  • The production of polyhydroxyalkanoate(PHA) from glucose by batch and fed-batch culture of Pseudomonas sp. HJ was studied. In batch culture using fermentor, 400 rpm of agitalion speed, 2 vvm of aeration rate, 18 hours of inoculum age, and 5% (vlv) of inoculum size were optimal. PHA production was not increased by deficiency of oxygen. In a batch culture, the final call mass was $6.251g/\ell$, and PHA content was 20% of dry cell weight. In a constant feeding fed-batch culture, cell mass increased to $33.24g/\ell$, and PHA content reached 48.9% of dry cell weight. In an intermittent feeding fed-batch culture, cell mass increased to $37.89g/\ell$, and PHA content reached 53.5% of dty cell weight.

  • In this study, a small punch creep(SP-Creep) test using miniaturized specimen$(10{\times}10{\times}0.5mm)$ has been described for the development of the newly semi-destructive creep test method for high temperature structural components such as headers and tubes of boiler turbine casino and rotor and reactor vessel. The SP-Creep testing technique has been applied to 2.25Cr-1Mo(STBA24) steel used widely as boiler tube material and the creep test temperature are varied at $550^{\circ}C{\sim}600^{\circ}C$. The overall deformations of SP-Creep curves are definitely depended with applied load and creep test temperature and show the creep behaviors of three steps like conventional uniaxial creep curves. The steady state creep rate${\delta}_{ss}$ of SP-Creep curve for miniaturized specimen increases with increasing creep temperature, but the exponential value with creep loading is decreased. The activation energy$(Q_{spc})$ during SP-Creep deformation with various test temperatures shows 605.7kJ/mol that is g.eater than 467.4kJ/mol reported in uniaxial creep test. This may be caused by the difference of stress states during creep deformation In two creep test. But from the experimental results, e.g. SP-Creep curve behaviors, the steady state creep rate${\delta}_{ss}$ with creep temperature, and the exponential value(n) with creep loading, it can be summarized that the SP-Creep test may be a useful test method to evaluate the creep properties of the heat resisting material.

  • The purpose of this study was to identify effects of the SP-6 acupressure on dysmenorrhea, skin temperature at th e CV2 acupoint. This study was a pretest-post test study design wit h a nonequivalent control group. Data were collected from May 1 to August 31, 2002. A total of 58 female student s from two universities participated in the study. Among them, 30 female students were assigned as the experimental group an d the other 28 students to the control group. Both groups were pretested before the intervention for three variables, the degree of dysmenorrhea, and skin temperature at the CV2 acupoint. Then, the SP-6 acupressure was provided for 20 minutes for students in the experimental group. The instruments used in this study included the Visual Analogue Scale developed by Johnson(1974), skin thermometer by Simson Eectric CO., Menstrual Attitudes Questionnaire Scale developed by Brooks-Gunn & Ruble(1980), and Stress scale developed by Chun and Kim(1990). The data were analyzed with the SAS program using Chi-square test, t-test, and ANCOVA and Bonferroni method were used to determine significant differences between the two groups. The results of this study are as follows ; 1. There was a statistically significant difference in the intensity of dysmenorrhea after the intervention with the experimental group having a lower intensity than the control group. 2. There was a statistically significant difference in skin temperature at the CV2 acupoint 30min (F=4.87, p=0.03) after the intervention with the experimental group having a higher temperature. In conclusion, the SP-6 acupressure has proved to be an effective nursing intervention to reduce dysmenorrhea. Therefore, it is recommended women suffering from dysmenorrhea use the SP-6 acupressure.

  • The purpose of this study was to investigate the influence of NK1 receptor antagonists on the pulpal blood flow (PBF) when applied iontophoretically through the dentinal cavity of the teeth in order to understand whether iontophoretically applied NK1 receptor antagonists can control the pulpal inflammation. Eleven cats were anesthetized with alpha-chloralose and urethane, and substance P (SP) was administered to the dental pulp through the catheterized lingual artery in doses that caused PBF change without the influence of systemic blood pressure. NK1 receptor antagonists were applied iontophoretically to the prepared dentinal cavity of ipsilateral canine teeth of the drug administration, and PBF was monitored. Data were analyzed statistically with paired t-test. PBF increase after iontophoretic application of the NK1 receptor antagonists followed by the intra-arterial administration of SP was significantly less than PBF increase after iontophoretic application of the 0.9% saline followed by the intra-arterial administration of SP as a control (p < 0.05). Iontophoretic application of the NK1 receptor antagonists (0.2~3.4 mM) following the intra-arterial administration of SP resulted in less increase of PBF than the iontophoretic application of the 0.9% saline following the intra-arterial administration of SP as a control (p < 0.05). Therefore. the results of the present study provide evidences that the iontophoretic application is an effective method to deliver drugs to the dental pulp. and that iontophoretically applied NK1 receptor antagonists block SP-induced vasodilation effectively. The above results show the possibility that the iontophoretical application of NK1 receptor antagonists can control the neurogenic inflammation in the dental pulp.

  • Fusarium wilt of radish (Raphanus sativus L.) is caused by the Fusarium oxysporum f. sp. raphani (FOR) which mainly attacks Raphanus spp. The pathogen is a soil-borne and forms chlamydospores in infected plant residues in soil. Infected pathogen colonizes the vascular tissue, leading to necrosis of the vascular tissue. Growth promoting beneficial organisms such as Pseudomonas fluorescens WCS374 (strain WCS374), P. putida RE10 (strain RE10) and Pseudomonas sp. EN415 (strain EN415) were used for microorganisms-mediated induction of systemic resistance in radish against Fusarium wilt. In this bioassy, the pathogens and bacteria were treated into soil separately or concurrently, and mixed the bacteria with the different level of combination. Significant suppression of the disease by bacterial treatments was generally observed in pot bioassy. The disease incidence of the control recorded 46.5% in the internal observation and 21.1% in the external observation, respectively. The disease incidence of P. putida RE10 recorded 12.2% in the internal observation and 7.8% in the external observation, respectively. However, the disease incidence of P. fluorescens WCS374 which was proved to be highly suppressive to Fusarium wilt indicated 45.6% in the internal observation and 27.8% in the external observation, respectively. The disease incidence of P. putida RE10 mixed with P. fluorescens WCS374 or Pseudomonas sp. EN415 was in the range of 10.0-22.1%. On the other hand, the disease incidence of P. putida RE10 mixed with Pseudomonas sp. EN415 was in the range of 7.8-20.2%. The colonization by FOR was observed in the range of $2.4-5.1{\times}10^3/g$ on the root surface and $0.7-1.3{\times}10^3/g$ in the soil, but the numbers were not statistically different. As compared with $3.8{\times}10^3/g$ root of the control, the colonization of infested ROR indicated $2.9{\times}10^3/g$ root in separate treatments of P. putida RE10, and less than $3.8{\times}10^3/g$ root of the control. Also, the colonization of FOR recorded $5.1{\times}10^3/g$ root in mixed treatments of 3 bacterial strains such as P. putida RE10, P. fluorescens WCS374 and Pseudomonas sp. EN415. The colonization of FOR in soil was less than that of FOR in root part. Based on soil or root part, the colonization of ROR didn't indicate a significant difference. The colonization of introduced 3 fluorescent pseudomonads was observed in the range of $2.3-4.0{\times}10^7/g$ in the root surface and $0.9-1.8{\times}10^7/g$ in soil, but the bacterial densities were significantly different. When growth promoting organisms were introduced into the soil, the population of Pseudomonas sp. in the root part treated with P. putida RE10 was similar in number to the control and recorded the low numerical value as compared with any other treatments. The population density of Pseudomonas sp. in the treatment of P. putida RE10 indicated significant differences in the root part, but didn't show significant differences in soil. The population densities of infested FOR and introduced bacteria on the root were high in contrast to those of soil. P. putida RE10 and Pseudomonas sp. EN415 used in this experiment appeared to induce the resistance of the host against Fusarium wilt.

  • An agar-degrading bacterium, designated as the GNUM08123 strain, was isolated from samples of red algae collected from the Yongil Bay near East Sea, Korea. The isolated GNUM08123 strain was gram-negative, aerobic, motile, and beige-pigmented, with $C_{16:0}$ (25.9%) and summed feature 3 (comprising $C_{16:1}{\omega}7c/iso-C_{15:0}2-OH$, 34.4%) as its major cellular fatty acids. A similarity search based on the 16S rRNA gene sequence revealed that it belonged to class Gammaproteobacteria and shared 97.7% similarity with the type strain Vibrio chagasii $R-3712^T$. The DNA G+C content of strain $GNUM08123^T$ was 46.9 mol%. The major isoprenoid quinone was ubiquinone-8. The results of DNA-DNA relatedness and 16S rRNA sequence similarity analyses, in addition to its phenotypic and chemotaxonomic characteristics, suggest that strain GNUM08123 is a novel species within genus Vibrio, designated as Vibrio sp. GNUM08123. Agarase production by strain GNUM08123 was induced by agar and sucrose, but was repressed probably owing to carbon catabolite repression by glucose and maltose.

  • Variovorax sp. PMC12 is a rhizobacterium isolated from tomato rhizosphere and enhanced the plant resistance to abiotic and biotic stresses. Here we present the complete genome sequence of strain PMC12. The genome is comprised of two circular chromosomes harboring 5,873,297 bp and 1,141,940 bp, respectively. A total of 6,436 protein-coding genes, 9 rRNAs, 64 tRNAs, 3 ncRNAs, and 80 pseudogenes were identified. We found genes involved in 1-aminocyclopropane-1-carboxylate (ACC) deaminase, antioxidant activity, phosphate solubilization, and biosynthesis of proline and siderophore. Those genes may be related to capability of improving plant resistance to various stresses including salinity, cold temperature, and phytopathogen.

  • The variations of species compositions, standing crops and seasonal succession of phytoplanktons including Cyanophyceae, Chlorophyceae and Bacillariophyceae were investigated with physico-chemical elements of water to clarify the causes of water bloom according to the water depth in the Lake Unmun. The increased amounts of turbidity, T-N, T-P and SS originated from heavy rain and typhoon in the middle of June provoked to produce phytoplanktons. In July and August, the dominant species was Peridinium sp., while the subdominant species was Microcystis aeruginosa. In October, Aulacoseira distans was dominant and Asterionella formosa was subdominant. During the period of water shortage, the concentrated pollutants caused the decrease of precipitation, the long stagnation time of water body and the mixed pollutants by upwelling as decreasing water temperature could accelerate the water bloom. The preventives to decrease microalgal generation should be controled by the stagnation time of water body by increasing outflow to intercept water bloom such as ulacoseira sp. in October as well as summer.
