Identification of Ran-binding protein M as a stanniocalcin 2 interacting protein and implications for androgen receptor activity

  • Stanniocalcin (STC), a glycoprotein hormone originally discovered in fish, has been implicated in calcium and phosphate homeostasis. While fishes and mammals possess two STC homologs (STC1 and STC2), the physiological roles of STC2 are largely unknown compared with those of STC1. In this study, we identified Ran-binding protein M (RanBPM) as a novel binding partner of STC2 using yeast two-hybrid screening. The interaction between STC2 and RanBPM was confirmed in mammalian cells by immunoprecipitation. STC2 enhanced the RanBPM-mediated transactivation of liganded androgen receptor (AR), but not thyroid receptor ${\beta}$, glucocorticoid receptor, or estrogen receptor ${\beta}$. We also found that AR interacted with RanBPM in both the absence and presence of testosterone (T). Furthermore, we discovered that STC2 recruits RanBPM/AR complex in T-dependent manner. Taken together, our findings suggest that STC2 is a novel RanBPM-interacting protein that promotes AR transactivation.

혈청 칼슘 농도에 미치는 넙치 유전자 재조합 스타니오칼신의 효과 (Effect of Recombinant Olive Flounder Stanniocalcin on Serum Calcium Levels)

  • Stanniocalcin 1 (STC1) is a glycoprotein hormone that is important in the maintenance of calcium and phosphate homeostasis in both fish and mammals. STC1 and its paralog STC2 are expressed in multiple tissues in fishes, although the physiological roles of piscine STCs are still unclear compared with those of mammals. In this study, we cloned olive flounder STC1 (ofSTC1) and ofSTC2 cDNAs into pET28a vector and used E. coli Rosetta (DE3) as the host strain for protein expression. Expression experiments were carried out using isopropyl-$\beta$-D-thiogalactoside (IPTG) and nickel affinity chromatography. We could identify the recombinant proteins as single 29.5 kDa (ofSTC1) and 33.2 kDa (ofSTC2) bands in the insoluble fraction on sodium dodecyl sulfate- polyacrylamide gel electrophoresis (SDS-PAGE). These results indicate that ofSTC1 and ofSTC2 were expressed as insoluble proteins in E. coli. Furthermore, the injection of ofSTC1 protein into juvenile tilapia resulted in a decrease of the serum calcium level. These results suggest that the purified fish STC1 and STC2 proteins may be used to elucidate the physiological role of STCs in fishes.

Effect of Sea Tangle (Laminaria japonica) and Charcoal Supplementation as Alternatives to Antibiotics on Growth Performance and Meat Quality of Ducks

  • A total of 150 growing ducks were assigned to five dietary treatments to study the effect of sea tangle and charcoal (STC) supplementation on growth performance and meat characteristics in a completely randomized design. There were six replicates and five ducklings in each replication. The five dietary treatments were control, antibiotic, and 0.1%, 0.5%, and 1% STC supplemented diets. No significant differences were found on ADG, ADFI, and gain:feed among treatments in different weeks. The overall (0 to 3 weeks) ADFI decreased in antibiotic treatment (p<0.05) whereas the gain:feed increased significantly upon 1.0% STC supplementation compared to control (p<0.05). No significant variation was found in meat chemical composition except crude fat content which was high in 1.0% STC dietary group (p<0.05). Meat cholesterol was reduced in 0.1% STC group (p<0.05) compared to other dose levels while serum cholesterol was unaffected. High density lipoprotein (HDL) content was high in 1.0% STC (p<0.05) and low density lipoprotein (LDL) was low in 0.1% and 1.0% STC dietary groups (p = 0.06). No significant effect was found on the thiobarbituric acid reactive substances (TBARS) of fresh meat, whereas the TBARS value of meat preserved for 1 week was reduced significantly in STC dietary groups (p<0.05). The 0.1% STC dietary group showed an increased myristic acid (p = 0.07) content whereas, the content of eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids increased in STC supplementation than antibiotic group (p<0.05). An increased concentration of omega-3 fatty acids and a reduced ratio of n-6/n-3 PUFA ratio was found upon 1.0% STC supplementation compared to antibiotic dietary group (p<0.05). Therefore, 1.0% STC dietary supplementation can be used as alternatives to antibiotics in duck production.

모형공에서의 음파실험을 통한 STC 기법의 적용과 특성 비교 (A Study on Slowness Time Coherence Application and its Characteristics from Sonic Log Experiment in Model Holes)

  • 소채널 소구경 음파검층에서 정확한 속도분석을 하기 위한 연구의 일환으로 다채널 석유검층에서 많이 이용되고 있는 STC 방법을 물성을 달리하는 콘크리트 재질로 제작된 3개의 NX 모형공에서의 3 채널 음파검층에 적용해 보고 그 결과를 분석하였다. 자료취득에 이용한 음원의 주 주파수가 측정된 음파파형 및 진폭스펙트럼 그리고 속도분석기법으로서의 STC 특성에 미치는 영향을 비교 분석하였으며, 적절한 음원의 주 주파수의 선택과 셈블런스 투영을 포함한 STC 기법 적용으로 모드별 속도를 효과적으로 결정할 수 있음을 보였다. 이론적 모형 및 모형 시추공 시험자료를 이용한 STC 기법에 대한 연구 결과, 소구경 천부검층에서 4개의 수진기 조합이 가장 바람직한 것으로 나타났으며, 초동피킹 방법을 효과적으로 병행한다면 3개 수진기에서의 적용도 효과적일 수 있음을 보였다.

  • Tuning of digital STC parameters for the system with unknown and time varying parameters as well as delays presents many preconditions and difficulties. In order to eliminate these difficulties and to enhance STC control functions, the present study employs the metbod of expert control for STC and to implement it in nod-minimum phase plants. In this paper, implimedtation of the Expert STC for process control, STC. parameters is carried out by tuning systematically and by applying these parameters to non-minimum phase plant. This controller supervises a status of the plant, and robust control function for time delay or change of the plant is demonstrated by computer simulations.
