Isolation, identification, and probiotic characteristics of Bacillus strains affecting the biogenic amine content in fermented soybean paste (발효 된장의 바이오제닉 아민 함량에 영향을 미치는 바실러스균의 분리 동정 및 프로바이오틱 특성)

  • The primary objective of this study was to determine the content of biogenic amines in Korean traditional fermented soybean pastes (doenjang) and to isolate potential probiotic Bacillus sp. with the ability to inhibit biogenic amines accumulation. There were significant differences in the bacterial cell counts, pH value, titratable acidity, salinity, and biogenic amine content between the samples. Among Bacillus strains isolated from doenjang, Bacillus (B.) licheniformis DB102, B. subtilis DB203, B. stearothermophilus DB206, Bacillus sp. DB209, Bacillus sp. DB310, B. coagulans DB311, B. cereus DB313, B. amyloliquefaciens DB714, Bacillus sp. DB917, B. cereus DB 915, B. subtilis DB1020, and Bacillus sp. DB1022 were found to be able to produce biogenic amines. On the other hand, biogenic amine-degrading strains were identified as Bacillus sp. DB403, Bacillus sp. DB407, B. subtilis DB517, B. licheniformis DB612, and B. subtilis DB821. In particular, Bacillus sp. DB407 and B. subtilis DB821 showed probiotic properties including tolerance to artificial digestive juices, adherence to intestinal epithelial cells, resistance to antibiotics, and antibacterial activity against biogenic amine-producing strains. In conclusion, the two probiotic Bacillus strains may be considered as the suitable starter for manufacture of fermented soybean foods with low biogenic amines content.

Characteristics of Raw Starch-Digesting Enzyme from Streptomyces sp. 4M-2 (Streptomyces sp. 4M-2가 생산하는 생전분 분해효소의 특성)

  • A raw starch-digesting enzyme from Streptomyces sp. 4M-2 was purified by ammonium sulfate fractionation, DEAE-Sephadex A-50 column chromatography and Sephadex G-100 gel filtration. The specific activity of the purified enzyme was 51.22 RSU/mg protein and the yield was 4.5% of the total activity of the culture broth. The purified enzyme was found to be homogeneous by polyacrylamide gel electrophoresis and its molecular weight was estimated to be about 102, 000 daltons by SDS-polyacrylamide gel electrophoresis, The optimal temperature and pH for the enzyme activity were 42$^{\circ}C$ and PH 5.5, respectively. The enzyme had Km, value of 44.44mg/$m\ell$ for raw corn starch. The enzyme was activated by addition of calcium and barium ions. Corn amylose was degraded by the enzyme very easily and raw potato starch was also degraded easily. Main products of the enzymatic hydrolysis of raw corn starch were analyzed to be maltose and maltotriose. The enzyme was considered as $\alpha$-amylase.

Two New Marine Tardigrades of Genus Batillipes (Heterotardigrada: Batillipedidae) from Korea (Batillipes속 (이완보강, Batillipedidae과)의 해양 완보류 2신종)

  • Two new marine tardigrade species, Batillipes longispinosus n. sp. and B. orientalis n. sp. are described on the basis of the specimens collected from the intertidal or shallow sublittoral sands of South Korea. Both the present species are characteristic in the absence of any caudal appendage throughout their life span, and bearing the long clava or cephalic appendages with the long spinous process on the femur of legs IV. B. longispinosus n. sp. most resembles B. tuvernatis Pollock, but is easily distinguished from it by the flattened caudal region and prominently developed spine of legs IV. B. orientalis n. sp. is much related to B. roscoffensis Kristensen, in having the wing-formed lateral body prohection between legs III and legs IV, the papillae on the head region, and the smooth caudal region, but discernible from it in the general body shape and the relatively longer spine of legs IV.

First report of interspecific facultative social parasitism by Polistes sp. on Polistes djakonovi Kostylev (Hymenoptera: Vespidae) in South Korea

  • Social parasitism is occasionally found in some Polistes and Vespa species, such as Vespa dybowskii. We report a discovey of interspecific facultative social parasitism by Polistes sp. (possibly P. mandarinus) on Polistes djakonovi Kostylev in two rural areas of South Korea. P. djakonovi is very similar to Polistes sp. in its body color patterns except that the mark on the clypeus is different. In nest 1 (65 cells), we found 5 females of P. djakonovi and 4 females of Polistes sp. on 30 July 2014, whereas nest 2 (102 cells) contained 12 females and 16 males of P. djakonovi, and 3 females of Polistes sp. on 28 August 2013. Although we found the two nests in July and August, P. djakonovi seems to have been exploited by Polistes sp. at the end of the preemergence period (early to mid-June). The two nests found in this study had mainly white cocoon caps of P. djakonovi with several yellow ones of Polistes sp. In most cases of social parasitism, intruders have a larger size of the body or some body parts than the host in order to usurp the host; in contrast, this study showed that the hosts had lager bodies than the intruders.

  • The conditions for the $\beta$-fructofuranosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at 50$\circ$C for 36 hrs with batch culture in the optimal medium containing 1.0% sucrose, 0.6% yeast extract, 0.1% each of KH$_{2}$PO$_{4}$, K$_{2}$HP0$_{4}$, NaH$_{2}$PO$_{4}$, and initial pH 9.5, and the final enzyme activity under the above condition was 102 units per ml of cell free extract.

The Distribution of Substance P and Calcitonin Gene-Related Peptides Immunoreactive Fibers in the Rat′s Larynx (흰쥐 후두에 분포된 신경에서 Substance P 및 Calcitonin Gene-Related Peptides의 발현양상)

  • The larynx has three major functions such as protective reflex, respiration and phonation, and is richly innervated by sensory, sympathetic and parasympathetic nerves. The sensory innervation of the laryngeal mucosa, which is involved in the perception of pain, mechanical and chemical irritation, prtects the airway via various laryngeal reflexes. We studied the distribution of Substance P (SP) and Calcitonin Gene-Related Peptide (CGRP) sensory fibers in the rat's larynx using the immuno-histochemical methods. Many SP and CGRP immunoreactive fibers were found in all regions of the laryngeal mucosa except the vocal cords. SP immunoreactive fibers showed a very similar distribution to the CGRP fibers in the epithelium and submucosa. But SP immunoreactive fibers were sparser than CGRP immunoreactive fibers in distribution density. Both reactive fibers were denser in the supraglottic region than subglottic region. Especially, intraepithelial fibers displayed the densest innervation to the laryngeal surface of the epiglottis. h the subepithelium, SP and CGRP immunoreactive fibers were distributed along the wall of vessels and around the glands. The present results suggest that the regional distribution of SP and CGRP immunoreactivity may be responsible for the protective reflex function of the laryngeal inlet.

Isolation and Characterization of Diesel Oil Degrading Bacterium, Pseudomonas sp. GENECO 1 Isolated from Oil Contaminated Soil (유류 오염 토양으로부터 분리한 디젤 분해 세균 Pseudomonas sp. GENECO 1의 분리 및 특성 규명)

  • With the enrichment culture technique, bacterial strains which degrade diesel oil were isolated from soil contaminated with diesel oil. One of the isolates named GENECO 1 showed the highest activity for emulsification of diesel oil as well as the highest growth rate. This strain, GENECO 1, was identified as a Pseudomonas sp. based on its biochemical, physiological characteristics and 16S rDNA sequences. The optimal cultural conditions for cell growth and oil emulsifying activity of its culture were as follow; $30^{\circ}C$ for temperature, 7.0 for pH. Diesel oil degradation was analysed by the gas chromatography. More than 95% of 1% treated diesel oil were converted into a form no longer extractable by mixed organic solvents after 96 hours incubation.
