Effects of spNab2 Deletion and Over-Expression on mRNA Export (분열효모에서 spNab2 유전자의 결실돌연변이 및 과발현에 대한 분석)

  • We constructed the deletion mutants of fission yeast Schizosaccharomyces pombe spNab2 gene that is homologous to poly(A)-binding protein NAB2 in budding yeast Saccharomyces cerevisiae, which plays crucial roles in mRNA 3' end formation and mRNA export from nucleus into the cytoplasm. A null mutant in an $h^+$/ $h^+$ diploid strain was constructed by replacing the spNab2-coding region with an $ura4^+$ gene using one-step gene disruption method. Tetrad analysis showed that the spNab2 is not essential for vegetative growth and mRNA export. However, over-expression of spNab2 cause the severe growth defects and intensive accumulation of poly(A) RNA in the nucleus. Also, the spNab2-GFP fusions were localized mainly in the nucleus. These results suggest that spNab2 is also involved in mRNA export out of the nucleus.

Fermentation Characteristics of Koji inoculated with Cordyceps sp. (동충하초를 이용한 코오지의 발효특성)

  • To investigate the quality of Koji produced by Cordyceps sp., a mixed culture of Aspergillus oryzae and the Cordyceps sp. were inoculated in Koji making. Viable cell counts and counts of fungi spore during Koji making for 120 hours were increased rapidly within 24-48 hours, after then their changes were not shown significantly during Koji making. The activities of amylase and protease in Koji inoculated with the mixed culture of A. oryzae and Cordyceps sp. were superior to one with A. oryzae only. The sensory evaluation on the Koji with 0.5% (w/w) A. oryzae and 0.5% (w/w) Cordyceps sp. were evaluated to be superior to ones. The best quality of Koji is prepared from the mixture of Cordyceps sp. and A. oryzae with the ratio of 0.5% and 0.5% (w/w).

Sphingobacterium sp. SW-09 Effectively Degrades Phenanthrene, a Polycyclic Aromatic Hydrocarbon, in a Soil Microcosm (Sphingobacterium sp. SW-09에 의한 토양환경에서의 다환 방향족탄화수소인 페난스렌의 분해)

  • We isolated a potent phenanthrene-degrading bacterium from oil-contaminated soils of Suzhou, China, and assessed the potential use of these bacteria for bioremediation of soils contaminated by polycyclic aromatic hydrocarbons (PAHs) in a microcosm. Based on 16S rDNA sequencing, we identified this bacteria as Sphigobacterium sp. SW-09. By PCR amplification, we also identified catechol 2,3-dioxygenase genes (nahH genes) mediating PAH degradation. Staphylococcus sp. KW-07, which has been identified in our previous study, showed potential for use in bioremediation of oil-contaminated soils. In this experiment, we compared the rate of phenanthrene-degradation between Staphylococcus sp. KW-07 and Sphingobacterium sp. SW-09 in a microcosm condition. Newly isolated Sphingobacterium sp. SW-09 showed a higher phenanthrene-degradation rate than that of Staphylococcus sp. KW-07 in soil microcosms. Together, our results suggest that the Sphingobacterim sp. SW-09 strain isolated from the Suzhou area may also be useful in bioremediation of PAH-contaminated soils.

Taxonomic Status of Acheilognathus sp. (Cyprinidae) found in the Geum River, Korea (금강에서 발견된 Acheilognathus sp. (Cyprinidae)의 분류학적 위치)

  • To examine the taxonomic status of Acheilognathus sp. specimens from the Geum River, morphological and genetical characteristics of A. sp., A. yamatsutae and A. majusculus were investigated and compared in detail. Specimens of A. sp. could be distinguished from the other two species by the combination of some morphological characters such as nuptial color, vertebrae, gillrakers and etc. Males of A. sp. had red bands on the outer margin of dorsal and anal fins and a white band on the outer margin of ventral fin in breeding season. A. sp. had larger maximum body length and somewhat more vertebrae than A. yamatsutae, and had fewer gillrakers than A. majusculus. A. sp. appeared as a monophyletic group with A. majusculus and A. cyanostigma based on genetic analysis. In addition, it had even more close relationship with other congeners than A. yamatsutae. Therefore it is presumed that A. sp. from the Geum River may be a distinct species in genus Acheilognathus.

Critical Review on the Microbiological Standardization of Salt-Fermented Fish Product (젓갈제품의 미생물학적 품질표준화에 관한 고찰)

  • Various problems in fermented fish products have been a major obstacle to manufacture the product in large scale, which is mainly concerned with the food safety. In this review, salt-fermented anchovy was selected to elucidate the characteristics of microorganisms involved in fermentation; thereby, it is suggested for research areas to achieve the quality improvement of tile product. Different microorganisms were involved in fermentation of anchovy. Dominant species were reported to be Bacillus sp., Pseudomonas sp., and Micrococcus sp., other microorganisms were Vibro sp., Clostridim sp., Serratia sp., Achromobacter sp., Streptococcus sp., Breuibacterium sp., Halobacterium sp., Flavobacterium sp., Corynebacterium sp., Acinetobacter sp., Sarcina sp., Staphylococcus sp., Torulopsis sp., and Saccharomyces sp. To standardize the quality of fermented fish products, screening and isolation of promising microorganisms should be carried out to develop different types of products; at the same time, proper sanitation control should be employed to keep the commercial value of the product by prolonging the shelf life.

Differences of Cesarean Section Rates according to San-Yin-Jiao(SP6) Acupressure for Women in Labor (삼음교 지압에 따른 산부의 제왕절개율 차이)

  • Purpose: The purpose of this study was to explain differences of cesarean section rates according to San- Yin-Jiao(SP6) acupressure for women in labor. Method: A noneqivalent control group pre test - post test design was used to explain differences of cesarean section rates according to SP6 acupressure. The participants were 209 women who were assigned to one of three groups SP6 acupressure(n=86), SP6 touch(n=47), and control group(n=76). For 30 minutes, the SP6 acupressure group received SP6 acupressure, and the SP6 touch group received SP6 touch for the duration of each uterine contraction. The Control group was encouraged to deep breath and relax for the duration of each uterine contraction for 30 minutes. Result: The rates of cesarean section were 12.8%, 29.8%, and 22.4% for the SP6 acupressure group, SP6 touch group, and control group respectively. There was a significant difference among groups (p=0.049). Cesarean section rateswere significantly different between the SP6 acupressure and non-SP6 acupressure group(p=0.035). Conclusion: This finding shows that 30 minutes of SP6 acupressure was effective in decreasing the cesarean section rate. Therefore, SP6 acupressure during labor could be applied as an effective nursing intervention.

Present Situation of Diseases Occurred with Cultured Marine Fishes in Kamak Bay (가막만 가두리 양식자의 어류질병에 관한 연구)

  • The pathogenic organisms occurred in cultured marine fishes in Kamak Bay were investigated from March to November in 1993. The samples were collected at 7 sampling stations once a month. Nine species of pathogenic organisms (Vibrio sp., Edwardsiella sp., Flexibacter sp., Streptococcus sp., Micrococcus sp., Caligus sp., Trichodina sp., Lymphocystis and Staphylococcus sp.) were identified as pathogenic organisms from four different species of fish (Sebastes schlegeli, Paralichthys olivaceus, lateolabrax japonicus and Pagrus major) collected in the study areas. Most of pathogenic organisms were found at over 20^{\circ}C$ of sea water temperature from June to October in 1993. On the test of drug sensitivity, Vibrio sp. (KS-9303) was sensitive to oxytetracycline and chloramphenicol ; Edwardsiella sp. (KP-9315) to oxytetracycline ; Flexibacter sp. (KP-9318) to oxytetracycline, chloramphenicol and oxolinic acid ; Streptococcus sp. (KP-9319) to erythromycin, chlorampheicol and oxytetracycline. However, all these 4 isolated bacteria were resistant to ampicilin, steptomycin, sulfamethoxazole and nitrofurazone.

A Study on a Modeling of the Inhibitory Reflex Mechanism of Jaw Muscle Induced by Electrical Stimulation (전기자극에 대한 턱근육의 억제반사 메카니즘의 모델링에 관한 연구)

  • EMG recordings of the electrical activity of muscle have proved to be a valuable tool in studying muscle function and reflex activity. SP(silent period) is elicited by a electrical stimulation on the chin during isometric contraction of jaw muscle. This paper proposes a model of the inhibitory reflex mechanism of jaw muscle after electrical stimulation. The SPs of jaw muscle after a electrical stimulation to the chin were divided into SP1 and SP2. SP1 is produced by the activation of periodontal receptors. The activation of nociceptors contributes to the SP2. As a result, the EMG signal generated by a proposed a model of inhibitory reflex mechanism is similar to real both EMG signal including SP1 and SP2. The present study have shown differences of SP1 and SP2 induced by inhibitory reflex mechanism.

A Study to Establish the Core Evaluation Items of SP Authentication using ANP(Analytic Network Process) (ANP 모형을 이용한 SP 인증 핵심 평가항목 도출 연구)

  • Software process (SP) authentication is a certification system designed in order to improve software process and ensure software reliability of the Korean small and medium enterprises (SMEs). It is composed of 3 grades, 17 evaluation items and 70 detailed evaluation items (software process). In order to certificate grade 2, 42 detailed evaluation items must be satisfied. In order to certificate grade 3, all 70 detailed evaluation items must be satisfied. SMEs are more vulnerable to manpower and material resources than the major company. Therefore, it is difficult for SMEs to invest in the long term to improve the software process. If we can provide the core evaluation items to SMEs, SMEs will be able to improve software quality more efficiently and effectively. It can be used as a short way to acquire SP authentication. The evaluation items of SP are defined as influencing each other. Therefore, the core evaluation items might be established using related techniques. Although we searched for previous researches and related informations, the associated study was insufficient. In this study, ANP (Analytic Network Process) technique was applied, and the core evaluation items of SP authentication were established. SMEs will be able to acquire SP authentication more easily using the core evaluation items. Finally, we expect that this study will be used as a basis material for creating the SP 2.0 version.

Antitumor Activities of Polysaccharudes fractuibuzed from Zoogloea sp. Against Meth A Cells (Zoogloea sp.의 다당체가 Meth A 세포에 의한 종양형성 억제 효과)

  • The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.
