메탄올로부터 Methylobacterium organophilum에 의한 Poly-$\beta$-hydroxybutyric Acid의 생산과 배지성분의 최적화 (Optimization of Growth Medium and Poly-$\beta$-hydroxybutyric Acid Production from Methanol in Methylobacterium organophilum)

  • Facultative methylotroph인 Methylobacterium organophilum을 유일한 탄소원으로 메탄올을 0.5% (v/v) 함유한 최소 합성배지를 이용하여 pH 6.8과 3$0^{\circ}C$에서 배양하였다. 세포의 증식은 배지 내의 여러 성분들에 의해 영향을 받았으며 이로부터 최대증식속도를 얻을 수 있는 최적 합성배지 조성을 확립하였다. 배지조성 중 질소원이 결핍되면 세포의 증식이 감소하며 Fe$^{+2}$ 또는 Mn$^{+2}$ 이온의 결핍은 세포의 비증식속도를 감소시켰다. 탄소원인 메탄올은 농도를 증가시킬수록 세포의 비증식속도가 감소하는 메탄올의 기질 저해성을 확인하였으며, 4%(v/v) 이상의 메탄을 농도에서는 세포의 증식이 완전히 저해되었다. 이러한 메탄올의 기질 저해성은 간헐식 유가배양법을 이용하여 극복하고 PHB의 축적을 촉진하는 영양분 제한(nutrient-deficiency) 실험을 수행하였다. 합성배지의 영양분 중 NH$_{4}^{+}$, SO$_{4}^{-2}$, $K^+$ 또는 PO$_{4}^{-3}$ 이온을 결핍시켰을 때 M. organophilum의 PHB 생산이 증가하였으며 칼륨이온을 결핍시켰을때 세포의 건조 중량에 대한 PHB 함량은 최대 58%를 얻었다.

GATA2-Mediated Transcriptional Activation of Notch3 Promotes Pancreatic Cancer Liver Metastasis

  • The liver is the predominant metastatic site for pancreatic cancer. However, the factors that determine the liver metastasis and the specific molecular mechanisms are still unclear. In this study, we used human pancreatic cancer cell line Hs766T to establish Hs766T-L3, a subline of Hs766T with stable liver metastatic ability. We performed RNA sequencing of Hs766T-L3 and its parental cell line Hs766T, and revealed huge differences in gene expression patterns and pathway activation between these two cell lines. We correlated the difference in pathway activation with the expression of the four core transcriptional factors including STAT1, NR2F2, GATA2, and SMAD4. Using the TCGA database, we examined the relative expression of these transcription factors (TFs) in pan-cancer and their relationship with the prognosis of the pancreatic cancer. Among these TFs, we considered GATA2 is closely involved in tumor metastasis and may serve as a potential metastatic driver. Further in vitro and in vivo experiments confirmed that GATA2-mediated transcriptional activation of Notch3 promotes the liver metastasis of Hs766T-L3, and knockdown of either GATA2 or Notch3 reduces the metastatic ability of Hs766T-L3. Therefore, we claim that GATA2 may serve as a metastatic driver of pancreatic cancer and a potential therapeutic target to treat liver metastasis of pancreatic cancer.

Anti-tumor activities of Panax quinquefolius saponins and potential biomarkers in prostate cancer

  • Background: Prostate carcinoma is the second most common cancer among men worldwide. Developing new therapeutic approaches and diagnostic biomarkers for prostate cancer (PC) is a significant need. The Chinese herbal medicine Panax quinquefolius saponins (PQS) have been reported to show anti-tumor effects. We hypothesized that PQS exhibits anti-cancer activity in human PC cells and we aimed to search for novel biomarkers allowing early diagnosis of PC. Methods: We used the human PC cell line DU145 and the prostate epithelial cell line PNT2 to perform cell viability assays, flow cytometric analysis of the cell cycle, and FACS-based apoptosis assays. Microarray-based gene expression analysis was used to display specific gene expression patterns and to search for novel biomarkers. Western blot and quantitative real-time PCR were performed to demonstrate the expression levels of multiple cancer-related genes. Results: Our data showed that PQS inhibited the viability of DU145 cells and induced cell cycle arrest at the G1 phase. A significant decrease in DU145 cell invasion and migration were observed after 24 h treatment by PQS. PQS up-regulated the expression levels of p21, p53, TMEM79, ACOXL, ETV5, and SPINT1 while it down-regulated the expression levels of bcl2, STAT3, FANCD2, DRD2, and TMPRSS2. Conclusion: PQS promoted cells apoptosis and inhibited the proliferation of DU145 cells, which suggests that PQS may be effective for treating PC. TMEM79 and ACOXL were expressed significantly higher in PNT2 than in DU145 cells and could be novel biomarker candidates for PC diagnosis.

소아 결절성 십이지장염의 임상적 및 조직병리학적 소견 (Clinical and Histopathologic Features and Their Correlations in Children with Nodular Duodenitis)

  • 목 적: 결절성 십이지장염은 내시경 검사상 십이지장에 두 개 이상의 홍반성 결절들이 보이며 주변에 염증 소견이 관찰되는 질환이다. 최근 위내시경 검사의 발달로 인해 결절성 십이지장염은 소아에서도 발생빈도가 증가하는데 아직 국내외적으로 전반적인 연구가 미미한 실정이다. 이에 저자들은 소아에서 결절성 십이지장염의 임상적 및 조직병리학적 소견을 조사하여 이 질환에 대한 전반적인 이해를 도모하고자 본 연구를 시행하였다. 방 법: 1995년 1월부터 1999년 12월까지 서울적십자병원 소아내시경실에서 시행된 상부위장관 내시경 검사상 결절성 십이지장염으로 진단된 39례를 대상으로 유병률, 성별 및 연령별 분포, 주증상, 내시경 소견과 조직병리학적 소견 및 이의 상관관계 등에 대해 후향적으로 연구를 실시하였다. 내시경 검사상 병변의 정도는 십이지장 결절의 크기와 분포에 따라 제 1, 2, 3 도로 분류하였고 십이지장의 조직병리학적 검사는 십이지장 고유층의 염증세포 침윤 정도, 상피세포 및 융모의 소실 정도에 따라 정상, 제 1, 2, 3 도로 분류하였다. 위의 조직학적 소견은 위점막의 고유판과 상피내에 염증세포의 침윤 정도 및 상피의 손상 정도에 따라 0에서 6까지 구분하였다. H. pylori의 감염 정도는 Prieto 등의 분류법에 따라 제 0, 1, 2, 3 도로 구분하였다. 십이지장에서 내시경적 소견의 정도와 조직병리학적 소견의 정도 및 H. pylori 감염 정도 간에 상관관계가 있는지 알아보기 위해 통계 프로그램으로 Graph PAD InStat의 linear regression module을 사용하여 correlation coefficient와 p-value를 구하였다. 결 과: 1) 결절성 십이지장염은 17.1%의 유병률을 나타내었고 남녀비는 1 대 2이었다. 7세부터 12세까지에서 가장 빈도(61.5%)가 높았고 주증상은 복통이 69.2%로 가장 많았다. 2) 본 질환의 내시경 등급은 제 1 도 45.7%, 제 2도 34.3%, 제 3 도 20.0%이었으며 동반된 내시경적 소견상 결절성 위염은 28.3%이었다. 3) 십이지장의 조직병리학적 검사상 제 1 도는 22.9%, 제 2 도는 54.3%, 제 3 도는 22.9%이었으며 위의 조직검사상 제 1 도는 22.9%, 제 2 도는 42.9%, 제 3 도는 25.7%, 제 4 도는 8.6%이었다. 4) 십이지장의 조직병리학적 검사상 H. pylori의 감염 정도는 제 1 도 5.7%, 제 2 도 8.6%, 제 3 도 22.9%로 37.1%의 십이지장 조직에서 H. pylori가 발견되었다. 위의 조직병리학적 검사상 H. pylori의 감염정도는 제 1 도 11.4%, 제 2 도 11.4%, 제 3 도 8.6%로 31.4%의 위 조직에서 H. pylori가 발견되었다. 5) 본 질환의 내시경적 등급과 조직병리학적 등급 간의 상관계수는 0.3983 (p=0.0178)이었고, 조직병리학적 등급과 십이지장 조직에서의 H. pylori 감염 등급간의 상관계수는 0.5154 (p=0.0018)이었다. 결 론: 결절성 십이지장염의 내시경적 등급과 조직병리학적 등급 사이에 통계적으로 의미있는 상관관계가 나타났으며(r=0.3983, p=0.0178), 또한 조직병리학적 등급과 십이지장 조직에서의 H. pylori 감염 등급간에도 통계적으로 의미있는 상관관계를보였다(r=0.5154, p=0.0018). 따라서 H. pylori 감염이 본 질환의 발생에 주요한 역할을 할 것으로 여겨지며 이외의 유발 원인에 대해서는 추후 지속적인 추적 관찰 및 보다 많은 연구가 시행되어야 하겠다.

선방활명음(仙方活命飮)이 알레르기 비염을 동반한 급성 비부비동염 마우스에 미치는 영향 (The Effects of Sunbanghwalmyung-eum Extract on Acute Sinusitis-induced Mouse)

  • Background and Objectives: Sinusitis, referring to an inflammatory disease that occurs on the mucus membrane inside the sinus, is one of the most common diseases in the otorhinolaryngology area. In oriental clinic, Sunbanghwalmyungeum (SHE) has been used as a primary prescription to treat inflammatory diseases and intumescence and to treat sinusitis. The aim of this study was to investigate the anti-inflammatory and anti-allergic effects of SHE on acute sinusitis induced mice. Materials and Methods: BALB/c mice were divided into three groups: the normal group, the group inoculated with S. pneumoniae which caused them allergic rhinitis (control group), and the group treated with the SHE extract after it was treated the same as the control group (sample group). We investigated the inhibition of Th 2 cell differentiation by SHE and the suppression of NF-${\kappa}B$ activation. Results: NF-${\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE in acute sinusitis. IL-4 and STAT 6 also appeared to be suppressed. The number of eosinophils in the sample group noticeably decreased when compared to the control group. In the general morphologic change, the increase of damaged respiratory ciliated epithelium & eosinophil's infiltration were decreased in the sample group. Goblet cells were maintained in the sample group. MIP-2 and HSP-70 decreased in the sample group. Apocrine secretion decreased in the sample group. Conclusion: The results suggest that SHE is significantly effective in the treatment of inflammation caused by acute sinusitis through the suppression of NF-${\kappa}B$ activation and the inhibition of Th 2 cell differentiation.

Combined Treatment with Stattic and Docetaxel Alters the Bax/Bcl-2 Gene Expression Ratio in Human Prostate Cancer Cells

  • Docetaxel, recognized as a stabilizing microtubule agent, is frequently administrated as a first line treatment for prostate cancers. Due to high side effects of monotherapy, however, combinations with novel adjuvants have emerged as an alternative strategy in cancer therapy protocols. Here, we investigated the combined effects of stattic and docetaxel on the DU145 prostate cancer cell line. Cytotoxicity was evaluated by MTT assay. To understand molecular mechanisms of stattic action, apoptotic related genes including Bcl-2, Mcl-1, Survivin and Bax were evaluated by real-time RT-PCR. Alteration in the expression of pro-apoptotic Bax and anti-apoptotic Bcl-2 genes and Bax/Bcl-2 ratio were investigated via the $2^{{\Delta}{\Delta}CT}$ method. The $IC_{50}$ values for docetaxel and stattic were $3.7{\pm}0.9nM$ and $4.6{\pm}0.8{\mu}M$, respectively. Evaluation of key gene expression levels revealed a noticeable decrease in antiapoptotic Bcl-2 and Mcl-1 along with an increase in pro-apoptotic Bax mRNA levels (p<0.05). Our results suggest that combination of a STAT3 inhibitor with doctaxel can be considered as a potent strategy for induction of apoptosis via increasing Bax mRNA expression.

Inflammatory Responses in a Benign Prostatic Hyperplasia Epithelial Cell Line (BPH-1) Infected with Trichomonas vaginalis

  • Trichomonas vaginalis causes the most prevalent sexually transmitted infection worldwide. Trichomonads have been detected in prostatic tissues from prostatitis, benign prostatic hyperplasia (BPH), and prostate cancer. Chronic prostatic inflammation is known as a risk factor for prostate enlargement, benign prostatic hyperplasia symptoms, and acute urinary retention. Our aim was to investigate whether T. vaginalis could induce inflammatory responses in cells of a benign prostatic hyperplasia epithelial cell line (BPH-1). When BPH-1 cells were infected with T. vaginalis, the protein and mRNA of inflammatory cytokines, such as CXCL8, CCL2, IL-$1{\beta}$, and IL-6, were increased. The activities of TLR4, ROS, MAPK, JAK2/STAT3, and NF-${\kappa}B$ were also increased, whereas inhibitors of ROS, MAPK, PI3K, NF-${\kappa}B$, and anti-TLR4 antibody decreased the production of the 4 cytokines although the extent of inhibition differed. However, a JAK2 inhibitor inhibited only IL-6 production. Culture supernatants of the BPH-1 cells that had been incubated with live T. vaginalis (trichomonad-conditioned medium, TCM) contained the 4 cytokines and induced the migration of human monocytes (THP-1 cells) and mast cells (HMC-1 cells). TCM conditioned by BPH-1 cells pretreated with NF-${\kappa}B$ inhibitor showed decreased levels of cytokines and induced less migration. Therefore, it is suggested that these cytokines are involved in migration of inflammatory cells. These results suggest that T. vaginalis infection of BPH patients may cause inflammation, which may induce lower urinary tract symptoms (LUTS).

수종의 Engine driven NiTi file과 stainless steel K-file을 이용한 근관형성 후 근관의 조직형태학적 변화에 관한 연구 (A STUDY OF HISTOMORPHOLOGICAL CHANGE OF CURVED ROOT CANAL PREPARATION USING GT ROTARY FILE, PROFILE AND STAINLESS STEEL K-FILE)

  • The purpose of this study was to compare the histomorphological change of curved root canal preparation using GT rotary File, Profile .04 taper and stainless steel K-file. 45 mesial canals(over 20 degree) of extracted human mandibular first molars were mounted in resin using a modified Bramante muffle system and divided into three groups. The roots were cross-sectioned at 2.5mm 5mm and 8mm levels from apical foramen. Tracings of the canals were made from preinstrumentation pictures of the cross section. The canals were prepared using a step-back technique with stainless steel K file(group 1), Profile .04 taper rotary file(group 2) and GT rotary file(group 3). Tracings of the prepared canals were made from postinstrumentation picture. Canal centring ratio. amount of transportation, area of dentin removed and shape of canal were measured and statistically were evaluated with Student-Newman-Keuls test using Sigma Stat(Jandel Scientific Software, USA). The results were as followings : 1 Amount of transportation of group 2 was the lowest at apical part, but there was no statistical difference. The direction of transportation was the outside of curvature at apical part. 2. Centering ratio at the apical part of group 1 was the highest, and there was statistical differences between apical and middle part, apical and coronal part(p<0.05). Centering ratio at the middle part of group 3 was the lowest, and there was statistical difference between apical and middle part(p<0.05). Centering ratio of group 2 was the lowest at apical part, but there was no statistical difference. 3. Amount of dentin removed of group 1 was the highest at coronal, middle and apical part among three groups, and there was statistical difference(p<0.05). 4. The majority of the cross-sectioned canal shape after instrumentation were irregular at coronal, middle and apical part. But there are more number of round shaped canals at group 3 than other group.

Th2 skewed condition 조절을 통한 대장점막 내 염증개선효과에 대한 두시 하태독법과 오메가3 비교 연구 (A Comparative Study of Douchi Hataedock versus Omega-3 on the Effect of Anti-inflammation in the Colonic Muscle through Th2 Skewed Condition Control)

  • Objectives Hataedock is an orally-administered herbal extract that used to treat toxic heat and help excrete meconium accumulates in the fetus from pregnancy. This study compared the effects of Hataedock using the extract of Douchi and omega-3 to control inflammation in the colonic mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), FGT group (given the extract of Douchi), and O3T group (given omega-3). The 3-week-old mouse from each group were given the extract of Douchi and omega-3. After 2 weeks, the colonic mucosa tissues of the mice were observed via Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF in the intestinal mucosa. Results In both FGT and O3T group, IL-4, IL-13, STAT-6 levels and CD40-positive reactions were decreased. However, EGF-positive responses were increased. FGT group showed 35% decrease in IL-4, 45% decrease in IL-13, 48% decrease in CD40, 50% decrease in $Fc{\varepsilon}RI$, 54% decrease in $p-I{\kappa}B$, 52% increase in EGF compared to the control group. O3T group also showed 14% decrease in IL-4, 23% decrease in IL-13, 22% decrease in CD40, 29% decrease in $Fc{\varepsilon}RI$, 41% decrease in $p-I{\kappa}B$, 39% increase in EGF compared to the control group. Conclusions Both early administration of Douchi extract and omega-3 were effective in regulating Th2, relieving inflammation and developing colonic mucosal tissues. Hataedock with Douchi extract may be more effective in modulating immune system in colonic mucosa than omega-3.

Nitric Oxide Dependency in Inflammatory Response-related Gene Transcripts Expressed in Lipopolysaccharide-treated RAW 264.7 Cells

  • Cytotoxic Nitric oxide (NO) overproduced by inducible NO Synthase (iNOS or NOS2), which was induced in inflammatory reactions and immune responses directly or indirectly affects the functions as host defense and can cause normal tissue damage. Microarray analysis was performed to identify gene profiles of both NO-dependent and -independent transcripts in RAW 264.7 macrophages that use selective NOS2 inhibitors aminoguanidine ($100\;{\mu}M$) and L-canavanine (1 mM). A total of 3,297 genes were identified that were up- or down-regulated significantly over 2-fold in lipopolysaccharide (LPS)-treated macrophages. NO-dependency was determined in the expressed total gene profiles and also within inflammatory conditions-related functional categories. Out of all the gene profiles, 1711 genes affected NO-dependently and -independently in 567 genes. In the categories of inflammatory conditions, transcripts of 16 genes (Pomp, C8a, Ifih1, Irak1, Txnrd1, Ptafr, Scube1, Cd8a, Gpx4, Ltb, Fasl, Igk-V21-9, Vac14, Mbl1, C1r and Tlr6) and 29 geneas (IL-1beta, Mpa2l, IFN activated genes and Chemokine ligands) affected NO-dependently and -independently, respectively. This NO dependency can be applied to inflammatory reaction-related functional classifications, such as cell migration, chemotaxis, cytokine, Jak/STAT signaling pathway, and MAPK signaling pathway. Our results suggest that LPS-induced gene transcripts in inflammation or infection can be classified into physiological and toxic effects by their dependency on the NOS2-mediated NO release.
