Background: Saponins are a major active component for the traditional Chinese medicine, Rubus parvifolius L., which has shown clear antitumor activities. However, the specific effects and mechanisms of saponins of Rubus parvifolius L. (SRP) remain unclear with regard to human chronic myeloid leukemia cells. The aim of this study was to investigate inhibition of proliferation and apoptosis induction effects of SRP in K562 cells and further elucidate its regulatory mechanisms. Materials and Methods: K562 cells were treated with different concentrations of SRP and MTT assays were performed to determine cell viability. Apoptosis induction by SRP was determined with FACS and DAPI staining analysis. Western blotting was used to detect expression of apoptosis and survival related genes. Specific inhibitors were added to confirm roles of STAT3 and AMPK pathways in SRP induction of apoptosis. Results: Our results indicated that SRP exhibited obvious inhibitory effects on the growth of K562 cells, and significantly induced apoptosis. Cleavage of pro-apoptotic proteins was dramatically increased after SRP exposure. SRP treatment also increased the activities of AMPK and JNK pathways, and inhibited the phosphorylation expression level of STAT3 in K562 cells. Inhibition of the AMPK pathway blocked the activation of JNK by SRP, indicating that SRP regulated the expression of JNK dependent oon the AMPK pathway. Furthermore, inhibition of the latter significantly conferred resistance to SRP pro-apoptotic activity, suggesting involvement of the AMPK pathway in induction of apoptosis. Pretreatment with a STAT3 inhibitor also augmented SRP induced growth inhibition and cell apoptosis, further confirming roles of the STAT3 pathway after SRP treatment. Conclusions: Our results demonstrated that SRP induce cell apoptosis through AMPK activation and STAT3 inhibition in K562 cells. This suggests the possibility of further developing SRP as an alternative treatment option, or perhaps using it as adjuvant chemotherapeutic agent for chronic myeloid leukemia therapy.
본 논문에서는 ECDLP(Elliptic Curve Discrete Logarithm Problem)를 이용한 EC-SRP (Elliptic Curve - Secure Remote Password) 프로토콜을 제안한다. 타원곡선 이산대수 문제를 SRP(Secure Remote Password) 프로토콜에 적용시킴으로써 타원 곡선이 갖는 높은 효율성과 보안성을 갖도록 하였으며, 이와 동시에 타원곡선의 스칼라 곱셈(scala. multiplication)의 회수를 최대한 줄임으로써 최적의 효율성을 갖도록 설계하였다. 또한 랜덤 오라클(random, oracle) 모델에서 EC-SRP 프로토콜이 안전한 AKC(Athenticated Key Agreement with Key Confirmation)프로토콜임을 증명하였다.
본 연구에서는 3상 SRP-PWM기법을 2상에 적용한 고정 스위칭 주파수의 공간벡터 SRP-PWM(Separately Randomized Pulse Position PWM)기법을 제안하고 이를 산업현장에 적용 가능한가를 검증하기 위하여, 속도 피드백 실험을 행하였다. 제안된 2상 SRP-PWM은 랜덤하게 배치된 a상의 펄스 범위 내에서 나머지 상의 펄스 위치를 완전히 랜덤하게 배치 할 수 있는 변조기법이다. 제안된 방법이 적용된 속도 피드백 시스템은 부하와 연결된 3상 유도모터의 v/f 일정 제어를 기반으로 하였다. 실험 결과, 제안된 방법이 적용된 기존의 2상 중앙정렬 SVM의 경우와 비교하여 모터의 v/f 일정 제어의 성능은 동일함을 확인할 수 있었으며, 모터의 전압 및 소음 스펙트럼의 광대역화 효과는 제안된 방법이 훨씬 우수함을 입증할 수 있었다.
본 연구는 발화범위 프로파일(Speech Range Profile, SRP) 과제를 개발하고, 개발된 SRP 과제가 최대발화범위를 측정하기에 적합한지 살펴보기 위해 45명의 18-29세 정상음성군을 대상으로 음성범위 프로파일(Voice Range Profile, VRP) 과제와 비교하였다. 이를 위해 한국어의 모든 말소리와 문장 유형을 포함하는 14개 문장으로 구성된 "불이야"문단을 개발하였다. SRP와 VRP 간의 차이를 비교하기 위해 SRP 과제로는 새롭게 개발된 문단으로 문단읽기와 21-30까지 숫자세기를 사용하였고, VRP 과제는 /a/ 모음을 낮은 음도부터 높은 음도까지 활창하기와 축약된 VRP를 사용하였다. SRP와 VRP의 변수로 최고기본주파수($F0_{max}$), 최저기본주파수($F0_{min}$), 기본주파수범위($F0_{range}$), 최대음성강도($I_{max}$), 최소음성강도($I_{min}$) 및 음성강도범위($I_{range}$)를 측정한 후 과제 간 차이를 비교하였다. 그 결과, $F0_{max}$, $F0_{min}$, $F0_{range}$, $I_{max}$ 및 $I_{range}$는 모두 문단읽기의 SRP와 활창하기의 VRP 간에 차이가 없었고, $I_{min}$은 숫자세기의 SRP가 가장 낮은 평균값을 보였다. 즉 새롭게 개발된 SRP 과제인 "불이야" 문단은 정상 음성 산출 화자에서 모음만을 통해 측정된 VRP의 음역대와 유사한 수준의 음역대를 산출할 수 있음을 알 수 있다. 이에 오랜 시간이 소요되거나 중증도의 음성장애에서 측정이 어려울 수 있는 VRP를 대신하여 기능적 말산출 과제인 SRP를 적용함으로써 국내 임상환경에서 비교적 짧은 시간 내에 음성평가를 효과적으로 실시할 수 있을 것으로 본다.
본 연구에서는 한국인 정상 성인에서 음성(VRP) 및 말소리 범위 프로파일(SRP)을 이용하여 문단 읽기 시 전기성문파형검사(EGG)를 이용하여 측정한 평균 발화 기본주파수(SFF)를 예측할 수 있는지 알아보고자 하였다. 또한 추정된 기본주파수(ESFF)와 실제 SFF 간 차이(DSFF)에 있어 성별 차이가 있는지 알아보고자 하였다. 연구대상은 정상 음성을 가진 한국어 모국어 화자 85명이었다. 각 대상자는 /a/ 발성으로 전체 음역대를 측정하는 VRP 과제, '가을' 문단의 첫 번째 문장을 읽어 말소리 산출 시 음역대를 측정하는 SRP 과제, 전체 문단을 읽어 SFF를 측정하는 문단 읽기 과제를 수행하였다. VRP와 SRP를 통해 측정된 음역대 관련 변수들와 연령, 성별이 EGG를 통해 측정된 SFF를 예측할 수 있는지 알아보기 위해 단계적 다중회귀분석을 시행하였고, 예측된 ESFF와 SFF 간 차이의 절대값(DSFF)과 그 합계를 구하였다. 연구 결과, SFF의 예측변인은 VRP에서는 최저음도, 음도범위, 성별, 연령(adjusted $R^2=.931$)이었으며, SRP에서는 반음 단위 음역대와 최고음도(adjusted $R^2=.963$)였다. VRP와 SRP를 통해 예측된 두 가지 ESFF와 실제 SFF 사이에는 강한 양의 상관관계가 있었다. VRP와 SRP를 이용한 DSFF와 그 합계에 있어 성별 차이는 없었다. 결론적으로 VRP와 SRP를 통해 문단 읽기 시 SFF를 예측할 수 있었으며, SFF의 이상을 보일 수 있는 음성장애 환자에서 후속 연구를 통하여 임상적 시사점을 탐색할 필요가 있을 것으로 여겨진다.
The effects of the agitation and aeration rates on the production of serratiopeptidase (SRP) in a 5-L fermentor (working volume 2-l) were systematically investigated using Serratia marcescens NRRL B-23112. The dissolved oxygen concentration, pH, biomass, SRP yield, and maltose utilization were all continuously measured during the course of the fermentation runs. The efficiencies of the aeration and agitation were evaluated based on the volumetric mass transfer coefficient ($K_La$). The maximum SRP production of 11,580 EU/ml with a specific SRP productivity of 78.8 EU/g/h was obtained with an agitation of 400 rpm and aeration of 0.075 vvm, which was 58% higher than the shake-flask level. The $K_La$ for the fermentation system supporting the maximum production (400 rpm, 0.075 vvm) was 11.3 $h^{-1}$. Under these fermentor optimized conditions, kinetic modeling was performed to understand the detailed course of the fermentation process. The resulting logistic and Luedeking-Piret models provided an effective description of the SRP fermentation, where the correlation coefficients for cell growth, SRP formation, and substrate consumption were 0.99, 0.94, and 0.84, respectively, revealing a good agreement between the model-predicted and experimental results. The kinetic analysis of the batch fermentation process for the production of SRP demonstrated the SRP production to be mixed growth associated.
In this paper, Inverter drives adopting 3-phase and 2-phase SRP-PWM (Separately Randomized Pulse Position PWM) with fixed switching frequency is proposed. In the proposed SRP-PWM scheme, each of 3 or 2 phase pulses Is located randomly in each switching interval. The experimental results show that the voltage / current harmonics and the switching noise harmonics are spread to a wide band area. Also, the performance of the 3-phase SRP-PWM and the 2-phase SRP-PWM are compared to each other. In result, the speed response is nearly similar to each other from the viewpoint of the v/f constant control.
Purpose: Full-mouth disinfection enables to reduce the probability of cross contamination from untreated pockets to treated ones, for completing the entire SRP under local anesthesia with chlorhexidine as a mouth wash in two visits within 24 hours. This study aimed to compare the clinical effects of modified full-mouth disinfection (Fdis) after 6 months with those of conventional SRP (cSRP). Methods: Thirty non-smoking chronic periodontitis subjects were randomly allocated two groups. The Fdis group underwent the entire SRP under local anesthesia in two visits within 24 hours, a week after receiving supragingival scaling. A chlorhexidine (0.1%) solution was used for rinsing and subgingival irrigation for Fdis. The cSRP group received SRP per quadrant under local anesthesia at one-week intervals, one week after they had received scaling. Clinical parameters were recorded at baseline, after 1, 3 and 6 months. Results: There are significant (P<0.05) decreases in the sulcus bleeding index, and plaque index, and the increases in gingival recession were significantly smaller with Fdis after six months compared with cSRP. There was significant improvement in the probing depth and clinical attachment level for initially medium-deep pockets (4-6mm) after Fdis compared with cSRP. Multi-rooted teeth showed significantly larger attachment gain up to six months after Fdis. Single-rooted teeth showed significantly more attachment gain, 1 and 6 months after Fdis. Conclusions: Fdis has more beneficial effects on reducing gingival inflammation, plaque level, probing depth, gingival recession and improving clinical attachment level over cSRP.
Polydnavirus genome is segmented and dispersed on host wasp chromosome. After replication, the segments form double- stranded circular DNAs and embedded in viral coat proteins. These viral particles are delivered into a parasitized host along with parasitoid eggs. A serine-rich protein (SRP) is predicted in a polydnavirus, Cotesia plutellae bracovirus (CpBV), genome in its segment no. 33 (CpBV-S33), creating CpBV-SRP1. This study explored its expression and physiological function in the diamondback moth, Plutella xylostella, larvae parasitized by C. plutellae. CpBV-SRP1 encodes 122 amino acids with 26 serines and several predicted phosphorylation sites. It is persistently expressed in all tested tissues of parasitized P. xylostella including hemocyte, fat body, and gut. Its physiological function was analyzed by injecting CpBV-S33 and inducing its expression in nonparasitized P. xylostella by a technique called SERI (segment expression and RNA interference). The expression of CpBV-SRP1 significantly impaired the spreading behavior and total cell count of hemocytes of treated larvae. Subsequent RNA interference of CpBV-SRP1 rescued the immunosuppressive response. This study reports the persistent expression of CpBV-SRP1 in a parasitized host and its parasitic role in suppressing the host immune response by altering hemocyte behavior and survival.
