터 분리된 단수체 효모 KH-12는 26.56 fg/cell, Saccharomyces diastaticus KCTC 1804 균주는 25.40 fg/cell이었고, 이들을 사용해서 얻어진 융합체 FA 776의 DNA함량은 42.67 fg/cell이었다. 전분배지 YPS24배지에서 60시간 발효하였을때 Saccharomyces cerevisiae KOY-1 균주는 알코올 생성하지 못하였지만 효모융합체 FA 776은 13 mg/mL의 알코올을 생성하였다. 이는 전분이용성 효모인 Saccharomyces diastaticus KCTC 1804 균주에 비해 1.86배나 향상된 알코올 발효능을 나타냈다.
본 연구는 효모의 세포벽에 존재하는 본 연구는 효모의 세포벽에 존재하는 ${\beta}-Glucan$을 대량생산하기 위해 Saccharomyces cerevisiae JUL3의 균체량을 증가시키기 위한 연구를 수행하였다. S. cerevisiae JUL3의 배양학적 특성을 알아보기 위해 kinetic parameter를 조사하였을 때 specific growth rate (${\mu}$)는 $0.145\;h^{-1}$, yield ($Y_{x/s}$)는 0.332 g/g, glucose 소모속도($q_{s}$)는 $0.437\;h^{-1}$이며 productivity (P)는 $0.4827\;g/{\ell}{\cdot}h$을 나타내었다. 균체량이 가장 높게 나타난 탄소원과 질소원은 고과당 (high fructose syrup)과 yeast extract이었다. 효모균체 대량생산 및 scale up을 위하여 반응표면 분석법을 통하여 고과당과 yeast extract에 대한 최적농도를 조사하였다. 균체량이 가장 많이 생산되어질 수 있는 조건으로 제시된 고과당과 yeast extract의 최적 농도는 각각 8.0 %와 5.2 %였으며, 이 때 예측되는 균체량은 $16.95\;g/{\ell}$ 이었다.
In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.
본 연구에서는 균주로 Saccharomyces ceremszae A Tee 24858을, 초기 당 농도로는 60, 90, 120 및 $150g/\ell$ 을, 1회 발효 시간으로는 1시간 또는 2시간을, 초기 균체농도로는 $17.5g/\ell$ 내지 $53.1g/\ell$ 사이의 값을 사용하여 10회 반복 회분식 실험을 수 행하였다. 이 결과로부터 반복 회분식 실험에서의 평균 초기 균체농도와 에탄올 생산성 사이의 함수 관계를 규명 하여 보았다. 초기 균체농도와 에탄올 생산성 사이의 함수 관계는 초기 균체농도가 증가함에 따라 초기 당 농도에 관계없이 에탄올 생산성이 비 션형적 으로 증가하는 함수 관계를 나타내였다. 초기 당 농도가 $60g/\ell$ 이고, 초기 균체농도가 $34.5g/\ell$ 인 경우 발효는 1시간 내에 완료되었으며 균체의 분리, 새로운 기질의 주입 등의 조작을 고려한 총 10회 반복 회분 실험의 에탄올 생산성은 $26.7g/\ell$.hr이었다. 초기 당 농도가 120gjP 이고 초기 균체농도가 5 50.3gj P 인 경우에도 1시간 내에 발효가 완료되었으 며 이때의 에탄올 생산성은 $48.8g/\ell$.hr이었다. 본 연구에서 수행한 8종류의 10회 반복 회분식 실험을 통하여 얻은 최대 에탄융 생산성은 $53g/\ell$.hr이였다.
