Hexon and fiber gene changes in an attenuated fowl adenovirus isolate from Malaysia in embryonated chicken eggs and its infectivity in chickens

  • Fowl adenovirus (FAdV) is distributed worldwide and causes economic losses in the poultry industry. The objectives of this study were to determine the hexon and fiber gene changes in an attenuated FAdV isolate from Malaysia in specific pathogen-free chicken embryonated eggs (SPF CEE) and its infectivity in commercial broiler chickens. SPF CEE were inoculated with 0.1 mL FAdV inoculum via the chorioallantoic membrane (CAM) for 20 consecutive passages. The isolate at passage 20 (E20), with a virus titer of $10^{8.7}TCID_{50}/mL$ ($TCID_{50}$, 50% tissue culture infective dose), was inoculated (0.5 mL) into one-day-old commercial broiler chicks either via oral or intraperitoneal routes. The study demonstrated that 100% embryonic mortality was recorded from E2 to E20 with a delayed pattern at E17 onwards. The lesions were confined to the liver and CAM. Substitutions of amino acids in the L1 loop of hexon at positions 49 and 66, and in the knob of fiber at positions 318 and 322 were recorded in the E20 isolate. The isolate belongs to serotype 8b and is non-pathogenic to broiler chickens, but it is able to induce a FAdV antibody titer. It appears that molecular changes in the L1 loop of hexon and the knob of fiber are markers for FAdV infectivity.

Reticuloendotheliosis virus의 닭에 대한 면역억제효과와 병원성 (Immunosuppressive effects and pathogenicity of a Korean isolate of reticuloendotheliosis virus in chickens)

  • Immunosuppressive effects of reticuloendotheliosis virus (REV) infection in chickens were investigated. Primary antibody responses to Newcastle disease virus (strain B1) and sheep red blood cells were significantly low in chickens inoculated with the local isolate 89-74 of REV compared to those of uninfected chickens. In chickens infected with REV strain T or 89-74, blastogenesis of spleen cells and peripheral blood lymphocytes (PBL) to concanavalin A (Con A) was severely suppressed. When specific pathogen free (SPF) chickens were inoculated with the isolate, the suppressive effect was observed up to 7 weeks of age while, in the contact infected chickens, the suppression was absent. Similar suppressive effects were observed in chickens inoculated with REV strain T at 2, 3 and 4 weeks of age. When spleen cells or PBL from uninfected chickens were co-cultured with spleen cells or PBL from chickens infected with REV at 1 day-old or 2 week-old, the blastogenesis of the normal cells was suppressed. The suppressive effect of PBL from REV-infected chickens on normal lymphocytes was abrogated by the treatment with trypsin. However the suppressive activity of the REV-infected PBL was not influenced at removing machrophage from the cell suspension by incubation in plastic petri dishes. In addition to the immunosuppression, chickens infected with the REV isolate showed abnormal feather development (nakanuke), anemia, paralysis and retarded growth. Three out of 11 chickens inoculated with the isolate at day-old died between 6 and 9 weeks of age by bacterial infections.

  • Avian reovirus (ARV) and fowl adenovirus (FAdV) were evaluated for pathogenicity in specific pathogen free (SPF) chickens. ARV was isolated from the broilers with history of malabsorption syndrome (MAS). FAdV was isolated from the layer breeders with inclusion body hepatitis and hydropericardium syndrome. Total 6 inoculated groups including 1 un-inoculated group were organized and inoculated with the ARV and/or FAdV by oral route. The minimal pathological lesions and lower viral gene detection rates were present in the ARV inoculated groups compared to those of FAdV or ARV/FAdV inoculated groups. Common gross lesions in the ARV inoculated group were distended intestine with foamy contents and in the FAdV group there were foamy cecal contents and hydropericardium among the evaluation methods such as gross and histological lesion, viral gene detection, body weight and serum chemistry, histopathological lesion score was reliable especially in the liver lesions such as hepatic necrosis and lymphocytic infiltration. However, we did not success to evaluate the synergetic effect of mixed infection of ARV and FAdV in this study. Therefore, we need further study to reproduce malabsorption syndrome of ARV infection using different viral agent such as rotavirus and using different dose of virus.

닭와포자충 재감염닭의 혈청1gG 항체가 추이 (Chronologic chrnge of serum IgG antibody response in chickens reinfected with Cryptosporidium baileyi)

  • 닭에 있어서 닭와포자충의 재감염에 대한 방어기전을 이해하기 위하여 장기간에 걸쳐 혈청IgG항체가를 경시적으로 조사하였다. 2일령의 SPF 병아리(Dekalb-Warren, Sex-Sal-Link)에 $5{\;}{\times}{\;}10^5$개 그리고 IgG가가 정상으로 복귀한 다음 10일인 340일령에 $1{\;}{\times}{\;}10^7$개의 닭와포자충의 오오시스트를 경구투여하고 나서 채혈하여 간점형광항체법을 이용하여 혈청IgG 항체가를 측정하였다. IgG 가는 감염후 3일부터 38일까지는 대조군과 차이가 없었으나 45일부터 1:21.33으로 상승하기 시작하여 120일에 1:512로 최고치에 이른다음 점점 떨어져 330일에 1:4로 정상치에 이르렀다. 도전 감염후 7, 14, 28일에 각각 1:16, 1:512, 1:1.024로서 점점 높아져 정점에 이른다음 급격히 떨어져 63일에 정상으로 복귀하였다. 한편, 대조군의 항체가는 모두 1:2~1:4의 범위에 있었으며 이 원충의 도전감염전 및 후에 있어서 분변으로부터 집충시켜 제작한 Kinyoun 항산염색표본에서도 오오시스트를 전혀 검출할 수 없었다. 분변내의 오오시스트의 소실 시기와 최초 감염에 있어서 IgG가의 피이크는 일치하지 않았다.

  • 닭에 있어서 닭와포자충 감염이 면역억제에 미치는 영항을 규명하기 위한 일환으로 150마리의 2 일령 SPF 병아리에 $5{\;}{\times}{\;}10^5$의 닭와포자충 오오시스트를 경구 투여하였다 경구투여 후 3일부터 95일까지 4일 간격으로 5마리씩의 병아리 정맥 내에 $1{\;}{\times}{\;}10^7$의 SRBC를 주입한 다음 각각 5일 후에 혈액을 채혈하여 SRBC의 응집소가를 측정하였다. 감염군에 있어서 분변 속의 오오시스트 배설은 정상적인 양상이었으며. 응집소가는 시일이 경과함에 따라 감염, 비감염 양군 모두 점점 높아져 52일에 최고에 이른 다음 점점 낮아졌다 한편. 실험기간을 통하여 응집소가는 오오시스트를 투여하지 않은 비감염군에 비하여 오오시스트를 투여한 감염군이 의의있게 낮았다. (P<0.05) 결과로 미루어 보아 닭와포자충 감염닭은 비감염닭베 비하여 항체반응이 저하되는 경향이 있다.

자연감염된 닭으로부터 chicken anemia agent (virus)의 분리 (Isolation of chicken anemia agent (virus) from naturally infected chickens)

  • Attempts to isolate chicken anemia agent (CAA) were made by inoculating tissue homogenates into MDCC-MSBl or LSCC-1104B1 cell lines and passaging the cells serially. CAA was isolated from the liver and thymus of 11 weeks old layer chickens and from the liver of 10 weeks old broiler breeder chickens. The layer flock experienced approximately 45% mortality during 9 to 14 week of age from gangrenous dermatitis and lymphoid organs of affected chickens were severely atrophied. The broiler breeder flock experienced approximately 7% mortality during 7 to 9 weeks of age and affected birds showed lesions of colibacillosis, staphylococcal arthritis, and coccidiosis together with atrophied lymphoid organs. The isolated viruses were identified as CAA by the indirect fluorescent antibody test and virus neutralization test using CAA immune sera including one to Gifu-1 strain of CAA. The CAA isolate 89-69, when inoculated into susceptible 1 day old SPF chicks, induced anemia 14 to 16 days after inoculation. It did not induce any cytopathic effects in chicken embryo liver and chicken embryo fibroblast cell cultures. Infectivity of the isolate was not affected by the treatment of chloroform or heat ($70^{\circ}C$ for 15 minutes).

  • Infectious coryza is one of the important acute respiratory diseases causing a significant egg drop and retarded growth in chicken. An attempt for the isolation of etiologic agent was made by utilizing SPF sentinel birds housed in commercial farms and the results obtained are as follows. Fifteen isolates of Haemophilus gallinarum were tested for their biological and serological characteristics with reference strains, 221 and Modesto and subsequently classified into two serotypes. Of them, isolates immunolocally identical to the standard strains were also selected as vaccine strains for future studies.

  • 마사츄셋형 전염성 기관지염 바이러스(IBV)를 SPF 발육란의 뇨막강내에서 증식시켜 Sucrose 밀도구배 초원심분리에 의해 정제한 다음 BALB/c 마우스에 면역시켰다. 면역 마우스에서 채취한 비장세포와 마우스 골수암세포와 여러 차례 융합시험을 실시하였다. 많은 융합세포 중에서 IBV에 특이적으로 작용하는 단클론항체(monoclonal antibody : MCA)를 산생하는 hybridoma클론은 2주밖에 얻지 못했다. 2주의 MCA는 모두 IgG형이었고 IBV중화능이나 혈구응집 억제능이 인정되지 않았다. 간접형광항체법으로 작성된 MCA를 이용하여 인공접종한 닭의 기관도말표본에서 10일간의 시험기간중 계속 IBV를 검출할 수 있었다.

  • 닭에 있어서 닭와포자충 갉염이 면역반응에 미치는 영향을 규명하기 위한 기초적 연구리 일환으로 파브리시우스낭의 병리조직학적 소견을 경시적으로 조사하고자 150마리의 2일령 SPF 병아리 (Drkalb-Warren, Sex-Sal-Link)에 닭와포자충의 오오시스트 $5{\;}{\times}{\;}10^5$를 한 번에 경구투여하였타. 분변 속의 오오시스트 배설양상은 정상적이었으며. 파브리시우스낭 지수는 전 실험기간에 걸쳐 거의 변동이 없얹다 많은 수의 원충체가 접종 후 4-16일에 이 낭상피의 미세융또 가장자리에서 관찰되었으며 많은 수의 비만세포가 출현한 다음 원충체가 급격하게 소실하염다. 원충체의 분포상황과 상피 및 인접점막 고유층의 위호산구 침윤은 일치하였다. 이 병소는 상피. 인접 점막고유층의 위호산구 침율과 점만상피 증식을 동반한 미만성 만성 표재성 화농성 파브리시우스낭영의 병리조직학적 소견이었다 이러한 파브리시우스낭염은 면역억제를 유발할 건으로 생각된다.

  • Mean death time of inoculated embryonated egg is one of the methods to determine the virulence of the Newcastle disease viruses (NDV). Evaluation of tissue tropism of NDV in the host has been proposed as an another way to determine the pathogenicity of NDV based on the principal site of viral replication. To evaluate the tissue tropism among NDV, an immunohistochemistry(IHC) technique using monoclonal antibody was applied in one-day-old SPF chickens inoculated with different ND vaccine strains such as Ulster 2C, VG/GA and B1 viruses by eye drop instillation. The tissues used for this comparison were trachea, intestine, Harderian gland and cecal tonsil, which were collected at 0.5, 1, 2, 3, 5, 8, 10, 14 days post inoculation. Among test groups, chickens inoculated with B1 viurs, which is known to replicate in the respiratory system, have IHC positive staining mainly in the trachea and those inoculated with Ulster 2C have IHC positive staining mainly in the intestine. However, chickens inoculated with VG/GA strain have IHC positive staining in both the trachea and intestine. Therefore, a differences in tissue tropism among ND vaccine strains has been proved by the IHC technique. Based on this results, the IHC staining technique could be used to classify the NDV or to study the pathogenesis of NDV in chickens.
