  • In the course of screening program for VHR DS-PTPase (dual-specificity protein tyrosine phosphatase) from natural sources, Gastrodia elata was selected. One compound showing potent inhibitory activity was isolated by the solvent extraction and column chromatography including silica gel, ODS RP-18, Sephades LH-20, and HPLC. This compound was identified as baicalein by several NMR techniques such as $^1H-NMR$, $^{13}C-NMR$, and DEPT. Baicalein showed selective inhibitory activity against VHR DS-PTPase with $IC_{50}=2.4\;{\mu}M$, and showed cytotoxicity against several human cancer cell lines with an $GI_{50}$ of $5.26{\sim}12.93\;{\mu}g/mL$ range, including, melanoma (LOX-IMVI), lung cancer (NCI H23 and A549), colon cancer (HCT 116 and SW 620), prostate cancer (PC-3), and leukemia (MOLT 4F).

Cytotoxic Effect of Extracts from Styela clava against Human Cancer Cell Lines (암세포주에 대한 미더덕 추출물의 세포독성 효과)

  • The present study describes the preliminary evaluation of the cytotoxic effect from Styela clava (Korean name : miduduk) extracts. S. clava was treated with methanol, ethanol, acetone, and water, then cytotoxic effect of the extracts were evaluated by the MTT reduction assay. The ethanol extracts from S. clava showed the cytotoxic activity on the HT-29 human colon cancer cells. The ethanol extracts was further fractionated with n-hexane, diethyl ether, ethyl acetate, and water according to the degree of polarity. The diethyl ether fraction showed high cytotoxic activity of HT-29 cells. however, the other fractions showed low cytotoxicity. The diethyl ether layer also showed the cytotoxic activity against SW620, HeLa, and MCF-7 cells. These studies support that extracts from S. clava may be a potential candidate as a possible chemotherapeutic agent against human cancer cells.

In vitro Cytotoxic Effect of Extracts from Styela plicata (오만둥이(Styela plicata) 추출물의 in vitro 세포독성 효과)

  • The present study describes the preliminary evaluation of the anticancer activity of Styela plicata. Freeze-dried S. plicata was extracted with methanol, ethanol, acetone, and water, and then anticancer effect of the extracts was measured by the MTT reduction assay and phase-contrast microscopy on the HT-29 human colon carcinoma cells. Among the extracts, acetone extract showed the highest anticancer activity. The cell proliferation rates markedly decreased by 94.0% at the concentration of 500 ${\mu}g/mL$ of acetone extract compared with control cells. The acetone extract was further fractionated with hexane, diethyl ether, ethyl acetate, and water layer according to the degree of polarity. The HT-29 cells with hexane layer extract (250 ${\mu}g/mL$) decreased the cell viability to 5.1% of untreated control. The growth of SW620, HeLa, and MCF-7 cells was decreased to about 10%, by the treatment of hexane layer extract 250 ${\mu}g/mL$. Theses results suggest extracts from S. plicata as possible natural cancer therapeutic material.

  • An automated minutes summary system is required to objectively summarize and classify the contents of discussions or discussions for decision making. This paper designs and implements a minutes summary system using word2vec model to complement the existing minutes summary system. The proposed system is further implemented with word2vec model to remove index words during morpheme analysis and to extract representative sentences with common opinions from documents. The proposed system automatically classifies documents collected during the meeting process and extracts representative sentences representing the agenda among various opinions. The conference host can quickly identify and manage all the agendas discussed at the meeting through the proposal system. The proposed system analyzes various agendas of large-scale debates or discussions and summarizes sentences that can be representative opinions to support fast and accurate decision making.

Comparison of CXCL10 Secretion in Colorectal Cancer Cell Lines

  • Established cancer cell lines are widely used for developing biomarkers for the patient-specific treatment of colorectal cancer and predicting prognoses. However, cancer cell lines may exhibit different drug responses depending upon the characteristics of the cell line. Therefore, it is necessary to select a tumor cell line suitable for the purpose of the study by considering the cell characteristics. This study investigated the levels of CXCL10, which were recently been reported to play an important role in the outcome of tumor treatment, secreted by colon cancer cells. 2 × 105 cells/mL of each colorectal cancer cell was seeded into a 35 mm cell culture dish. After 24 h incubation, culture supernatant was used to determine the secreted CXCL10 levels. Among six colorectal cancer cell lines (HT-29, HCT116, CaCo-2, SW620, SW480, and CT26), Caco-2 cells showed the highest level of CXCL10 secretion. HT-29 cells showed the second-highest level of CXCL10 secretion. No significantly measurable level of CXCL10 secretion was detected in HCT116 cells. These results will be helpful in investigating the molecular basis of colorectal cancer.

  • A series of 2-alkyl-2,3-dihydro-1H-2-azacyclopenta[b]anthracene-5,10-diones ($3a{\sim}h$) were synthesized and evaluated in vitro cytotoxicity against colon cancer cell lines (HCT116 and SW620) and nuroblastoma cell lines (SK-N-SH and SK-N-MC). Among them, compound 3f showed significant cytotoxic activity ($IC_{50}$ against SK-N-SH; $14.8{\mu}M$, $IC_{50}$ against SK-N-MC; $11.3{\mu}M$).

Antioxidant and Antiproliferative Activity of Extracts from Water Chestnut (Trapa japonica Flerow) (마름 추출물의 항산화활성 및 암세포증식 억제 활성)

  • Background : This study aimed to investigate the antioxidant and antiproliferative activities of extract from different parts of water chestnut (Trapa japonica Flerow). Methods and Results : The total polyphenol content of pericarp and seed extract was 438.31 mg/g and 25.32 mg/g respectively. DPPH radical scavenging assay showed that the half maximal inhibitory concentration ($IC_{50}$ values) of pericarp and seed extract were $5.28{\mu}g/m{\ell}$ and $355.51{\mu}g/m{\ell}$ respectively. In addition, the pericarp extract showed strong reducing power. In the MTT assay, the pericarp extract significantly inhibited the viability of A549, AGS, HeLa, PC-3, HCT116, HT29 and SW620 cell lines compared with the seed extract. Conclusions : These results suggest that T. japonica Flerow extracts have significant antioxidant and antiproliferative activity.

Cytotoxic Activity of the Inula japonica Extracts Against Several Human Cancer Cell Lines In Vitro (선복화 (Inula japonica)추출물의 암세포주에 대한 In Vitro 세포독성)

  • The present study describes the preliminary evaluation of the cytotoxic activity of the extracts from Inula japonica. I. japonica was extracted with methanol, ethanol, acetone, and water, and then cytotoxic activity of these extracts were evaluated. The cytotoxic activity of each extract was assessed by the MTT-dye reduction assay. Both ethanol and acetone extracts from I. japonica showed the cytotoxic activity against the HT-29 human colon cancer cells. Furthermore, the ethanol extract was fractionated with n-hexane, diethyl ether, ethyl acetate, and water according to degree of Polarity, The diethyl ether fraction showed the highest cytotoxic activity against HT-29 cells, but the other fractions showed low cytotokic activity. In addition, diethyl ether layer also showed the cytotoxic activity against various tumor cells, such as human colon carcinoma SW620, human cervix adenocarcinoma HeLa, and human breast adenocarcinoma MCF-7 cells as well as HT-29 cells. These studies support that extracts of I. japonica may be a potential candidate as possible chemotherapeutic agent against human cancer.

변성암석학과 구조지질학에서 석류석 반상변정의 중합적인 의의: 조산운동의 온도-압력-변형-시간 경로 해석

  • 석류석은 변성 온도와 압력을 계산하는 지온지압계에 널리 사용되며, 또한 이들 내부엽리와 외부엽리와의 접촉 관계를 이용하여 상대적인 변형작용의 시기를 밝히는 데 이용되어져 왔다. 석류석 반상변정 내의 내부 엽리 조직에 대한 정량적인 해석과 내부 미세 구조와 화학적 누대구조와의 관계에 대한 접근 방법은 석류석이 형성되는 동안에 온도-압력-변형작용의 진화 과정을 보다 정량적으로 해석할 수 있을 뿐만 아니라, 절대 연령 자료와 결합은 조산 운동 동안 발생하는 다변성/다변형 작용의 특징 보여주는 온도-압력-변형-시간 경로를 규명할 수 있다. 미국 메사추세츠 주 북중부 지역에서 십자석, 남정석과 함께 산출되는 석류석 반상변정은 온도-압력이 증가(540-570 $^{\circ}C$, 4.0-5.0 kbar에서 620-637 $^{\circ}C$, 7.9-8.8 kbar 까지) 하면서 성장하였다. 또한 이와 같은 변성작용 동안 압축 변형 방향은 다음과 같은 순서로, NE-SW, NW-SE, N-S 그 다음 E-W 방향으로 변화하였다. 결론적으로 이와 같은 변성작용과 변형작용은 주로 고생대 중기 데본기 동안에 일어났던 아카디안 조산운동으로 특징 지워 지며, 후기 알레게니안 조산운동에 부분적으로 영향을 받은 것으로 판단한다.

Antioxidant and Anticancer Properties of the Extracts from Lepisorus thunbergianus (Kaulf.) Ching (일엽초 추출물의 항산화 및 항암 효과)

  • Lepisorus thunbergianus (Kaulf.) Ching has been used in folk medicine in Korea. In this study, a L. thunbergianus methanol extract and its fractions were investigated for their antioxidant properties. The results showed that the ethyl acetate and butanol fractions of L. thunbergianus possess potent DPPH radical scavenging activities. Both fractions also possessed reducing power and inhibited reactive oxygen species formation. In addition, the cytotoxic activity of the L. thunbergianus n-hexane fraction (HF) was investigated. The results suggested that the HF remarkably suppressed proliferation of human breast, liver and colon cancer cells. These results demonstrate, for the first time, that L. thunbergianus extract induces apoptosis in SW620 cells, suggesting that L. thunbergianus may have potential as a therapeutic agent for colon cancer.
