  • The growth characteristics of Rhodotorula sp. Y-55 were examined on minimal medium containing ethanol, acetic acid or acetadehyde as a sole carbon source by batch culture. The increased concentration of substrate reduced overall growth yield and prolonged lag time. The specific growth rate of the yeast was changed, depending upon the initial concentrations of ethanol and acetaldehyde during the exponential period, but was constant on acetic acid without regard to the initial substrate concentrations, giving a value of 0.l07h-1. The highest ${\mu}$ value was obtained on ethanol and acetadehyde substrates and the respective values were 0.270 at 20g/L and 0.041h-1 at 0.2g/L. The maximum overall growth yields were appeared to be 32.6% for ethanol of 10g/L, 25.6% for acetic acid of 20g/L, and 45% for acetaldehyde of 0.2g/L. The respective cellular contents of crude protein and nucleic acids were determined to be 41.5 and 4.9wt% on ethanol and 40.2 and 4.7wt% at the concentration revealing maximal growth yield.

Isolation of Strains that Produce Ethanol Efficiently from Cellulosic Materials (섬유질 가수분해물로부터 효율적인 Ethanol 생산균주의 분리)

  • Three strains able to efficiently produce ethanol from cellulosic hydrolysates were isolated from soil samples by enrichment culture in liquid saccharified wheat bran medium. The profiles of physiological and biochemical properties of two yeasts KM-09 and KM-402 and a bacterium Hg-225 were almost identical from those of Candida sp. and Klebsiella sp., respectively. Strains KM-09 and HG-225 used xylose and cellobiose as fermentable sugars, and HG-225 had a wide range of sugar utilization for ethanol fermentation. The optimal pH and temperature for growth of KM-09, KM-402 and HG-225 were 5.8, 5.6 and 6.8 and 32t, $30^{\circ}C$~ and $38^{\circ}C$, respectively. During the ethanol fermentation in saccharified wheat bran by the isolated strains, optimal temperature for ethanol production was more or less higher than those for growth, and addition of 0.2% (w/v) $MgSO_4$, into the medium enhanced ethanol productivity. Of the three strains ethanol content of KM-09 was the highest with about 2.3% (v/v), and ethanol production rate of HG-225 was faster than the others and maximum productivity was after 4 days. KM-09 (1.42% v/v) and HG-225 (1.05%, vlv) produced ethanol from 4% (wIv) xylose but growth rate was slower than on glucose. Otherwise KM-402 showed the highest ethanol productivity on glucose, but no ethanol was detected on xylose and cellobiose.

Microbacterium elymi sp. nov., Isolated from the Rhizospheric Soil of Elymus tsukushiensis, a Plant Native to the Dokdo Islands, Republic of Korea

  • Microbacterium elymi KUDC0405T was isolated from the rhizosphere of Elymus tsukushiensis from the Dokdo Islands. The KUDC0405T strain was Gram-stain-positive, non-spore forming, non-motile, and facultatively anaerobic bacteria. Strain KUDC0405T was a rod-shaped bacterium with size dimensions of 0.3-0.4 × 0.7-0.8 ㎛. Based on 16S rRNA gene sequences, KUDC0405T was most closely related to Microbacterium bovistercoris NEAU-LLET (97.8%) and Microbacterium pseudoresistens CC-5209T (97.6%). The dDDH (digital DNA-DNA hybridization) values between KUDC0405T and M. bovistercoris NEAU-LLET and M. pseudoresistens CC-5209T were below 17.3% and 17.5%, respectively. The ANI (average nucleotide identity) values among strains KUDC0405T, M. bovistercoris NEAU-LLET, and M. pseudoresistens CC-5209T were 86.6% and 80.7%, respectively. The AAI (average amino acid identity) values were 64.66% and 64.97%, respectively, between KUDC0405T and its closest related type strains. The genome contained 3,596 CDCs, three rRNAs, 46 tRNAs, and three non-coding RNAs (ncRNAs). The genomic DNA GC content was 70.4%. The polar lipids included diphosphatydilglycerol, glycolipid, phosphatydilglycerol, and unknown phospholipid, and the major fatty acids were anteiso-C17:0 and iso-C16:0. Strain KUDC0405T contained MK-12 as the major menaquinone. Based on genotypic, phylogenetic, and phenotypic properties, strain KUDC0405T should be considered a novel species within the genus Microbacterium, for which we propose the name M. elymi sp. nov., and the type strain as KUDC0405T (=KCTC 49411T, =CGMCC1.18472T).

  • An investigation has been done to study the evolution of the microstructure, mechanical and electrical properties of AlMgSi alloy destined for the transport of electric energy, in function of the deformation caused by the cold drawing process. We identified that drawing of aluminum wire causes development of a fibrous texture of type <111> and <100>. We notice also that the electrical resistivity and mechanical resistance increases with the increasing of the deformation level. Characterization methods used in this work is: The Electron Back Scattered Diffraction EBSD, X-Ray diffraction, Vickers microhardness, Tensile test, Measuring electrical resistivity, the Scanning Electron Microscope (SEM) and Energy Diffraction Spectrum (EDS).

Purification and Physiochemical Characterization of Melanin Pigment from Klebsiella sp. GSK

  • A bacterium capable of producing melanin pigment in the presence of L-tyrosine was isolated from a crop field soil sample and identified as Klebsiella sp. GSK based on morphological, biochemical, and 16S rDNA sequencing. The polymerization of this pigment occurs outside the cell wall, which has a granular structure as melanin ghosts. Chemical characterization of the pigment particles showed then to be acid resistant, alkali soluble, and insoluble in most of the organic solvents and water. The pigment got bleached when subjected to the action of oxidants as well as reductants. This pigment was precipitated with $FeCl_3$, ammoniacal silver nitrate, and potassium ferricynide. The pigment showed high absorbance in the UV region and decreased absorbance when shifted towards the visible region. The melanin pigment was further charecterized by FT-IR and EPR spectroscopies. A key enzyme, 4-hydroxyphenylacetic acid hydroxylase, that catalyzes the formation of melanin pigment by hydroxylation of L-tyrosine was detected in this bacterium. Inhibition studies with specific inhibitors, kojic acid and KCN, proved that melanin is synthesized by the DOPA-melanin pathway.

Isolation and Characterization of Cellulolytic Anaerobic Fungi from the Guts of the Hanwoo Cattle and the Korean Native Goat (한우 및 산양의 장내 섬유소 분해 혐기 곰팡이의 분리 및 특성 구명)

  • The study was conducted to isolate and identify highly fibrolytic anaerobic fungi from the guts of a Hanwoo steer and a Korean native goat, and then investigate the characterization of cellulolytic activity of an anaerobic fungus. Twenty-one anaerobic fungal colonies were isolated in the study, in which 16 colonies were isolated from the rumen contents of the Hanwoo steer and 5 colonies from the duodenal fluids of the Korean native goat. Four anaerobic fungi were selected based on higher cellulolytic enzyme activities to identify under a optical microscope. NLRI-M003 and -T004 belong to Neocallimastix genus and NLRI-M014 belongs to Piromyces genus based on the morphology of their thallus, sporangia, rhizoid and the number of flagella. NLRI-M001 appeared to be an unknown strain of anaerobic fungi due to its different morphology from existing types of anaerobic fungi, though the morpholgoy is similar to Orpinomyces sp. Supplementation of 2% anaerobic fungal culture(NLRI-M003) in rumen-mixed microorganisms increased in vitro DM degradability of rice straw and filter paper up to 4 and 11%, respectively, compared with non-supplementation(control). CMCase and xylanase activities in in vitro culture were also higher in 2% fungal supplementation than controls in both rice straw and filter paper substrates.

Bionomics of Caloglyphus sp.(Acarina : Acaridae) (가루응애류 Caloglyphus sp.의 생태에 관한 연구)

  • Developmental characteristics and reproductive ability of Caloglyphus sp. were examined under the constant temperature conditions. This mites has five stages ; egg, larva, protonymph, deutonymph and adult. All active instars preyed on larvae of pine gall midge, Thecodiplosis japonensis Uchida et Inouye. The developmental period from the egg to the adult decreased with temperature increase at a range between $15^{\circ}C$ and $30^{\circ}C$. The threshold of temperature and the thermal constant for the development from the egg to the adult were $8.2^{\circ}C$ and 122.0 day-degrees, respectively. At $25^{\circ}C$, survival rate from egg to deutonymph was estimated as 66.4%, and longevity of the adult was 12.3 days for male and 10.0 days for female. After a preoviposition period of 1 and 2 days female laid an average of 360.6 eggs each, almost all during the first half of their life time. The value of the net reproduction(Ro), the mean length of a generation(T) and the intrinsic rate of natural increase($r_m$) were calculated as 101.1, 9.3 days and 0.494 per female per day, respectively.

Novosphingobium ginsenosidimutans sp. nov., with the Ability to Convert Ginsenoside

  • A Gram-negative, strictly aerobic, non-motile, non-spore-forming, and rod-shaped bacterial strain designated FW-$6^T$ was isolated from a freshwater sample and its taxonomic position was investigated by using a polyphasic approach. Strain FW-$6^T$ grew optimally at $10-42^{\circ}C$ and at pH 7.0 on nutrient and R2A agar. Strain FW-$6^T$ displayed ${\beta}$-glucosidase activity that was responsible for its ability to transform ginsenoside $Rb_1$ (one of the dominant active components of ginseng) to Rd. On the basis of 16S rRNA gene sequence similarity, strain FW-$6^T$ was shown to belong to the family Sphingomonadaceae and was related to Novosphingobium aromaticivorans DSM $12444^T$ (98.1% sequence similarity) and N. subterraneum IFO $16086^T$ (98.0%). The G+C content of the genomic DNA was 64.4%. The major menaquinone was Q-10 and the major fatty acids were summed feature 7 (comprising $C_{18:1}{\omega}9c/{\omega}12t/{\omega}7c$), summed feature 4 (comprising $C_{16:1}{\omega}7c/iso-C_{15:0}2OH$), $C_{16:0}$, and $C_{14:0}$ 2OH. DNA and chemotaxonomic data supported the affiliation of strain FW-$6^T$ to the genus Novosphingobium. Strain FW-$6^T$ could be differentiated genotypically and phenotypically from the recognized species of the genus Novosphingobium. The isolate that has ginsenoside converting ability therefore represents a novel species, for which the name Novosphingobium ginsenosidimutans sp. nov. is proposed, with the type strain FW-$6^T$ (= KACC $16615^T$ = JCM $18202^T$).

  • Compared to their terrestrial and marine counterparts, little is known about the capacity of freshwater-derived actinomycete bacteria to produce novel secondary metabolites. In the current study, we highlight the disparities that exist between cultivation-independent and -dependent analyses of actinomycete communities from four locations in Lake Michigan sediment. Furthermore, through phylogenetic analysis of strains isolated from these locations, we identified a Streptomyces sp., strain B025, as being distinct from other Streptomyces spp. isolated from sediment. Upon fermentation this strain produced a rare class of eight-membered lactone secondary metabolites, which have been for their antitumor properties. We used spectroscopic and chemical derivitization techniques to characterize octalactin B (1) in addition to its corresponding novel, unnatural degradation product (2).
