Fucoidan, a sulfated polysaccharide is found in several types of edible brown algae. It has shown numerous biological activities; however, the molecular mechanisms on the activity against atopic dermatitis have not been reported yet. We now examined the effects of fucoidan on chemokine production co-induced by TNF-α/IFN-γ, and the possible mechanisms underlying these biological effects. Our data showed that fucoidan inhibited the TNF-α/IFN-γ-induced production of thymus and activation-regulated chemokine (TARC) and macrophagederived chemokine (MDC) mRNA in human keratinocytes HaCaT cells. Also, fucoidan suppressed phosphorylation of nuclear factor kappa B (NF-κB) and activation of signal transducer and activator of transcription (STAT)1 in a dose-dependent manner. In addition, fucoidan significantly inhibited activation of extracellular-signal-regulated kinases (ERK) phosphorylation. These data indicate that fucoidan shows anti-inflammatory effects by suppressing the expression of TNF-α/IFN-γ-induced chemokines by blocking NF-κB, STAT1, and ERK1/2 activation, suggestive of as used as a therapeutic application in inflammatory skin diseases, such as atopic dermatitis.
Experimental studies have demonstrated that the triple combination of amantadine (A)/ ursodeoxycholic acid (UDCA, U)/ biphenyl dimethyl dicarboxylate (DDB, D) might have a preferential antiviral effect compared with that observed in interferon-induced antiviral signal pathways, such as those of $STAT1\alpha$ and the 6-16 genes. To confirm the results, this study examined whether th signal transduction for the antiviral activity in HepG2 2.2.15 was induced dependently or independently of interferon. To accomplish this, the correlation between the $STAT1\alpha$ and 6-16 genes, and nitric oxide, for the mediation of the antiviral activity was assessed. The increase in nitric oxide in the UDCA groups suggests that the inhibition of viral gene replication was enhanced by the amantadine combinations (AU and AUD), and might be more effective if incubated for longer periods. It was found that $STAT1\alpha$ was activated by the amantadine combination, although to a lesser extent than that of $interferon-\alpha$, and the primary endpoints examined for the inhibition of gene expression (HBsAg and HBcAg) were remarkably well regulated. This suggests that the amantadine triple, or at least the double, combination had better clinical benefits than those of $IFN-\alpha$ and the nucleoside analogue single treatment. This demonstrates that the amantadine combination might be a substitute for the existing HBV therapy if the results of in vivo and in vitro studies concur.
페놀화합물의 함량이 높다고 알려진 카카오(powder)로부터 카카오 추출물(CE)을 획득한 후, CE가 pancreatic lipase의 활성에 미치는 영향과 CE를 함유한 emulsion의 aging에 의한 가수분해율 변화를 살펴보기 위하여 두 가지 형태의 기질을 사용한 pH-stat digestion model을 이용하였다. Type I 의 경우 소화액에 CE와 콩기름을 첨가하여 emulsion을 제조하였으며 aging time(0, 5, 24시간)에 따른 가수분해 변화를 살펴보았다. CE를 첨가하였을 때의 가수분해율은 CE를 첨가하지 않았을 때와 큰 차이를 보이지 않아 CE가 pancreatic lipase 활성에 영향을 주지 않았다고 판단된다. 그러나 aging time이 지남에 따라 CE를 첨가하였을 때와 CE를 첨가하지 않았을 때의 가수분해율 모두 감소하였다. 이는 CE의 영향보다는 aging time이 길어짐에 따라 emulsion의 안정도가 낮아져 지방구 크기가 증가하여 가수분해율이 모두 감소한 것으로 생각되고, 따라서 이 모델에서는 CE에 의한 가수분해 저하가 크지 않았다. 한편 type II의 경우 먼저 콩기름과 CE, Tween 20를 혼합하고 고압균질기를 사용하여 micro-emulsion을 제조한 후, 이를 기질로 하여 aging time(0, 2, 4, 7, 18, 43일)에 따른 가수분해율 변화를 살펴보았다. 그 결과 aging time에 따라 CE를 첨가하지 않은 control과 CE를 첨가한 CE-emulsion의 가수분해율이 감소하였는데, 특히 control보다 CE-emulsion이 더 많이 감소하여 43일에는 control의 ${\Phi}$ max가 92.13%(0일, 96.53%)였으나 CE-emulsion은 77.69%(0일, 97.91%)를 보이면서 CE-emulsion의 가수분해 반응이 control에 비해 낮았다. 다른 kinetic parameter(k value, $t_{1/2}$ 등)에서도 이와 유사한 경향을 나타냈다.
본 논문에서는 SRC-Stat 통계패키지를 이용하여 변량효과를 적합하는 방법에 대해서 소개하고자 한다. 본 패키지를 통하여 단변량 평균 뿐만 아나리 산포 및 분산에도 변량효과를 고려하는 이중 다단계 일반화 선형모형을 적합할 수 있다. 고정효과 및 변량효과의 추정치는 다단계 우도 방법을 이용하고 있으며, 실제 자료 적합을 통해 패키지의 사용법에 대해서 설명하고자 한다.
Cryptotanshinone (CPT), is a quinoid diterpene isolated from the root of the Asian medicinal plant, Salvia miotiorrhiza bunge. Numerous researchers have found that it could work as a potent antitumor agent to inhibit tumor growth in vitro, buith there has been much less emphasis on its in vivo role against breast tumors. Using a mouse tumor model of MCF7 cells, we showed that CPT strongly inhibited MCF7 cell growth in vivo with polarization of immune reactions toward Th1-type responses, stimulation of naive CD4+ T cell proliferation, and also increased IFN-${\gamma}$ and perforin production of CD4+ T cells in response to tumor-activated splenocytes. Furthermore, data revealed that the cytotoxic activity of CD4+ T cells induced by CPT was markedly abrogated by concanamycin A(CMA), a perforin inhibitor, but not IFN-${\gamma}$ Ab. On the other hand, after depletion of CD4+ T cells or blocked perforin with CMA in a tumor-bearing model, CPT could not effectively suppress tumor growth, but this phenomenon could be reversed by injecting naive CD4+ T cells. Thus, our results suggested that CPT mainly inhibited breast tumor growth through inducing cytotoxic CD4+ T cells to secrete perforin. We further found that CPT enhanced perforin production of CD4+ T cells by up-regulating JAK2 and STAT4 phosphorylation. These findings suggest a novel potential therapeutic role for CPT in tumor therapy, and demonstrate that CPT performs its antitumor functions through cytotoxic CD4+ T cells.
B-32 is one of a panel of monoclonal anti-idiotypic antibodies to growth hormone (GH) that we developed. To characterize and identify its potential role as a novel growth hormone receptor (GHR) agonist, we determined that B-32 behaved as a typical $Ab2{\beta}$ based on a series of enzyme-linked immunosorbent assay assays. The results of fluorescence-activated cell sorting, indirect immunofluorescence and competitive receptor binding assays demonstrated that B-32 specifically binds to the GHR expressed on target cells. Next, we examined the resulting signal transduction pathways triggered by this antibody in primary porcine hepatocytes. We found that B-32 can activate the GHR and Janus kinase (2)/signal transducers and activators of transcription (JAK2/STAT5) signalling pathways. The phosphorylation kinetics of JAK2/STAT5 induced by either GH or B-32 were analysed in dose-response and time course experiments. In addition, B32 could also stimulate porcine hepatocytes to secrete insulin-like growth factors-1. Our work indicates that a monoclonal anti-idiotypic antibody to GH (B-32) can serve as a GHR agonist or GH mimic and has application potential in domestic animal (pig) production.
In this study, we investigated the potential protective effects of (+)-afzelechin (AZC), a natural compound that is derived from Bergenia ligulata, on lipopolysaccharide (LPS)-induced inflammatory responses. AZC is known to have antioxidant, anticancer, antimicrobial, and cardiovascular protective properties. However, knowledge regarding the therapeutic potential of AZC against LPS-induced inflammatory responses is limited. Thus, we investigated the protective attributes of AZC against inflammatory damage caused by LPS exposure. We examined the effects of AZC on heme oxygenase (HO)-1, cyclooxygenase (COX)-2, and inducible nitric oxide synthase (iNOS) in LPS-activated human umbilical vein endothelial cells (HUVECs). In addition, the effects of AZC on the expression of iNOS, tumor necrosis factor (TNF)-α, and interleukin (IL)-1β were analyzed in the lung tissues of LPS-injected mice. Data revealed that AZC promoted the production of HO-1, inhibited the interaction between luciferase and nuclear factor (NF)-κB, and reduced the levels of COX-2/PGE2 and iNOS/NO, thereby leading to a decrease in the signal transducer and activator of transcription (STAT)-1 phosphorylation. Moreover, AZC facilitated the nuclear translocation of Nrf2, increased the binding activity between Nrf2 and the antioxidant response elements (AREs), and lowered the expression of IL-1β in the LPS-treated HUVECs. In the animal model, AZC significantly reduced the expression of iNOS in the lung tissue structure and the TNF-α level in the bronchoalveolar lavage fluid. These findings demonstrate that AZC possesses anti-inflammatory properties that regulate iNOS through the inhibition of both NF-κB expression and p-STAT-1. Consequently, AZC has potential as a future candidate for the development of new clinical substances for the treatment of pathological inflammation.
목 적: 단백질 인산화는 세포신호전달에 매우 중요한 현상으로서, 수많은 조절인자들이 난자성숙에 관여하게 된다. 그러나 이들 중에서 어떤 단백질이 인산화되어 난자성숙을 조절하는지는 잘 알려져 있지 않다. 따라서 체세포의 신호전달과정에서 인산화를 통해 중요한 기능을 한다고 알려져 있는 일곱 가지 단백질들이 생쥐의 난자성숙과정에서 어떻게 인산화 되고 있는지 알아보고자 한 개 샘플에서 일곱 개의 변화를 한꺼번에 측정할 수 있는 bead-based multiplex phosphorylation assay를 이용하여 본 연구를 수행하였다. 연구방법: ICR 생쥐에 PMSG를 주사하고 46시간 후에 cumulus-oocyte complex (COCs) 형태로 미성숙 난자를 채취한 후 체외배양 하면서, 배양 2시간 후에 GVBD를, 배양 8시간 후에 MI을, 배양 16시간 후에 MII 단계의 난자를 얻었고 체내에서 배란한 MII 단계의 난자는 수란관에서 얻었다. 각 단계의 난자를 100개씩 모아서 mitogen-activated protein kinase (MAPK)에 속하는 세가지 단백질인 ERK1/2, JNK, p38 MAPK와 Akt, GSK-$3{\alpha}/{\beta}$, $I{\kapa}B{\alpha}$, STAT3 등 총 일곱 단백질의 인산화를 Bio-Plex System을 이용하여 같은 시료에서 동시에 측정하였으며 세 번의 반복실험을 통하여 얻어진 결과를 통계적으로 분석하였다. 결 과: 생쥐의 난자성숙과정에서 측정된 일곱 가지 단백질 중에서 인산화가 현저히 증가하는 단백질로는 ERK1/2, JNK, p38 MAPK와 STAT3로서 미성숙 난자에 비해서 3배에서 20배까지 인산화되는 결과를 보였다. 반면에 GSK-$3{\alpha}/{\beta}$, $I{\kappa}B{\alpha}$의 인산화의 변화는 미약하였으며, Akt의 경우에는 변화가 전혀 없었다. 난자성숙 과정에서 분석 대상 단백질들의 인산화는 GVBD 단계에서 활성화되기 시작하여 MI에서 현저히 높게 증가하며 MII까지 높게 유지되었다. 결 론: 본 연구는 난자성숙과정에서 일곱 가지 단백질의 인산화를 동시에 측정한 최초의 보고로서 이 방법은 난자와 같이 적은 양의 시료에서의 여러 개의 단백질 인산화를 동시에 분석하는데 유용할 것으로 생각된다. 본 연구결과, 세 가지 MAPK 단백질인 ERK1/2, JNK, p38 MAPK 외에도 STAT3가 난자성숙에 있어서 매우 중요한 조절자로 생각되었다. 또한 Akt의 473번 serine기의 인산화는 난자성숙에 관여하지 않음을 알 수 있었다.
목적 : 이 연구는 $Vipera$$lebetina$$turanica$ 사독(蛇毒)이 인간 대장암 세포주인 HCT116 세포에서 세포주기진행, death receptor 의존적 세포자멸사 경로 관련단백질 발현 및 NK-${\kappa}B$와 STAT3 활성에 미치는 영향을 규명함으로써 대장암 세포 성장에 대한 억제와 그 기전에 대하여 살펴보고자 하였다. 방법 : 사독을 처리한 후 HCT116의 세포주기를 분석하기 위해서 FACS analysis를 시행하였고, apoptosis 평가에는 TUNEL assay를 시행하였으며 death receptor 의존적 세포자멸사 경로 관련단백질 및 NF-${\kappa}B$와 STAT3 활성 변동 관찰에는 RT-PCR 및 western blot analysis를 시행하였다. 결과 : 1. 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 등의 사독을 처리한 결과 농도 의존적으로 HCT116 대장암 세포활성의 억제가 나타났다. 2. 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 등의 사독을 처리한 결과 농도의존적으로 세포자멸사 활성세포의 증가가 나타났고, SVT $1{\mu}g/m{\ell}$에서는 60-70%의 대장암세포 억제 효과가 나타났다. 3. 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 등의 사독을 처리한 결과 약한 G1 arrest와 강한 G2/M arrest가 나타났고, G0/G1 또는 G2/M 관련 cyclin D, E 및 B1의 증가가 나타났다. 4. 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 등의 사독을 처리한 결과 death receptor4, 5의 발현증가와 그에 따른 세포자멸사 촉진 Bax, PARP, caspase-3, -8, -9 발현 증가 및 세포자멸사 억제의 Bcl-2의 발현 감소 등이 나타났다. 6. 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 등의 사독을 처리한 결과 NF-${\kappa}B$와 STAT3의 활성변동은 관찰되지 않았다. 결론 : 이상의 연구에서 사독은 death receptor 의존적인 세포자멸사를 촉진하여 대장암의 화학치료 내성을 극복할 수 있는 하나의 대안이 될 것으로 생각되지만 보다 심화된 연구가 필요할 것으로 사료된다.
알콜증류폐액에 빵효모를 배양하는 중에 glucose와 ammo-nium을 자동첨가함으로써 폐액배양액의 균체농도를 높여서 알콜증류폐액의 이용성을 증가시키고자 하였다. 배양중에 glucose를 공급하여 줌으로서 공급하지 않은 경우보다 1.3배 더 높은 균체농도를 얻었으며, glucose와 (NH4)2SO4를 공급하여 줌으로써 5.8배 더 높은 균체농도를 얻었다. Glucose의 경우에는 배양액의 DO를 제어파라미터로 하는 제어방법을 사용하여 자동첨가하고, ammonium의 경우에는 pH조정제로서 NH4OH을 사용함으로서 pH제어기에 의하여 자동으로 공급되도록 배양하였다. 배양결과 증식의 정체없이 배양 22.5시간후에 최대 12.0 g/L의 건조균체량에 도달하였으며, 균의 최대비증식속도는 0.18 hr-1였고, 대당균체수율이 약 0.54g/gdldjTdmamfh glucose의 첨가가 경제적이라고 생각되었다. 증균배양으로 폐액배양액의 COD를 약 22% 더 감소시킬 수 있었다.
