Effects of Prunellae Herba on the H2O2-Treated LLC-PK Cell's Redox Status and NF-κB Signaling (H2O2 처리된 LLC-PK₁세포에서 Redox Status 및 NF-κB Signaling에 대한 하고초(夏枯草)의 효과)

  • This study was to investigate the anti-inflammatory effects of Prunellae Herba(PH). The generation of superoxide anion radical (․O2-), nitric oxide (NO), peroxynitrite (ONOO-) and Prostaglandin E₂(PGE2) were measured in the H2O2-Treated renal epithelial cells(LLC-PK1 cell) of mouse. And the effects of Prunellae Spica on the expression of NF-κB (p50, p65), IKK-α, phospho-IκB-α and inflammation-related proteins, COX-2, iNOS, IL-1β and VCAM-1, were examined by western blot. The fluorescent probes, 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein (DAF-2) and dihyldrorhodamine 123 (DHR 123) were used to estimate the scavenging effect of Prunellae Spica on ․O2-, NO, ONOO-. Western blot was conducted to assess the protein expression levels of NF-κB (p50, p65), IKK-α, phospho-IκB-α, inflammation-related proteins, COX-2, iNOS, IL-1β, VCAM-1. PH inhibited H2O2-treated cell death dose-dependently. It reduced the generation of ·O2-, NO, ONOO- and PGE₂ in the H2O2-treated renal epitheial cells(LLC-PK1 cell) of mouse in vitro. PH reduced the expression of NF-κB, IKK-α, phospho-IκB-α, COX-2, iNOS, IL-1β and VCAM-1 genes through means of decreasing activation of NF-κB signaling as well. According to these results, PH has an antioxidative activity and anti-inflammatory effect by regulating the NF-κB pathway. This suggest that PH is expected to be used to regulating inflammatory process and treating inflammation-related disease.

Experimental study of Gagam-Cheongsang BangPungTang on the anti-inflammatory effects (가감청상방풍탕(加減淸上防風湯)의 항염증 효과에 대한 실험적 연구)

  • Background and Object : This study was carried out to investigate the effects of GCSBPT (Gagam-Cheongsang BangPungTang) on the in vitro and in vivo anti-inflammatory reactions. Methods : Vascular permeability and Cyclooxygenase inhibition assay are examined in vitro and nitric oxide inhibition assay, radical scavenging activity test, $TNF-{\alpha}$, COX-2 inhibition test are examined in vivo. Results : GCSBPT showed inhibitory effects on vascular permeability and leukocyte migration in animal test. In cyclooxygenase 2 inhibition assay, an ethanol extract of GCSBPT inhibited prostaglandin E2 generation at a concentration of $10{\mu}g/ml$. Among the herbal ingredients of GCSBPT, ethanol extracts of Nepetae Spica exhibited potent inhibitory activities. Ethanol extract of GCSBPT inhibited the release of nitric oxide and the gene expression of inducible nitric oxide synthase in RAW 246.7 cells stimulated by lipopolysaccharide. Ethanol extract of GCSBPT exhibited radical scavenging activity of 54% at $100{\mu}g/ml$. Among the herbal ingredients of GCSBPT. Conclusions : According to the above results, I expected that GCSBPT was a potent anti-inflammatory prescription.

  • Twenty-nine cases of unicameral bone cyst developed in long bone of children have been treated and followed up for 4.5 years in average form Department of Orthopaedic Surgery, Guro Hospital, Korea University, College of Medicine since September, 1983, Treatment for those lesions differed to form largely two groups, one of which consised of insillation of Methyl-prednisolone for non-weight bearing bones(12 humeri) and the other of curettage and autogenous bone graft for weight-bearing bones(7 femur). Methl-prednisolone group required repetition of instillation for 3.5 time in average spanning over 4 years until cloudy obliteration occurs. Curettage and bone graft had healed in 3 year 6 months' time in average. There were neither recurrence nor pathologic fractures of the lesions with the latter group. Immobilization period was virtually non with Methyl-prednisolone group and 4-6 weeks by hip spica with curettage and bone graft group. As conclusions, It seems confirmed that treatment strategy of unicameral bone cyst consisted of Methyl-prednisolone instillation for humerus lesions and early curettage and bone graft for femur lesions is applicable as guideline having solid ground in clinical experiences.

Acute Oral Toxicity Study on BDR-29 in Rats (랫트에서 한약재 복합물 BDR-29의 단회 경구투여 독성에 관한 연구)

  • Acute toxicity of BDR-29, a combined preparation of Cassia Semen, Prunellae Spica, Tribuli Fructus, and Uncariae Rhamulus et Uncus was examined using male and female Sprague-Dawley rats. Rats were treated with the BDR-29 intra-gastrically at 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. At the doses used no mortality or abnormal clinical signs in animals were shown during at the observation period. Also there was no difference in net body weight gain, gross pathological findings, and urine analysis at the terminal sacrifice among the groups rats treated with different doses of the test substance. The results suggested that acute oral toxicity of BDR-29 in rats is very low at the conditions employed in this study.

Study for Thirteen Weeks Subacute Toxicity of BDR-29 in Rats (BDR-29의 랫트에 대한 13주 반복투여 독성에 관한 연구)

  • The subcronic toxicity of BDR-29, a herbal preparation of Cassiae Semen, Prunellae Spica, Tribuli Fructus, and Uncariae Rhamulus et Uncus, was examined in male and female Sprague-Dawley rats. Rats were treated with the test substance at a dose 5 mg/kg, 50 mg/kg and 500 mg/kg intragastrically for 13 weeks. No death and abnormal clinical signs were observed throughout the administration period. There were not significantly different from control group in net body weight gain, food and water consumption, organ weight, gross pathological findings, and urine analysis among the groups rats treated with different doses of the BDR-29. Hematological findings and biochemical examination revealed no evidence of specific toxicity related to BDR-29. From these results, no observation effect level (NOEL) of BDR-29 is 500 mg/kg/day under the condition employed in this study.

  • Objective: The purpose of this study is to find out Herbal medicine effective to the hypertensive patients. Methods: The present study was done over 42 chinese papers about hypertension from 2000 to 2006. We calculated frequency of the herbs used to treatment for hypertension. Result & Conclusion: 1. In 42 herbal medicines, Gamibanhabaekchulcheonmatang and Gamitaeksatang showed high frequency. 2. In 125 herbs, RADIX SALVIAE MILTIORRHIZAE(丹蔘) showed highest frequency. And RAMULUS ETUNCUS UNCARIAE(釣鉤藤), RADIX ACHYRANTHIS BIDENTATAE(牛膝), RADIX PAEONIAE ALBA(白芍藥), RHIZOMA ALISMATIS(澤瀉), RHIZOMA CNIDII(川芎), PORIA(白茯苓), RHIZOMA GASTRODIAE(天麻), SPICA PRUNELLAE(夏枯草), CONCHA HALIOTIDIS(石決明) showed high frequency.

Effects of Herbal Extracts Used in Oriental Medicines on Heme Oxygenase-1 Expression

  • Effects of twenty-three aqueous herbal extracts used in oriental medicines on heme oxygenase (HO)-1 expression were estimated in a mouse hippocampal cell line, HT22. HO-1 is one of the cytoprotective enzymes activated various stimuli, and Western blot analysis was used for evaluated HO-1 expression. Six aqueous extracts such as Rhei Rhizoma, Paeoniae Radix, Uncariae Ramulus et Uncus, Theae Folium, Prunellae Spica, and Coptidis Rhizoma significantly increased HO-1 expression in HT22 cells at the concentration of 300 ${\mu}$g/ml. In Addition, four aqueous extracts including Eucommiae Cortex, Moutan Cortex Radicis, Ginseng Radix Rubra, and Scutellariae Radix moderately increased HO-1 expression. These results would be usefulfor the isolation and identification of their neuroprotective principles.

LABORATORY EXPERIMENTS OF OFF-AXIS MIRROR OPTICS OF ALUMINUM FOR SPACE INFRARED MISSIONS

  • We report our research on aluminum mirror optics for future infrared astronomical satellites. For space infrared missions, cooling the whole instrument is crucial to suppress the infrared background and detector noise. In this aspect, aluminum is appropriate for cryogenic optics, because the same material can be used for the whole structure of the instrument including optical components thanks to its excellent machinability, which helps to mitigate optical misalignment at low temperatures. We have fabricated aluminum mirrors with ultra-precision machining and measured the wave front errors (WFEs) of the mirrors with a Fizeau interferometer. Based on the power spectral densities of the WFEs, we confirmed that the surface accuracy of all the mirrors satisfied the requirements for the SPICA Coronagraph Instrument. We then integrated the mirrors into an optical system, and examined the image quality of the system with an optical laser. As a result, the total WFE is estimated to be 33 nm (rms) from the Strehl ratio. This is consistent with the WFEs estimated from the measurement of the individual mirrors.

Protective effects of Jinnoe-san, a novel herbal formula in experimental in vitro models of Parkinson's disease (파킨슨병의 세포모델에서 진뇌산(鎭腦散)의 보호효과)

  • Objectives : Jinnoe-san (JNS) is a novel herbal formula consisting of five oriental medicinal herbs including Polygalae Radix, Prunellae Spica, Perillae Herba, Betulae Cortex, and Lonicerae Flos. In this study, we investigated the effects and molecular mechanism of JNS on Parkinson's disease in vitro model. Methods : The effects of JNS on 1-methyl-4-phenylpyridinium ($MPP^+$)-induced cell death in SH-SY5Y cells were evaluated with a cell viability assay, flow cytometry, and western blots analysis. The effects of JNS on lipopolysaccharide (LPS)-stimulated BV2 microglia were determined with a nitric oxide (NO) assay, enzyme linked immunosorbent assays, and western blots analysis. Result : $MPP^+$-induced cell death in SH-SY5Y cells was significantly reduced by JNS pre-treatment in a dose-dependent manner. JNS inhibited the production of reactive oxygen species, mitochondria dysfunction, and apoptosis induced by $MPP^+$ in SH-SY5Y cells. Furthermore, JNS significantly activated Akt and ERK in SH-SY5Y cells and the ability of JNS to prevent mitochondria dysfunction by $MPP^+$ was antagonized by pre-treatment of LY294002 and PD98059, an Akt and ERK inhibitor, respectively. In addition, JNS inhibited LPS-induced NO and $PGE_2$ production as well as iNOS expression and secretion of TNF-${\alpha}$, pro-inflammatory cytokines without affecting the cell viability. JNS also suppressed LPS-induced ERK activation. Conclusions : These results demonstrate that JNS has a protective effect on the dopaminergic neurons against $MPP^+$-induced neurotoxicity and anti-inflammatory effect on the LPS-stimulated microglia. These findings provide evidences for JNS to be considered as a new prescription for treating Parkinson's disease.

Effects and molecular mechanisms of Noemyeong-san, a novel herbal prescription for treating Alzheimer's disease on microglia (미세아교세포에서 알츠하이머형 치매 치료 처방인 뇌명산(腦明散)의 효능 및 기전연구)

  • Objectives : Noemyeong-san (NMS) is a novel herbal prescription composed of five oriental medicinal herbs including Prunellae Spica, Betulae Cortex, Foeniculi Fructus, Asiasari Radix, and Clematidis Radix for treating Alzheimer's disease. In the present study, we investigated the effects and molecular mechanisms of NMS on BV2 microglia to evaluate the potential action of this formula for preventing or treating neurodegenerative disease such as Alzheimer's disease. Methods : To determine the cytotoxicity of NMS on BV2 microglia, the MTT assay was performed. The effects of NMS on lipopolysaccharide (LPS)-stimulated BV2 microglia were determined with a nitric oxide (NO) assay and western blots for inflammatory mediator-related proteins, mitogen activated protein kinases (MAPKs), nuclear factor kappa B (NF-${\kappa}B$) pathway-related proteins, and heme oxygenase-1 (HO-1). Result : NMS inhibited induction of iNOS and COX-2 as well as NO production without affecting the cell viability in LPS-stimulated BV2 microglia. NMS also suppressed activation of ERK and p38 MAPK among main kinases of MAPKs as well as NF-${\kappa}B$ by LPS stimulation. Furthermore, NMS dose-dependently induced the expression of HO-1 and the inhibitory effect of NMS on the production of NO were blocked by pretreatment with an HO-1 inhibitor, Snpp. Conclusions : These results demonstrate that NMS has potent anti-neuroinflammatory effect on the LPS-stimulated microglia. These findings provide evidences for NMS to be considered as a new prescription for preventing or treating neurodegenerative disease such as Alzheimer's disease.
