  • In the present study, we have evaluated cytotoxic effects of Taraxaci Herba extract on human skin melanoma cells. The light microscopic study showed morphological changes AG-NOR (argyrophylic nucleolar organizer region) by silver chloride stain, and glycoprotein by PAS reaction of the treated cells. Disruptions in cell organelles were determined by SRB assay.

  • The phytochemical study of Cacalia koraiensis Nakai (Compositae) led to the isolation of eight compounds, $\beta$-sitosterol (1), stigmasterol (2), phytol (3), $\beta$-sitosterol-3-Ο-glucopyranoside (4), adenostylol (5), campesterol (6), germacradien-4$\alpha$-ol (7), and quercetin-3-rhamnoside (8). Their structures were established by chemical and spectroscopic methods. The cytotoxicity of the isolated compounds was evaluated by SRB assay against five cultured human tumor cell lines.

  • We have focused on various biological activities of acharan sulfate (AS) isolated from the giant African snail Achatina fulica. In a previous study, AS showed antiangiogenic and immunomodulating activity. We also investigated antitumor activity of AS. In vitro AS had no cytotoxicity within 0 to 200 ug/ml in tumor cells such as Lewis lung carcinoma(LLC) , KM1214 (human colon cancer cell) and Caki-1 (human kidney cancer cell) by both MTT and SRB assay. In vivo AS was used to treat C57BL/6 mice bearing LLC by subscutaneous injection. (omitted)

  • This study was carried out to develop antitumor effect of the n-butanol soluble fraction of Perilla frutescens on (KB cells) human oral epitheloid carcinoma cells. The cytotoxictty of methanollc extract of Perilla frutescens on KB cells was evaluated by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) assay. The antitumor activity of various fractions obtained from n-butanol soluble fraction of Perilla frutescens was evaluated in human oral epithelold carcinoma cells. The antitumor acavity of the n-butanol soluble fraction on human oral epitheloid carcinoma cells was evaluated by MTT assay of colorimetric method. The light microscopic study was carried out to observe morphological changes of cultured human oral epitheloid carcinoma cells. These results were obtained as follows; 1. The fractions 1,2 and 3 of the n-butanol soluble fraction of Perilla frutescens were shown significant antitumor activities. 2. The number of human oral epitheloid carcinoma cells were decreased and tend to form cell cluster by treatment with fractions 1,2,3 and 4 of the n-butanol soluble fraction of Perilla frutescens. 3. The fraction 1 of the n-butanol soluble fraction of Perllla frutescens showed the highest antitumor activity on Perilla frutescens. It has been selected as a lead fraction for further examinations.

오매 에탄올 추출물의 항산화 활성 및 항암 활성 연구 (Study on Antioxidant and Cytotoxic Activities in Ethanol Extract from Prunus mume)

  • Prunus mume is well known contain many functional materials that play beneficial roles in the human body. Studies have found that many organic acids and polyphenol compounds exist in Prunus mume. In this study, content of total polyphenols and flavonoids, antioxidative activity and cytotoxicity of ethanol extract from Prunus mume were measured Contents of total polyphenolic and total flavonoid compounds in ethanol extract from Prunus mume were 16.92 mg/g and 59.95 mg/g, respectively. The $IC_{50}$ of ethanol extract from Prunus mume were $237.72 {\mu}g$/assay and $239.58 {\mu}g$/assay by DPPH and ABTS radical cation scavenging test, respectively. Additionally, ferric ion reducing antioxidant power (FRAP) value of ethanol extract from Prunus mume was 0.94 mM ($FeSO_4$ eq.) by $800 {\mu}g$/assay. Cytotoxicity of Prunus mume against five kinds of cancer cell lines increased as the extract concentration increased Especially, cytotoxicity of the ethanol extract against A-549 (lung cancer line) was higher than that against any other cancer cell line by both MTT and SRB assay. These results show that ethanol extract of Prunus mume has considerably high antioxidative and cytotoxic activities.

소목의 열수 및 에탄올 추출물의 항산화 및 항암활성 (Antioxidant and Cytotoxic Activities of Hot Water and Ethanol Extracts From Caesalpinia sappan)

  • 소목은 인도, 말레이시아, 중국 남부 등의 열대 아시아에 분포하는 낙엽 관목의 콩과 식물로써, 한방에서는 항통증, 항염증의 용도로 사용된다. 본 연구에서는 국내산 소목의 열수 및 에탄올 추출물의 총 폴레페놀, 총 플라보노이드의 함량 분석과 함께 항산화 및 항암활성 분석을 통해 기능성 소재로의 이용가능성을 살펴보고자 하였다. 소목 열수 추출물은 건소목 100 g에 증류수 2L를 가하여 $65^{\circ}C$의 수조에서 4시간 동안 추출하고 에탄올 추출물은 건소목 100 g에 70% 에탄올 2L를 가하여 $25^{\circ}C$, 108 rpm으로 24시간 동안 진탕 추출하였다. 추출물은 모두 여과, 농축한 후 동결 건조하여 시료로 사용되었다. 총 폴리페놀 함량은 열수 추출물 및 에탄올 추출물에서 각각 22.6 mg GAE/g과 17.6 mg GAE/g으로 나타났으며 총 플라보노이드 함량은 14.5 mg QE/g, 13.2 mg QE/g으로 에탄올 추출물에서 더 높은 함량을 보였다(p<0.05). DPPH 실험결과, 10-800 ug/mL의 농도에서 에탄올 추출물은 36.5-82.8%, 열수 추출물은 26.7-75.5%의 radical 소거능을 각각 나타내었다. DPPH실험과 동일한 농도에서 ABTS radical 소거능 측정 결과, 에탄올 추출물은 5.5-94.9%, 열수 추출물은 5.8-88.8%의 소거능을 각각 나타내었는데, 결과적으로 소목 에탄올 추출물이 열수 추출물에 비해 우수한 항산화력을 가지고 있음을 보여주었다(p<0.05). 소목의 항암 활성 측정을 위해 인체 암세포주인 A-549, HeLa, Hep3B cell에 대해 MTT assay와 SRB assay를 실시한 결과, 소목 에탄올 추출물의 농도가 증가함에 따라 각 세포주에 대한 세포 독성이 증가하는 경향을 보였다. MTT assay와 SRB assay의 모든 실험농도에서 폐암 세포인 A-549에 대해 가장 높은 세포 독성을 나타내었으며, 특히 MTT assay에서는 300 ug/mL의 소목 에탄올 추출물이 A-549 cell에 대해 90% 이상의 높은 암세포 억제효과를 보여주었다. 따라서, 우수한 항산화 활성과 항암 활성을 나타낸 소목 추출물은 향후 다양한 기능성 식품 및 의약 소재로서의 활용이 가능할 것으로 보인다.

아가리쿠스 버섯 균사체 추출물 및 분획물의 항돌연변이원성 및 세포 독성 효과 (The Antimutagenic Effects and Cytotoxic Activities of Agaricus blazei Murill Mycelium Extracts and Fractions)

  • This study was performed to observe the antioxidative effects, antimutagenic capacity, and cytotoxic activity of the 70% ethanol extract, and fractions, of Agaricus blazei Murill mycelium, using DPPH free radical scavenging ability, the Ames test, and SRB assay, respectively. Among the fractions, ethyl acetate showed the most effective antioxidative capacity according to the $RC_{50}$(73.6 $\mu$g/mL) of the scavenging effect on the DPPH radical. The inhibition rate of both the aqueous fraction and 70% ethanol extract(200 $\mu$g/plate) toward the Salmonella typhimurium TA100 strain was 94.6%, and it was 89.4% against the mutagenesis induced by MNNG(0.4 $\mu$g/plate). In addition, an identical concentration of the 70% ethanol extract in the TA98 strain, and the ethyl acetate fraction in the TA100 strain, showed inhibition rates of 80.3% and 76.9%, respectively, the highest activities against the mutagenesis induced by 4NQO(0.15 $\mu$g/plate). The cytotoxic effects of the 70% ethanol extract and its fractions increased with increasing sample concentration against human cervical adenocarcinoma(HeLa), human hepatocellular carcinoma(Hep3B), human breast adenocarcinoma(MCF-7), human stomach adenocarcinoma(AGS), and human lung carcinoma (A549). A 1 mg/mL concentration of the ethyl acetate fraction showed cytotoxicities of 77%, 83.8%, 82.1%, 83.1%, and 92.6% against HeLa, Hep3B, MCF-7, AGS and A549, respectively.

천화분의 카드뮴에 대한 독성억제효과(III) (The Inhibitory Effects of Trichosanthes kirilowii Root against Cadmium Induced Cytotoxicity (III))

  • This study was conducted to investigate the antitoxic components in the water extract of the roots of Trichosanthes kirilowii (Cucurbitaceae). The results were as follows: Generally, detoxication effects by the water extract of T. kirilowii increased in proportion to the concentrations. Experimental animals were treated with cadmium and T. kirilowii water extract by oral administration. When 40 mg/kg dosage of T. kirilowii extract was administrated it showed the highest antitoxic effects in metallothionein induction. After the water extract treatment, body weights did not increase in proportion to the extract concentrations. These results suggest that T. kirilowii extract increased metallothionein concentration and decreased the toxicity of cadmium in rats. In vitro the antitoxic activity of water extract of T. kirilowii on NIH 3T3 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treated cells. These results were obtained as follows; The concentration of $10^{-2}\;mg/ml$ of T. kirilowii extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were increased and tend to regenerate. These results suggest that T. kirilowii extract retains a potential antitoxic activity.

Detoxication effects By H. cordata $T_{HUNB}$ extract increased in proportion to the extract concentrations. When 40 mg/kg dosage of H. cordata $T_{HUNB}$ extract was administered, it showed the highest antitoxic effects in metallothionein induction.
