Wild Boar (Sus scrofa corranus Heude ) Habitat Modeling Using GIS and Logistic Regression (GIS와 로지스틱 회귀분석을 이용한 멧돼지 서식지 모형 개발)

  • Accurate information on habitat distribution of protected fauna is essential for the habitat management of Korea, a country with very high development pressure. The objectives of this study were to develop a habitat suitability model of wild boar based on GIS and logistic regression, and to create habitat distribution map, and to prepare the basis for habitat management of our country s endangered and protected species. The modeling process of this restudyarch had following three steps. First, GIS database of environmental factors related to use and availability of wild boar habitat were built. Wild boar locations were collected by Radio-Telemetry and GPS. Second, environmental factors affecting the habitat use and availability of wild boars were identified through chi-square test. Third, habitat suitability model based on logistic regression were developed, and the validity of the model was tested. Finally , habitat assessment map was created by utilizing a rule-based approach. The results of the study were as folos. First , distinct difference in wild boar habitat use by season and habitat types were found, however, no difference in wild boar habiat use by season and habitat types were found , however, ho difference by sex and activity types were found. Second, it was found, through habitat availability analysis, that elevation , aspect , forest type, and forest age were significant natural environmental factors affecting wild boar hatibate selection, but the effects of slope, ridge/valley, water, and solar radiation could not be identified, Finally, the habitat at cutoff value of 0.5. The model validation showed that inside validation site had the classification accuracy of 73.07% for total habitat and 80.00% for cover habitat , and outside validation site had the classification accuracy of 75.00% for total habitat.

The influence of root surface distance to alveolar bone and periodontal ligament on periodontal wound healing

  • Purpose: The purpose of this animal study was to perform a 3-dimensional micro-computed tomography (micro-CT) analysis in order to investigate the influence of root surface distance to the alveolar bone and the periodontal ligament on periodontal wound healing after a guided tissue regeneration (GTR) procedure. Methods: Three adult Sus scrofa domesticus specimens were used. The study sample included 6 teeth, corresponding to 2 third mandibular incisors from each animal. After coronectomy, a circumferential bone defect was created in each tooth by means of calibrated piezoelectric inserts. The experimental defects had depths of 3 mm, 5 mm, 7 mm, 9 mm, and 11 mm, with a constant width of 2 mm. One tooth with no defect was used as a control. The defects were covered with a bioresorbable membrane and protected with a flap. After 6 months, the animals were euthanised and tissue blocks were harvested and preserved for micro-CT analysis. Results: New alveolar bone was consistently present in all experimental defects. Signs of root resorption were observed in all samples, with the extent of resorption directly correlated to the vertical extent of the defect; the medial third of the root was the most commonly affected area. Signs of ankylosis were recorded in the defects that were 3 mm and 7 mm in depth. Density and other indicators of bone quality decreased with increasing defect depth. Conclusions: After a GTR procedure, the periodontal ligament and the alveolar bone appeared to compete in periodontal wound healing. Moreover, the observed decrease in bone quality indicators suggests that intrabony defects beyond a critical size cannot be regenerated. This finding may be relevant for the clinical application of periodontal regeneration, since it implies that GTR has a dimensional limit.

Development of Drainage Water Disinfection System by Electric Shock in Recirculating Soilless Culture (순환식 수경재배에서 배액의 전기충격살균법 연구)

  • This study was conducted to develop the disinfection system using electric shock for recycled nutrient solution in recycling soilless culture. Stainless steel (SUS 316) was found as the most appropriate electrode material for electrical disinfection system from the view of high electrical conductivity, low electric resistance, and low price. There were no changes in nutritional elements when the electric shock passed through the nutrient solution by stainless steel electrode. The amount of electric current increased with width than thickness of the electrode. The farther the distance between the electrodes was increased the time to reach out the aimed amount of current. The electric shock was applied to Ralstonia solanacearum and Fusarium oxysporum as representative bacteria and also fungi. Any of those pathogens were killed with the percentage of higher than 97% in the condition of 15VDC or 24VDC.

  • The heavy moderator acts as the ultimate heat-sink in an operating CANDU reactor. HUKINS has been developed to investigate moderator flow patterns. HUKINS consists of a 38.4-mm-thick cylindrical shell with a 0.95 m inner diameter and 88 sus-tubes that produce a total heat of 10 kW. A chemical visualization method was selected to estimate the occurrence of typical moderator flow patterns. Momentum-dominated flow, mixed flow, and buoyancy-dominated flow are detected under conditions of a heat load of 7.7 kW and input mass flow rates of 4, 7, and 11 L/min. The experimental results are similar to the results of a CFD simulation that consisted of approximately 1.9 million grids and was conducted using the k-${\varepsilon}$ turbulence model. Therefore, both the present experiments and simulations using HUKINS, a 1/8-scale model, represent all three important flow patterns expected in the real CANDU6 reference reactor. Thus, it has been demonstrated that HUKINS could be useful in the study of CANDU6 moderator circulation.

An Analysis on Aspects of Farm Lands Damaged by the Wild Boar (Sus scrofa) in Gyeongnam Province, Korea (경상남도 농경지에서 멧돼지에 의한 피해 경향 분석)

  • Wild boars are one of the major wild life animals of which the number has increased a lot because there has been any predator and/or competitor in the Korean ecosystem. The increase of their number was a cause damaging crops in farm lands during the growing season in South Korea. This study was done to recognize the spatial pattern of farm lands damaged by wild boars. Totally 2,342 farms were known damaged by wild boars in 2012, and used to statistically analyze the perspectives of the farm land damages by wild boars in ArcMap v. 9.3. Damages by wild boars frequently happened in the western part of Gyeongnam Province including Jinju city, Tongyoung city and Namhae county. Most farm lands damaged were located nearby large mountains in this area. It might cause the number of wild boars increased in this area, which could finally stimulate the increase of farm land damaged by the species. Farm land damages by wild boars were also coincident with the preference of wild boars on their food. They preferred crops (e.g., sweet potato and corn) in uplands and rice paddies and orchards. The reason of their preference on rice, upland crops and fruits was related to the efficiency of their getting much more energy in a unit area. Another reason for the species to come into a rice paddy would be that they enjoy mud bath in there for scraping off parasites such as ticks and lice. Wild boars were seemed much overcrowded during the period from July to October when most of crops and fruits get ripen. About three-quarters of total farm land damages happened in this period. This analysis also said that 1,915 fields (81.8% of total targets) appeared within the 100-meter buffer from boundaries of mountain areas. This meant that wild boars were more sensitive to the anthropogenic land uses than we expected. They seemed to conservatively try their feeding activities in farm lands with paying attention to the human activity.

Cloning and sequence analysis of Wild Argali short palate, lung and nasal epithelium clone 1 cDNA

  • Objective: Experiments were conducted to clone the sequence of Wild Argali short palate, lung and nasal epithelium clone 1 (SPLUNC1) cDNA, and to lay the foundation for further study the biological function of Wild Argali SPLUNC1. Methods: The complete sequence of Wild Argali SPLUNC1 cDNA was generated by rapid amplification of cDNA ends. The entire coding sequence was inserted into the pPIC9K vector and expressed in Pichia pastoris (P. pastoris) GS115. The recombinant SPLUNC1 protein was detected by Western blot and purified by $Ni^{2+}$ chelate affinity chromatography. The test of effect of the protein on Mycoplasma ovipneumoniae (MO) was performed with real-time polymerase chain reaction. Results: The Wild Argali SPLUNC1 cDNA was 1,076 bp with an open reading frame of 768 bp, which encoded a 26.49 kDa protein composed of 255 amino acids. Its amino acid sequence shared 98.4%, 96.9%, 94.5%, 90.2%, 80.8%, 78.4%, 78.3%, 72.5%, 72.3%, 68.8% identity with those of SPLUNC1 cDNA from Ovis aries (accession no. NP_001288334.1), Capra hircus (accession no. XP_005688516.1), Pantholops hodgsonii (accession no. XP_005979709.1), Bos taurus (accession no. NP_776851.1), Felis catus (accession no. XP_006929910.1), Homo sapiens (accession no. NP_001230122.1), Sus scrofa (accession no. NP_001005727.1), Chinchilla lanigera (accession no. NP_001269294.1), Mus musculus (accession no. NP_035256.2), and Rattus norvegicus (accession no. NP_742028.1), respectively. The recombinant protein corresponded to the expected molecular mass of 25.47 kDa as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and it was detected in the supernatant of P. pastoris, and it could be purified. The results from the test of inhibition effect of argali recombinant SPLUNC1 protein on MO showed that the product could inhibit MO very well (p<0.01). Conclusion: The amino acid sequence of Wild Argali SPLUNC1 was different from other organisms. The recombinant SPLUNC1 protein has good biological activity.

Feeding Effect of Oriental Medicine on the Functional Properties of Pig Meat (한약재 부산물 투여가 돈육의 기능성에 미치는 영향)

  • To evaluate the feeding effect of oriental medicine on the functional properties of pork, male pigs(Sus scrofa var. domesticus L.) were fed commercial basic diets containing 1.0%, 3.0% and 7.0% of oriental medicine complex from 30 or 45 days before slaughter. The growth pattern and physical conditions of pigs during the feeding period were checked, and after slaughter, the taste of pork and biological characteristics of serum were analyzed. Body weight gain was significantly increased in case of 45 day feeding groups of 3.0 and 7.0% compared with control group (p<0.05), whereas food intakes were slightly decreased in these groups. Triglyceride and total cholesterol levels were effectively decreased in the same feeding groups compared with control group (p<0.01~0.001). Three percent feeding group not only effectively decreased the LDL-cholesterol levels, but also sig-nificantly decreased the atherogenic index in 30 days(p<0.001). Malondialdehyde levels and hy-droxyl radical formations were effectively inhibited in all oriental medicine feeding groups. Superoxide dismutase activities were effectively increased only in 3.0% feeding groups, HDL-cholesterol levels almost did not change in 3.0% and 7.0% feeding groups in 30 days. External and sensory evaluations make satisfactory results in all oriectal medicines feeding groups. According to the experimental results, the growth pattern and physical nomditions of the pigs fed oriental medicine without feeding any antibiotics were relatively superior to those of control group. The authors suggest that, if more than 3.0% of oriental medicine were fed to the pigs from more then 30 days before slaughter the pork is reatively better than those of general pork not only for the modulating the chronic degenerative diseases, but also for its qualities and tastes.

Genome-wide association study reveals genetic loci and candidate genes for average daily gain in Duroc pigs

  • Objective: Average daily gain (ADG) is an important target trait of pig breeding programs. We aimed to identify single nucleotide polymorphisms (SNPs) and genomic regions that are associated with ADG in the Duroc pig population. Methods: We performed a genome-wide association study involving 390 Duroc boars and by using the PorcineSNP60K Beadchip and two linear models. Results: After quality control, we detected 3,5971 SNPs, which included seven SNPs that are significantly associated with the ADG of pigs. We identified six quantitative trait loci (QTL) regions for ADG. These QTLs included four previously reported QTLs on Sus scrofa chromosome (SSC) 1, SSC5, SSC9, and SSC13, as well as two novel QTLs on SSC6 and SSC16. In addition, we selected six candidate genes (general transcription factor 3C polypeptide 5, high mobility group AT-hook 2, nicotinamide phosphoribosyltransferase, oligodendrocyte transcription factor 1, pleckstrin homology and RhoGEF domain containing G4B, and ENSSSCG00000031548) associated with ADG on the basis of their physiological roles and positional information. These candidate genes are involved in skeletal muscle cell differentiation, diet-induced obesity, and nervous system development. Conclusion: This study contributes to the identification of the casual mutation that underlies QTLs associated with ADG and to future pig breeding programs based on marker-assisted selection. Further studies are needed to elucidate the role of the identified candidate genes in the physiological processes involved in ADG regulation.

Non-linear tensile behavior of high manganese steel based on elasto-plastic damage model (탄-소성 손상모델을 활용한 고망간강의 인장거동 모사에 관한 연구)

  • High manganese steel exhibits excellent mechanical properties with respect to strength and durability at low temperatures. Recently, high manganese steel has been considered as an alternative to existing materials, such as nickel steel and SUS304L for application as tank material for Liquefied Natural Gas (LNG) cargo containment systems. In the present study, tensile tests were performed at room and cryogenic temperatures in order to investigate the mechanical properties and non-linear tensile behavior of high manganese steel. In addition, elasto-plastic damage model was applied using the finite element analysis software ABAQUS via a user defined material subroutine (UMAT) to describe the material behavior. Finally, the results of the finite element simulations using the UMAT were compared to those of the tensile tests in order to validate the proposed UMAT. It has been demonstrated that the UMAT can effectively describe the non-linear tensile behavior of high manganese steel.

Mad1p, a Component of the Spindle Assembly Checkpoint in Fission Yeast, Suppresses a Novel Septation-defective Mutant, sun1, in a Cell Division Cycle

  • Schizosaccharomyces pombe is suited for the study of cytokinesis as it divides by forming a septum in the middle of the cell at the end of mitosis. To enhance our understanding of the cytokinesis, we have carried out a genetic screen for temperature-sensitive S. pombe mutants that show defects in septum formation and cell division. Here we present the isolation and characterization of a new temperature-sensitive mutant, sun1(septum uncontrolled), which undergoes uncontrolled septation during cell division cycle at restrictive temperature $(37^{\circ}C)$. In sun1 mutant, actin ring and septum are positioned at random locations and angles, and nuclear division cycle continues. These observations suggest that the sun] gene product is required for the proper placement of the actin ring as well as precise septation. The sun] mutant is monogenic recessive mutation unlinked to previously known various cdc genes of S. pombe. In a screen for $sunl^+$ gene to complement the sun] mutant, we have cloned a gene, $susl^+$(suppressor of sun1 mutant), that encodes a protein of 689 amino acids. The predicted amino acid sequence of $susl^+$ gene is similar to the human hMadlp and Saccharomyces cerevisiae Mad1p, a component of the spindle checkpoint in eukaryotic cells. The null mutant of $susl^+$ gene grows normally at various temperatures and has the increased sensitivity to anti-microtubule drug, while $susl^+$ mutant shows no sensitivity to microtubule destabilizing drugs. The putative S. pombe Sus1p directly interacts with S. pombe Mad2p in yeast two-hybrid assays. These data suggest that the newly isolated susr gene encodes S. pombe Mad1p and suppresses sun] mutant defective in controlled septation in a cell division cycle.
